1.STUDY ON THE ANTIOXIDATIVE AND ANTIAGING EFFECT OF THE BIOACTIVE EXTRACT FROM AGROCYBE AEGERITA

Objective To investigate the antioxidative and antiaging effects of the bioactive extract from Agrocybe aegerita. Method Fruit flies and D-galactose induced aging mice were fed with the bioactive extract from Agrocybe aegerita. The lifespan of fruit fly and spleen index, brain index,SOD activity in serum and lipofuscin level in 1iver of the mice were measured, and its acute toxic effect was studied. Results The bioactive extract from Agrocybe aegerita could significantly increase the mean lifespan and half death time of fruit flies. The percentage of life prolongation was 41.81% compared with the control. Compared to the models, SOD activity in serum,spleen index and brain index of male mice fed with 800mg/(kg.d) of the bioactive extract from Agrocybe aegerita were respectively increased by 42.36%, 33.33% and 17.85%,while lipofuscin content of liver decreased by 50.06%. 20g/(kg.d) of the bioactive extract from Agrocybe aegerita was safe for mouse. Conclusion The bioactive extract from Agrocybe aegerita have obviously antioxidative and antiaging effects with little side effect.

3.Effects of antagonistic peptides binding specifically with first and second extracellular loops of CCR5 on colitis rats induced by TNBS

AIM:To study the effects of antagonistic peptides binding specifically with the first and second extracellular loops (ECL1 and ECL2) of C-C chemokine receptor 5 (CCR5) on the colitis rats induced by trinitrobenzenesulfonic acid (TNBS) and the mechanisms.METHODS:The colitis model of SD rats was induced by TNBS (100 mg/kg).The effects of 2 antagonistic peptides at different doses (ECL1:25, 35 and 45 mg/kg;ECL2:15, 25 and 35 mg/kg) on the model rats including the changes of disease activity index (DAI), colon macroscopic damage index (CMDI) and histological grading were observed.The mRNA and protein expression levels of TNF-α and COX-2 in the colonic mucosa were detected by real-time PCR and Western blot, respectively.RESULTS:Compared with model group, the changes of DAI, CMDI and histopathological injury of the rats treated with ECL2 antagonistic peptide HY at an appropriate dose were significantly reduced (P<0.05), and the protein and mRNA expression levels of TNF-α and COX-2 were significantly decreased (P<0.05).However, the effects of ECL1 antagonistic peptide GH on all scores and the expression levels of TNF-α and COX-2 were not obvious.CONCLUSION:ECL2 antagonistic peptide HY relieves TNBS-induced colitis in SD rats via down-regulating the expressions of TNF-α and COX-2 in the colonic mucosa, while the effect of ECL1 antagonist peptide GH was not obvious.
4.Performance of osteoporosis self-assessment screening tool for middle aged and elderly healthy Asians men in Chengdu

Objective To evaluate the screening effect of osteoporosis self-assessment tool for Asians (OSTA) in middle aged and elderly healthy men in Chengdu.Methods A total of 4042 healthy men aged 40 to 106 years received dual energy X-ray absorptiometry (DXA) assay,and OSTA index evaluation.Measurement sites included lumbar spine (L1-4),left femoral neck,trochanter,Ward's area,total hip and femoral shaft.All persons were classified into highosteoporosis-group (OSTA≤-4),mediumosteoporosis-group (-4 < OSTA≤≤-1),low osteoporosis-group (OSTA>-1),or the low risk-group (OSTA>-1) and high risk-group (OSTA≤-1) by OSTA scores.T-scores were compared between different measurement sites detected by DXA.The sensitivity,specificity,Kappa value and the area under receiver operating characteristic (ROC) curve (AUC) of OSTA in screening osteoporosis were evaluated.Results The prevalence of osteopenia and osteoporosis in lumbar spine,proximal femur were gradually increased along with aging.The detection rate of osteoporosis in lumbar spine and proximal femur were 16.2% and 24.0% respectively in subjects aged over 80 years.OSTA index in low-risk,medium-risk group,high-risk group were 85.0%,11.0%,4.0% respectively.The detection rate of osteoporosis in lumbar spine and proximal femur were 2.6% and 1.6% in low-risk group,10.4% and 10.4% in medium-risk group,and 29.3% and 30.5% in high-risk group,respectively.Taking OSTA ≤-1 as the cut-off value,the sensitivity and specificity of OSTA in screening osteoporosis in lumbar spine and femur by T-score<-1 were 28.1%,28.7 %,89.0% and 92.4% respectively,and by T-score≤-2.5 were 51.6%,63.2%,86.7% and 86.8% respectively.The consistency of diagnosis result between T-score and OSTA index according to the three versus two risk levels was 0.153 and 0.197 versus 0.195 and 0.243 Kappa value,respectively.The AUC of OSTA index for lumbar spine and femur by T-score<-1 and T-score≤-2.5 were 0.689 and 0.823,and for different age groups and different measurement sites were 0.639 and 0.899 (all P<0.001).Conclusions OSTA index has a certain ability in screening osteoporosis in men aged over 50 years.There are different screening results on osteoporosis among the different age groups.
5.Automated breast volume imaging with color doppler ultrasound in evaluating effect of neoadjuvant chemotherapy for breast cancer

Objective: To explore automatic breast full volume imaging (ABVS) and color doppler ultrasound (CDFI) for breast cancer neoadjuvant chemotherapy (NCT) in the evaluation of curative effect. Methods:The application of a CDFI and ABVS 42 cases of breast cancer patients receiving NCT lesions imaging characteristics before and after observation and analysis. Results:ABVS from coronal distribution and audio-visual change according to the primary tumor, and CDFI images showing the primary tumors and blood flow changes within all have significant difference in patients with complete remission after NCT (P<0.01);ABVS from coronal distri-bution and audio-visual change according to the primary tumor, and CDFI showed lesions in blood flow changes, etc. in patients with the absence of change and easing some lesions have no significant difference (P>0.05). Conclusion:We use ABVS and CDFI for breast cancer before and after the NCT curative effect evaluation of great clinical significance and value.
6.X-ray diagnostic site selectivity studies of skeletal fluorosis

Objective To study the X-ray signs of forearm and leg in skeletal fluorosis and its diagnostic value,aim at finding the easy examination parts.Methods One thousand four hundred and forty subjects were examined using developed shield,darkroom and other portable dedicated device combined with a small X-ray machine.A total of 384 cases were diagnosed skeletal fluorosis.All patients were divided into different groups and the time,degree and range of X-ray to the forearm and calf elbow,knee,and long bone were compared.Results The X-ray change in the forearm elbow was earlier than that of the leg knee,and trabecular bone change was the earliest indicator,197 cases and 157 cases,respectively,and the difference was statistically significant (x2 =28.006,P < 0.01).Membrane ossification of forearm backbone was earlier than that of the leg,and most of them were degree Ⅰ photos,213 cases and 126 cases respectively.The difference was statistically significant (x2 =17.626,P < 0.01).The direction of the interosseous membrane ossification was from the forearm radius to the ulna,then to the fibula and tibia,and was accompanied by changes in the aggravation of forearm.A variety of indicators were observed,especially the membrane ossification in bone and joint trabecular bone and the long bone was the most active,and the forearm was more sensitive,obviously than that of the calf.Conclusion In the X-ray screening or detection of endemic fluorosis,the forearm radiography is a simple,economical,and effective diagnostic method.
7.Predictability of multi-slice CT perfusion in the restorability of renal function of hydronephrotic kidneys

Objective To evaluate the predictability of MSCT perfusion in the restorability of renal function of hydronephrotie kidneys with unilateral partial ureteric obstructed rabbit model as to explore a method to predict the restorability of renal function of hydronephrotic kidneys and to investigate the changes of MSCT perfusion parameters during the course of the restore of renal function.Methods Establish a unilateral partial ureteric obstructed rabbits hydronephrotie model.Hydronephrotie rabbits were grouped as control,2,4 and 8 week(G_2w,G_4w and G_8w)after obstruction and the later 3 groups of rabbits were reared for further 4 weeks after the obstruction was released.MSCT perfusion scanning was performed and the specimen was made into histological slices with HE staining.Results BF and BV value of renal cortex and medulla of G_2w after obstruction [(864?32)ml?100 g~(-1)?min~(-1),(19.5?0.9)ml/100 g (cortex ); (182.1?7.5)ml?100g~(-1)?min~(-1),(8.37?0.51)ml/100g(medulla)]was released restored in substance and approached that of control[(899?63)ml?100g~(-1)?min~(-1),(21.6 + 1.4)ml/100 g (cortex);(193.5?16.5 )ml?100g~(-1)?min~(-1),(8.50?0.54 )ml/100 g (medulla)]while there was no significant restore in that of G_4w and G_8w after obstruction[(525?15)ml?100g~(-1)?min~(-1),(12.8? 0.6)ml/100g (G_4 w);(512?10)ml?100g~(-1)?min~(-1),(9.4?1.0)ml/100 g (G_8w)] was released. Histologically,there was a positive correlation between the duration of obstruction and the seriousness of pathologic changes.Conclusion MSCT perfusion can provide information not only morphologically but also about renal perfusion of hydronephrotic kidneys.
8.Study of multi-slice helical CT perfusion imaging in cerebral neoplasms

Objective To study the potential of multi-slice spiral CT perfusion imaging in the assessment of cerebral neoplasms.Methods Multi-slice helical CT perfusion imaging was performed in 38 patients with cerebral tumors.The perfusion imaging of the tumor was carried out by cine scan technique (ls/1 rotation) with slice thickness 5 mm/4i,reconstructed slice thickness 10 mm/2i.Contrast injection was done by using 50 ml nonionic contrast agent(300 mg I/ml),at a flow rate of 3.5 ml/s with a power injector,and 5 seconds delay,and data acquisition lasted for 45 seconds.The scanning images were processed in ADW 4.0 workstation.BF、BV、PS values of tumors were calculated and statistically analysed. Results 38 patients with cerebral neoplasms included 9 cases of gradeⅠ—Ⅱgliomas(group 1),10 cases of gradeⅢ—Ⅳgliomas (group 2),9 eases of metastases (group 3)and 10 cases of meningiomas (group 4).All raw data was transformed to square root so as to be consistent with normal distribution.BF~(1/2) values for groupl to group 4 were (5.99?1.03)、(7.55?1.57)、(7.72?2.02 )、(11.40?2.13)ml?rain~(-1)?kg~(-1).The differences in BF~(1/2) were statistically significant between group 1 and group 2,between group 1 and group 3,between group 1 and group 4,between group 2 and group 4,between group 3 and group 4(t_(1,2)=6.89,t_(1,3)=4.59,t_(1,4)=11.03,t_(2.4)=10.58,t_(3,4)=7.65,P<0.05),and there was no statistically significant difference between group 2 and group 3(t_(2.3)=1.17,P>0.05);BF~(1/2) values for groupl to group 4 were (1.01?0.19)、(1.42?0.38)、(1.25?0.33)、(1.60?0.24)ml?kg~(-1).The differences in BF~(1/2) were statistically significant between every two groups (t_(1,2)=7.15,t_(1.3)=3.71, t_(1.4)=5.93,t_(2.3)=2.94,t_(2,4)=2.72,t_(3.4)=4.46,P<0.05 );PS~(1/2) values for groupl to group 4 were (1.70?0.37)、(3.63?0.95)、(4.29?1.30)、(5.69?1.03)ml?min~(-1)?kg~(-2).The differences in PS~(1/2) were statistically significant between every two groups(t_(1.2)=11.53,t_(1.3)=10.61,t_(1.4)=16.77,t_(2.3)=3.69, t_(2,4)=9.94,t_(3,4)=5.52,P<0.05).Conclusion Multi-slice helical CT perfusion imaging is very useful to evaluate tumor vessels of cerebral neoplasmas and it can provide information of incremental benefit in diagnosis,in staging of tumor grade,in the distinction of benign from maglignant cerebral neoplasmas and in differentiating intracerebral neoplasmas from extracerebral neoplasmas.
9.Effects of hyperbaric oxygenation combined with As(2)O(3) on proliferation of K562 cells and associated mechanism.

This study was aimed to explore the effects of hyperbaric oxygenation (HBO) alone or combined with As(2)O(3) on proliferation, apoptosis and expression of HIF-1a, VEGF, caspase-3 mRNA of K562 cells, and the molecular mechanism of As(2)O(3) enhancing the anti-leukemic effect of HBO so as to provide a scientific basis for clinical treatment of chronic myeloid leukemia. The effects of drugs on proliferation of K562 cells was assayed by MTT method, the apoptosis rate of K562 cells was detected by flow cytometry with Annexin V/PI double staining, the expressions of HIF-1a, VEGF, caspase-3 mRNA of K562 cells were determined by real-time quantitative PCR. The results showed that as compared with As(2)O(3) alone, HBO combined with As(2)O(3) could increase inhibitory rate of K562 cell proliferation, and enhance apoptotic effect, obviously down-regulate expressions of HIF-1a and VEGF mRNA, up-regulate expression of caspase-3 mRNA. The effect of HBO combined with As(2)O(3) was higher then effect of As(2)O(3) alone, and their effects were synergistic (P < 0.05). It is concluded that HBO combined with As(2)O(3) can increase the expression of caspase 3 mRNA and decrease the expression of HIF-1a and VEGF mRNA, which may be one of molecular mechanisms underlying their synergistic antileukemia efficacy.

10.Effects of arsenic trioxide combined with bortezomib on proliferation and apoptosis of K562 cells and their mechanism.

This study was aimed to investigate the effect of arsenic trioxide (As(2)O(3)) alone and in combination with bortezomib (Bor) on proliferation and apoptosis of leukemia cell line K562, and to analyze the potential mechanism. K562 cells were treated with different concentrations of As(2)O(3) or Bor (alone or combination) for 24, 48 h. MTT method was used to detect the cell proliferation. After K562 cells were treated with 0.5 µmol/L As(2)O(3) alone or in combination with 10 nmoL/L Bor, the apoptosis rate and cell cycle were measured by flow cytometry, and the activity of NF-κB was analyzed by SP immunohistochemistry. The results indicated that the different concentrations of As(2)O(3) and Bor could inhibit the K562 cell proliferation in a time- and dose-dependent manners (P < 0.05). The IC(50) of Bor and As(2)O(3) in 48 h were 20 nmol/L and 0.6 µmol/L respectively. When K562 cells were treated with As(2)O(3) or Bor alone for 24 h, the apoptotic rate of K562 cells increased, and the apoptotic rate in combination group was higher than that in As(2)O(3) or Bor group. The cells were apparently arrested in G(2)/M phase in Bor group and G(0)/G(1) phase in As(2)O(3) group. The activity of NF-κB decreased significantly in As(2)O(3) or Bor group (P < 0.05), this effect was most significant in the combination group (P < 0.01). It is concluded that both As(2)O(3) and Bor can inhibit the proliferation and induce apoptosis of K562 cells, a synergistic effect can be observed when a low dose of As(2)O(3) combined with low dose of Bor. The different cell cycle block site and the decrease of activity of NF-κB may be one of the mechanisms underlying their synergic effect.
