1. The role of microtubule regulatory protein Nur77 in metastasis of prostate cancer and its mechanism

Objective: To investigate the clinical significance of Nur77 expression in prostate cancer tissues, and to explore the mechanism of its effect on the metastasis of prostate cancer. Methods: The expression of Nur77 in 72 cases of prostate cancer and 24 cases of benign prostatic hyperplasia was detected by immunohistochemistry. The relationship between Nur77 expression and the clinicopathological features of prostate cancer was analyzed. The co-localization of Nur77 and P-tubulin in prostate cancer PC-3 cells was detected by immunofluorescence. The interaction between Nur77 and β-tubulin was verified by co-immunoprecipitation assay. PC-3 cells were transfected with the different plasmids including pcDNA3.1-Nur77, Nur77ΔDBD, Nur77ΔTUBD and Nur77-shRNA by liposome, respectively. The soluble and polymerized tubulin fractions were differentially extracted and detected by Western blotting. The migration ability of PC-3 cells was detected by Transwell chamber assay. Results: The expression of Nur77 protein in prostate cancer tissues was higher than that in the benign prostatic hyperplasia tissues (P < 0.01). The expression level of Nur77 was downregulated with the ascending of N staging, M staging and Gleason score in prostate cancer (all P < 0.05). The Nur77 and β-tubulin were co-located and interacted with each other in the cytoplasm of PC-3 cells. Compared with the blank control group, the ratio of polymerized/soluble (P/S) tubulin was significantly increased in pcDNA3.1-Nur77 and Nur77ΔDBD groups, while the count of migratory cells was significantly decreased in pcDNA3.1-Nur77 and Nur77 ΔDBD group (all P < 0.05). In contrast, P/S ratio was significantly decreased, and the count of migratory cells was significantly increased in Nur77-shRNA group (both P < 0.05). Conclusion: Nur77 can inhibit the migration of PC-3 cells, which may be related to promoting tubulin polymerization into microtubule.

3.Efficacy and safety of vardenafil in difficult-to-treat erectile dysfunction men.

This paper reviewed the efficacy and safety of vardenafil, a highly selective phosphodiesterase type 5 (PDE 5) inhibitor, in men with erectile dysfunction who were difficult to treat. Several large-scale studies indicated vardenafil was effective and safe in the treatment of these difficult-to-treat ED patients, including ED with depression or diabetes, ED after radical retropubic prostatectomy, ED caused by spinal cord injury, and sildenafil nonresponders. Vardenafil provides a rational treatment alternative.

4.Effects of midazolam and estazolam as hypnotics in hypertensive patients with chronic insomnia: a multicentre, open labeled, randomized clinical trial.

OBJECTIVETo compare the safety and efficacy of midazolam and estazolam in hypertensive patients with chronic insomnia.

RESULTS(1) Blood pressure was equipotent reduced after both treatments (-11.8/7.3 mm Hg for midazolam group, and -9.1/5.6 mm Hg for estazolam group, all P < 0.05 vs. before treatment). (2) The total sleep score was also significantly decreased in both groups after medication (P < 0.01) and midazolam was significantly superior to estazolam in shortening sleep latency, reducing awakening frequency, improving objective sleep evaluation and decreasing daytime sleepiness, but there were no differences in dream frequency and total sleep time. (3) The adverse reactions such as dizziness, headache and nausea was similar in midazolam (3%) and estazolam group (7%, P > 0.05).
CONCLUSIONSIt is safe to take midazolam or estazolam for hypertensive patients with chronic insomnia and both drugs reduced blood pressure. Midazolam is superior to estazolam in shortening sleep latency, reducing awakening frequency, improving objective sleep evaluation and decreasing daytime sleepiness.

5.Research progress of the inhibitory effect of deuterium-depleted water on cancers.

Deuterium is an important predisposing factor for cancer. Deuterium-depleted Water, also known as low deuterium water, ultra-light water or no deuterium water, can be obtained by removing deuterium from natural water. Studies have shown that water with a low deuterium concentration (<65% percent of volume) can inhibit cancer growth. Clinical trials demonstrated that drinking DDW (10-20 ppm) caused growth arrest of malignant cells in cancer patients and significantly prolonged the patient survival with also improved quality of life. A wide range of anti-cancer drugs in current use are associated with severe adverse effects, while deuterium-depleted water appears to have virtually no pharmacological side effects and is convenient to administer. The authors review the advances in the researches of anti-cancer effects and the underlying mechanisms of deuterium-depleted water.

6.The research on time-frequency detection method of respiratory component in ballistocardiogram signal.

Based on the fact that the respiratory component modulates the cardiac cycle component in the ballistocardiogram (BCG) signal, we propose a method that detects respiratory with time-frequency analysis for the sitting ballistocardiography system. Firstly, we demodulated the BCG signal by using the variable frequency complex demodulation (VFCDM) to obtain the output for different center frequency of interest. Then we calculated the instantaneous frequencies and the instantaneous amplitudes by the time-frequency representation. We reconstructed the time-domain waveform of respiratory at last. In order to verify the feasibility and accuracy of this method, we applied wavelet transform and nasal thermistor signal to compare qualitatively and quantitatively. The simulation results showed that the proposed method could detect the respiratory rate from BCG signal more accurately, which provided meaningful attempt for monitoring the multiple physiological parameters synchronously and unconsciously.

7. The impact of job burnout on perception of risk from adverse events among clinical nurses

Objective:
To determine the impact of job burnout on perception of risk from adverse events among clinical nurses.
Methods:
A total of 480 clinical nurses were recruited from four Secondary B-class hospitals in Tianjin by convenience sampling method,between December 2017 to December 2018. They were investigated with Clinical Nurses Adverse Events Risk Perception Structure Scales and Maslach Burnout Inventory. The relationship between job burnout and perception of risk from adverse events was analyzed.
Results:
The total score of clinical nurses adverse events risk perception structure scales was 304.60±59.20. There were significant negative correlations between score of job burnout and perception of risk form adverse events among clinical nurses (
8.Synthesis and cytotoxicity of longistylin C derivatives.

The present study was designed to identify potent anti-tumor compounds from a series of new longistylin C derivatives. Ten longistylin C derivatives were synthesized and their structures were confirmed by (1)H NMR, MS, and elemental analyses. Their cytotoxicity in vitro against three human cancer cell lines (A549, HepG2, and MCF-7) were evaluated by the MTT assay. Among these compounds, DT-6 and DT-9 displayed much better cytotoxicity against A549, HepG2, and MCF-7 cells, DT-1 exhibited selective cytotoxicity against HepG2, and the structure-activity relationships were investigated. In conclusion, Compounds DT-6 and DT-9 may serve as potential lead compounds for the discovery of new anti-cancer drugs.
