AIM:To study the protective effect of aqueous extract of 2-branched and 3-branched velvet antler on cisplatin (CDDP)-induced nephrotoxicity in mice .METHODS:The mouse model of renal injury was induced by intra-gastric administration of CDDP at the dose of 15 mg/kg.After treatment, kidney index (KI), serum creatinine (SCr), blood urea nitrogen (BUN), the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), and the content of malondialdehyde ( MDA) in the kidney were determined .The renal pathological changes were observed with HE staining.RESULTS:Aqueous extract of velvet antler at the tested dose markedly decreased BUN , SCr and the content of MDA, and elevated the activity of SOD and GSH-Px in the mice pretreated with CDDP ( P<0.05) .The pathological chan-ges of the renal tissues were improved obviously , and the injury of the epithelial cells of renal tubules was mitigated .The effect of the aqueous extract of 2-branched velvet antler on renal function and cisplatin-induced nephrotoxicity was better than that of 3-branched one at the same concentration .CONCLUSION: The aqueous extract of 2-branched and 3-branched velvet antler has a certain protective effect on cisplatin-induced nephrotoxicity , which may be associated with in-creasing the anti-oxidative capability of mouse renal tissue .

BACKGROUND:Studies have funded that reduced Wnt/β-catenin signaling is involved in the onset and/or progression of bone erosion in rheumatoid arthritis. It can lead to potential new treatment approaches of bone erosion by enhancing Wnt/β-catenin signaling pathway. R-spondin 1 may act as a Wnt agonist, but there is no study in human osteoblasts. OBJECTIVE:To verify the effect of R-spondin 1 on promoting differentiation and maturation of human osteoblasts by inhibiting DKK1. METHODS:S40-transfected human osteoblast lines, hFOB1.19, were treated with R-spondin 1, Wnt-3a and DKK1 to detecting the proliferation, alkaline phoshpatase activity and osteoprotegerin concentration. RESULTS AND CONCLUSION:R-spondin 1 had no effects on hFOB1.19 cel s. Wnt-3a upregulated the activity of alkaline phoshpatase, which could be enhanced by addition of R-spondin 1. R-spondin 1 could reduce the DKK1-mediated inhibition of alkaline phoshpatase activity in hFOB1.19 cel s. R-spondin 1 increased the concentration of osteoprotegerin, and moreover, the promotion of osteoprotegerin by R-spondin 1 alone was stronger than the inhibition by DKK1. These findings suggest that R-spondin 1 can inhibit DKK1 by Wnt/β-catenin signal pathway to promote the differential and maturation of human osteoblasts to excrete osteoprotegerin.

Objective To investigate the concentration of 17β-estradiol in bone marrow stem cells (BMSCs) and different tissues of rats, and study the function of estradiol produced in extragonadal sites prelimi-narily. Methods (1) The concentration of 17β-estradiol in lysate of BMSCs and in supernatant cell culture media were detected by Elisa method after rats BMSCs were serum-free cultured in 0 h , 24 h and 48 h respective ly . (2) The tissues of organs were grinded and broken under ultrasonic wave , then washed and weighed. The con-centration of 17β-estradiol in different tissues was detected by Elisa method. Results Compared with the concen-tration of 17β-estradiol at 24 h, the concentration of 17β-estradiol at 48h significantly increased (P < 0.05). The concentration of 17β-estradiol among different tissues were higher, but the difference is not significant (P > 0.05). Conclusions (1) 17β-estradiol can be secreted by BMSCs , and the concentration is proportional to the time to some degree. But whether the extragonadal estrogen can function locally is still unclear. (2) The concentration of 17β-estradiol is non- gonad dependent but whether it is secreted locally remains to be elucidated. The non- gonad estrogen could be the estrogen source of bone metabolism in order to sustain bone health after menopause.

Objective To compare the characteristics of lymph nodes on 18F-FDG imaging in patients with NHL and widespread lymph node metastases carcinoma (WLNMC) for helping the differential diagnosis. Methods A total of 40 NHL patients(22 males,18 females, average age 51 years) and 42 WLNMC patients (19 males,23 females, average age 61 years) confirmed by histopathology from November 2010 to May 2015 were included in this retrospective study. The regions and the metabolism features of the lesions on PET/CT images were observed and recorded with routine visual method.Two-sample t test and x2 test were used for statistical analysis. Results Lymph nodes of NHL were kidney-shaped (47.2%,142/301),which was significantly different from that of WLNMC with circular form (47.6%,140/294;x2 =36.261,P<0.05).Lymph nodes of NHL mostly showed homogeneous density with no necrosis (51.8%,156/301);however,that of WLNMC showed uneven density (20.4%,60/294) or with necrosis (32.3%,95/294;x2 =26.266,P<0.05).Both of the two kinds of lymph nodes showed the characteristic of fusion (30.6% (92/301) vs 36.1% (106/294);x2 =2.019,P>0.05).The ratios of lymph nodes with clear boundary in NHL and WLNMC were significantly different:28.2%(85/301) vs 19.4%(57/294);x2 =6.413,P<0.05.Lymph nodes with symmetric distribution were more in NHL patients (52.5%,158/301)than that in WLNMC patients (42.2%,124/294;X2=6.347,P<0.05).Sizes of lymph nodes in NHL patients and WLNMC patients were not significantly different:(2.08±0.65) cm vs (1.97±0.81) cm;t=0.316,P>0.05.SUVmax of lymph nodes in NHL patients (9.02±3.97) was much higher than that in WLNMC patients (6.92± 1.34;t=0.370,P<0.05).Conclusions Some characteristics of lymph nodes on 18F-FDG PET/CT imaging, such as the distribution, morphology, density and glucose metabolism features, are different in patients with NHL and WLNMC.Those differences may be helpful for the differential diagnosis.
