1.NADPH oxidase participation in cell proliferation of human lens epithelial cells and its expression

Objective To study whether the inhibitor of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase can inhibit the proliferation of human lens epithelial cells and to investigate which isoforms of NOX, one of the subunits of NADPH oxidase, are expressed in human lens epithelial cells at mRNA level. Design Experimental study. Participants Human lens epithelial cells (SRA 01/04). Methods The cells were divided into four groups: EGF group was added with EGF in SRA 01/04, DPI group was added with the inhibitor of NADPH oxidase-diphenylene iodonium (DPI), DPI +EGF group was added with EGF after DPI and control group was added with nothing. Using Microplate reader and Cell Counting Kit-8 to determine OD450 value of SRA 01/04 of each group. The ex- pression of NOX family, including NOX1, NOX2 or gp91phox, NOX3, NOX4 and NOX5, was detected with RT-PCR. The RT-PCR products were sequenced to confirm their identities by NCBI BLAST. Main Outcome Measures OD450 of SRA 01/04, expression of NOX and their sequence alignments with other human tissues. Results After added with CCK-8 for 3.5 h, the OD450 value of EGF group increased 12.0% compared with control group (P=0.000). The OD450 value of DPI group decreased 25.5% compared with control group (P=0.000). The OD450 value of DPI+EGF group decreased 26.1% compared with EGF group (P=0.000). RT-PCR using primers specific for mRNAs of the five isoforms of the NOX proteins documented that mRNA encoding NOX1 through NOX5 were constitutively present in SRA 01/04 cells. The similarity of sequences of NOX1-NOX5 in human lens epithelial cells with other human tissues was 100%, 100%, 99.7%, 100% and 100%, respectively. Conclusions NADPH oxidase complex could promote cell proliferation in human lens epithelial cells. SRA 01/04 cells constitutively produced mRNA encoding five isoforms of NOX proteins, NOX1 was much weaker than the other four NOXs.
2.Study on Electroacupuncture in Improving Discomforts During and after Oocyte Retrieval

Objective To observe the efficacy of electroacupuncture in improving discomforts during and after oocyte retrieval, for providing scientific and significant clinical evidence for the application of electroacupuncture to oocyte retrieval.Method A hundred and thirty-four outpatients receiving IVF-ET fertility treatment were randomized into a treatment group and a control group, 67 in each group. The two groups both received muscular injection with Sauteralgyl 50 mg. Additionally, the treatment group was also given acupuncture at Baihui (GV20), Uterus (MA-TF, right ear), Tengtong (Extra, right), Sanyangluo (TE8, right), and Zusanli (ST36). When needling qi arrived, Tengtong (Extra) and Sanyangluo (TE8) were connected to SDZ-2 electroacupuncture apparatus. The needles were removed at the end of oocyte retrieval. For patients in the control group, oocyte retrieval was conducted 30 min after injection with Sauteralgyl. The operation duration was recorded; blood pressures before injection and at the end of surgery were measured; discomforts including dizziness, nausea, vomiting, perspiration, dry mouth, palpitation, and abdominal distension during operation were observed; discomforts (dizziness, nausea, vomiting, abdominal pain, and lassitude) 0.5 h, 1.5 h, and 2 h after operation were observed.Result The difference in comparing oocyte retrieval duration was statistically significant between the two groups (P<0.05); there were significant differences in comparing the occurrence rates of dizziness, perspiration, nausea, and palpitation during operation between the two groups (P<0.05); there were significant differences in comparing dizziness, vomiting, nausea, abdominal pain, and lassitude between the two groups 0.5 h, 1 h, 1.5 h, and 2 h after operation (P<0.05). The inter-group differences in dizziness, vomiting, nausea, abdominal pain, and lassitude were statistically significant after operation (P<0.05).Conclusion Electroacupuncture is effective in relieving discomforts during and after oocyte retrieval under transvaginal ultrasound.
3.Expression of plate-reactive protein in cervical cancer and its effect on biological behavior of cervical cancer Hela cells

Objective:To investigate the expression of F-spondin (SPON1) in cervical cancer tissues and its effects on the biological behavior of Hela cell line, including proliferation, apoptosis, invasion, and migration.Method:Select 80 samples of cervical cancer tissue and 80 samples of cervical inflammation tissue stored in the Pathology Department of Xuzhou Medical University from January 2022 to May 2023. Immunohistochemistry was used to detect the expression of SPON1 in cervical cancer and chronic cervicitis tissues, and the corresponding clinicopathological data were collected for analysis. Small disturbance RNA(siRNA) was used to interfere with the expression of SPON1 in Hela cell lines. Untransfected cells were set as normal control group (Ctrl group), negative control group (NC siRNA group), and siRNA knockout group (SPON1 siRNA group). The interference effect was detected by qPCR and Western blot. Cell proliferation assay (CCK-8), flow cytometry, Transwell, and scratch assay were used to detect the proliferation, apoptosis, invasion, and migration of each group of cells. Each experiment was repeated three times. Quantitative data that conforms to a normal distribution are represented by xˉ± s. The comparison of means among the three groups was conducted using one-way analysis of variance; Compare pairwise using LSD- t test. Calculate the percentage based on qualitative data, and compare the rates between groups using the chi square test. P<0.05 indicates a statistically significant difference. Results:Compared with the chronic cervicitis group, the positive expression rate of SPON1 in cervical cancer tissues (72.5% (58/80)) was higher than that in chronic cervicitis tissues (20.0% (16/80)), and the difference was statistically significant ( χ2=44.35, P<0.001). The positive expression rate of SPON1 was positively correlated with the clinical stage, pathological differentiation and tumor diameter of cervical cancer ( χ2 values were 4.10, 4.98, and 4.40, respectively; P values were 0.043, 0.026, and 0.036, respectively), and were not related to the age and tissue type of patients ( χ2 values were 0.72 and 0.14, P values were 0.386 and 0.713, respectively). The relative expressions of SPON1 mRNA were 1.000±0.014, 0.966±0.082, and 0.365±0.036, respectively. The expression levels of SPON1 protein were 1.000±0.013, 1.022±0.031, and 0.655±0.026, respectively. The number of cells that penetrated the model in the three groups was (113.3±4.1), (107.0±3.1), and (80.3±3.2), respectively. The wound healing rates of the three groups were (56.00±3.45)%, (55.00±5.03)%, and (35.33±3.76)%, respectively, and the apoptosis rates of the three groups were (5.88±0.44)%, (6.27±0.38)%, and (10.50±0.39)%, respectively. Compared with the Ctrl group and the NC-siRNA group, the proliferation ability of cells in the siRNA interference group decreased (all P<0.001), invasion ( P values were 0.001 and 0.004) and migration ( P values were 0.029 and 0.035), and the apoptosis rate was increased ( P=0.001). Conclusion:SPON1 is highly expressed in cervical cancer tissue and is associated with the occurrence and development of cervical cancer. Inhibiting SPON1 expression significantly reduces the proliferation, invasion, and migration ability of Hela cells, and promotes their apoptosis.
4.Seven cases of severe pneumocystis jirovecii pneumonia in children with non-human immunodeficiency virus infection and a literature review

Objective:To summarize the clinical features and treatment of pneumocystis jirovecii pneumonia(PCP) in children with non-human immunodeficiency virus(HIV) infection.Methods:A retrospective study was performed on seven cases of severe PCP children with non-HIV infection who were admitted to PICU of The University of Hong Kong-Shenzhen Hospital and PICU of Xianyang Rainbow Hospital from May 1, 2015 to May 1, 2021.The risk factors, clinical manifestations, laboratory results, pulmonary radiological features, treatment and outcomes were observed.Results:Seven children with PCP, including four males and three females, aged from 13 months to 85 months[(42.4±26.8) months], were all associated with underlying diseases, and most of which was hematological malignancies.Six children had a history of using TMP-SMX for PCP prevention, but four of them stopped by themselves and infected PCP in 2 to 4 weeks.All children had hypoxic respiratory failure, whose OI was 30.6±3.4, and presented with fever, dry cough, progressive dyspnea but no lung rales in the early stage.LDH[(745.7±317.0) U/L] and β-D-glucan[(513.8±225.0) pg/mL] increased in all patients.Chest CT showed diffused interstitial changes in bilateral lung fields associated with multiple exudative lesions.Among the anti-Pneumocystis Jirovecii treatment regimens, all cases began the treatment in the first three days during the admission, five cases were treated with intravenous TMP-SMX, and two cases were treated with oral TMP-SMX + caspofungin, with a course of 21 days.All children were also treated with glucocorticoid at the same time.Three days after the treatment of PCP, two children were worsened and one of them died, while another one started to recover on the 6th day of the regimen.The remaining five cases began to show clinical improvement after 3~7 days of PCP treatment.Finally six children were cured and one was died.Conclusion:PCP infection of children without HIV has high risk of destruction in immune system.TMP-SMX can prevent PCP effectively.In the severe PCP cases, early commencement of intravenous TMP-SMX can reduce the mortality rate.In the absence of intravenous TMP-SMX, oral TMP-SMX can be used with caspofungin.
5.Clinical assessment for motor cognitive risk syndrome in patients with enlarged perivascular spaces

Objective To investigate the incidence of motoric cognitive risk(MCR)syndrome and declines in both motor and cognitive functions in elderly patients with enlarged perivascular spaces(EPVS).Methods A total of 113 patients(≥60 years old)with EPVS identified by head MRI admitted to our department from March 1 to November 30,2023 were consecutively recrui-ted,and according to having MCR syndrome or not,they were divided into MCR group(44 cases)and non-MCR group(69 cases).Cognitive function and motor function were assessed with aid of Montreal Cognitive Scale(MoCA),Simple Mental State Examination Scale(MMSE),trail making test,and motor function assessments,including Tinetti Balance and Gait Scale,Short Physical Performance Battery(SPPB),8-meter walk,Timed up and Go test(TUGT),walking and contin-uous subtraction task(WSS),walking and semantic fluency task(WSF),and TUGT and serial subtraction task(TUGSS).The motor and cognitive functions were compared between the two groups,and the risk factors related to MCR syndrome in EPVS patients were analyzed by multi-variate logistic regression.Results Compared with the non-MCR group,the MCR group had low-er MoCA score,total SPPB score,8-meter walk speed,and longer times of TMT-A,TMT-B,WSS,WSF and TUGSS(P<0.05,P<0.01).Multivariate logistic regression analysis showed that in Model 1(including variables with differences in univariate analysis),MoCA score,8-meter walk speed,and TUGSS time were independent risk factors for MCR syndrome in EPVS patients(P<0.05,P<0.01);in Model 2(adjusting for age,sex,education level,and comorbidities),MoCA score(OR=0.621,95%CI:0.432-0.929,P=0.010),8-meter walk speed(OR=1.332,95%CI:1.115-1.591,P=0.002),and TUGSS time(OR=0.393,95%CI:0.232-0.665,P=0.001)re-mained independent risk factors for MCR syndrome in EPVS patients.Conclusion Among elderly EPVS patients,those with MCR syndrome show significantly worse cognitive and motor function compared to those without.MoCA score,8-meter walk speed,and TUGSS time are related factors for MCR occurrence in EPVS patients.
6.Establishment and evaluation of a high-throughput and rapid method for detection of fluorine content in water based on microplate reader

Objective:To study and establish a high-throughput and rapid detection method for fluorine ion content in water based on 96-well plate and microplate reader, and to evaluate its performance.Methods:According to the principle of blue ternary complex formed by reaction of fluorine ion with fluorine reagent and lanthanum nitrate, the absorbance of the complex at the wavelength of 650 nm was proportional to the concentration of fluoride ion, and the content of fluoride ion in water samples was quantitatively determined by microplate reader. The accuracy, precision and detection limit of the method were evaluated by the standard recovery rate method and the reference material method.Results:The linear correlation coefficient of the standard curve of this method was > 0.999; the standard recovery rates of different concentrations were 99.00% - 103.33% in the accuracy experiment of this method, and the average recovery rate was 101.08%. In the standard material method, the measured values are all within the calibration range of the standard solution. The relative standard deviation of water samples with different concentrations was ≤10% in the precision experiment and the detection limit was 0.06 mg/L.Conclusion:The method has good accuracy and precision, low detection limit and small sample consumption, and this method can greatly improve the detection efficiency.
7.Taking "Seven Emotions Scale" as an Example to Explore the Suitability of Four Qualitative Methods in the Development of TCM Scale

ObjectiveTo explore the suitability of four qualitative research methods in the development of TCM scale. MethodsTaking the development of "Seven Emotions Scale" as an example, we conducted semi-structured interviews with 31 patients of emotional disorders and 10 healthy people by objective sampling, and collected psychological feelings and emotional cognition data related to seven emotions according to the interview outline. Two qualitative methods, descriptive qualitative research and descriptive phenomenology, were used to analyze the data and construct the item library of the scale. The conceptual framework of the scale was constructed by using commonly used grounded theory and frame analysis. ResultsDuring data analysis, it is found that the themes extracted from descriptive phenomenology were not easily understood by the interviewees, and it is difficult for the researchers to truly achieve the "suspension" required by phenomenology. Considering the feasibility and convenience of the researchers' actual operation, as well as whether the initial purpose of the scale research can be intuitively included in the interviewees' views and feelings, descriptive phenomenology is not suitable for the formation of scale items. Using descriptive qualitative research method to analyze the interview data of healthy people and patients with emotional disorders, 306 and 476 scale items were obtained respectively. Through grounded theory, five selective codes were obtained: physical symptoms, external manifestations, psychological feelings, behaviors and emotional control. Using frame analysis, four themes including physical symptoms, psychological feelings, behavior and emotional cognition were constructed. Both methods can be used to construct the conceptual frame of scale, but the framework analysis is more convenient and can better ensure the transparency of the research. ConclusionDescriptive qualitative research methods can be used to form the item library of TCM scales. Framework analysis is more suitable for the construction of the conceptual framework of the scale than grounded theory, while descriptive phenomenology is not suitable for the development of TCM scales.
8.Clinical risk factors for early adverse cardiovascular events after surgical correction of supravalvar aortic stenosis: A retrospective cohort study

Objective To identify clinical risk factors for early major adverse cardiovascular events (MACEs) following surgical correction of supravalvar aortic stenosis (SVAS). Methods Patients who underwent SVAS surgical correction between 2002 and 2019 in Beijing and Yunnan Fuwai Cardiovascular Hospitals were included. The patients were divided into a MACEs group and a non-MACEs group based on whether MACEs concurring during postoperative hospitalization or within 30 days following surgical correction for SVAS. Their preoperative, intraoperative, and postoperative clinical data were collected for multivariate logistic regression. Results This study included 302 patients. There were 199 males and 103 females, with a median age of 63.0 (29.2, 131.2) months. The incidence of early postoperative MACEs was 7.0% (21/302). The multivariate logistic regression model identified independent risk factors for early postoperative MACEs, including ICU duration (OR=1.01, 95%CI 1.00-1.01, P=0.032), intraoperative cardiopulmonary bypass (CPB) time (OR=1.02, 95%CI 1.01-1.04, P=0.014), aortic annulus diameter (OR=0.65, 95%CI 0.43-0.97, P=0.035), aortic sinus inner diameter (OR=0.75, 95%CI 0.57-0.98, P=0.037), and diameter of the stenosis (OR=0.56, 95%CI 0.35-0.90, P=0.016). Conclusion The independent risk factors for early postoperative MACEs include ICU duration, intraoperative CPB time, aortic annulus diameter, aortic sinus inner diameter, and diameter of the stenosis. Early identification of high-risk populations for MACEs is beneficial for the development of clinical treatment strategies.
