1.Expression of HMGB1, MMP-9 and VEGF-C in papillary thyroid carcinoma

Objective To study the expression of high mobility group box 1 (HMGB1),matrix metalloproteinase-9(MMP-9) and vascular endothelial growth factor-C (VEGF-C)gene in papillary thyroid carcinoma (PTC) and their significance.Methods Expression of HMGB1,MMP-9,VEGF-C proteins and HMGB1 mRNA was detected by technology of immunohistochemistry(SP) and in situ hybridization in 58 cases of PTC tissues,20 cases of thyroid adenoma tissues,25 cases of nodular goiter tissues and 10 cases of normal thyroid tissues.Results Expression of HMGB1,MMP-9,VEGF-C proteins and HMGB1 mRNA in PTC tissues was much higher than that in the 3 non-cancerous tissues (P < 0.05).The positive expression rate of HMGB1,MMP-9 proteins and HMGB1 mRNA was closely associated with the thyroid carcinoma size and lymph node metastasis (P <0.05).There was also closely association between the positive expression rate of VEGF-C protein and lymph node metastasis(P < 0.05).The positive expression rate of HMGB1,MMP-9,VEGF-C proteins and HMGB1 mRNA had no association with patients' sex or age(P > 0.05).The expression of HMGB1,MMP-9 and VEGF-C proteins had positive correlation with each other(P < 0.05).The expression of HMGB1 protein and HMGB1 mRNA was also positively correlated with each other(P <0.05).Conclusion Expression of HMGB1,MMP-9,VEGFC proteins and HMGB1 mRNA in PTC is correlated with the progress,invasion,metastasis of PTC,by detecting them may help to predict the clinical progress and prognosis.
2.Analysis of rhFK506-Bimding Protein 12 by Matrix-assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry

The primary structure of rhFK506-Binding Protein (FKBP) 12 was studied with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) by measuring its molecular weight and tryptic peptide mass fingerprinting.The calculated molecular weight of rhFKBP12 was 11819.5 and the measured value was 11818.0. The relative error was only 0.01%. The measured peptide mass fingrprinting of rhFKBP12was identical with that deduced from its amino acid sequence. It proved that the primary structure of rhFKBP12 was correct and there was no amino acid deletion, mutation and modification in its expression, refolding andpurification.

To obtain active hFKBP52 protein for screening novel neu rotrophic drugs. Semi-nested and overlap PCR and affinity chromatography were u sed. hFKBP52 gene was cloned successfully from human fetal brain cDNA library, a nd then highly expressed (about 30%) as fusion protein in pET28a(+) vector syste m. The recombinant protein was purified as one band on SDS-PAGE. The purified h FKBP52 showed peptidyl-prolyl cis-trans isomerase (PPIase) activity, simil ar to the wild type.
4.Relationship between HBV genotype and liver damage in northern China

Objective To discuss the relationship between hepatitis B virus (HBV) genotype and HBV DNA ,liver fibrosis ,liver function and HBeAg .Methods HBV genotypes ,HBV DNA ,liver fibrosis indicators and alanine aminotransferase(ALT ) ,aspartate aminotransferase(AST ) ,total bilirubin(TBIL) ,albumin(ALB) and HBV e antigen(HBeAg) were detected in patients with serum hepatitis .All data were statistically analyzed .Results There was no significant difference of HBV DNA ,ALT ,AST ,TBIL ,ALB , procollagen- Ⅲ -peptide ,type Ⅳ collagen ,hyaluronic acid and laminin between patients with B and C genotype infection (P> 0 .05) . However ,HBeAg level in patients with C genotype infection was higher than that in patients with B genotype infection (P< 0 .05) . Conclusion There might be no significant difference of HBV DNA ,liver function and liver fibrosis between patients with B and C genotype infection ,but HBeAg level in patients with C genotype infection could be higher than patients with B genotype infection .
5.Gait and surface electromyography characteristics in hurdle athletes in a total gait cycle

BACKGROUND:In recent years, the technical parameters about hurdle athletes are mainly obtained through video analysis and DLT algorithm. However, the gait and surface electromyography (sEMG) characteristics during normal walking are little reported. OBJECTIVE: To explore the changes of the gait and lower limb sEMG signals relative to gait period in hurdle athletes. METHODS:Eight male professional hurdlers were selected to perform gait and lower limb sEMG tests on the trail, and the differences in gait and sEMG signals were analyzed by mathematical statistics. RESULTS AND CONCLUSION: The gait parameters of hurdlers showed no significant differences (except step length). In the total gait cycle, along with the gait changing, the right and left side muscles of the same name moved alternately. The median frequency and average power frequency of the tapping leg in the lower limb muscles were greater than those of the swinging leg (except biceps femoris, tibialis anterior and lateral gastrocnemius), but the mean EMG and EMG integral values of the tapping leg were smaller than those of the swinging leg. At the stand phase, the median frequency and average power frequency of the tapping leg in the lower limb muscles were greater than those of the swinging leg (except tibialis anterior), but the mean EMG and EMG integral values of the tapping leg were smaller than those of the swinging leg (except soleus). At the swing phase, the median frequency and average power frequency of the tapping leg in the lower limb muscles were greater than those of the swinging leg (except tibialis anterior and lateral gastrocnemius), but the mean EMG and EMG integral values of the tapping leg were smaller than those of the swinging leg (except soleus). To conclude, there are different degrees of differences in the frequency domain and time domain of the lower limb muscles between tapping and swinging legs. Additionally, the muscle strength of the tapping leg is less than that of the swinging leg.
6.Heat-killed Staphylococcus aureus induces a common early response in human monocytes in the presence of high concentration glucose

Objective To investigate the effects of heat-killed Staphylococcus aureus (HKSA) on the apoptosis and expression of iNOS and IL-1β in THP-1 monocytes in the presence of high concentration of glucose.Methods THP-1 cells were cultured in medium containing 25.0 mmol/L(HG) or 5.5 mmol/L (LG,control) D-glucose for 12 h-8 d.The THP-1 cells cultured for 6 d were extracted on the 0-48 h with or without HKSA,then apoptosis and expression of iNOS and IL-1β were examined.Apoptosis was analyzed by flow cytometry and expressions of IL-1β and iNOS were quantitated by real-time PCR.Results The expression of iNOS and IL-1β in THP-1 monocytes was increased significantly in the presence of high concentration of glucose for 12-48 h(P<0.05),reaching the highest level at 24 h and returned to baseline after 4 d.The expression was significantly lower than that of control after 4-6 d.Apoptosis rate was also increased significantly after 48 h to 4 days.HKSA infection enhanced apoptosis,but inhibited the expression of iNOS and IL-1 β in the presence of high concentration of glucose.The expression of iNOS and IL-1β increased significantly at 6 h(P<0.01),reaching the highest level at 12 h,but the levels were significantly lower than those in control groups (P<0.05).Conclusion These data suggest that high concentration of glucose can interfere with the anti-bacterial function of monocytes by reducing their expression of iNOS and IL-1β and enhancing their apoptosis.

8.Expression of aldehyde dehydrogenase 1 in breast cancer and its clinical significance

Objective:The purpose of this study is to investigate the expression of tumor stem cell marker aldehyde dehydrogE-nase 1 (ALDH1) in breast cancer and its clinical significance. Methods:The expression of ALDH1 protein was examined by immunohistochemical staining in 92 breast cancer tissues. The correlation analysis and diseasE-free survival analysis of patients was evaluated based on the clinical follow-up data. Results:Expression of ALDH1 protein had a significant correlation with progesterone receptor (PR) and cerb-B2 (P<0.05), but had no significant correlation with age, tumor size, clinical staging, and lymph node metastasis (P>0.05). The 2-year diseasE-free survival rate of AlDH1-positive patients was lower than that of ALDH1-negative patients (P<0.05). ALDH1-positive patients, who received CEF regimen chemotherapy and hormone therapy, had lower 2-year diseasE-free survival rate than that of ALDH1-negative patients (P<0.05). Conclusion:The decreased diseasE-free survival rate of ALDH1-positive patients is related with drug resistance. ALDH1 could be used as an independent factor for predicting the prognosis of breast cancer.
9.Primary Hodgkin disease of the supermaxilla: a case report and review of the literatures

Objective To intensify the diagnosis and treatment of primary osseous Hodgkin disease (HD) and reinforce the impression of its features of pathology and imaging. Methods The clinical manifestation, laboratory examination, treatment and outcome of a patient with primary Hodgkin disease of the supermaxilla were first reported and the pertinent literatures were reviewed. Results Pain of the right supermaxilla was the first clinicM symptom. Plain X-rays showed mixed osteolytic and partially osteosclerotie lesions in the right supermaxiUa. The tumor was removed and the pathohistology was HD lymphocyte-depletion. The clinical diagnosis was primary HD of the supermaxilla (Stage Ⅰ ). The case was treated with ABVD regimen and no obviously adverse reaction appeared. Conclusion Primary osseous HD rarely presents as a malignant bone lymphoma and is easily misdiagnoed. Pathological and immunohistologic studies can be useful to confirm the diagnosis of primary osseous HD. Early diagnosis and the differentiation from other disease should be performed and the prognosis of the present-day chemotherapy regimen appears good.
10.Observation of the tape after TVT-O surgery using two and three dimensional ultrasonography in the patients with stress urinary incontinence

Objective To observe the position and mobility of the tape after tension-free vaginal tape-obturator(TVT-O) surgery using transperineal two and three dimensional ultrasonography in patients with stress urinary incontinence.Methods A total of 32 patients with stress urinary incontinence who had TVT-O surgery and cured were enrolled.Transperineal two and three dimensional ultrasonography were performed after operation 3 months later.Both the position,morphology and mobility of the tape at rest and during Valsalva menuaver were observed.Results The tapes were mainly located in the 1/3 connection of the middle and distal urethra.The length of the urethra and the length of the urethra between the bladder neck and the proximal tape at valsalva menuaver were longer than those at rest.The tapes moved front-down at valsalva manuaver according to the mean distance between the tape and the symphysis pubis.The angles of the tapes were smaller at valsalva manuaver without significant difference.Conclusion It is useful to observe the position,morphology and mobility of the tape using transperineal two and three dimensional ultrasonography.The changes of the position and morphology of the tapes at valsalva menuaver were not significant.But the tapes can strengthen the urine control in the middle of urethra.
