1.Mechanism of breviscapine entrapped by polymethacrylate nanospheres

AIM: To prepare polymethacrylate nanospheres and study the mechanism of breviscapine entrapped by polymethacrylate nanospheres. METHODS: Polymethacrylate nanospheres were prepared by microemulsion polymerization with SDS as surfactant, n-butanol as cosurfactant, MAA and BMA as monomer, TRIM as cross-linker and AIBN as initiator. Breviscapine added would be divided into two ways:prior to polymerization (encapsulation) and after polymerization (sorption), respectively. RESULTS: The size of the nanospheres was found to be 50nm by measuring with TEM. The ? potential of the nanospheres was -27.2 mv and it was increased after breviscapine being entrapped. The drug content entrapped in nanospheres was proportional to the drugs amount added in encapsulation method while the drug content in nanospheres was increased step by step in sorption method. CONCLUSION: The polymethacrylate nanospheres prepared by microemulsion polymerization could be applied to encapsulate hydrophobic traditional Chinese medicine extracts.

Objective To study the effect of baicalin on the content of glutamate acid (Glu), aspartic acid (Asp), ?-aminobutyric acid (GABA) in the brain of rat with experimental intracerebral hemorrhage. Methods The intracerebral hemorrhage rat model was induced by collagen enzyme + heparin sodium. The rats were randomly divided into sham group, model group and high, middle, low dose group of baicalin. Rats were treated with the physiological saline and different concentration of baicalin respectively. The brain of each group was taken out three days later and the brain homogenate was prepared. The content of Glu, Asp and GABA was measured with the high performance liquid chromatography (HPLC). Chronic pancreatitis (CP) is a kind of disease with the sustainable and irreversible damage of the tissue structure and function of pancreas,which may be caused by alcohol,gene,gallstone,metabolism,deformity and other factors.The clinical manifestations are intractable abdominal pain and disorder of the internal and external secretion of pancreas.At present,the main purpose of the treatment for chronic pancreatitis is to relieve patients' pain,and to maintain the secretion function of pancreas as far as possible.The main surgical procedures for chronic pancreatitis includes the pancreatic resection,nerve block and decompression drainage of the pancreatic duct.In recent years,people hve paid more attenntion to the whole pancreas resection combined autologous transplantation due to retaining some functions of the islet cells.After whole pancreas resection combined autologous transplantation,the abdominal pain will relieve and the life quality will improved significantly.At the same time,it will effectively reduce the occurrence of pancretogenic diabetes since the surgery retains some function of the islet cells.
5.Diagnosis and treatment of 13 cases of autoimmune pancreatitis

Objective To investigate and summarize the clinical characteristic of autoimmune pancreati-tis(AIP).Methods Clinical data of 13 patients with AIP, admitted to the West China Hospital from 2009 to 2013, were retrospectively analyzed .Results The median age of the 13 patients was 48 years old.The main clinical manifestations included epigastric pain or discomfort (8 cases), obstructive jaundice(7 cases)and weight loss(10 cases).Diffuse enlargement of pancreas and localized pancreatic enlargement were observed in 6 cases, pseudocyst and pancreatic stones were observed in 1 case.Dilation of pancreatic duct was found in 4 cases.Bili-ary stricture and thickened wall of bile duct were detected in 8 cases, dilated gallbladder with delayed enhance-ment of the thickened wall in 7 cases, retroperitoneal fibrosis surrounding the aorta in 1 case, abdominal lymph-adenopathy in 6 cases, stenosis of splenic vein in 4 cases, and Sjogren's disease and ulcerative colitis in 1 case. The positive rate of serum IgG, RF, ANA was 71.43%(5/7), 42.86%(3/7), and 42.86%(3/7), respec-tively.8 patients were misdiagnosed and underwent surgery .Steroid therapy was administered in all patients with satisfactory response.Conclusions AIP is a rare and autoimmune disease ,without speci? c symptoms,and often be misdiagnosed as pancreatic cancer or bile duct carcinoma .We must give more attention to AIP and keep pa-tients from undergoing unnecessary surgery .
6.Effects of Zinc sulfate on preventing vascular endothelial cells from oxidative stress

Objective:To investigate the effects of Zinc sulfate on preventing human umbilical veins endothelial cells ( HUVECs) from oxidative stress. Methods:Hydrogen peroxide was used to stimulate HUVECs to build the oxidative stress model in vitro. HUVECs were divided into normal group, H2 O2 group, Zinc sulfate group and Zinc sulfate treated group. Method of nitrate reductase was used to detect the content of nitric oxide ( NO ) and ELISA was used to detect the content of endothelin ( ET ) in supernatant in different groups. The level of HO-1,SOD and CAT of HUVECs were measured by Western blot. Apoptosis of HUVECs was examined by TUNEL as well. Results: Zinc sulfate could enhance the content of NO and decrease the content of ET in the supernatant,which induced by hydrogen peroxide on HUVECs. Zinc sulfate could also increase the level of HO-1, SOD and CAT obviously and decrease the apoptosis cells significantly induced by H2 O2 . Conclusion: Zinc sulfate play an important role in resisting oxidative stress in HUVECs, and maybe prevent HUVECs from oxidative stress damage. Zinc sulfate is expected to be an effective medicine on improving endothelial cells anti-oxidative ability.
7.Peritoneal vibration increases peritoneal permeability and damages the peritoneal surface layer

AIM: To investigate the effect of peritoneal vibration on the peritoneal permeability and the peritoneal surface layer. METHODS: Peritoneal transport rate was examined in twelve male SD rats. Six (S group) were put on an electronic shaker and the other six were used as control (C group). After that, the peritoneum was examined by electron microscopy (EM). RESULTS: The net ultrafiltration volume (NUF) in the S group was lower than that in the C group. This difference in NUF was due to both a significantly higher peritoneal fluid absorption rate and a significantly lower transcapillary ultrafiltration rate in S group as compared to C group. The peritoneal direct lymphatic absorption rate was higher in S group. The transport rates of small solutes were also significantly higher in S group. EM showed that the thickness of the peritoneal surface layer was significantly decreased in S group. CONCLUSION: Our results suggest that the peritoneal surface layer may be an important layer in modulating the peritoneal transport rate.
8.Screening potentially drug resistant and multidrug resistant genes in NPC cells with metastatic ability

Objective To screen potential genes associated with drug resistance and multidrug resistance. Methods Microarray with 8000 genes was used to detect the different expression of 5-8F cells and 6-10B cells. Subsquently, genes of drug resistance and multidrug resistance were screened by MILANO online programme. Semiquantitative RT-PCR was utilized to confirmed the reliability of differentially expressed genes. Results 283 genes were identified the differential expression. Of these, 85 genes were shown to be upregulated and 98 downregulated. After the analysis of MILANO, 4 genes including UGT1A9 (15.85 folds),MVP(6.77 folds), CAV1(2.49 folds) and HIF1A(2.67 folds) with higher expression in 5-8F cells were found to be likely associated with drug resistance and multidrug resistance. Subsquently, semiquantitative RT-PCR confirmed the reliability of differential expression of these 4 genes. Conclusion Differentially expressed genes shown in NPC 5-8F cells compared to 6-10B cells with the identification of online MILANO program analysis are likely associated with drug resistance and mnltidrug resistance of NPC cells with the ability of metastasis.
9.A study on the pathological type and surgical therapy of chronic pancreatitis

Objective To evaluate surgical therapies for chronic pancreatitis.Method The clinical data of 229 patients admitted for chronic pancreatitis during March 2009 to November 2013 in our hospital was retrospectively analyzed,different operation method and their clinical outcome were compared.Results Drainage operations or resection operations were made to all these 229 patients according to different types.57 patients underwent longitudinal pancreaticojejunostomy (LPJ procedure).118 patients received local resection of the head of pancreas combined with longitudinal pancreaticojejunostomy (Frey procedure in 105 cases,Berne procedure in 6 cases,Beger procedure in 7 cases).7 patients received pancreaticoduodenectomy,21 patients received distal pancreas resection,26 patients received other procedures.Post-operative pain relief rate was 89.3%,overall morbidity was 19.6%.Conclusions In cases of chronic pancreatitis,different surgical types are adopted according to their individual indications.Operation in accordance with pathological types guarantees clinical outcome.
10.Prognosis and staging of non small ceil lung cancer that extends across the fissure into adjacent Iobe

Objective To determine the prognosis and staging non small cell lung cancer (NSCLC) that extends across the fissure into adjacent lobe after surgery.Methods 3752 patients with histopathologically confirmed non small cell lung cancer (NSCLC) received surgical reeessetion from January,1997 to April,2007.Among them,163 patients have a tumor invasion beyond fissure.After matching by pathologic TNM staging (7th),326 patients whose tumor defined in a single lobe were eligible for analysis.Results Histopatholngic staging of matched patients was I a:10 patiens(6.1% ),I b:79 patients (48.5%),Ⅱa:5 patients (3.1% ),111:44 patients (27.0%) and Ⅲa:25 patients( 15.3% ).5 years survival in patients with stage 1 tumors crossing the interlobar fissure was 51%,while in patients not cross the interlobar fissure was 63% ( P <0.05 ).There was no difference in survival for tumors stage Ⅱa and above with regard to importance of interlobar extension.The T2 tumor extending across a lung fissure had a reduction in survival compared with T2 tumor not cross the lung fissure and similar to the T3 tumor without the fissure invasion.Conclusion Our results suggest that TNM staging should be modified for tumor extends the fissure into adjacent lobe.
