1.Characteristics in frequency and phenotype of myeloid and plasmacytoid dendritic cells in chronic HCV infection

Objective To explore the frequencies and phenotype of myeloid and plasmacytoid dendritic cells (mDC and pDC) in chronic HCV infection and to investigate the relationships between DC frequencies and HCV viral load and serum ALT level. Methods PBMC were isolated from chronic HCV infected patients and healthy control. Multi-color flow cytometry was used to analyze the frequencies and surface marker expression on mDC and pDC. The relationship between DC frequencies and viral load and ALT level was also calculated. Results In comparison with healthy control, frequencies of mDC and pDC in chronic HCV infection were significantly decreased (0. 37 ± 0. 19 and 0. 19 ± 0. 12 vs 0. 51 ± 0. 18 and 0. 29 ± 0.13, P<0.05). The frequency of mDC was negatively correlated with HCV viral load (r= -0.5878, P < 0. 0001 ) and serum ALT level ( r = - 0. 4628 , P = 0. 003 ). Both costimulatory markers ( HLA-DR, CD83, CD86, and CD40) and coinhibitory marker (PD-L1) expression on mDC and pDC in HCV infection were increased (P<0.01 for costimulatory marker, P<0.05 or F<0.01 for coinhibitory marker). Conclusion The frequencies of mDC and pDC in chronic HCV infection were decreased, while the expression of costimulatory markers and coinhibitory marker were increased or not decreased in HCV infection. The decreased frequency of mDC was probably related to persistance of HCV infection.
2.Optimization of extraction method of total flavones in Lonicera japonica by orthogonal design

Objective To optimize and establish the extraction of total flavones in Lonicera japonica. Methods The ultraviolet spectrophotometry was used to determinate the total flavonoids. The effect of extraction time, the amount of solvent and extraction times,pH were tested by the orthogonal design.ResultsThe total flavonoids showed a linear relationship,the optimal extraction condition of total flavones in Lonicera japonica was with 16 times volume of sample weight water for three times, each time was 2 h.Conclusions The method is simple and applicable to the extraction of total flavones in Lonicera japonica.
3.Study on the technology of flocculation of water-extraction solution inLonicerae Japonicae Flos by uniform design method

Objective To study the technology of flocculation of water-extraction solution in Lonicerae Japonicae Flos by uniform design method.Methods The liquid concentration ratio, chitosan dosage, temperature and pH were studied with the ratio of the precipitation and the rate of the transformation of valid target as index.Results The optimal flocculation process was: dosage of chitosan was 0.14%, pH was 6, the concentration of the solution was 1:3 and the temperature was 30℃.Conclusions The effect of purification is good, and the flocculation process can replace the traditional precipitation process.
4.Optimized the preparation process of thermo-sensitive Yinhuang ophthalimic gel by central composite design-response surface method

Objective To optimize the preparation technology of thermo-reversible Yinhuang opthalimic gel by the central composite design-response surface method.Methods The poloxamer 407 and poloxamer 188 were used as the investigation factors, and the evaluation index was the gel temperature. Central composite design-response surface method was used to evaluate the mathematic relation between the evaluation index and tow investigation indexes to identify the optimum prescription.Results According to the quadratic models, it was found that there was reliable quantitative relation between the evaluation index and two investigation indexes,among which the optimun dosage was 20% for P407, 1% for P188, 0.1% for hyaluronic acid, 0.01% for benzalkonium chloride, 0.02% for EDTA.Conclusions This method is reliable and feasible, whicn can realize the prescription optimization of the gel.
5.Treatment to high-risk acute non-lymphocytic leukemia with sequential induction

Objective To investigate the outcome of high-risk acute non-lymphocytic leukemia treated with sequential low-dose cytarabine and harringtonine(LD-HA) and standard induction. Methods 50 high-risk ANLL. patients (LD-HA group) who were regarded as unfit for intensive chemotherapy were chosen to receive LD-HA. Reinductive treatments with standard regimens would be given for those who did not achieve complete remission. 23 patients DA/HA group given two cycles of standard inductive regimens were taken as the control. Results In LD-HA group 80.0 %. (40/50) reached CR, 2 patients died shortly after inductive therapy. The median leukemia-free survival(LFS) was 19.6 months, and the median overall survival (OS) was 12.2 months. Overall survival was 57.0 % at 1 year, 24.1% at 3 years, and 18.8 % at 5 years. While the CR rate was 73.9 % for DA/HA group, and none died during the inductive therapy. LFS and OS was 19.8 months and 12.1 months, respectively. OS rate was 56.58 % at 1 year, 27.1% at 3 years, and 27.1% at 5 years.There were no difference on OS rates between 2 groups (x2 were 0.009, 0.237 and 1.807, respectively,P >0.05). Conclusion In patients who were unfit for intensive chemotherapy, sequential therapy with LD-HA and standard induction improved the rate of complete remission and the duration of survival.
