Aim To study the effect of Echinacoside on immune function and the hepatocyte mitochondrial DNA relative content of experimentally aging model mice. Methods Mice were given sc 10%D-galactose 10 ml?kg-1,once daily for 8 weeks to establish the model of aging mice. Vitamin E group ,and ECH groups were given ig Vitamin E 40 mg?kg-1 and ECH 20,40,60 mg?kg-1,respectively at one time. The interleukin-2(IL-2) and interleukin-6(IL-6) content in peripheral blood was measured by ELISA method.Phagocytosis of peritoneal macrophages was tested by neutral red test. Proliferation and transformation of spleen lymphocytes induced by ConA were determined by MTT method. The mitochondrial DNA relative content of liver cells was measured by the means of SDS alkali decomposition from liver. Results The IL-2 content, phagocytosis of peritoneal macrophages and proliferation of spleen lymphocytes were obviously decreased in the aging model group as compared with those in the normal control group. The IL-6 content and mitochondrial DNA relative content of liver cells were significantly increased. ECH 20,40,60 mg?kg-1 and Vitamin E 40 mg?kg-1 could improve the IL-2 content, phagocytosis of peritoneal macrophages and transformation of spleen lymphocytes, and reduce the IL-6 content and mitochondrial DNA relative content of liver cells. Conclusion ECH can enhance immune function and reduce the mitochondrial DNA relative content of liver cells, which may be one of the mechanisms of anti-aging.

This study is to investigate the therapeutic effect of syringin on adjuvant arthritis (AA) in rats and its mechanisms. Complete Freund's adjuvant (FCA) was used to induce AA in rats. Secondary paw swelling of AA rats was measured with volume meter. Pain response and polyarthritis index were scored. Meanwhile, splenic lymphocyte proliferation response induced by concanavalin A (ConA) or lipopolysaccharide (LPS) was examined with MTT assay. IL-2 production of splenic lymphocytes and IL-1 beta, TNF-alpha production of peritoneal macrophage (PM phi) were estimated by enzyme linked immunosorbent assay (ELISA). The secondary inflammation of AA rats appeared on the 14th day after injection of FCA. Syringin and tripterygium glycosides (TG) were given by intragastric administration for 16 days from the 14th day. Treatment of AA rats with syringin and TG from the 22th day significantly attenuated the secondary hind paw swelling, as well as relieved the pain response and the polyarthritic symptoms of the whole body as compared with that of the AA model group. The suppressed lymphocyte proliferation and IL-2 production of splenic lymphocytes in AA rats were reversed by treatment with syringin. Meanwhile, syringin remarkably down-regulated IL-1 beta, TNF-alpha productions from PM phi. These results indicate that anti-inflammatory effects of syringin on AA rats are mediated by modulating the immune function of abnormal cells and the balance of cytokines.

BACKGROUND:Autologous shed blood washing with the autologous transfusion system involving recovery, anticoagulation, centrifugation, concentrating and washing has been widely used in clinical practice.
OBJECTIVE:To clarify the characteristics of erythrocytes washing with autologous transfusion system, including recovery rate and hematocrit, the changes of shape, deformability, flow properties and in vivo half-life, oxygen carrying and delivering capacity and erythrocyte immunity and immunereceptor expression.
METHODS:The literatures published from January 1987 to January 2013 were retrieved by the first author in Wanfang and PubMed databases. Key words were“blood transfusion, autologus, blood preservation, erythrocytes”in English and Chinese. A total of 200 literatures relating to the erythrocyte characteristics in autologous blood transfusion were found by the computer, 60 of which were retained for further analysis after eliminating repetitive researches.
RESULTS AND CONCLUSION:Because of the mechanical force, such as negative pressure suction, centrifugal separation, and inflammatory mediators, enzymes, activated complements released by various damaged tissues and cells, the col ected erythrocytes were damaged to some extent. As a result, the total recovery rate of erythrocytes depended on the recovery rate, storage breakage rate and cleaning loss rate. The oxygen carrying capacity of erythrocytes was not influenced significantly by this procedure, so the recycled erythrocytes had the same oxygen carrying capacity with normal erythrocytes. To some extent, the number of surface receptors and immune function of recycled erythrocytes descended, but they were better than the erythrocytes preserved for 2 weeks. Studies suggested that blood recovery technology should be improved to reduce the functional decline in immune adherence of the recycled erythrocytes.

Objective To evaluate lower extremity deep venous hemodynamic changes and lower extremity deep venous disease in abdominal obesity. Methods To compare venous flow parameters of the lower limbs as assessed by Duplex ultrasound scanning in obese and nonobese individuals according to body mass index(BMI).Venous hemodynamics were studied in a prospective cohort study in nonobese( BMI < 25 kg/m2) and obese individuals( BMI > 30 kg/m2 ).Diameter, peak, mean, and minimum velocities were assessed. Results The study examined 45 limbs in 24 nonobese individuals and 44 limbs in 22 obese individuals. The diameter of the femoral vein was significantly greater in obese vs nonobese limbs ( 10.4 ± 1.3) mm vs (7.1 ± 1.0) mm,P < 0.01 ).Calculation of venous amplitude and shear stress showed significantly higher values in nonobese vs obese limbs [ ( 11 ± 5 ) vs ( 8 ± 3 ) cm/s ],( P < 0.01 ) and [ (0.50 ± 0.17 ) vs ( 0.30 ± 0.09 ) ] dyn/cm2,( P < 0.01 ).Spearman rank correlation revealed a significant inverse correlation between waist-to-hip ratios and waist circumference and velocities amplitude (peak ve locity-minim velocity),and shear stress. Conclusions Lower limb venous flow parameters differ significantly between obese and nonobese individuals. These findings suggest the mechanical role of abdominal adipose tissue potentially leading to elevated risk for both chronic venous insufficiency and venous thromboembolism.

Limb rehabilitation is one of the key issues for improving the quality of life in patients with stroke. Studies have demonstrated that the retention and recovery of active movement of hemiplegic limb have predictive value for the prognosis in acute stroke. The responsible lesion sites and rehabilitation training in the acute phase may be an important factor in influencing the recovery of active movement ability of hemiplegie upper limb. However, there is still lack of a large, multi-center study, and further related studies are needed.

The Journal of Practical Medicine 2015;(20):3324-3327
Objective To explore the relationship between plasma mannose-binding lectin (MBL), lipopolysaccharide (LPS) and body mass index in pregnant women with gestational diabetes mellitus (GDM). Methods 45 newly diagnosed GDM pregnant women and 45 healthy women (control group) were selected in this study. Plasma concentration of MBL and LPS were measured. All the groups were sub-divided into obesity (BMI ≥ 25 kg/m2) and non-obesity (BMI < 25 kg/m2) subgroup. The relationships of levels of plasma LPS, MBL with BMI were analyzed. Results The level of plasma MBL in GDM group was significantly lower than that in control group(P < 0.05), while the concentration of LPS in serum was much higher than that in healthy pregnant women (P < 0.05). Pearson analysis showed the level of MBL in GDM group was negatively correlated with BMI(r=-0.28, P<0.05), serum LPS concentration was positively correlated with BMI(r = 0.62, P < 0.05) and LPS was negatively correlated With MBL(r = -0.43, P < 0.05) . The above correlations were not found in control group. Conclusion Serum MBL and LPS maybe two important risk factors for those pregnant women being overweight, and could offer great significance for the prevention and treatment of GDM.

Serum 25-(OH)D3concentration was determined in subjects with different types of obesity. The serum 25-(OH)D3levels in overweight or obese and central obese subjects were lower than that in non-obese subjects.(P<0.01). In a multiple linear regression analysis serum 25-(OH)D3was independently and negatively correlated with body mass index and waist circumference.
