1.Effects of maternal deprivation at different lactation periods on depression behavior and brain catecholamine of rat offsprings

Objective To investigate the effects of maternal deprivation at different time periods in rat pup development on depressions in adults.Methods Newborn SD rat pups were randomly divided into 5 maternal deprivation groups(n=5 in each group) and one control group (n=7).The rat pups in the maternal deprivation groups were separated from their mother for 6 hours in one day,at p0,p5,p 10,p15,p20 respectively,lasting for five days.Then the rats will be screened for depression at the age of 10 weeks,using different behavioral tests,which included forced swim,coat score,sucrose preference test.Norepinephrine concentration in the cerebral spinal fluid was also measured through high performance liquid chromatography (HPLC).Results Maternal deprivation for rat pups induced depression like behavior in adults.The floating time during forced swim test increased from(58.5 ±7.5) s in control group to(p0(75.6±9.4) s,p5(150.4±12.5) s,p10(174.5±14.3) s,p15(155.4±11.4)s,p20 (112.4±10.3) s respectively in maternal deprivation groups;and it was significant different among the groups(P< 0.01).Compared with the control group(0.87±0.1),the sucrose preference in maternal deprivation groups decreased(p0 (0.73±0.12),p5 (0.53±0.09),p10 (0.5±0.06),p15 (0.54±0.07),p20 (0.67±0.05)).And the coating scores in maternal deprivation groups decreased from 4.5±0.6 in control group to p0(3.3±0.6),p5 (2.3±0.4),p10 (1.9±0.3),p15 (2.4±0.4),p20 (4.4± 0.4).Finally,maternal deprivation also induced low concentration of catecholamine in the cerebral spinal fluid compared with that in control group (p0 (94.5± 11.5) %,p5 (78.5± 9.6) %,p10(67.6±±7.4)%,p1S(64.5±6.6)%,p20(88.5±7.5)%,P<0.01,n=5-7).Conclusion Maternal deprivation at different lactation periods may induce depression behaviors in rat offspring,and also lead to lower concentration of catecholamine in the cerebral spinal fluid.
2.Immobilized enzyme reactors in HPLC and its application in inhibitor screening: A review

This paper sets out to summarize the literatures based on immobilized enzyme biochromatography and its application in inhibitors screening in the last decade.In order to screen enzyme inhibitors from a mass of compounds in preliminary screening,multi-pore materials with good biocompatibility are used for the supports of immobilizing enzymes,and then the immobilized enzyme reactor applied as the inmobilized enzyme stationary phase in HPLC.Therefore,a technology platform of high throughput screening is gradually established to screen the enzyme inhibitors as new anti-tumor drugs.Here,we briefly summarize the selective methods of supports,immobilization techniques,co-immobilized enzymes system and the screening model.
3.Cause analysis of the screw loosening after the internal fixation with thoracolumbar vertebral pedicle screw in the elderly

Objective To explore what cause the screw loosening after the internal fixation with thoracic and lumbar pedicle screws in the elderly and to develop the preventive measures.Methods Retrospective analysis were used to collect the clinical data from patients ager 60 years and over who had undergone surgeries of thoracolumbar pedicle screw internal fixation in our hospital during May 2011 and May 2013.Totally 187 cases were included in this study.According to whether there was a loosening of the screws, patients were divided into fixtion-loose and-well groups.The operation time and blood loss volume in the two groups were recorded.Visual analogue scale (VAS), Oswestry disability index (ODI), the rate of screw loosening were compared at 6 months after operation and the last follow-up.The causes of screw loosening were analyzed.Results The main causes for the thoracic and lumbar pedicle screw internal fixation were thoracolumbar kyphoscoliosis, fractures,spondylolisthesis, cancer and degenerative diseases, etc.Patients were followed up for an average of (35.2±8.7) months.13 patients (7.0%) were found screw loosening, 5 males and 8 females, with the mean age of (78.4 ± 3.5) years.The screw loosening occurred 3 months to 1 year after the surgery, (7.6±3.7) months on average.Among the 13 patients with screw loosening, 1 patient suffered screw loosening in single segment, 3 patients in two segments, and 9 patients in three segments or above.Osteoporosis combined with screw loosening.appeared in 10 patients VAS and ODI scores in all subjects had statistically differences at 6 months after operation and the last follow up versus preoperation (P<0.05 for all), but no statistical difference versus the last follow-up(P> 0.05).There were significant differences in gender and preoperative osteoporosis rate between the two groups (P<0.05 for both).Conclusions Osteoporosis is one of the reasons for the screw loosening in elderly patients after pedicle screw internal fixation.The increased probability of the screw loosening in long segment may be related with the change of the older spine mechanics.Further randomized controlled trials are still required to confirm whether elderly osteoporosis patients need to receive bone cement augmentation treatment.
4.Cloning and sequence analysis of dystonin(Dst) variant X1 in Cricetulusbarabensis and albino mutant Cricetulusbarabensis

Objective To clone and sequence the dystonin variant X1 gene of Cricetulusbarabensis and the albino mutant Cricetulusbarabensis so as to find out the difference of encoding arear of the muscular ribosome between Cricetulusbarabensis and the albino mutant Cricetulusbarabensis.Methods According to the same type of abnormal muscle tone protein of the mice and rats, we designed 6 pairs of primers, and got their cDNA genes from skins of the Cricetulusbarabensis and the albino mutant Cricetulusbarabensis by RT-PCR amplification, then cloned and sequenced. Results Sequence alignment showed 17 variances in coding areas, and 24 in amino acid, but no in key nucleic acid and protein.Conclusion The variances in coding areas will not lead to the albino, and its mechanism requires further investigation.
5.Regulatory function of long noncoding RNA MAGI2-AS3 in tumor development: a review

Abstract
Long noncoding RNA (lncRNA) is closely related to the pathogenesis of cancer,representing a burgeoning field in tumor research in recent years. MAGI2-AS3, a tumor-associated lncRNA, exerts pivotal roles in epigenetics, transcription and post-transcriptional regulation. Studies have suggested that MAGI2-AS3 may be involved in multiple stages of tumor development, and has potential applications for tumor diagnosis, therapy and prognosis. This review summarizes the expression, regulatory mechanism and clinical application value of MAGI2-AS3 in common malignant tumrs, providing the reference for tumor prevention and treatment.
6.Expression of VEGF, COX-2 mRNA in esophageal cancer

Objective To study the expression of VEGF isoforms, COX-2 in esophageal cancer Methods VEGF, COX-2 mRNA expression in 40 paired samples (tumor and adjacent normal tissue) were determined by using real time RT-PCR. Results VEGF165 was overexpressed in 25 of 40(62.5 %) tumor tissues compared with in 6 of 40 (15 %) adjacent normal tissue; COX-2 was overexpressed in 28 of 40(70 %) tumor tissues compared with in 5 of 40(12.5 %) adjacent normal tissue. Conclusion This result suggests that VEGF165 and COX-2 overexpression in esophageal cancer.
7.Iodine nutrition and goiter prevalence among children at ages of 8 to 10 years in Zhejiang Province

Objective:
To investigate the status of iodine nutrition and prevalence of goiter among children at ages of 8 to 10 years in Zhejiang Province in 2021, so as to provide insights into the management of iodine deficiency in children.
Methods :
A total of 90 counties (districts) were sampled as field survey sites from 11 cities of Zhejiang Province using a multi-stage stratified random sampling method in 2021, and non-residential children at ages of 8 to 10 years in these sites were sampled as study subjects. Subjects' household edible salt samples and random urine samples were collected. The iodine content in salt and urinary iodine level were determined using the direct titration method and arsenic-cerium catalytic spectrophotometry for evaluation of iodine nutrition among children. In addition, the lateral lobe of the thyroid gland was measured using ultrasound, and the prevalence of goiter was estimated.
Results:
A total of 19 363 children were recruited, including 9 710 male children and 9 653 female children, with a male to female ratio of 1.01︰1, and there were 10 704 urban children (55.28%) and 8 659 rural children (44.72%), 9 149 children living in coastal areas (47.25%) and 10 214 children in inland regions (52.75%). The mean iodine content was (19.79±9.25) mg/kg in salt, and the coverage of qualified iodized salt (15 252) was 78.77%. The coverage of qualified iodized salt was significantly lower in urban children than in rural children (76.70% vs. 81.21%; χ2=68.301, P<0.001), and was lower in children living in coastal regions than in inland regions (68.05% vs. 88.27%; χ2=1 270.769, P<0.001). The median urinary iodine concentration was 195.1 μg/L, and the household iodine content in salt correlated positively with urinary iodine concentration in children (rs=0.383, P<0.001). There were 2 885 children with iodine deficiency (14.90%), 7 137 children with adequate iodine (36.86%), 5 414 children with excessive iodine intake (27.96%), and 3 927 children with iodine overdose (20.28%), and the distribution of iodine nutrition in children varied significantly in regions (χ2=283.277, P<0.001) and gender (χ2=126.349, P<0.001). The prevalence of goiter was 2.45% among 7 195 children receiving ultrasound examinations, and a higher prevalence rate of goiter was detected in urban children than in rural children (2.76% vs. 2.00%; χ2=3.962, P=0.047).
Conclusions
The overall urinary iodine nutrition was adequate among children at ages of 8 to 10 years in Zhejiang Province in 2021, and the prevalence of goiter in children fell within the threshold defined in the criteria of elimination of iodine deficiency. However, the supervision of the iodized salt quality remains to be improved and iodine deficiency control remains to be reinforced.
8.Study on Electroacupuncture in Improving Discomforts During and after Oocyte Retrieval

Objective To observe the efficacy of electroacupuncture in improving discomforts during and after oocyte retrieval, for providing scientific and significant clinical evidence for the application of electroacupuncture to oocyte retrieval.Method A hundred and thirty-four outpatients receiving IVF-ET fertility treatment were randomized into a treatment group and a control group, 67 in each group. The two groups both received muscular injection with Sauteralgyl 50 mg. Additionally, the treatment group was also given acupuncture at Baihui (GV20), Uterus (MA-TF, right ear), Tengtong (Extra, right), Sanyangluo (TE8, right), and Zusanli (ST36). When needling qi arrived, Tengtong (Extra) and Sanyangluo (TE8) were connected to SDZ-2 electroacupuncture apparatus. The needles were removed at the end of oocyte retrieval. For patients in the control group, oocyte retrieval was conducted 30 min after injection with Sauteralgyl. The operation duration was recorded; blood pressures before injection and at the end of surgery were measured; discomforts including dizziness, nausea, vomiting, perspiration, dry mouth, palpitation, and abdominal distension during operation were observed; discomforts (dizziness, nausea, vomiting, abdominal pain, and lassitude) 0.5 h, 1.5 h, and 2 h after operation were observed.Result The difference in comparing oocyte retrieval duration was statistically significant between the two groups (P<0.05); there were significant differences in comparing the occurrence rates of dizziness, perspiration, nausea, and palpitation during operation between the two groups (P<0.05); there were significant differences in comparing dizziness, vomiting, nausea, abdominal pain, and lassitude between the two groups 0.5 h, 1 h, 1.5 h, and 2 h after operation (P<0.05). The inter-group differences in dizziness, vomiting, nausea, abdominal pain, and lassitude were statistically significant after operation (P<0.05).Conclusion Electroacupuncture is effective in relieving discomforts during and after oocyte retrieval under transvaginal ultrasound.
9.Construction of eukaryotic vector of monkey B virus glycoprotein D gene and the gD gene expression

Objective To establish an eukaryotic vector of monkey B virus glycoprotein D gene and analyze the expression of gD gene in human embryonic kidney 293T cells.Method First, the protein of monkey B virus glycoprotein D was obtained by gene synthesis.The gene fragments were digested with Pst I and Not I, and ligated to pEGPF-N3. Then, the recombinant plasmid pEGPF-N3-GD was transfected into 293T cells.The expression of gD protein in the cells was detected by Western blot, and the expression localization was investigated using laser scanning confocal microscopy. Results The recombinant plasmid pEGPF-N3 carrying gD gene was successfully constructed, and normally expressed in the 293T cells.Conclusions Glycoprotein D of monkey B virus is expressed successfully in the 293T cells and the protein is located on the cell surface.It may be useful for the preparation of specific recombinant antigen to the glycoprotein D of monkey B virus on cell surface, and can be also used for preparation of antigen slide for detection of monkey B virus.
10.Effects of leukocyte-depleted allogeneic blood transfusion on perioperative cellular immunity in children

Objective To evaluate the effects of leukocyte-depleted allogeneic blood transfusion on perioperative cellular immunity in children.Methods Three hundred and fifty-nine ASA Ⅰ or Ⅱ children (aged 3 months-14 years and weighing 5-74 kg) requiring allogeneic blood transfusion during operation were randomly divided into two groups:163 children receiving normal allogeneic blood transfusion (control group,group C) and 196 children receiving leukocyte-depleted allogeneic blood transfusion (group D).Blood samples were collected from the peripheral vein before blood transfusion,and 2 and 6 days after blood transfusion for determination of the levels of CD3+,CD4+,CD8 +,and CD56+ by flow cytometry.CD4+ /CD8+ ratio was calculated.The volume of allogeneic blood transfusion during operation,the duration of operation,postoperative drainage,antibiotic administration,hospital stay and the incidence of postoperative infection were recorded.Rssults The levels of CD3+,CD4+,CD56+ and CD4+/CD8+ ratio significantly increased at 6 days after blood transfusion while the duration of postoperative drainage,postoperative antibiotic administration,hospital stay and the incidence of postoperative infection significantly decreased in group D compared with group C.Conclusion Leukocyte-depleted allogeneic blood transfusion is helpful in improving the postoperative cellular immunity in children.
