2.Effects of CurTn on proliferation of VSMC

Aim To explore the anti-proliferation effects of curcumin trinicotinate ( CurTn ) on vascular smooth muscle cell ( VSMC ) and its mechanism. Methods The cells were cultured in DMEM supple-mented with 10% fetal bovine serum. MTT assay was used to examine cell proliferation. FCM was used to observe cell cycle. The expressions of PCNA, Cy-clinD1 and p-ERK1/2 were analyzed using Western blot. Results CurTn could inhibit the proliferation of VSMC and showed a certain amount-time relationship. What’ s more, CurTn could increase the G1 phase pro-portion of cell, decrease the S phase proportion and the expression level of PCNA protein. It was also found that CurTn significantly inhibit the protein expression of p-ERK1/2 and Cyclin D1 . Conclusion CurTn may inhibit the proliferation of VSMC via downregulating the expression of CyclinD1 and p-ERK1/2 .
3.Expression and clinical significance of plasma long non-coding RNA MALAT1 in patients with oral squamous cell carcinoma

Objective: To investigate the expression of long non-coding RNA (lncRNA) metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) in plasma of patients with oral squamous cell carcinoma (OSCC) and its clinical significance. Methods: 70 OSCC patients and 50 healthy controls were included. The relative expression of MALAT1 in plasma was examined by quantitative realtime PCR. The expression of MALAT1 in plasma in 15 OSCC patients was analyzed retrospectively 30 days after operation. Results: The expression level of MALAT1 in plasma of OSCC patients was significantly higher than that of healthy controls(P< 0. 001). The expression level of MALAT1 in OSCC patients was significantly correlated with TNM stage, tumor differentiation and lymph node metastasis(P< 0. 05). After operation the expression level of MALAT1 in plasma of OSCC patients was significantly decreased(P< 0. 001). The AUC of the diagnosis of OSCC with MALAT1 was 0. 814, and the sensitivity and specificity were 87. 43% and 72. 00% respectively. Conclusion: MALAT1 can be used as an auxiliary diagnostic marker for OSCC.

Applied principle of the evidence was: systematic reviews, RCTs, the results from multiple factors ana-lysis, consensus, especially combined with Chinese experience and some lung cancer guidelines used in USA or Europe. All doctors who use the guideline in making therapeutic strategy must combine patients' conditions with the knowledge of biological behavior, dynamic change and response to treatment of lung cancer.
5.Differential expression of circular RNA in oral squamous cell carcinoma

Objective :
To analyze circular RNA (circRNA) expression profiles in oral squamous cell carcinoma (OSCC) and its clinical significance.
Methods :
The expression of circRNA was detected with circRNA microarray assay in three samples of OSCC tumor and matched adjacent tissues. Quantitative real-time PCR (RT-qPCR) was used to verify the expression of circRNA in 45 pair OSCC tissues and normal adjacent tissues. The relationship between the expression of circRNA and the clinicopathological characteristics of OSCC was analyzed. circRNAs/miRNAs interaction were predicted using Arraystar' s home-made miRNA target prediction software.
Results :
155 circRNAs were differentially expressed between the OSCC tissues and matched adjacent tissues, of which 45 circRNAs were up-regulated and 110 circRNAs were down-regulated in OSCC tissues (fold changes ≥1.5 and P < 0.05). In the selected three circRNAs that were most significantly upregulated or downregulated in OSCC, the RT-qPCR results showed that hsa_circ_0001874, hsa_circ_0001971 and has_circ_0067934 were increased, while hsa_circ_0000520, hsa_circ_0023944 and hsa_circ_0000140 were decreased in OSCC tissues versus normal adjacent tissues (P < 0.001). The results were generally consistent with the microarray data. Among the circRNA expression profiles in OSCC, the up-regulation of hsa_circ_0001874 was the highest and the expression of hsa_circ_0001874 was significantly correlated with TNM stage and tumor grade. The result of Arraystar' s home-made miRNA target prediction software indicated that miR-103a-3p, miR-107, miR-593-5p, miR-661 and miR-662 may be potential target genes of hsa_circ_0001874.
Conclusion
The differentially expressed circRNAs in OSCC tissues and normal adjacent tissues were identified, and these dysregulated circRNAs and their potential target gents may play important roles in the development of OSCC.
6.Construction and validation of a simple model for predicting the risk of prenatal depression

BackgroundMental illness during pregnancy has become a major public health problem in China over the recent years, and depression is the most common psychological symptom during pregnancy. Current research efforts are directed towards the therapy on prenatal depression, whereas the construction of prediction model for prenatal depression risk has been little studied. ObjectiveTo construct a simple model for predicting the risk of prenatal depression, thus providing a valuable reference for the prevention of maternal depression during pregnancy. MethodsA total of 803 pregnant women attending three hospitals in Nanchong city were consecutively recruited from May 2021 to February 2022. A self-administered questionnaire was developed for the assessment of social demographic variables, obstetrical and general medical indexes and psychological status of all participants, and Self-rating Depression Scale (SDS) was utilized to screen for the presence of maternal depression. Subjects were randomly assigned into modelling group (n=635) and validation group (n=168) at the ratio of 8∶2 under simple random sampling with replacement. The candidate risk factors of maternal depression during pregnancy were screened using binary Logistic regression analysis, and the predictive model was constructed. Then the performance of the predictive model was validated using receiver operating characteristics (ROC) curve. Results① Lack of companionship (β=-0.692, OR=0.501, 95% CI: 0.289~0.868), low mood during the last menstrual period (β=-1.510, OR=0.221, 95% CI: 0.074~0.656), emotional stress during the last menstrual period (β=-1.082, OR=0.339, 95% CI: 0.135~0.853), unsatisfactory relationship between mother-in-law and daughter-in-law (β=-1.228, OR=0.293, 95% CI: 0.141~0.609), and indifferent generally relationship between mother-in-law and daughter-in-law (β=-0.831, OR=0.436, 95% CI: 0.260~0.730) were risk factors for prenatal depression in pregnant women (P<0.05 or 0.01). ② Model for predicting the prenatal depression risk yielded an area under curve (AUC) of 0.698 (95% CI: 0.646~0.749), the maximum Youden index was 0.357 in modelling group with the sensitivity and specificity was 0.606 and 0.751, and an AUC of 0.672 (95% CI: 0.576~0.767) and maximum Youden index of 0.263 in validation group with the sensitivity and specificity of 0.556 and 0.707. ConclusionThe simple model constructed in this study has good discriminant validity in predicting of the risk of prenatal depression. 7.Application of diffusion tensor imaging scanning of conus medullaris in lower urinary tract dysfunction

Objective To investigate the signal abnormality of conus medullaris in patients with overactive bladder(OAB)and un-deractive bladder(UAB)by MRI diffusion tensor imaging(DTI). Methods From May,2021 to April,2023,23 patients with lower urinary tract dysfunction without trauma and supraspi-nal lesions were enrolled(case group).All patients underwent imaging urodynamics and pelvic floor electromy-ography.Based on the bladder contraction during the filling phase of urodynamics,the patients were divided into UAB group and OAB group.Eight healthy subjects were included as the control group.All participants under-went T10 to L5 spinal segment MRI scans and DTI scans.The position of conus medullaris was determined by comparing the DTI sequences with the MRI scans.The fractional anisotropy(FA),apparent diffusion coefficient(ADC),and relative anisotropy(RA)of the conus medullaris intermediate segment were compared. Results Twelve cases were in UAB group,and eleven in OAB goup.Abnormalities were found in the pelvic floor elec-tromyography in the case group.There was significant difference in sacral reflex arc nerve conduction testing be-tween UAB and OAB groups(P = 0.036).Compared with the control group,ADC increased(t = 2.185,P = 0.037)in the case group;FA decreased(t = 3.439,P = 0.005)and ADC increased(t = 4.582,P<0.001)in UAB group. Conclusion DTI is helpful to find the potential lesion of spinal cord in patients with lower urinary tract dysfunction.FA and ADC are valuable indicators for the diagnosis of conus medullaris injury.
8.Comparison of esophageal motility and reflux characteristics of patients with endoscopic-negative heartburn based on the Lyon Consensus

【Objective】 To explore the characteristics of esophageal motility and reflux of endoscopic-negative heartburn patients based on the Lyon Consensus Diagnostic Criteria and discuss the differential diagnosis value of the mean nocturnal baseline impedance (MNBI) and the postreflux swallow-induced peristaltic wave (PSPW) index for reflux hypersensitivity (RH) and functional heartburn (FH) patients. 【Methods】 We enrolled 132 patients with heartburn as the main symptom who visited the Gastroenterology Department of our hospital from January 2017 to June 2021, including 24 in the non-erosive reflux disease (NERD) group, 24 in the RH group, and 84 in the FH group. All the patients completed gastroscopy, esophageal high-resolution manometry and 24h-pH impedance monitoring. We analyzed and compared the related indexes of esophageal motility and reflux. The receiver operating characteristic (ROC) curve was used to analyze the esophageal proximal MNBI, distal MNBI, and PSPW indexes for the differential diagnosis of RH and FH. 【Results】 The lower esophageal sphincter (LES) average resting pressure, LES residual pressure and intact relaxation pressure (IRP) in NERD were lower than those in RH and FH (P<0.05). The number of acid reflux, the upright acid exposure time (AET), the supine position AET, the total AET, and the DeMeester score were higher in NERD than in RH and FH (P<0.05). The non-acid reflux, distal MNBI and PSPW indexes were lower in NERD than in RH and FH (P<0.05). The distal MNBI and PSPW indexes were lower in RH than in FH (P<0.05). However, there was no significant difference in proximal MNBI among the three groups (P>0.05). When the proximal MNBI, distal MNBI, and PSPW indexes were used alone respectively to diagnose RH, the area under the ROC curve was 0.480, 0.810, 0.682, respectively, with the sensitivities being 87.5%, 100% and 91.7%, and the specificities being 26.2%, 66.7% and 51.2%, respectively. 【Conclusion】 NERD patients have obvious abnormal esophageal dynamics, mainly manifested as LES relaxation, which further aggravates the pathological acid reflux, while patients with RH and FH are mainly exposed to physiological acid or non-acid reflux. In patients with endoscopic negative heartburn, distal MNBI value can improve the clinical diagnosis rate of RH and help distinguish RH from FH.
9. Circular RNA circHIPK3 acts as the sponge of microRNA-124 to promote human oral squamous cell carcinoma cells proliferation

Objective:
To explore the expression and clinical significance of circular RNA circHIPK3 in oral squamous cell carcinoma (OSCC), analyze the effect of circHIPK3 on the proliferation of OSCC cells.
Methods:
The expression of circHIPK3 in OSCC tissues, adjacent non-cancerous tissues and OSCC cell lines were detected by quantitative real-time polymerase chain reaction (qPCR). The correlations between the expression of circHIPK3 in OSCC tissues and the clinicopathological features were analyzed as well. circHIPK3-specific siRNA si-circHIPK3 and negative control siRNA si-NC were designed and synthesised and used to transfect CAL27 and SCC15 cells respectively. The proliferation capacity of CAL27 and SCC15 cells after transfection with si-circHIPK3 was detected by CCK-8 assay. The expression of miR-124 in OSCC was detected by qPCR, and the correlation between expression of circHIPK3 and the expression of miR-124 was analyzed. Using qPCR to detect the expression of miR-124 in CAL27 and SCC15 cells after transfection with si-circHIPK3 and si-NC respectively. Furthermore, using CCK-8 assay to detect the proliferation capacity of CAL27 and SCC15 cells after transfection with si-NC, si-circHIPK3, miR-124 mimic, si-circHIPK3+miR-124 inhibitor.
Results:
The expression of circHIPK3 in OSCC tissues [2.23 (1.86, 3.00)] was significantly higher than that of the adjacent non-cancerous tissues [1.05 (0.85, 1.26)] (
