1.Treatment of murine hypoparathyroidism with transgenetic hemopoietic stem cell transplantation

Objective To investigate the gene therapy of hypoparathyroidism (HPT) with hemopoietic stem cell s(HSCs) integrated with parathyroid hormone (PTH) gene.Methods Human PTH gene was amplified by polymerase chain reaction from pcDNA 3.1 -PTH templates and inserted into murine stem cell virus (MSCV) vectors. The plasmids recombinated with PTH gene were transfected into PA317 packaging cells and screened for positive clones with Geneticin. The condensed recombinant retroviruses were obtained and used to infect HSCs coming from umbilical cord blood. The infected HSCs were injected into the mice suffering from HPT. The change of symptoms and serum level of PTH and calcium in each group of mice was observed.Results Human PTH gene was proved to be inserted into MSCV vectors and the titres were up to 2?10 7 colony forming unit (CFU)/ml in condensed retroviral solution. The secretion of PTH reached 15 ng per 10 6 cells per 48 h. The wild type viruses were not detected by PCR amplification. The mice suffering from HPT recovered quickly and the serum level of calcium and PTH remained normal for about 3 months after the HSCs recombinated with PTH gene were injected into them. In the groups of mice subject to the injection of the recombinant retroviruses containing PTH gene, the serum level of PTH and Calcium was significantly increased in a short time, and then declined slowly. And the mice took on the manifestation of hypoparathyroidism again. The mice only subject to the implantation of HSCs were all dead within 20 days.Conclusion A long term therapeutic effect could be obtained by infusing the HSCs which were integrated with PTH gene into the mice suffered from hypoparathyroidism.
2.The imaging changes of premature infants with periventricular leukomalacia and its influence on prognosis

Periventricular leukomalacia ( PVL) is the most distinctive and characteristic brain injury in premature infants, which may result in nervous system sequelae, and affect motor development and quality of life of the infants. PVL lacks specific symptoms and the diagnosis of PVL depends on imagine. Transcranial ultrasound could make initial diagnosis and prognostic evaluation for PVL. Combined with MRI, it can assess the degree of the white matter damage and predict the possible adverse consequences, which could provide evi-dence for early treatment. The changes of brain imaging and their impact on the prognosis of PVL in preterm newborn was reviewed in this article.
3.Effect of changes in the anterior arch dimension on precision of Bolton analysis

Objective:To investigate whether the changes of anterior arch dimension have an impact on the precision of Bolton analysis. Methods:A mathematic-geometric model was used to evaluate the data. Results:The ideal anterior tooth size ratio may need adjustments, depending on the dimensions in radii of the upper and lower anterior dental arches. The ideal ratio is lower for dental arches with a high anterior curvature. There is a deficiency in the upper arch, a flatter anterior segment may compensate for some of the discrepancy and vice versa. Conclusion:An ideal Bolton value may not guarantee an ideal occlusion. Anterior tooth size ratio is not precise in predicting the anterior inter relationship post-treatment.
4.Analysis of bacterial spectrum in diabetes ulcer

Objective To evaluate the features of bacterial spectrum and drug resistance in diabetic ulcer.Methods Excretion specimen of diabetic ulcer from 130 patients admitted into our hospital from Mar.2010 to Sep.2011 were collected for bacterial culture and drug sensitivity test.28 antibiotics were chosen from 35commonly used antibiotics to test antibiotic sensitivity to decide whether the antibiotic was sensitive (S),intermediate sensitive (I),or resistant (R).Results 19 strains were isolated from 130 cases.Staphylococcus aureus with 14.62 % (19 cases)rated the first,followed by escherichia coli with 13.85% (18 cases),proteus mirabilis with 13.08% (17 cases),staphylococcus epidermidis with 12.31% (16 cases),enterococcus faecalis with 12.31% (16 cases),acinetobacter baumannii with 10.77% (14 cases).Antibiotics with high sensitivity to Gram-positive bacteria were tigecycline and vancomycin while with high sensitivity to Gram-negative bacteria were imipenem and amikacin.Conclusions Bacteria of diabetic ulcer are commonly resistant to antibiotics.It is of great importance for clinical rational drug use to know the distribution and resistance of commonly encountered bacteria.
5.Predictive accuracy of different Partin tables in Chinese prostate cancer patients

Objective To validate the Partin table 1997,2001 and 2007 for their accuracy in predicting pathologic stage in Chinese prostate cancer patients.Methods From January 1997 to June 2007,109 consecutive patients with clinically localized prostate carcinoma underwent open retropubic or laparoscopic radical prostatectomies and met all inclusion criteria well enrolled.Receiver operating characteristic(ROC)analysis was performed tO test the predictive accuracy of organ confined disease (0CD),extraprostatic extension(EPE),seminal vesicle involvement(SVI)and lymph node involvement(LNI). Results OCD,EPE,SVl and LNl were noted in 70%,17%,13%and 0%of cases respectively.The area under curve(AUC)of ROC for Partin table 1997 was 0.727,0.654 and 0.811for 0CD.EPE and SVl respectively,and was 0.693,0.633 and 0.835 for Partin table 2001 and 0.669.0.611 and 0.778 for Partin table 2007.Conclusions Partin tables 1997,2001 and 2007 are able to accurately predict the pathologic feature of seminal vesicle involvement.However,only Partin table 1997 can more accurately predict organ confined disease in this external validation for Chinese patients.
6.PTEN inhibits cell growth and increases chemosensitivity to doxorubicin of human breast cancer cells

Objective To study whether transient overexpression of tumor suppressor gene PTEN could lead to growth suppression and up-regulate the sensitivity to doxorubicin of human breast cancer MCF-7 cells. Methods The eukaryotic expression plasmid pEGFP-C 1-PTEN containing whole cDNA of PTEN was constructed and transfected into MCF-7 cells by Lipofectamine 2000 in vitro. Growth inhibition of the cells was observed by phase contrast microscope and flow cytometry. The clonogenic cell survival ability was studied by clony forming assay. MCF-7 cells′ chemosensitivity to adriamycin was studied with MTT assay. Results PTEN overexpression led to morphological changes characteristic of apoptosis of MCF-7 cells. PTEN overexpression also resulted in a significant increase in G 0/G 1 cell population (14.79%) and apoptosis (10.60%) detected by flow cytometry. The clonogenic survival rate of cells transfected with PTEN was significantly decreased after doxorubicin treatment compared with control. 7.Performance evaluation of orthotopic bladder reconstruction with ileocecum-appendix evaluated by imaging urodynamics

Objective To evaluate the effect of the urinary reservoir constructed with ileocecum-appendix in the elderly with bladder cancer. Methods From March 2002 to June 2008, 12 cases were treated using ileocecum-appendix as the reservoir after radical cystectomy. They all received the imaging urodynamics examination and were followed up for 1 year. Results The 11 of 12 patients had urinary continence completely. Only 1 case had incontinence and 3 cases had incontinentia urinae at night. Times of uresis were 8-10/day and 3-5/night within 3 months after surgery, and 4-6/day and 0-2/night 6 months after surgery. The urinary output was 150-350 ml/time. Urodynamics showed that mean urinary flow rate was 10.5 ml/s, mean initial bladder pressure was 27 cm H2O, the maximum filling pressure was 35 cm H2O. The average reservoir capacity was 152 ml and 420 ml, respectively. The out let pressure of posterior urethra was 52 cm H2O. The volume of residual urine was 0-65 ml. No evidence of ureteral reflux occurred, no hyperchloremic acidosis was observed. Conclusions Orthotopic bladder reconstruction is considered as an ideal form of urinary diversion characterized by low pressure, larger capacity and continence.
8.Evaluation of application effect of evidence- based nursing in reducing ventilator - associated pneumonia of patients with severe head injury

Objective To evaluate the application effect of evidence-based nursing in reducing ventilator-associated pneumonia (VAP) of patients with severe head injury. Methods 100 patients with severe head injury using mechanical ventilation were randomly divided into the control group and the observation group with 50 patients in each group.The observation group explored evidence from nursing of ventilator tube,oropharyngeal and sound sects care,airway care,balloon sleeves care to prevent biofilm on the endotracheal tube(ETT-BF) to fall off,enteral nutrition care,decubitus care,ward environment and the hand disinfection of medical staff and was given evidence- based nursing.The control group took routine care.The incidence of VAP,off-line time and mortality rate of the two groups were compared during the same time period. Results The incidence of VAP,off-line time,mortality rate showed significant difference between the two groups.The observation group showed better effect. Conclusions Implementation of evidence-based nursing can significantly reduce the off-line time of patients with severe traumatic brain injury using mechanical ventilation.It can decrease the incidence of VAP and mortality rate,improve the prognosis and reduce medical cost.
9.Photoprotective effect of a self-made sunscreen cream: an evaluation in vitro and in vivo by using guinea pigs

[Objective] To determine the sun protection factor (SPF) and protection factor of ultraviolet A (PFA) of a self-made sunscreen cream.[Methods] The sample of a self-made sunscreen cream and a commercial sunscreen cream were painted to an artificial skin (3M film) at a dose of 2 μl/cm2,and a tester was used to evaluate their photoprotective effect.A total of 48 albinism guinea pigs were classified into 8 groups to remain unprotected or be protected by the self-made or commercial sunscreen cream.A solar ultraviolet light simulator (SUV-1000) was used to simulate the ultraviolet rays in sunlight to irradiate the guinea pigs,and the photoprotecfive effect of these sunscreen creams was determined.[Results] The in vitro evaluation revealed that the SPF value of the self-made sun-screen cream and commercial cream was 32.26 ± 2.42 and 30.87 ± 2.57respectively (n =5,t =0.94,P > 0.05),and the PFA value was 24.28 ± 2.44 and 17.53 ± 2.28 respectively (n =5,t =4.52,P< 0.01 ).As the in vivo experiment showed,the SPF value of the self-made sun-screen cream and commercial cream was 30.39 ± 6.65 and 28.79 ± 7.36,respectively (n =12,t =0.38,P> 0.05),and the PFA value was > 8.91 and > 8.93 respectively.[Conclusion] The photoprotective effect of the self-made sunscreen cream is similar to that of the commercial cream.
10.A study on the relationship between neurodevelopment and early nutritional status of very low birth weight infants

Objective To study the relationship between neurodevelopment and early nutritional status of very low birth weight(VLBW)infants in NICU.Methods VLBW infants admitted to NICU of our hospital from January 2013 to December 2014 and received regular follow-up management at our high-risk infant outpatient clinic were retrospectively studied. All infantsˊdevelopmental quotient ( DQ) were evaluated at 3 months of corrected gestational age ( cGA) . DQ ≥130 was defined as excellent, 115-129 above medium, 85-114 medium, 70 -84 below medium and ≤69 abnormal. According to their DQ scores, infants were assigned into two groups, normal neurodevelopment group ( DQ≥85 ) and abnormal neurodevelopment group ( DQ<85 ) . Nutritional status during hospitalization between the two groups were compared.Results A total of 125 VLBW infants were recruited.At three months of cGA, 2 cases ( 1. 6℅) had excellent DQ score; 3 ( 2. 4℅) above medium; 63 ( 50. 4℅) medium;57 (45. 6℅) below medium. 68 cases (54. 4℅) in the normal development group and 57 (45. 6℅) in the abnormal group. The total energy intake, the proportion of enteral energy supply on 4 d, 7 d, 14 d after birth and amino acid supply on 4 d, 7 d were significantly higher in the normal neurodevelopment group than the abnormal group (P<0. 05). Comparing with the abnormal group, weight loss of VLBW infants in the normal neurodevelopment group was less; the time needed to reach birth weight and exclusive enteral nutrition in the normal neurodevelopment group was shorter. The differences were statistically significant between the two groups(P<0.05).Conclusions VLBW infantsˊearly nutritional status may influence their neurodevelopmental outcome.
