1.Effects of cisplatin,oxymatrine on the cell growth and telomerase activity of QGY hepatoma cells

Objective:To investigate the anti-cancer mechanism of cisplatin,oxymatrine by observing their effects on telomerase activity.Methods:The inhibition of proliferation in QGY cells were estimated by MTT-test.Distribution of cell cycle and apoptosis were analysed using flow cytometry.The changes of telomerase activity were detected by PCR-ELISA method.Results:These two drugs could inhibit the proliferation of QGY cells,block the cell cycle,induce its apoptosis and inhibit the activity of telomerase.Conclusion:The inhibition of telomerase activity may be one of the mechanisms for the anti-cancer effect of the two drugs.
2.Growth-inhibiting effects of Taxol on QGY cells:G_2/M arrests and apoptosis

Objective:To investigate the effects of Taxol on QGY human hepatoma and its mechanism.Methods:The hepatoma cell growth was measured by MTT-microculture tetrazolium assay.Cell-cycle kinetics and apoptosis were analyzed by flow cytometry and microscopic examination.Results:Taxol inhibited the hepatoma cell growth in concentration- and time-dependent manners.Marked cells accumulated in G 2/M phase. The rate of cell growth inhibition was 78.6% after intervention of 56?mol/L Taxol for 72 hours. The cells showed G 2/M arrest and apoptosis.Conclusion:Taxol suppresses the growth of QGY human hepatoma cells in vitro by causing cell-cycle arrest, and apoptosis of the human hepatoma cells. The mechanisms of Taxol provide theoretical guidance for clinical treatments.
3.Study on establishment of training courses for emergency nursing

Objective To investigate the present status of the emergency nursing training to form the curricula system for emergency nursing training of nurses (including course projects,class hour,teaching form,training aim).Methods Based on the actual investigation,literature research,using Delphi method,the training course system of emergency nursing was established.Results Cronbach's α coefficient was 0.943 and the expert authority coefficient was 0.868.Emergency training should be included in the nursing training program,and the four modules courses were built.With reference to emergency management requirements of home and abroad and the training model of foreign troops as well as combining the reality of our country to set up the training objectives,to improve class hour,enrich teaching form,and adjust the training content to set up the emergency training evaluation model.Conclusions The results of this study supply basic guarantee for nurses to increase coping ability of emergency affairs as well as training basis and reference with clear structure and content,reasonable design and reliable results.
4.Study of HBV-X gene mutation among patients with HBV-related chronic hepatitis,liver cirrhosis,and primary liver cancer

Objective To study the relationship between hepatocarcinogenesis and the mutation in X gene among patients with chronic hepa-titis B virus (HBV)infection,such as chronic hepatitis B (CHB),liver cirrhosis (LC)and primary liver cancer (PLC).Methods The serum samples from 89 patients with chronic HBV infection who visited the Second Affiliated Hospital of Chongqing Medical University from 201 1 to 2013 were collected.PCR was used to amplify the X gene of HBV DNA extracted from the serum samples.After sequencing,the HBV-X genome was compared with those reported in GenBank to find the variable sites and variant forms.Chi -square and one -way ANOVA were used for the statistical analysis afterwards,whereas genotypes were determined by the genotyping tool of the National Center for Biotechnology Information.Results All patients were genotype B or C.Among HBeAg-positive patients,46.2% were genotype B,and 53.8% were genotype C;among HBeAg-negative patients,81.2%were genotype B,and 18.8%were genotype C (P=0.001).PLC pa-tients had a significantly higher risk of mutation in the basic core promoter (BCP)region than the CHB and LC groups (69.2%vs 34.4%and 61.3%,P<0.05);in addition,an evident T-base deficiency was observed at nt1821 site (88.5% vs 53.1% and 71%,P=0.014).Among CHB and LC patients,those with genotype C had a significantly higher risk of BCP double mutation than those with geno-type B (61.5%vs 15.8%,P=0.007;83.3%vs 47.4%,P=0.045).The incidence of BCP double mutation was significantly higher in the low-viral load group (≤106 copies/ml)than in the high-viral load group (>106 copies/ml)(81.3% vs 47.9%,P=0.015). Conclusion The BCP double mutation and T-base deficiency at nt1821 site may play important roles in the development of PLC.
5.Effect of Xingnaojing Injection Combined with Head Temperature-decreasing Instrument for the Treatment of Patients with Acute Hemorrhagic Stroke

Objective To observe the therapeutic effect of Xingnaojing Injection(XI) combined with head temperature-decreasing instrument in the treatment of patients with acute hemorrhagic stroke(AHS).Methods Sixty AHS patients were randomized into two groups: the control group received routine western treatment of decreasing intracranial pressure,controlling cerebral edema,protecting brain cells,and preventing infection and complications;the treatment group received XI combined with lowering head temperature based on the routine western treatment.Twenty-one days constituted one treatment course.After one treatment course,therapeutic effect was evaluated,and the volume of intracerebral hematoma and the cerebral edema was observed.After two treatment courses,the scores of neurological deficit and the activities of daily life(ADL) were observed.Results(1) After one treatment course,the total effective rate was 80.0% in the treatment group and 63.3% in the control group,the difference being significant(P

7.Clinical analysis of 134 patients with ileocecal disease

Objective To investigate the clinical characteristics and diagnosis and treatment of ileocecal disease.Methods The general data,clinical manifestations,imaging examinations,colonoscopy and pathological examination and diagnosis of 134 patients in our hospital with ileocecal disease from September 2009 to March 2016 were collected.The clinical characters were summarized by retrospective analysis.Results A total of 134 cases with ileocecal disease were collected,and 36(26.87%)of them were ileocecal cancer,30(22.39%)of them were inflammatory bowel disease,26(19.40%)of them were intestinal tuberculosis.The main clinical manifestations of ileocecal lesion were abdominal pain,abdominal distention,diarrhea,bloody stool,etc.The colonoscopy imaging showed mucosal hyperemia,edema,erosion,ulcer and tumors on the intestinal tract.The benign lesions mainly showed ulcer with mucosal erosion and edema,and the ileocecal cancer showed neoplasm.CT examination was the common imaging method,and 93(69.40%)cases got tested.49 cases had operation,in which 7 cases had misdiagnosed before operation,and the rate of misdiagnosis was 14.28%.Conclusion The ileocecal disease is mainly benign lesions,and the process of diagnosis is complex;the rate of misdiagnosis is a little high.
8.In vitro Corneal Permeation and Antibacterial Activity of N-Trimethyl Chitosan-coated Sparfloxacin Lac-tate Nanoliposomes

Objective:To study the in vitro corneal permeation and antibacterial activity of N-trimethyl chitosan(TMC)-coated sparfloxacin lactate(SL)nanoliposomes. Methods:Franz diffusion cells were used with rabbit cornea as the barrier to study the in vitro corneal permeation of TMC-coated SL nanoliposomes,and the permeation parameters were calculated. Eseheriehia eoli,staphylo-eoeeus aureus,pseudomonas aeruginosa and baeillus subtilis were used as the tested bacterial strains and the in vitro antibacterial activity of the SL preparations was investigated to obtain the minimum inhibitory concentration(MIC),the minimum bactericidal concentration (MBC)and the relationship of bacterial inhibitory rate and time. Results:The order of steady permeation rate(J),corneal perme-ation coefficient(P)and corneal retention rate was TMC-coated SL nanoliposomes > SL nanoliposomes > SL eye drop. The order of lag time(τ)and corneal diffusion coefficient(D)was SL nanoliposomes > SL eye drop > TMC-coated SL nanoliposomes. The in vitro an-tibacterial activity showed the order of TMC-coated SL nanoliposomes > SL nanoliposomes > SL eye drop. Conclusion:Compared with the eye drop,liposomes can enhance the corneal permeation and storage of SL with improved antibacterial activity,and TMC-coating can further improve the permeation and antibacterial activity,which is worthy of further study.
9.Effect of free radicals induced by electrolysis on endothelium derived relaxing factor in rabbit cerebral basilar artery

The effect of free radicals induced by electrolysis on endothelium derived relaxing factor (EDRF) was studied in the isolated rabbit basilar artery using bioassaytechnique. It was found that after the injury was made by the free radicals, the relaxa-tion of the artery rings induced by Ach was decreased markedly while the contractioninduced by NE was very much augmented. Meanwhile, in the basilar arteries MDA con-tent was increased and SOD activity was decreased. Pretreated the basilar artery with SOD,led to the recovery of decreased relaxation and attenuation of changes in MDA and SOD.These results suggested that during subarachnoid hemorrhage, the release of EDRF wasdecreased which was closely related to the free radicals injury of the basilar arteries.
10.EXCISION OF EXTENSIVE EYELID MALIGNANT TUMOR AND PRIMARY PLASTIC SURGERY REPAIP

The optimal distance from tumor to incision varied according to the cellular type of the tumor. In each case the cutting edge of the excised tissue was checked with frozen sections for cancer cells. Multiple neighboring skin flaps in full thickness raised from the periosteum were used to cover the large defects (the smallest defect is 25 20mm, the largest one is 75 65mm). The results showed that good cosmetic results were obtained in all cases, visual function was preserved in 35 of the 36 patients, and no recurrence of the tumor was found in 5 years' follow-up. One patient with squamous cell carcinoma that disseminated to nasal cavity and sinuses had a complication of meningitis after the operation,and it was cured with proper treatment.
