1.Evaluation of dissolution consistency of Iloperidone Tablets

Objective To compare the dissolution curves of reference preparation and self-prepared Iloperidone Tablets in four different pH dissolution media (0.1 mol/L HC1 solution,pH 4.5 acetate buffer solution,pH 6.8 phosphate buffer solution,and water).Methods The solubility of Iloperidone in different pH solutions was measured,the dissolution curves of two preparations in four different pH dissolution media were determined by HPLC,and the similarity was investigated according to the f2 factor method.Results The f2 factors between reference preparation and self-prepared Iloperidone Tablets in four different media were more than 50.Conclusion The two preparations are equivalent in four different pH dissolution media in vitro.
2.Ionization interference and its suppressing in determination of heavy metal elements in iron dextran by inductively coupled plasma optical emission spectrometry

Objective To establish a method for the determination of eight heavy metal elements ofPb,Cd,As,Hg,Co,V,Se,Mo in iron dextran by inductively coupled plasma optical emission spectrometry (ICP-OES).Method Through selection of detection wavelengths,optimization of instrument parameters,correction of spectral interference,verification of ionization interference and investigation of suppression methods,the eight heavy metal elements were analyzed by ICP-OES.Results The recovery rate of the detected elements increased 5% to 10% by suppression of ionization interference.The accuracy of the method was good.The spiked recovery rates of the detected elements were from 95.7% to 101.1%.The precision of the method were good (RSD < 3.6%,n =6).The linearities of the detected elements were good,and the correlation coefficients were all greater than 0.999.The detection limits were from 0.15 to 8.09 ng/mL.The quantization limits were from 0.46 to 24.26 ng/mL.Conclusion The method was accurate,sensitivity,rapid and reliable,which can be applied to the determination of contents of eight heavy metal elements in iron dextran.
3.Simultaneous Determination of 8 Heavy Metal Elements in Iron Dextran by ICP-OES

Objective:To establish a method for the simultaneous determination of 8 heavy metal elements Pb, Cd, As, Hg, Co, V, Se and Mo in iron dextran by inductively coupled plasma optical emission spectrometry ( ICP-OES) . Methods:Through the detec-tion wavelength selection,the detection wavelength was confirmed as follows:220. 353 nm for Pb, 228. 802 nm for Cd, 188. 980 nm for As, 194. 164 nm for Hg, 228. 615 nm for Co, 311. 070 nm for V, 196. 026 nm for Se and 204. 598 nm for Mo. Through optimizing the instrument parameters, the optimal radio frequency power was 1. 3 kW, the nebulizer gas flow rate was 0. 7 L·min-1, and the pump speed was 10 r·min-1. By applying the above detection parameters, the 8 heavy metal elements were analyzed by ICP-OES simultaneously. Results: The linearity of the detected elements was good, and the correlation coefficients were all greater than 0. 9990. The detection limits were from 0. 12 to 7. 26 ng·ml-1 , the quantitation limits were from 0. 40 to 23. 96 ng·ml-1 and the recoveries were from 94. 1% to 103. 4% (RSD<3%, n=9). Conclusion: The method is specific, sensitive, rapid and accurate, which can be applied in the simultaneous determination of 8 heavy metal elements in iron dextran.
4.Determination of eight heavy metal elements in iron dextran by inductively coupled plasma optical emission spectrometry and correction of spectral interference

Objective To establish a method for the determination of eight heavy metal elements (Pb,Cd,As,Hg,Co,V,Se,and Mo) in iron dextran by inductively coupled plasma optical emission spectrometry (ICP-OES).Methods Through detection wavelength selection,optimization of instrument parameters and applying interference element correction (IEC) method to correction of spectral interference,the eight heavy metal elements were analyzed by ICP-OES.Results The elements recoveries were from 88.7% to 101.3% by correction of spectral interference with IEC method.The accuracy of the method was good.The linearities of the detected elements were good,and the correlation coefficients were all greater than 0.999.The detection limits were from 0.12 to 7.26 ng/rnL.The quantization limits were from 0.40 to 23.96 ng/mL.The precision of the method was good (RSD<3.5%,n=6).Conclusion The results of the spectral interference correction by IEC method are greatly superior to the results of conventional method.The established method is accurate,sensitive,and rapid,which can be applied to the determination of contents of eight heavy metal elements in iron dextran.
5.Construction of eukaryotic vector of monkey B virus glycoprotein D gene and the gD gene expression

Objective To establish an eukaryotic vector of monkey B virus glycoprotein D gene and analyze the expression of gD gene in human embryonic kidney 293T cells.Method First, the protein of monkey B virus glycoprotein D was obtained by gene synthesis.The gene fragments were digested with Pst I and Not I, and ligated to pEGPF-N3. Then, the recombinant plasmid pEGPF-N3-GD was transfected into 293T cells.The expression of gD protein in the cells was detected by Western blot, and the expression localization was investigated using laser scanning confocal microscopy. Results The recombinant plasmid pEGPF-N3 carrying gD gene was successfully constructed, and normally expressed in the 293T cells.Conclusions Glycoprotein D of monkey B virus is expressed successfully in the 293T cells and the protein is located on the cell surface.It may be useful for the preparation of specific recombinant antigen to the glycoprotein D of monkey B virus on cell surface, and can be also used for preparation of antigen slide for detection of monkey B virus.
6.Synthesis and eukaryotic expressing of the gene encoding extracellular domains of gB protein B virus

Objective To get the gene encoding extracellular domains of gB protein of B virus and analyze its expression in the eukaryocyte cell.Methods synthesizing gene fragment encoding extracellular domains of gB protein of B virus was by using synthesis gene, then digested with the restriction endonucleases BamHⅠand NotⅠand inserted into eukaryotic expressing vector pEGFP-N3.pEGFP-N3-GB合 was transfected into 293 cells.After protein extraction, the expression of gene was detcted by western blotting, and the cellular localization of the gene was analyzed by immunofluorescence and laser scanning confocal microscopy.Results pEGFP-N3-GB合were expressed in 293 cells and on the cell membrane.Conclusion eukaryotic expressing system can produce specific antigen recombination protein of B virus gB protein and express on the cell membrane.
7.Research progress of immune regulation in antidepressant treatment

In recent years, the role of inflammation in depression has received people's attention.Studies have suggested that immune disorder may play an important role in depression, and patients with depression exhibit characteristic immunophenotypes.Inflammation seems to interact with a variety of pathogenesis of depression.Therefore, immunoregulation is becoming an adjuvant therapy for depression.Clinically, not only antidepressants show anti-inflammatory effects, but also anti-inflammatory drugs show antidepressant effects, and they mainly including non-steroidal anti-inflammatory drugs, statins, and cytokine inhibitors.In addition, some non-drug treatment methods are also given immunomodulatory effects, such as electric shock therapy, vagus nerve stimulation, acupuncture and exercise therapy.However, there are still some problems in immunomodulation therapy, such as immunomodulation therapy may be only effective for some subgroups of patients, and its efficacy and safety need to be evaluated.In the future, looking for more effective biomarkers and identifying immune-inflammation-related subtype of depression, will serve to explore new diagnosis and treatment strategies.
8.Study of Symptom Burden and Symptom Clusters at the End of Thoracic Radiotherapy in 220 Patients with Lung Cancer

Objective To investigate the characteristics of symptom burden and extract symptom clusters in lung cancer patients at the end of thoracic radiotherapy.Methods Lung cancer patients receiving Thoracic Radiotherapy were collected from 20 tertiary hospitals in China,including Guang'anmen Hospital of the Chinese Academy of Traditional Chinese Medicine,during November 2014—September 2017,and the MDASI-TCM scale assessed patient symptoms at the end of radiotherapy,and descriptive statistics and exploratory factor analysis explored symptom burden and the distribution of TCM symptom clusters.Results At the end of radiotherapy,45.1%of patients reported two or more moderate to severe symptoms,with fatigue being the most prevalent and severe symptom.Fatigue(31.4%),cough(30.9%),expectoration(24.1%),distress(23.2%)and shortness of breath(20.9%)were the top five symptoms from the highest to the lowest in the incidence of moderate to severe symptoms.The enjoyment of life(36.4%)had the highest incidence of interference with moderate to severe symptoms.Exploratory Factor analysis extracted three symptom clusters,namely the excessive heat and damaged yin-Spleen and stomach disorders related symptom cluster,the heart and spleen-deficient related symptom cluster,and the lung qi-unfriendly related symptom cluster,with a cumulative variance contribution of 70.1%.Conclusion Lung cancer patients undergoing thoracic radiotherapy have a certain symptom burden at the end of radiotherapy.The identification of symptom burden and symptom group characteristics is conducive to promoting standardization of TCM syndrome differentiation and laying a preliminary foundation for formulating reasonable and effective TCM intervention measures in the future.
9.Analysis of the association between threat and deprivation childhood traumas and dysfunctional attitudes in patients with major depressive episode

Objective This study aims to explore the associations between threat and deprivation childhood traumas and dysfunctional attitudes among patients with major depressive episode,based on the dimensional model of adversity and psychopathology(DMAP).Methods Data from 970 patients with major depressive episode were collected and categorized based on the childhood trauma questionnaire-short form(CTQ-SF)scores into no trauma(426),threat-only(86),deprivation-only(210),and mixed groups(248).Cognitive distortions were assessed using the dysfunctional attitudes scale(DAS),with scores above the upper quartile indicating significant distortions.Multifactorial logistic regression analysis was employed to examine the association between threat and deprivation and dysfunctional attitudes.Results There were significant differences in the proportion of cognitive distortions among the no trauma,threat-only,deprivation-only,and mixed groups(19.25% vs.30.23% vs.20.00% vs.35.89%,P<0.001),with differences across all dimensions(P≤0.001).Multifactorial logistic regression revealed a positive correlation between threat and overall cognitive distortions(OR=2.031,P<0.001),and specific positive correlations with dimensions such as vulnerability(OR=1.709,P<0.001),appeal and repulsion(OR=2.088,P<0.001),perfectionism(OR=2.321,P<0.001),compulsivity(OR=1.765,P=0.001),approval seeking(OR=1.837,P<0.001),dependency(OR=1.554,P=0.006),and autonomous attitudes(OR=1.864,P<0.001).In contrast,deprivation was only positively correlated with the cognitive philosophy dimension(OR=1.495,P=0.015).Conclusion Childhood trauma is closely associated with dysfunctional attitudes in patients with major depressive episode.Threat correlates positively with cognitive distortions in all dimensions except cognitive philosophy,whereas deprivation correlates only with the cognitive philosophy dimension.
10.Evaluation for the performance of HIV Ag/Ab assay based on ECLIA in blood screening

【Objective】 To evaluate the performance of electrochemiluminescence immunoassay (ECLIA) in detecting HIV antigen/antibody in blood screening. 【Methods】 A total of 128 donors, reactive to anti-HIV(ELISA)testing, from September 2016 to September 2020 were enrolled, and seven samples were reactive to double anti-HIV reagents, among which 6 were confirmed by WB, 1 confirmed by NAT as negative.Two group of donors, reactive to solo anti-HIV reagent but being confirmed negative by WB(n=121) vs.randomly selected donors non-reactive to ELISA + NAT(June to September 2020, n=1360), were subjected to HIV antigen/antibody testing using ECLIA to compare the testing results, including concordance rate, sensitivity and specificity. 【Results】 The ECLIA results remained non-reactive for 1360 samples initially non-reactive to both ELISA and NAT.The concordance rate of anti-HIV reactivity by ECLIA and ELISA+ WB were 100%(6/6). For 122 samples, reactive to ELISA anti-HIV testing but nonreactive to confirmatory testing, 4(3.28%)of them were reactive to HIV antigen/antibody testing and 118(96.72%) nonreactive, with the concordance rate of ECLIA and ELISA at 96.88%(124/128). The sensitivity, specificity and false positive rate of ECLIA and ELISA were 100% vs 100%, 99.73% vs 91.77%, and 0.27% vs 8.23%, respectively. 【Conclusion】 ECLIA for HIV antigen/antibody detection has good sensitivity and specificity, which can meet the requirements of blood screening, and the false positive rate is lower than that of ELISA, adopted commonly in blood bank at present.
