1.Determination of Astragaloside Ⅳ in Xin'an Granules by HPLC-ELSD

OBJECTIVE: A HPLC-ELSD method was developed for determination of the content of astragaloside Ⅳ in Xin'an granules.METHODS: The separation was performed on Kromasil ODS C18 column(150 mm?4.6 mm,5 ?m) with the mobile phase consisted of acetonitrile-water(35∶65).The temperature of the drift tube was 40 ℃ and the flow rate of gas(air) was 2.1 L?min-1.RESULTS: The linear range of astragaloside Ⅳ was 1.32~7.95 ?g(r=0.998 9) and the average recovery was 97.84(RSD=1.01%,n=9).CONCLUSION: The method is simple,accurate and reproducible,and it can be used for the quality control of Xin'an granules.
2.Pharmacodynamic Study of Yunkang Capsule

Objective To investigate the pharmacodynamic action of Yunkang Capsule(YKC).Methods The animals were divided into YKC groups(at high-,moderate-and low-dosage respectively),diphenidol control group,model control group and blank control group.The action of YKC on vertigo and thrombosis,hemorrheologic indexes,piamatral microcirculation and free radicals of the animal models were observed.Results YKC could significantly prolong the latency of vertigo,reduce wet weight of thrombus,decrease blood viscosity,plasma viscosity and hematocrit,promote the dilation of piamatral micro-vessels,increase the amount of interweave microdot,increase plasma SOD activity and decline plasma MDA content of animal models.Conclusion YKC has actions of counteracting vertigo and thrombosis,improving hemorrheology and microcirculation,and clearing away free radicals.
3.Expression of STAT6 in human nasal polyps and the relation between STAT6 and eosinophil infiltration

Objective:To study the distribution and expression of STAT6 and to examine the suggested roles of STAT6 in the pathogenesis of eosinophil infiltration in nasal polyps and to evaluate the role of STAT6 in the pathogenesis of nasal polyps.Method:All selected cases met the enrollment criteria.Thirty samples of nasal polyps were obtained from patients undergoing nasal polypectomy, and 10 samples of inferior turbinate tissues were from patients undergoing nasal septal reconstruction.STAT6 in nasal polyp tissues from 30 nasal polyposis patients and 10 samples of inferior turbinate tissues were detected with immunohistochemistry(SP) method.SPSS13.0 system was used to perform the statistical analysis.Result:The positive expression of STAT6 was significantly higher in epithelium of nasal polyps than that of the control. The number of eosinophils was significantly higher in epithelium of nasal polyps than that of the control. The difference between these two groups was statitically significant(P<0.05). STAT6 positive cell were localized on epithelium, gland cells and on inflammatory cell of nasal polyps. STAT6 expression was positively correlated with the recruitment of eosinophils in nasal polyps.Conclusion:The high expression of STAT6 protein and the suggested roles of STAT6 in the recruitment of eosinophils in nasal polyps may contribute to the initiation and progression of nasal polyps.
4.Evaluating the relationship between intracranial atherosclerotic plaque stability and stroke recurrence risk with high-resolution MRI

Objective To investigate the relationship between intracranial atherosclerotic plaque stability and stroke recurrence risk.Methods Forty-eight patients with acute ischemic stroke caused by intracranial atherosclerosis in Guangdong Province Hospital of TCM were analyzed retrospectively.After the Essen Stroke Risk Scale (ESRS) was used to assess the risk factors for the patients,they were divided into either an ESRS ≥3 group (n=21 in the high-risk recurrence group) or an ESRS <3 group (n=27 in the low-risk recurrence group).Both groups of patients underwent high-resolution MR imaging (HR-MRI) examinations of the intracranial guilty vessels (basilar artery or unilateral middle cerebral artery).According to the signal intensity of HR-MRI on the T1-weighted imaging,T2-weighted imaging,and T1 fat suppression sequences,the intracranial atherosclerotic stable plaques and unstable plaques were distinguished.The stabilization of intracranial atherosclerotic plaques was compared in patients of both groups.Results There were significant differences in the age and incidences of hypertension,diabetes mellitus,and unstable plaques in patients of both groups (P<0.05).Further multivariate logistic regression analysis of the four factors showed that the age,hypertension,diabetes,and intracranial atherosclerotic unstable plaques were the high-risk factors for recurrent ischemic stroke (ORs,87.114,159.423,8.942,and 11.551,respectively;95%CIs 4.218-1 799.078,3.235-7 855.957,1.054-75.857,and 1.011-132.043,respectively;all P<0.05).Conclusion In addition to the traditional risk factors such as age,hypertension,and diabetes,the intracranial atherosclerotic unstable plaque is an independent risk factor for high-risk recurrence of ischemic stroke.
5.Evaluation of the dinical curative effect of using 153Sm-EDTMP in painful bone metastasis of malignant tumor

Objective To evaluate the clinical curative effect of using 153Sm-EDTMP in painful bone metastasis of malignant tumor. Methods Eighty patients with bone metastasis of malignant rumor underwent radionuclide bone palliation therapy were analysed.The treatment efficacy was evaluated by visual analogue scale (VAS) and Kamofsky performance scale.Results 68.75% of patients had a positive response.A better analgesic effect was found in cases of lung,prostate and breast carcinoma metastasm compared to metastasis from other malignant lesions.Improvement of performance in Karnofsky scale was found in cases of midrange and heavy range patients.Conclusion The analgesic effects of 153Sm-EDTMP is obvious in painful bone metastasis of malignant tumor.Improvement of life quality is significant in cases of midrange and heavy range patients.The therapeusis is a beneficial supplement of radiotherapy and odynolysis therapy.
6.The role of 131Ⅰ in the apoptosis of thyrocytes of patient with Graves' disease

The changes of Fas.FasL and Bcl-2 expression in thyrocytes of patients with Graves'disease were investigated before and 2 weeks after 131Ⅰ administration. The results showed that 131Ⅰ couhl induce thyrocytes to express the apoptotic protein Fas, FasL and the anti-apoptotic protein Bet-2 in patients with Graves'disease. A statistically significant correlation was found between the dose of 131Ⅰ and the expression levels of Fas and FasL but not Bet-2 ,suggesting that early onset of hypothyroid after 131Ⅰ administration may be due to the increased expression of Fas and FasL in thyrocytes.
7.Effect of Cordyceps Sinensis from Different Origins on Immune Response in Mice

Objective To investigate the effect of cordyceps sinensis from different origins on immune response in mice. Methods Cordyceps sinensis from two origins were prepared into powder, and then the mice were divided into high, middle and low dose(0.4,0.2,0.1 g.kg-1)groups, respectively.In addition, purified water was given as the normal control group.Effects of cordyceps from two different origins were observed by detecting spleen lymphocyte proliferation induced by ConA, delayed type hypersensitivity ( DTH) in mice induced by sheep red blood cells ( SRBC ) , the number of antibody-producing cells, carbon clearance and peritoneal macrophages Swallow fluorescent microspheres, as well as the activity of NK cells. Results The ability of spleen lymphocyte proliferation induced by ConA, carbon clearance and peritoneal macrophages Swallow fluorescent microspheres, and the activity of NK cells were significantly enhanced in the middle and high dose group of two different origins cordyceps, compared with normal control group (P<0.05).Additionally, the number of antibody-producing cells was obviously increased in medium dose group of both origins cordyceps and decreased in the high dose group (P<0.05).The middle and high dose Qinghai cordyceps significantly improved DTH in mice, while Tibet cordyceps sinensis had no obvious effect, and there was significant difference (P<0.05) between the high dose group of Qinghai and three dose groups of Tibet Cordyceps sinensis.In addition, levels of serum hemolysin in mice were significantly increased in the middle and high dose group of Qinghai and high dose group of Tibet Cordyceps sinensis (P<0.05), and the differences of corresponding medium and high doses of two origins were significant ( P<0.05) . Conclusion Cordyceps sinensis of both different regions significantly improved the immune response of mice.However, the efficacy between the two origins was roughly equivalent and had no significant difference.
8.Clinical characteristics and gene mutation analysis of one pedigree with infantile glycogen storage disease type II.

The clinical data of 2 infants with infantile glycogen storage disease type II (GSD II) from one pedigree were collected. The method of dried blood spots (DBS) was applied to collect peripheral blood samples, and the activity of acid alpha-D-glucosidase (GAA) in leukocytes was measured. The coding region of GAA gene in this pedigree was amplified by polymerase chain reaction and then direct sequencing was used to analyze mutations in GAA gene. The two infants were twins, who were admitted to the hospital due to feeding difficulties, generalized muscle weakness and hypotonia, cardiomegaly, and cardiac insufficiency when they were 10 months old. The GAA activity in leukocytes in the two infants was significantly lower than in normal controls. Gene sequencing revealed 2 compound heterozygous mutations in the two infants, i.e., G1942A and G2214A, respectively. G1942A had been proved pathogenic, and the latter one, G2214A, was a nonsense mutation, resulting in the change of tryptophan, the 738th amino acid of GAA, into a stop codon. The two infants were diagnosed with GSD II by gene detection and no enzyme replacement therapy could be provided to them. Follow-up visits showed that the two infants died at home at the age of 15 months and 17 months, respectively. GSD II is caused by deficiency of GAA activity resulting from mutation of GAA gene. The detection of GAA activity in peripheral blood by DBS and GAA gene detection are effective and feasible methods for diagnosis of GSD II.

9.Clinical significance of the expression of p53, p63 and p73 in nasal and paranasal sinus carcinoma.

OBJECTIVE:
To study the clinical significance of the expression of p53, p63 and p73 protein and the correlation of p53 and p63 in nasal and paranasal sinus carcinoma (NPSC).
METHOD:
The immunohistochemical technique was used to detect the expression of p53, p63 and p73 in 67 cases of NPSC, para-cancer tissues and non-cancer tissues.
RESULT:
The positive rate of p53 and p63 protein in NPSC was significantly higher than that in park cancer tissues and non-cancer tissues (P<0.01), and there were a positive correlation between the expression of p53 and p63 protein in NPSC (P<0.01). But the expression of p73 had no significant difference among NPSC, para-cancer tissues and non-cancer tissues (P>0.05).
CONCLUSION
There are positive correlation between p53 and p63 protein in NPSC, and they may play an important role in the tumorgenesis of NPSC. The p73 may not be associated with tumorgenesis of NPSC.

10.Expression and significance of STAT3 and VEGF with MVD in the nasal polyps.

OBJECTIVE:
To investigate microvessel density (MVD) and expressions of phosphor-STAT3 and vascular endothelial growth factor (VEGF) in nasal polyps, to explore their pathogenetic correlations.
METHOD:
All selected cases were measured up the standard of enrollment. Thirty samples of nasal polyps were obtained from patients undergoing nasal polypectomy, and 10 samples of inferior turbinate tissues were from patients undergoing nasal septal construction. p-STAT3,VEGF and MVD in nasal polyp tissues from 30 nasal polyposis patients and 10 samples of inferior turbinate tissues were detected with immunohistochemistry (sp) method.
RESULT:
In nasal polyp tissues, The positive rates of p-STAT3 and VEGF expression were 60% and 70% respectively, all were significantly higher in nasal polyps than in control inferior turbinates (P < 0.01). MVD was significantly higher in nasal polyps than in control inferior turbinates (22.78 +/- 7.89 vs 7.12 +/- 2.84, P < 0.01). p-STAT3 expression was positively correlated with VEGF expression and MVD.
CONCLUSION
p-STAT3, VEGF and MVD were overexpressed in the tissues of nasal polyps, and p-STAT3 expression was positively correlated with VEGF expression and MVD; p-STAT3 was positively correlated with the neovascularization of nasal polyps, The inhibition of STAT3 pathway may inhibit the neovascularization, and then inhibit the formation and recurrence of the nasal polyps.
