1.Clinical study on serum soluble CD8 antigen of patients with Graves disease

Objective:To study the clinical value of serumsoluhle CD8 antigen(sCD8)inpatients withGraves disease(GD).Methods:The level of serum sCD8 was detected by ELISA in40 healthy controls and 46 GD Patients.Serial measurement were carried out in 28 patientsbefore and after trea treatment with PTU. Results: Serum sCD8 levels were increased signifiantly in untreated GD patients as compared with conrols(21.91 ±9.13 kU/Lvs9.84±3.08kU/L, P <0.001).After treatment with PTU for 2 to 7 months,sCD8 levels tetumed to normal when eu-thyroidism was esta established in 22 patients(9.67 ± 5. 14 kU/L) but remained high in 6 unmiproved patients(38.74 ± 9.89 kU/L). No statisticalrelationship was found between sCD8 and T3、T4、TSH、TGA、 TMA.Conchusion: The level of serum sCD8 correlate well with disease activityand may be of value in disease prognosis. PTU show some suppressor effect on immune.
2.Expression of synapsin and dopamine transporter mRNA in hippocampus of chronic stress depression model rats

Objective To study the mechanism of depression by explore the expressional differences of synapsin and dopamine transporter(DAT) mRNA of hippocampus in depression model rats. Methods Depression model were replicated by chronic stress. 20 rats were randomly divided into normal control group(n=10) and de-pression model group(n=10). Synapsin and DAT mRNA expressions in hippocampns were detected with RT-PCR technique. Result The values of synapsin and DAT mRNA in hippocampus of normal control group were 0.84±0.08 and 1.24±0.08, respectively, and 1.24±0.12 and 1.85±0.09 of model group, their t values were 8.87 and 16.50 (P<0.01). Conclusion The expressions of synapsin and DAT mRNA in hippecampus of depression model rats were increased significantly.
3.Preparation and Drug Release in vitro of N-Trimethyl Chitosan-coated Sparfloxacin Nanoliposomes in Situ Gels

Objective:To prepare N-trimethyl chitosan (TMC)-coated sparfloxacin (SL) nanoliposomes in situ gels(ISG)and in-vestigate the drug release in vitro.Methods:SL liposomes were prepared by a pH gradient method , and then homogenized to nanolipo-somes by high pressure .TMC was used as the coating material to prepare TMC-coated SL nanoliposomes .Poloxamer 407 was used as the gel base, and the optimal amount was selected according to the gelation temperature .TMC-coated SL nanoliposomes ISG was pre-pared using a cold method , and the morphology , size, zeta potential and entrapment efficiency of TMC-coated SL nanoliposomes were studied.A membraneless model was used to study the drug release in vitro, and the result was compared with that of TMC-coated SL nanoliposomes.Results:The optimal amount of poloxamer 407 in the formula was 25%, and the gelation temperature was 23.6℃in the artificial tears and 33 .5℃in the diluted artificial tears .The morphology of TMC-coated SL nanaoliposomes in the ISG was spherical with the mean diameter of (96.8 ±1.5) nm, zeta potential of (46.2 ±1.4) mV and entrapment efficiency of (76.6 ±2.4) %, and the indices had no significant difference when compared with those of TMC-coated SL nanoliposomes .Both the drug release in vitro and gel dissolution profile of TMC-coated SL nanoliposomes ISG exhibited the characteristics of zero-order kinetics, and compared with that of TMC-coated SL nanoliposomes , the sustained release property of the ISG was more significant .Conclusion:TMC-coated SL nanoli-posomes ISG has promising gelation temperature and notable sustained release property .
4.Disputes revolved about surgeries of macular vitreoretinal interface abnormalities in highly myopic eyes

Macular vitreoretinal interface abnormalities in highly myopic eyes are among the most vision-threatening diseases associated with macular retinal schisis and macular holes.To relieve the traction of the posterior vitreous cortex and to recover the anatomy of fovea for good central vision are the keys to successful repair.However,there are many controversial issues in the efficacy of the surgerical procedures including gas injection,scleral buckling and vitrectomy.How to evaluate these different surgeries and to establish standard surgical procedure options for macular vitreoretinal interface abnormalities in highly myopic eyes needs to be explored.
5.Determination of Globin Beta-chains and Comparison of Genetic Type of SMMC/B Mice of Three Generations by Peptide Map Analysis

The separation and purification of beta chains of globm from the inbreeding SMMC/B mice were performed by the dissociation of chains in urea, CMhcellulose column chromatography for the separation of alpha and beta chains, and Sephadex G25 column chromatography for removing salts. The pure beta-chain was digested by trypsin. Digested fragments were resolved by cellulose membrane two-way electrophoresis, thus obtaining the map of spots of beta-chain fragments for B33, B34, Bp(), Bp and Bf. The spots,BTp3, which have the same positions ,were separated and micro-sequences were assayed by DABITC/PITC double coupling technique. According to the genetic code, we might deduce the nucleotide sequence of their mRNA from the known amino acid sequence of BTp3, a piece of beta-chain. By analyzing the altered genetic loci of beta-chain of globin, we might determine the genetic purity of the strain and distinguish objectively the genetic types between species and subspecies.
6.Analysis of risk factors of pneumonia in patients with non ST segment elevation-acute cononary syndromes

Objective To investigate the risk factors of pneumonia in patients with non ST segment elevation-acute cononary syndromes (NSTE-ACS).Methods A total of 183 cases with NSTE-ACS was divided into pneumonia and non-pneumonia groups.The possible risk factors were screened firstly,then nonconditional logistic model multivariate analysis was used to investigate risk factors including age,sex,weight,smoking,chronic obstructive pulmonary disease (COPD),hypertention,diabetes,left ventricular failure,renal failure,low serum albumin,early revascularization and early rehabilitation exercise on the occurrence of pneumonia in NSTE-ACS.Results The differences were statistically significant in factors including age,smoking,COPD,diabetes,left ventricular failure,renal failure,low serum albumin,early revascularization,and early rehabilitation exercise between two groups (P < 0.05).By analysis of logistic,risk factors on the occurrence of pneumonia in NSTE-ACS were age,smoking,COPD,diabetes,left ventricular failure,renal failure,and low serum albumin(P <0.05).Early revascularization and early rehabilitation exercise were the protective factors to avoid pneumonia.Conclusions Left ventricular failure,age,smoking,COPD,diabetes,renal failure,and low serum albumin were the risk factors of pneumonia occurred in NSTE-ACS,but early revascularization and early rehabilitation exercise were the protective factors to avoid pneumonia.Actively treating the risk factors and early initiating the protective factors can effectively reduce the incidence of pneumonia.

8.Effect of lentivirus mediated HER2-shRNA on chemosensitivity of human ovarian cancer SKOV3 cells

Objective To investigate the lentivirus-mediated RNA interference(RNAi) reduced human epidermal growth factor 2 (HER2) gene expression in ovarian cancer chemosensitivity.Methods Specifically targeted short hairpin RNA HER2 gene ( shRNA) lentiviral expression vector and negative control sequences lentiviral vector were constructed.stable expression of HER2-shRNA in SKOV3 cell line ( KD) and negative control cell line (NC) were established.The uninfected SKOV3cell lineserved as blank control(CON)group, sensitivity to changes in three kinds of cells to cisplatin chemotherapy in the HER2 gene expression was detected by RT-PCR and Western blot technique, CCK-8 assay CON and KD cells strains were established xenograft model in nude mice in each group to detect the expression of the HER2 protein, as well as by cisplatin chemotherapy tumor size. Results Expression results showed that a stable cell line HER2-shRNA and negative control cell lines had been successfully establish by fluorescence and flow cytometry.RT-PCR results showed KD cells relative HER2 mRNA expression levels compared with the NC group, the difference was statistically significant (P<0.01), whereas no significant difference when compared with the CON group, HER2 mRNA KD Group inhibition rate was (95.5 ± 0.12)%.After transduction specific HER2-shRNA ovarian cancer tumor cells, HER2 protein expression was significantly reduced, growth had slowed down significantly after cisplatin treatment ( P<0.01 ) .Conclusion Lentivirus-mediated HER2-shRNA can effectively inhibit the expression of the HER2 gene, increase chemosensitivity to cisplatin in ovarian cancer.
9.Clinical features and pathogenesis of paroxysmal dyskinesias

Objective To investigate the clinical characteristics and pathogenesis of paroxysmal dyskinesias.Methods Retrospective analysis was performed in 5 patients suffering from paroxysmal kinesigenic choreoathetosis(PKC) and 2 patients with paroxysmal persistent exercise-induced dystonia(PED).Results The episodes of all cases of PKC were induced by sudden movements.3 cases showed rigidty and hypertonia.3 cases presented with twist of limbs and dystonia.2 PED cases were induced by persistent movement,manifested involuntary movements of limbs,and the duration of the attack usually last seconds to minutes.5 patients showed epileptic discharges in EEG or AEEG.2 patients had abnormal findings of brain CT or MRI.4 PKC cases responded well to carbamazepine and 1 PED patients to large dose of valproate sodium.Conclusions Paroxysmal dyskinesias are usually induced by sudden movement and present paroxysmal extrapyramidal symptoms.Most of the patients show epileptic discharges in EEG and responded well to antiepileptic drugs.This implies the underlying relationship of pathogenesis between paroxysmal dyskinesia and epilepsy.
