Aim To explore the mechanism of E.coli heat-labile enterotoxin B subunit(LTB)as adjuvant by analysis of cellular proteins interacting with LTB. Methods Whole cell proteins were purified from RAW 264.7 cell after treated with LTB or NaCl 12 h, respectively. The cellular proteins were interacted with LTB and the interacting proteins were purified by pull-down assay and identified by mass spectrography. The LTB interaction proteins were conformed with Western blot and immunofluorescence assay. Results 25 LTB interaction proteins were found, and their interaction network was mapped;four proteins (Jup,Dsp,Ddx5 and Vimentin)were indicated to be related with LTB adjuvant activity;immunofluorescence assay indicated that GM130 interacted with LTB,however,Vimentin had no interaction with LTB in vivo. After treated by LTB,the expression of β-actin was upregulated obvi-ously in RAW 264.7 cell,whereras,Hspd1 did not show any change. Conclusions LTB exerts adjuvant activity through binding to GM1 of immune cells,cau-sing endocytosis and transporting to the Golgi apparatus by vesicles. Then LTB might bind to Jup and affect TCF/LEF activity, regulating the expression of Bcl 2, IL-6,and Runx3.The result is promoted T cell and B cell proliferation, differentiation and activation by se-cretion of cytokines and immunoglobulins.

Objective To analyze clinical and genetic risk factors of refractory or relapsed acute myeloid leukemia (AML) patients, and evaluate the efficacy of reinduction of chemotherapy. Methods 296 newly diagnosed AML patients, including 89 refractory or relapsed cases, were observed with clinical characteristics. And the efficiency of different reinduction chemotherapy regimens were compared. Results Compared with the non-refractory or relapsed AML,age, complex karyotype and Fms like tyrosine kinase 3 internal tandem duplication (FLT3-ITD) gene mutations were risk factors of relapsed or refractory AML (P < 0.05).Seventy-eight refractory and relapsed AML patients received reinduction therapy. The overall response rate (the complete response rate and the partial response rate) was 44.90 % (30/78).All reinduction regimens were divided into three categories:using the initial induction scheme or using new induction scheme including some chemotherapeutics without cross-resistance (regimen A),using the induction regimen containing medium-or high-dose cytarabine (regimen B),and using priming regimen containing of G-CSF,cytarabine,aclacinomycin or homoharringtonine (regimen C).Their overall response rate were 35.12 % (13/37),61.90 % (13/21) and 45.00 % (9/20),respectively,in which the overall response rate of regimen B was statistically higher than regimen A (P < 0.05).Conclusions Age,complex karyotype and FLT3-ITD mutation were important causes of relapsed or refractory AML.The overall response rates were different among three different reinduction regimens. It is helpful to improve the overall response rate of reinduction therapy to use the regimen containing medium-or high-dose cytarabine, which was more suitable for young patients. For patients with poor tolerance, the priming regimen suit was more helpful to improve the overall response rate.

American medical insurance is mainly composed of public medical insurance borne by the government and private medical insurance. The main cause of death in this country are ischemic heart disease, Alzheimer’s disease, and lung cancer. The motality rate mainly caused by drug use disorders, chronic kidney disease and Alzheimer’s disease. The development of Chinese herbal medicine is relatively slow and difficult. The education system has covered Traditional Chinese Medicine (TCM). However, there still exist certain challenges of the development of TCM acupuncture faces the challenge of localization; Chinese herbal medicine still lacks standardization and TCM education needs to be standardized. Based on the current situation, it is suggested to focus on the development of acupuncture and moxibustion, to promote the registration and declaration of Chinese herbal medicine products for treating difficult diseases, and to strengthen the international exchange of TCM education, so as to promote the development and spread of TCM in the United States.

OBJECTIVETo evaluate the association of glutathione S-transferase M1 (GSTM1) and glutathione S-transferase T1 (GSTT1) polymorphisms with noise-induced hearing loss.
METHODSThe Cochrane library, PubMed, OVID, EMBASE, Springer, Wanfang Data, VIP, CNKI, and CBM were searched to collect case-control studies on GSTM1 or GSTT1 polymorphism and noise-induced hearing loss. The articles meeting the inclusion criteria were reviewed systematically, and the reported data were aggregated using Revman 5.0.
RESULTSFive studies were included in the meta-analysis. The meta-analysis and subgroup analysis showed that the persons with GSTM1 null genotype had an increased risk of noise-induced hearing loss compared with those with GSTM1 wild genotype (OR = 1.37, 95%CI: 1.13∼1.66); in the Chinese population, the risk of noise-induced hearing loss was higher in persons with GSTM1 null genotype than in those with GSTM1 wild genotype (OR = 1.5, 95%CI: 1.2∼1.86); there was no significant difference in the risk of noise-induced hearing loss between persons with GSTT1 null and wild genotypes.
CONCLUSIONGSTM1 polymorphism is related to noise-induced hearing loss, but GSTT1 polymorphism is unrelated to this condition.

Objective:
To compare the clinical effects of laparoscopic repair and open repair of gastroduodenal ulcer perforation.
Methods:
Retrospective analysis was performed on 117 patients with perforated gastroduodenal ulcer admitted to Sijing Hospital of Shanghai Songjiang District from October 2005 to February 2018, including 86 males and 31 females. The average age was 35.56 years with a range from 17 to 68 years. Patients were divided into two groups according to different surgical methods: laparoscopic group (

β-N-acetylglucosaminidases (NAGases) can convert natural substrates such as chitin or chitosan to N-acetyl-β-D glucosamine (GlcNAc) monomer that is wildly used in medicine and agriculture. In this study, the BcNagZ gene from Bacillus coagulans DMS1 was cloned and expressed in Escherichia coli. The recombinant protein was secreted into the fermentation supernatant and the expression amount reached 0.76 mg/mL. The molecular mass of purified enzyme was 61.3 kDa, and the specific activity was 5.918 U/mg. The optimal temperature and pH of the BcNagZ were 75 °C and 5.5, respectively, and remained more than 85% residual activity after 30 min at 65 °C. The Mie constant Km was 0.23 mmol/L and the Vmax was 0.043 1 mmol/(L·min). The recombinant BcNagZ could hydrolyze colloidal chitin to obtain trace amounts of GlcNAc, and hydrolyze disaccharides to monosaccharide. Combining with the reported exochitinase AMcase, BcNagZ could produce GlcNAc from hydrolysis of colloidal chitin with a yield over 86.93%.
