1.Determination of Astragaloside Ⅳ in Xin'an Granules by HPLC-ELSD

OBJECTIVE: A HPLC-ELSD method was developed for determination of the content of astragaloside Ⅳ in Xin'an granules.METHODS: The separation was performed on Kromasil ODS C18 column(150 mm?4.6 mm,5 ?m) with the mobile phase consisted of acetonitrile-water(35∶65).The temperature of the drift tube was 40 ℃ and the flow rate of gas(air) was 2.1 L?min-1.RESULTS: The linear range of astragaloside Ⅳ was 1.32~7.95 ?g(r=0.998 9) and the average recovery was 97.84(RSD=1.01%,n=9).CONCLUSION: The method is simple,accurate and reproducible,and it can be used for the quality control of Xin'an granules.
2.The Protective Effect of Xinan Granule on Viral Myocarditis in Mice

Objective To investigate the protective effect of Xinan granule on viral myocarditis in mice. Methods Viral myocarditis model was made by intraperitoneal inoculation with Coxsackie virus B3 in BALB/C mice. Mice were randomly devided into six groups:the normal control group, the model control group, the Xinan granule10、20、40 g/(kg?d) groups and the Binduzuo 0.1 g/(kg?d) group.After CVB3 infection 2 h, one set of mice were given drugs for 7 d.Blood and heart samples were obtained on the 8th day after CVB3 infection.The myocardial histopathologic changes were observed. The serum cardiac enzyme (LDH) were measured.The others were given drugs for 2 d before CVB3 infection, and for 7ds after CVB3 infection. The survival time were recorded for 20 ds. Results The myocardial histopathologic damage, LDH, cardiac index, punctate hemorrhage on heart surface in Xinan group reduced more significantly than that of the model control group. The survival time in Xinan group were longer than that of the model control group. Conclusion Xinan granule may have the preventional and therapeutic effect on viral myocarditis in mice.
3.Clinical observation of the treatment based on Peitushengjin theory in patients with AECOPD with Ⅱ respiratory failure

Objective to observe the effect of lung and spleen mixture on lung spleen deficiency AECOPD with Ⅱ respiratory failure of TCM symptoms integral,nutrition,blood gas and related indicators.Methods in accordance with AECOPD with Ⅱ type of respiratory failure need to use the noninvasive or have a respirator assisted ventilation,and conform to the type of TCM for patients with spleen-deficiency type,40 cases were divided into two groups,control group to the conventional western medicine therapy and Chinese medicine group on the basis of western medicine treatment for reasons of Yifeijianpi mixture,oral or tube feeding,a week for a period of treatment,observation of patients before and after treatment of TCM symptoms and physical signs integral,APACHE Ⅱ score,serum albumin,pre-albumin,neutrophil percentage,CRP and blood gas analysis and related indicators was conducted.Results The results show that the TCM group can improve the patients' TCM symptoms and physical signs integral,there was significant difference between the two groups after treatment (P< 0.05);compare the serum albumin,pre-albumin,transferrin,neutrophil percentage,CRP and arterial blood gas between two groups of patients after treatment,Chinese medicine group is superior to the control group,with statistical significance (P< 0.05);And compared with control group,the APACHE Ⅱ improvement situation of Chinese medicine group were superior to control group after the treatment after a week,with statistical significance (P< 0.05).Conclusion on the basis of using conventional western medicine treatment,treatment based on the theory of ridging raw gold formulae of Yifeijianpi mixture has good effect in the treatment of AECOPD combined type Ⅱ respiratory failure,and can improve the nutritional status of patients and the indicators of arterial blood gas analysis.
4.Pharmacodynamic Study of Yunkang Capsule

Objective To investigate the pharmacodynamic action of Yunkang Capsule(YKC).Methods The animals were divided into YKC groups(at high-,moderate-and low-dosage respectively),diphenidol control group,model control group and blank control group.The action of YKC on vertigo and thrombosis,hemorrheologic indexes,piamatral microcirculation and free radicals of the animal models were observed.Results YKC could significantly prolong the latency of vertigo,reduce wet weight of thrombus,decrease blood viscosity,plasma viscosity and hematocrit,promote the dilation of piamatral micro-vessels,increase the amount of interweave microdot,increase plasma SOD activity and decline plasma MDA content of animal models.Conclusion YKC has actions of counteracting vertigo and thrombosis,improving hemorrheology and microcirculation,and clearing away free radicals.
5.Cloning and bioinformatics analysis of Giardia lamblia C2 strain SENP gene and prokaryotic expression of SENP catalytic domain

SUMOylation is a post‐translational modification involved in various cellular processes .SUMO‐specific protease (SENP) regulates SUMOylation by removing SUMO from conjugated substrates (deSUMOylation) and promoting maturation of SUMO precursor .In order to express Giardia lambia (C2 strain) SENP catalytic domain in E .coli ,the full‐length open reading frame of SENP was amplified by PCR from Giardia lamblia genome DNA .The PCR product about 1 620 bp was cloned into cloning vector pGM‐T .Sequencing result showed the sequence of SENP in C2 strain was identical with that in Gi‐ardia WB strain .Bioinformatics analysis showed that SENP protein possessed a 372 aa discontinuous ULP catalytic domain at C‐terminal .The catalytic domain of SENP was cloned into prokaryotic expression vector pET‐28a(+ ) .The recombinant vector pET‐28a(+ )‐SENPc was transformed into E .coli Rosetta(DE3) ,then the recombinant SENPc protein was expressed by IPTG induction .SDS‐PAGE and Western blot using anti‐His Tag antibody showed that the expression product of SENPc was a fusion protein with a molecular weight of 43 kD .The successful prokaryotic expression and bioinformatics analysis of Giardia lamblia SENP protein provide basis for further functional study of SENP .
6.Effect of Cordyceps Sinensis from Different Origins on Immune Response in Mice

Objective To investigate the effect of cordyceps sinensis from different origins on immune response in mice. Methods Cordyceps sinensis from two origins were prepared into powder, and then the mice were divided into high, middle and low dose(0.4,0.2,0.1 g.kg-1)groups, respectively.In addition, purified water was given as the normal control group.Effects of cordyceps from two different origins were observed by detecting spleen lymphocyte proliferation induced by ConA, delayed type hypersensitivity ( DTH) in mice induced by sheep red blood cells ( SRBC ) , the number of antibody-producing cells, carbon clearance and peritoneal macrophages Swallow fluorescent microspheres, as well as the activity of NK cells. Results The ability of spleen lymphocyte proliferation induced by ConA, carbon clearance and peritoneal macrophages Swallow fluorescent microspheres, and the activity of NK cells were significantly enhanced in the middle and high dose group of two different origins cordyceps, compared with normal control group (P<0.05).Additionally, the number of antibody-producing cells was obviously increased in medium dose group of both origins cordyceps and decreased in the high dose group (P<0.05).The middle and high dose Qinghai cordyceps significantly improved DTH in mice, while Tibet cordyceps sinensis had no obvious effect, and there was significant difference (P<0.05) between the high dose group of Qinghai and three dose groups of Tibet Cordyceps sinensis.In addition, levels of serum hemolysin in mice were significantly increased in the middle and high dose group of Qinghai and high dose group of Tibet Cordyceps sinensis (P<0.05), and the differences of corresponding medium and high doses of two origins were significant ( P<0.05) . Conclusion Cordyceps sinensis of both different regions significantly improved the immune response of mice.However, the efficacy between the two origins was roughly equivalent and had no significant difference.
7.Relationship between a novel polymorphism of C5L2 gene C698T and type 2 diabetes in Uygur population of Xinjiang region

Objective To investigate the relationship between a novel polymorphism of C5L2 gene and type 2 diabetes mellitus (T2DM) in Uygur population from Xinjiang region.Methods A novel single nucleotide polymorphism(SNP),698C>T(P233L) was found using a polymerase chain reaction direct-sequencing method.C5L2 gene 698C > T variant from 252 patients with T2DM and 747 healthy control subjects was detected by polymerase chain reaction and restriction fragment length polymorphism.Result Heterozygote carriers of the 698CT genotype were more frequent among T2DM patients than that among controls (0.107 vs 0.036,x2 =18.576,P<0.01) in the Uygur population. After adjustment of confounding factors such as sex,age,smoking,alcohol consumption,and hypertension,as well as serum levels of triglyceride,total cholesterol,low-density lipoproteincholesterol,and high-density lipoprotein-cholesterol,the difference remained significant ( P<0.01,OR =3.373,95% CI 1.736-6.553 ).Conclusion The CT genotype of the C5L2 gene might be a risk factor of T2DM in Uygur nationality population in Xinjiang.

Objective To investigate the prevalence and associated risk factors of type 2 diabetes mellitus in populations of Uigurs and Kazaks in Xinjiang.Methods Four-stage selected random samples aged 35 years and over were used to analyze the prevalence and associated risk factors of type 2 diabetes mellitus in populations of Uigurs and Kazaks from 6 localities(Urumqi,Kelamayi,Fukang,the Turfan Basin locality,Hetian locality,Yili Hazakh autonomous prefecture)in Xinjiang according to different nationality,age,and sex.The data were collected according to questionnaire,and physical examination and laboratory test were performed.Results A total of 8 539 Uigurs and Kazaks adults were surveyed.The prevalences of type 2 diabetes mellitus were 5.01%and 3.16%in Uigurs and Kazaks.The prevalence in male was higher than that in female(5.22%vs3.89%,P<0.01).The prevalences of type 2 diabetes mellitus were2.45%,4.92%,5.94%,7.49%,and 5.64%in 35-44,45-54,55-64,65-74,>75 yeats of age groups,respectively.Age,gender,nationality,systolic blood pressure,body weight,triglyceride,and total cholesterol were the risk factors for type2 diabetes mellitus,with the ORs being0.742,1.025,0.673,1.050,1.005,1.517,1.126,respectively.Conclusion It is important to culture healthy life style,enhance the control of hypertension and hyperlipidemia in Uigurs and Kazaks patients for preventing type 2 diabetes mellitus in Xinjiang.
9.(99)Tc(m)N-NOET dual-phase SPECT in differential diagnosis of benign and malignant lung tumors.

OBJECTIVETo investigate the value of (99)Tc(m)N-NOET dual-phase SPECT in differential diagnosis of benign and malignant lung tumors.
METHODSCT scan, early (20 to 30 min) and delayed (2 h) imaging of NOET SPECT were performed on 61 patients suspected of lung lesions before operation. The results were compared with the pathological findings. All cases were not treated with radiotherapy, chemotherapy or surgery before checks. Moreover, all patients had pathological diagnosis. To determine the value in differential diagnosis of tumors by analyzing the tumor uptake and excretion of (99)Tc(m)N-NOET, and the results were compared with that of CT.
RESULTSThe value of early T/N ratio (ER) in the malignant (G1) and benign (G2) groups was 1.25 ± 0.15 and 1.09 ± 0.11 (P < 0.001), respectively, and delayed T/N ratio (DR) was 1.40 ± 0.17 and 1.18 ± 0.21 (P < 0.001). The retention index (RI) of groups G1 was (12.22 ± 6.38)% and group G2 was (8.3 ± 10.91)%, with a non-significant difference between them (P > 0.05). The ER, DR and RI of NOET SPECT in the malignant patients were not significantly correlated with TNM staging, pathological types, tumor diameter, cavity in the lung tumor mass, history of smoking, tumor size and patient gender (P > 0.05). The sensitivity of NOET dual-phase SPECT and CT in the differential diagnosis of benign and malignant lung tumors was 94.1% vs. 90.2%, specificity was 70.0% vs. 80.0% , positive predictive value (PPV) was 94.1% vs. 95.8%, negative predictive value (NPV) was 70.0% vs. 61.5 %, and accuracy was 90.2%. vs. 88.5% (P > 0.05 for all).
CONCLUSIONS(99)Tc(m)N- NOET dual-phase SPECT could be used in differential diagnosis of benign and malignant lung tumors, with no significant differences compared with the efficacy of CT imaging. The semiquantitative indexes (ER, DR and RI) of NOET SPECT can also be used in differential diagnosis of benign and malignant lung tumors, and are not significantly correlated with TNM staging, pathological types, tumor diameter, cavity of the lung tumor mass, history of smoking, tumor size and patient gender.

10.The Influence of Pediatric Nutrition Support Team on Hospitalized Pediatric Patients Receiving Parenteral Nutrition

Purpose:
Malnutrition is a common problem in hospitalized patients that can increase the risk of complications, including infections and length of hospitalization. Appropriate nutritional support is important, particularly in pediatric patients, because growth and development are closely related to the nutritional supply. This study examined the status of nutritional support for pediatric patients in general wards to determine if interventions of the pediatric nutrition support team (pNST) contribute to appropriate nutritional support and help improve their nutritional status.
Methods:
Between July 2016 and June 2017, all pediatric inpatients who received parenteral nutrition support at the Seoul National University Bundang Hospital were recruited and divided into the NST group and non-NST group according to the activities of pNST. The nutritional status was assessed in all subjects, and the calories and proteins delivered through nutritional support in each group were calculated and then compared with the recommended requirements in pediatric patients.
Results:
The number of patients recruited was 174: 100 (57.5%) in the NST group and 74 (42.5%) in the non-NST group. Significantly more calories and protein were supplied in the NST group than the non-NST group in hospitalized children aged between four and 17 years (P<0.05). In the non-NST group, the amounts of calories supplied were insufficient compared to the recommended requirements. The proportion of patients supplied with the appropriate number of calories was 60.0% in the NST group and 20.0% in the non-NST group (P<0.001), and the proportion of cases supplied with the appropriate amount of protein was 87.0% in the NST group and 62.2% in the non-NST group (P<0.001).
Conclusion
Interventions of the pediatric nutrition support team contributed to the sufficient supply of calories and protein and the improvement of clinical outcomes in hospitalized children on parenteral nutrition therapy.
