1.Adiponectin attenuates H2 O2-induced SH-SY5 Y cell injury and tau hyper-phosphorylation via activating PP2 A

AIM:To study the effects of adiponectin on H 2 O2-induced cell injury and tau hyperphosphorylation in human neuroblastoma SH-SY5Y cells.METHODS:Cell viability was determined by MTT assay .H2 O2-induced cell in-jury and morphological changes in the SH-SY5Y cells with or without adiponectin treatment were observed .The level of tau phosphorylation as well as the activities of protein phosphatase 2A (PP2A) and of glycogen synthase kinase-3β(GSK-3β) were examined by Western blotting . RESULTS: Adiponectin significantly attenuated H 2 O2-induced cell injury (P<0.01).Adiponectin upregulated the activity of PP2A and decreased phosphorylation levels of tau under the stimula-tion with H2O2(P<0.01).Okadaic acid, a specific inhibitor of PP2A, blocked the protective effects of adiponectin (P<0.01).Adiponectin increased the phosphorylation level of GSK-3βat Ser9 site under H2O2stimulation (P<0.01).CON-CLUSION:Adiponectin protects SH-SY5Y cells against H2O2-induced cell injury and decreases tau hyperphosphorylation by activating PP2A and inactivating GSK-3β.
2.MicroRNA-122 expression during isoniazid anti-tuberculosis drug-induced liver injury in mice

OBJECTlVE To study the reIationship between microRNA(miRNA)-122 and Iiver injury in-duced by anti-tubercuIosis drugs,and to discover the new biomarkers for earIy diagnosis of this type of Iiver injury. METHODS mice were given 2 mL isoniazid oraIIy at 90 mg·kg-1 . BIood and Iiver tissue sampIes were coIIected at 1,3,5,7,14,21 and 28 d after administration of isoniazid. Serum gIutamic-pyruvic transaminase( GPT)and gIutamic-oxaIoacetic transaminase( GOT)IeveIs were determined using an automatic biochemicaI anaIyzer. Cu/ Zn-superoxide dismutase( Cu/ Zn-SOD ) activity and maIondiaIdehyde( mDA)content were detected by biochemicaI method. ReaI-time qPCR was used to measure the expression of miRNA-122. RESULTS GPT and GOT IeveIs were significantIy higher at 14 and 21 d(P﹤0.05)than in the controI. Cu/ Zn-SOD began to decIine whiIe mDA began to increase after 5 d(P﹤0.05). miRNA-122,which progressiveIy decreased after administration,was reduced to the mini-mum 0.58 ±0.02 at 14 d. There were good correIations between miRNA-122 and GPT,Cu/ Zn-SOD, mDA(the correIation coefficients were -0.370,0.268,and -0.298,respectiveIy),but no correIation with GOT was observed. CONCLUSlON The tissue miRNA-122 profiIe can be used as a sensitive marker for anti-tubercuIosis drug-induced Iiver injury,which couId contribute to the earIy diagnosis of Iiver injury.
4.Application of COI-based DNA Barcoding for Identifying Animal Medical Materials in the Chinese Pharmacopoeia

Medicinal animals are important part of Traditional Chinese medicine resources in China. Cytochrome c oxidase subunit I (COI) was selected as the standard DNA barcoding sequence for animal medical materials. In this study, the 51 animal species from 45 animal medical materials in the Chinese Pharmacopoeia were selected and the intra-specific variation and the inter-specific divergence, the barcoding gap, the identification efficiency of their COI sequences were analyzed. The results showed that the inter-specific divergence is higher than intra-specific distance. The barcoding gap existed between inter-specific sequence divergence and intra-specific dis-tance. The identification efficiencies were 100% both at the genus and species level except the Arthropoda. The cluster dendrogram exhibited that different species distinguished from others. Therefore, COI sequence as a bar-code is suitable to identify the species of animal medical materials in Chinese Pharmacopoeia.
5.The centric relation-maximum intercuspation discrepancy in adult angle's class II pretreatment patients.

OBJECTIVEThis study investigates the differences in condylar position between centric relation (CR) and maximum intercuspation (MI) in Angle's Class II orthodontic patients before treatment.
METHODS80 cases, who were Angle's Class II pretreatment patients, and 50 cases, who were normal(ideal) occlusion were accepted. Dental casts were mounted on Panadent articulator with CR bite record, taken by bilateral manipulation and load testing. The differences in condylar position between CR and MI in all three spatial planes were measured using the Panadent condylar position indication (CPI).
RESULTS(1) The positive rate of CR-MI discrepancy was 92.50%(74 cases) in the group of Angle's Class II malocclusion and 10.00% (5 cases) in the group of normal occlusion(P< 0.001). 2)74.32% (55 cases) CR-MI discrepancy in 74 cases in the group of Angle's Class II pretreatment patients were coincidence discrepancy. (3)91.25% patients in the group of Angle's Class 11 malocclusion and 66.00% in the group of normal occlusion present occlusion interferences which located at the posterior teeth.
CONCLUSIONThe results suggested that orthodontists should be aware of a high incidence of condylar displacement in Angle's Class ii pretreatment patients, and measure condylar displacement before the start of comprehensive orthodontic treatment to unmask real jaw relationships and avoid possible misdiagnoses.

6.Value of perfusion CT quantitative analysis for predicting tumor regression grade after chemoradiotherapy in patients with rectal cancer

Objective To explore the value of perfusion CT quantitative analysis for predicting tumor regression grade (TRG) after chemoradiotherapy in patients with rectal cancer.Methods From June 2016 to June 2018,94 rectal cancer patients diagnosed and treated in Cangzhou Central Hospital of Hebei Province were selected and were divided into reaction group (TRG 3-4) and non-reaction group (TRG 0-2) according to the results of surgical specimens.Perfusion CT was performed in both groups before treatment,and chemoradiotherapy and surgery were used.Baseline data and perfusion CT results including blood flow,blood volume,mean transit time (MTT),permeability surface (PS) were compared between the two groups,and receiver operating characteristic (ROC) curve was used to evaluate the predictive efficacy of perfusion CT indexes for chemoradiotherapy responsiveness.Results In this study,a total of 23 cases (24.47%) were responsive to chemoradiotherapy,and 71 cases (75.53%) were not responsive to chemoradiotherapy.Blood flow in reaction group [(38.60 ±7.13) ml · 100 g-1 · min-1] was significantly lower than that in non-reaction group [(67.39 ± 11.33) ml · 100 g-1 · min-1,t =3.273,P =0.001].MTT in reaction group was significantly longer than that in non-reaction group [(11.12 ±2.19) s vs.(6.88 ± 1.32) s,t =4.500,P <0.001].There was no significant difference in blood volume [(4.62 ±0.73) ml/100 g vs.(5.01 ± 1.04) ml/100 g] and PS [(13.72±3.82) ml · 100 g-1 · min-1 vs.(11.40 ±2.59) ml · 100 g-1 · min-1] between the two groups (t =0.818,P =0.415;t =0.409,P =0.683).The best cut-off points of blood flow and MTT for predicting chemoradiotherapy responsiveness were 50.89 ml · 100 g-1 · min-1 and 8.99 s,the area under the curve (AUC) was 0.825 and 0.922,and the AUC of combined prediction of chemoradiotherapy responsiveness was 0.982,which was significantly better than that of single prediction (Z =2.868,P =0.004;Z =2.051,P =0.004).The accuracy (91.49%) and specificity (90.14%) of combined prediction of chemoradiotherapy responsiveness were significantly better than those of single prediction (blood flow:accuracy 75.53%,specificity 73.24%;MTT:accuracy 79.79%,specificity 78.87%),and the differences were statistically significant (x2 =8.800,P =0.012;x2 =6.766,P =0.034).Conclusion Blood flow and MTT in perfusion CT have great predictive value for chemoradiotherapy responsiveness in patients with rectal cancer.

8.Efficacy of endoscopic variceal ligation and its correlation with liver function.

OBJECTIVETo analyze the efficacy of endoscopic variceal ligation and its correlation with liver function.
METHODS322 patients received EVL (endoscopic variceal ligation) and 34 patients with PDP (pericardial devascularization procedure) were retrospectively analyzed and divided into groups A, B and C. These patients were then subdivided into bleeding and non-bleeding subgroups according to Child-Pugh scores of liver function and history of upper gastrointestinal bleeding. The bleeding rate and mortality were contrasted between EVL and PDP. Liver function, Platelet count, leucocyte count and spleen thickness of before and after ligation were contrasted in EVL.
RESULTSThe bleeding rate and mortality were 1.7%, 3.4%, 7.0%; 0%, 5.1%, 8.1% in EVL group and 9.1%, 14.3%, 100.0%; 0%, 9.5%, 50.0% in PDP group, respectively. Variceal obliteration needed means of 2.1+/-0.7, 3.1+/-0.8 and 4.2+/-1.2 sessions in A, B and C ligation groups, respectively (F = 41.2, P is less than 0.01). On subgroup analysis, the numbers of ligation session were 2.6+/-0.7, 3.2+/-0.9 and 4.3+/-1.1 in A, B and C bleeding subgroup (F = 39.3, P value is less than 0.01) and 2.0+/-0.6, 2.7+/-0.6, and 2.9+/-0.4 in A, B and C non-bleeding subgroup, respectively (F = 17.0, P value is less than 0.01). ALT, AST, Platelet count and leucocyte count reduced significantly, spleen thickness increased remarkably in bleeding subgroup after ligation.
CONCLUSIONThe efficacy of EVL was significantly negatively correlated with liver function and prior to pericardial devascularization procedure. EVL had no effect on liver function but might increase spleen thickness and aggravate hypersplenism. EVL was recommended especially for the bleeding liver cirrhosis patients with Child B and C scores.

9.Influence of clinical characteristics on health-related quality of life after mid-low rectal cancer surgery.

OBJECTIVEThe present study aims to investigate health-related quality of life (HRQOL) in disease-free survivors after radical surgery for mid-low rectal cancer.
METHODSA retrospective cross-sectional study was performed in patients with rectal cancer who underwent primary surgery between August 2002 and February 2011 by use of the European Organization for Research and Treatment of Cancer QLQ-C30 and CR-38 questionnaires (n = 330). The impact of clinical characteristics on HRQoL were assessed and compared by univariate and multivariate regression analyses.
RESULTSTwo hundred and four effective responses were received. Patients with stoma were more impaired in HRQoL than those without stoma, especially in the field of social psychology, such as emotional function (M(50) = 91.67, U = 2668.5, P = 0.026), social function (M(50) = 83.33, U = 2095.5, P < 0.001), financial difficulties (M(50) = 0, U = 2240.5, P < 0.001) and body image (M(50) = 88.89, U = 2507.0, P = 0.013). Only in the constipation scale (M(50) = 14.29, U = 2376.0, P = 0.001), nonstoma patients had a better score. The analysis in different types of surgical procedure paralleled those of stoma. Patients with complication had a poorer function in some symptom scales such as dyspnoea (M(50) = 0, U = 1505.0, P < 0.001), gastro-intestinal symptom (M(50) = 6.67, U = 1766.0, P = 0.034) and financial difficulties (M(50) = 33.33, U = 1795.5, P = 0.044), and in some functioning scales such as emotional function (M(50) = 83.33, U = 1608.5, P = 0.009), cognitive function (M(50) = 66.67, U = 1612.5, P = 0.010) and body image (M(50) = 66.67, U = 1617.0, P = 0.012). In our study, HRQoL after rectal cancer surgery improved with time. Our multivariate analysis displayed that stoma and postoperative time were the most significant characteristics. Variables associated with worse financial status were less postoperative months, occurrence of complications and presence of stoma.
CONCLUSIONSDifferent scales of HRQoL in patients of China after curative surgery for mid-low rectal cancer are significantly influenced by different clinical characteristics.

10.Effects of mesenchymal stem cells on expansion potential and adhesion molecules expression of cord blood CD34+ cells.

OBJECTIVETo explore the effects of bone marrow mesenchymal stem cells (MSCs) on in vitro expansion potential, the adherent molecules expression of cord blood (CB) CD34(+) cells.
METHODSMSCs were obtained from human bone marrow and their differentiation function and phenotype were identified. CB CD34(+) cells were expanded in culture systems with or without MSC layer. Hematopoietic progenitor cells and adhesion molecules expression were assessed by semisolid culture assay and flow cytometry.
RESULTSThy-1, SH2, SB10, CD44, CD13, CD49e and CD29 were highly expressed on MSCs with no expressions of CD34, CD45, HLA-DR, CD14 and CD31. The MSCs could differentiate into adipocytes and osteoblasts under specific induction conditions. After culturing on MSCs layer with supplement of cytokines for 8 days, the absolute numbers of nuclear cells, CD34(+), CD34(+)CD38(-), CD34(+)CD62L(+) cells and CFU-Cs were increased by 145.57 +/- 17.89, 37.47 +/- 13.78, 69.78 +/- 50.07, 10.74 +/- 5.89 and 20.73 +/- 5.54-folds, respectively, being significantly higher than that cultured with cytokines alone. The expression of ALCAM, VLA-alpha4, VLA-alpha5, VLA-beta1, HCAM, PECAM and LFA-1 on CD34(+) cells remained unaffected. The expressions of ICAM-1 and L-selectin were downregulated during expansion, while the absolute numbers of CD34(+)CD62L(+) and CD34(+)CD54(+) cells were increased.
CONCLUSIONSMSCs layer improves expansion of CB CD34(+) cells while inhibiting their differentiation and retaining their homing ability.
