Journal of Biomedical Engineering 2014;31(3):648-651
To study the effect of the proliferation and apoptosis of Survivin-T34A mutant on breast cancer MCF-7 cell, we adopted the method of cell culture in vitro to observe the proliferation and apoptosis of the cell. In the experiment, MCF-7 cells were randomly divided into three groups and transfected with normal saline, PORF-9-null and Survivin-T34A, respectively. Breast cancer nude mouse models were established to study anti-tumor effect of Survivin-T34A in vivo. The activity of the cells in the Survivin-T34A-transfected group was lower than that in PORF-9-null group. The increase of cell apoptosis was observed under electron microscopy, meanwhile the apoptotic rate was obviously higher than that in PORF-9-null control by flow cytometry. Tumor inhibition effects of the mouse which received the injection of Survivin-T34A intratumoral injection were apparent, and the inhibition ratio was as high as 47.1%. In conclusion, Survivin-T34A mutant has anti-tumor effect through efficiently inhibiting the growth of breast cancer MCF-7 cell and actively promoting apoptosis of cancer cells.

Chinese Journal of Tissue Engineering Research 2010;14(11):2022-2025
BACKGROUND:An increase in activity of muscle contraction can induce transcription of a variety of signaling molecules to activate a large number of gene expression within the nucleus through proprietary signaling pathway OBJECTIVE:To review studies related the peroxisome proliferator-activated receptor-gamma coactivator 1(PGC-1)and exercise induced skeletal muscle adaptations.METHODS:A computer-based online search of Pubmed was performed for relevant English articles published from January 1995 to January 2009 with the keywords of"PGC1,skeletal muscle, exercise, adaptations"The relevant articles about PGC-1 and exercise induced skeletaI muscle adaptations were included.and repetitive contents were excluded.PGC1 and mitochondrial oxidative metabolism, and exercise-induced skeletal muscle adaptation and PGC1s changed served as the evaluation indicators.RESULTS AND CONCLUSION:Of 57 collected articles,34 were classified and sorted according to the criteria.Endurance training results in profound adaptations for skeletal muscle, including mitochondrial biogenesis, capillary density, and fiber composition.Transcription factors are highly dependent on coactivator molecules to regulate training physiologic adaptation processes. The majority of transcription factors target genes are involved in mitochondrial biogenesis and metabolism. These transcriptional patterns may provide a basic framework for understanding the integration of mitochondrial biogenesis and function with signaling events that exercise induced energetic properties Post-transcription of PGC-1 alpha protein, there are a variety of protein modifications, with a variety of biological processes closely related to the possible adaptive mechanism of exercise-induced skeletal muscle.

BACKGROUND: The augmenter of liver regeneration (ALR) is a kind of important secondary liver regeneration factors, and it can particularity promote the liver regeneration. OBJECTIVE: To clone the RNA extracted from neonatal rat liver to ALR gene, and to analyze the gene using the bioinformatics. DESIGN, TIME AND SETTING: The study, gene molecule in vitro experiment, was performed at the Laboratory of Neurobiology, College of Life Science of Henan University between March 2008 and May 2008. MATERIALS: The liver tissues were harvested from 3-day-old Wistar rat of SPF grade. METHODS: Total RNA was exacted from rat liver tissues using guanidinium isothiocyanate-phenol-chloroform extraction method. According to the guideline of molecular biology experiment, the target fragment was amplified using reverse transcription-polymerase chain reaction. Alter purified, the target gene was recovered with gel recovery kit and subcloned to pGEM-T easy, and then was transformed to E.coli. The extracted piasmid was sequenced by Shanghai Sangon Biological Engineering Technology And Service Co., Ltd. The sequence was analyzed with molecular biological software. MAIN OUTCOME MEASURES: Polymerase chain reaction amplification band; analysis of gene sequence; sedne, threonine and tyrosine phosphorylation predictions; phytogenetic analysis of ALR. RESULTS: The potymerase chain reaction product was in concordance with target fragment. The analysis of the gene sequence proved it is the ALR gene. The overall length of the ALR gene was 378 bp, it was highly conservative in many species, for instance the Homo sapiens, the Mus musculus, the Bos Taurus and the Danio redo. CONCLUSION: In this experiment, we have cloned the ALR gene successfully.

As targeted drugs to B-cell malignancies, anti-CD20 monoclonal antibodies have been proved to be important in therapeutic antibody field. With three generations in more than ten years' development, the structures of these drugs have been improved, and many new indications have been found. Nowadays, these kinds of antibodies are not only used in the treatment of lymphoid malignancies, but also been proved to be useful in some autoimmune diseases treatment, and their new indications are still being expanded. With the optimization of their clinical dosage regimens, drug reaction has been increased, thus, therapeutic and side effects of anti-CD20 monoclonal antibody have been further improved as well. However, the exact mechanism of action of their combination therapy with other chemical drugs is still unclear, which remains to be further studied. This article reviewed new development of anti-CD20 therapeutic monoclonal antibodies research in recent years.

Posterior circulation ischemia (PCI) is a common disease and frequent encountered disease in clinical practice. It refers to the transient ischemic attack and the cerebral infarction in vertebral-basilar artery system. Vertebral aretery atherosclerosis is one of the common causes of PCI. Imaging examinations for PCI include transcranial Doppler (TCD), digital subtraction angiography (DSA), computerized tomographic angiography (CTA), magnetic resonance angiography (MRA), color Doppler ultrasound (CDU) and so on. TCD can serve as initial screening tool in diagnosis of PCI. DSA is still thegold standardin diagnosis of vertebral-basilar abnormalities, although the operation injury and photography drugs poisonousness have not been resolved. CTA has almost the same defects as DSA, while MRA is noninvasive. Some researches have reposed that MRA can exaggerate the vertebral artery depression level. The inner diameter and hemodynamics of the vertebral artery can be accurately measured by CDU, and it can be seen as a noninvasive examination for PCI,which is direct, accurate, convenient and repeatable. It can provide positive evidence for diagnosis and treatment of PCI. This article reviews the progess of CDU in diagnosis of vertebral aretery atherosclerosis.
