1.Cyberknife treatment of pancreatic carcinoma

Cyberknife,a newly developed stereotactic operation platform in radiosurgery system,has many advantages such as accurate positioning,image-guided and hyperfractionated radiotherapy.It has made very good effect in the treatment of locally advanced pancreatic carcinoma.At the same time,it has been applied in the preoperation and postoperation treatment of pancreatic carcinoma,and the palliative treatment of metastatic pancreatic carcinoma.So it can be a very good option for patients with pancreatic carcinoma.
2. Detection of Th17 cells in peripheral blood of patients with multiple myeloma and its clinical significance

Objective: To examine the Th17 cells (helper T cells 17) in peripheral blood of patients with MM (multiple myeloma) and explore its clinical significance. Methods: The peripheral blood samples from 39 patients with MM and 30 healthy volunteers were collected. The proportion of Th17 cells in CD4 + T cells was determined by flow cytometry. The expression of ROR-γt (retinoid-related orphan receptor-γt) mRNA, a specific transcription factor of Th17 cells, was detected by RT-PCR. The plasma concentration of IL-17 (interleukin-17) was measured by ELISA assay. Results: The proportion of Th17 cells, the expression level of ROR-γt mRNA and the concentration of IL-17 in peripheral blood in patients with MM were higher than those in the healthy volunteers [(3.509±0.594)% vs (2.913±0.846)%, 0.568±0.177 vs 0.468±0.163, 27.200±4.666 vs 23.758±7.410 pg/mL; all P values < 0.05). The proportion of Th17 cells was not correlated with the gender, age, disease type, globulin, immune globulin, light chain, M-protein and the proportion of plasmocytes of the patients with MM (P > 0.05), but correlated with ISS (international staging system) stage, β2-microglobulin level and the therapeutic response (P < 0.05). Conclusion: Th17 cells may be involved in the occurrence and development of MM. Detection of proportion of Th17 cells may contribute to the ajudgement of disease severity, therapeutic response and the prognosis of patients with MM. Copyright © 2013 by TUMOR.
3.Expression and significance of miR-219 in tongue squamous cell carcinoma

Objective To investigate the expression of miR -219 and its role in tongue squamous cell carcinoma(TSCC).Methods The relative expression of miR -219 in the 26 samples of surgically resected paired TSCC and normal tumor -adjacent tissues was detected by qRT -PCR.The expression of miR -219 in SCC-15 and normal oral mucosa cell were also tested by qRT -PCR.The miR-219 mimic was transferred into SCC-15 cell.Cell proliferative rate and the ability of colony formation were analyzed .The abilities of migration and invasion in SCC-15 cell were evaluated by Transwell test .Results Expression of miR -219 was signifi-cantly lower in TSCC tissues(P<0.001)and SCC-15(P<0.05)in comparison with normal tumor adjacent tis-sues and normal oral mucosa cell.The abilities of proliferation(P<0.05),colony formation(P<0.05),migratio-nand invasion(P<0.05)were inhibited in miR-219-transferred SCC-15 cells.Conclusion The expression of miR-219 was downregulated in TSCC .The miR-219 could inhibit cell proliferation ,colony formation ,migra-tion and invasion in TSCC .
4.Study on TLC and Content Determination for Xiangshuyin Dispersible Tablets

Objective To establish the quality standard for Xiangshuyin Dispersible tablets. Methods Fructus citrus sarcodactylis and Borneolum syntheticum in the prescription were identified by TLC. The content of the total Naringin was determined by HPLC. The column of AichromBond-AQ-C18 (4.6 mm?150 mm,5 ?m) was used. The mobile phase was mobile phase A-mobile phase B (84∶16),mobile phase A was Water-Acetic Acid (100∶1),mobile phase B was Acetonitrile,at the flow rate of 1.0 mL/min,the column temperature of 30 ℃,and peaks were detected at 283 nm. Results The study on the quality control showed that the characteristic of identification by TLC was distinct and highly specific. The linear range of Naringin was 0.120 6~0.603 ?g,r =0.999 9,the average recovery was 97.90%,RSD=1.76% (n=5). Conclusion The method was easy to operate with accurate result and good reproducibility. It can be used effectively for the quality control of the preparation.
5.Evaluation of performance of the machine vision based on microscope

Objective To evaluate performance of the AVE‐766 automated urine sediment analyzer(AVE‐766) based on the ma‐chine vision for detecting erythrocytes(RBCs) ,leukocytes(WBCs) ,epithelial cells(ECs)and CASTs in urine specimen .Methods The within‐run variable coefficients(CVs) ,linearities and carryover rates for RBC ,WBC ,EC ,and CAST in urine specimens detected by using the AVE‐766 were analyzed ,the results of RBC ,WBC ,EC ,and CAST count in urine specimens detected by using AVE‐766 and Fast‐Read102 counting plate(Fast‐Read102) were compared .Results The within‐run CVs for RBC ,WBC ,EC ,and CAST detected by using AVE‐766 and Fast‐Read102 were increased by decreases of concentration of urine sediment .Good linearities (R2 >0 .97 ,P<0 .05) were observed for RBC(in the range of 60 -1 255/μL) ,WBC(in the range of 68 -2 718/μL) ,EC(in the range of 28-296/μL) and CAST(in the range of 5-86/μL) detected by using the AVE‐766 .The carryover rates for RBC ,WBC , EC ,and CAST detected by using AVE‐766 was 0 .9% or less .The values detected by using AVE‐766 were correlated well with those detected by using Fast‐Read102 for RBC ,WBC ,and EC in urine specimens(0 .67< r<0 .75) .However ,for CAST ,the values detected by using AVE‐766 were poorly correlated with those detected by using Fast‐Read102(r=0 .183) .There were statistically significant differences between manual and automated urinalysis for RBC ,WBC ,EC ,and CAST in urine specimens(P<0 .05) .Con‐clusion The AVE‐766 could not take over microscopic examination ,only is suitable for the first screening to detect RBC ,WBC , EC ,and CAST in urine specimen .
6.Effect of adipose stem cells derived conditioned medium on fibrogenesis of dermal fibroblasts co-stimulated by transforming growth factor-β1

Objective · To clarify effect of adipose stem cells derived conditioned medium (ASCs-CM) on fibrogenesis of dermal fibroblasts costimulated by transforming growthfactor-β1 (TGF-β1), and explore the possible paracrine pathway of ASCs in regulating the dermal tissue rehabilitation.Methods · Co-stimulated by TGF-β1, dermal fibroblasts were treated with different concentrations of ASCs-CM. Variouscellular events including cell proliferation, apoptosis, and mRNA and (or) protein levels of α-smooth muscle actin (α-SMA), type Ⅰ collagen (COL-1) , type Ⅲ collagen (COL-3), hepatocyte growth factor (HGF), basic fibroblast growth factor (FGF-2) were explored. Furthermore, the effect of HGF antibody on apoptosis of fibroblasts caused by ASCs-CM and TGF-β1 were also observed. Results · ASCs-CM inhibited fibroblasts proliferation caused by TGF-β1 and lead to cell apoptosis. α-SMA expression in fibroblasts was attenuated by 10% ASCs-CM +TGF-β1. It was demonstrated that 100% ASCs-CM with TGF-β1 had promoted collagens (especially COL-3) expression in fibroblasts, and increased HGF mRNA level as well. HGF antibody inhibited fibroblasts apoptosis produced by 100% ASCs-CM and TGF-β1. Conclusion · ASCs may have an effect on fibrogenesis of dermal fibroblasts co-stimulated by TGF-β1 through a paracrine way.High concentration of ASCs-CM not only increases collagen production and secretion, but also inhibits fibroblasts proliferation and accelerates apoptosis.
7.Treatment of non-small cell lung cancer comes to the age of immunotherapy

Immune checkpoint inhibitors have become an important alternative for advanced non-small cell lung cancer (NSCLC) patients to surgery, chemotherapy, radiotherapy and targeted therapy. Monoclonal antibodies directed against immune checkpoint have shown better results in the application of first- or second-line treatment of NSCLC and for both squamous and non-squamous cell carcinoma patients, especially for those with positive PD-L1 tumor cells. Some comments will be made in present paper about the efficacy, biomarker, combined therapy and the resistant mechanism of immune checkpoint inhibitors.
10.Risk factors for deep vein thrombosis in lower extremity after hip or knee arthroplasty and hip fracture internal fixation

Objective To determine the factors contributing to the development of deep vein thrombosis (DVT) in the lower extremity in patients after hip or knee arthroplasty and hip fracture internal fixation.Methods One hundred and forty-seven consecutive patients receiving hip or knee arthroplasty and hip fracture internal fixation from 2004 to 2005 were included in this study. Their age ranged from 33 to 92 years. Duplex color ultrasonic inspection was performed on veins of the bilateral lower extremities before operation and 2 weeks after operation for detection of DVT. The patients were divided into a DVT group and a DVT-free group based on the development of DVT after operation. Detailed perioperative clinical information about the patients, surgery and anesthesia was collected.Results Lower extremity DVT was found in 42.2% of the patients after operation, while the incidence of proximal DVT was 2.7%. Compared with the DVT-free group, the usage rate and dosage of ephedrine increased significantly, the duration of anesthesia was significantly longer, and the white blood cell count (WBC) on the 1st postoperative day and the highest WBC count were significantly higher in the DVT group(P<0.05). Logistic regression analysis indicated that the above factors were closely related to DVT.Conclusion Duration of anesthesia > 3 h, ephedrine administration and a marked increase in WBC count after operation are the risk factors for DVT in the lower extremities in patients after hip or knee arthroplasty and hip fracture internal fixation.
