1.Expression of STAT6 in human nasal polyps and the relation between STAT6 and eosinophil infiltration

Objective:To study the distribution and expression of STAT6 and to examine the suggested roles of STAT6 in the pathogenesis of eosinophil infiltration in nasal polyps and to evaluate the role of STAT6 in the pathogenesis of nasal polyps.Method:All selected cases met the enrollment criteria.Thirty samples of nasal polyps were obtained from patients undergoing nasal polypectomy, and 10 samples of inferior turbinate tissues were from patients undergoing nasal septal reconstruction.STAT6 in nasal polyp tissues from 30 nasal polyposis patients and 10 samples of inferior turbinate tissues were detected with immunohistochemistry(SP) method.SPSS13.0 system was used to perform the statistical analysis.Result:The positive expression of STAT6 was significantly higher in epithelium of nasal polyps than that of the control. The number of eosinophils was significantly higher in epithelium of nasal polyps than that of the control. The difference between these two groups was statitically significant(P<0.05). STAT6 positive cell were localized on epithelium, gland cells and on inflammatory cell of nasal polyps. STAT6 expression was positively correlated with the recruitment of eosinophils in nasal polyps.Conclusion:The high expression of STAT6 protein and the suggested roles of STAT6 in the recruitment of eosinophils in nasal polyps may contribute to the initiation and progression of nasal polyps.

OBJECTIVE:
To study the clinical significance of the expression of p53, p63 and p73 protein and the correlation of p53 and p63 in nasal and paranasal sinus carcinoma (NPSC).
METHOD:
The immunohistochemical technique was used to detect the expression of p53, p63 and p73 in 67 cases of NPSC, para-cancer tissues and non-cancer tissues.
RESULT:
The positive rate of p53 and p63 protein in NPSC was significantly higher than that in park cancer tissues and non-cancer tissues (P<0.01), and there were a positive correlation between the expression of p53 and p63 protein in NPSC (P<0.01). But the expression of p73 had no significant difference among NPSC, para-cancer tissues and non-cancer tissues (P>0.05).
CONCLUSION
There are positive correlation between p53 and p63 protein in NPSC, and they may play an important role in the tumorgenesis of NPSC. The p73 may not be associated with tumorgenesis of NPSC.

4.Expression and significance of STAT3 and VEGF with MVD in the nasal polyps.

OBJECTIVE:
To investigate microvessel density (MVD) and expressions of phosphor-STAT3 and vascular endothelial growth factor (VEGF) in nasal polyps, to explore their pathogenetic correlations.
METHOD:
All selected cases were measured up the standard of enrollment. Thirty samples of nasal polyps were obtained from patients undergoing nasal polypectomy, and 10 samples of inferior turbinate tissues were from patients undergoing nasal septal construction. p-STAT3,VEGF and MVD in nasal polyp tissues from 30 nasal polyposis patients and 10 samples of inferior turbinate tissues were detected with immunohistochemistry (sp) method.
RESULT:
In nasal polyp tissues, The positive rates of p-STAT3 and VEGF expression were 60% and 70% respectively, all were significantly higher in nasal polyps than in control inferior turbinates (P < 0.01). MVD was significantly higher in nasal polyps than in control inferior turbinates (22.78 +/- 7.89 vs 7.12 +/- 2.84, P < 0.01). p-STAT3 expression was positively correlated with VEGF expression and MVD.
CONCLUSION
p-STAT3, VEGF and MVD were overexpressed in the tissues of nasal polyps, and p-STAT3 expression was positively correlated with VEGF expression and MVD; p-STAT3 was positively correlated with the neovascularization of nasal polyps, The inhibition of STAT3 pathway may inhibit the neovascularization, and then inhibit the formation and recurrence of the nasal polyps.

6.Low- and moderate-volume intracerebral hemorrhage at acute stage treated by Qufeng Tongxuan method: a multicenter, randomized, controlled study

Objective:To verify the clinical efficacy and safety of Qufeng Tongxuan method in treating low- and moderate-volume intracerebral hemorrhage at acute stage.Methods:A prospective, multicenter, randomized, double-blind, placebo-controlled study was performed; patients with low- and moderate-volume intracerebral hemorrhage at the basal ganglia and/or thalamus accepted treatment in 16 hospitals from September 2019 to April 2022 were enrolled. These patients were divided into experimental and control groups with a block randomized method by SAS software. Patients in control group were given conventional western medicine treatment; those in experimental group accepted Qufeng Tongxuan method (sequential therapy of Shexiang Huayu Xingnao granules and Zhilong Huoxue Tongyu granules) besides conventional western medicine treatment. NIHSS was used to assess neurological function before treatment and on 7 th, 14 th, 30 th, and 90 th d of treatment. Prognoses of these patients were assessed by modified Rankin scale (mRS) before treatment and on 180 th d of treatment. Brain CT was performed before treatment and on 7 th and 14 th d of treatment to calculate the hematoma volume. Before treatment and on 14 th d of treatment, changes of coagulation function, liver and kidney functions of the 2 groups were compared. Adverse reactions during treatment in the 2 groups were recorded. Results:No significant differences in NIHSS scores were noted between the 2 groups before treatment, on 7 th, 14 th, and 30 th d of treatment ( P>0.05); NIHSS scores in experimental group on 90 th d of treatment were signficantly lower than those in control group ( P<0.05); NIHSS scores in experimental group decreased gradually before treatment and on 7 th, 14 th, 30 th and 90 th d of treatment, with statistical significances ( P<0.05). No significant differences in mRS scores were noted between the 2 groups before treatment ( P>0.05); mRS scores in experimental group on 180 th d of treatment were signfciantly lower than those in control group ( P<0.05). No significant difference in hematoma volume was noted between the 2 groups before treatment and on 7 th and 14 th d of treatment ( P>0.05); both groups had gradually decreased hematoma volumes before treatment and on 7 th and 14 th d of treatment, respectively, with significant differences ( P<0.05); the volume difference of hematoma between 14 th d of treatment and before treatment in experimental group (6.42[4.10, 11.73]) was significantly higher than that in control group (4.00[1.25, 10.58], P<0.05). No significant differences in liver and kidney function indexes or coagulation function indexes were noted between the 2 groups before treatment and on 14 th d of treatment ( P>0.05). Adverse reaction incidence was 9.52% ( n=12) in experimental group and 10.34% ( n=12) in control group, without statistical difference ( P>0.05). Conclusion:Under premise of conventional western medicine treatment, Qufeng Tongxuan method can promote hematoma absorption and improve neurological deficit symptoms in low- and moderate-volume intracerebral hemorrhage at acute stage, without obvious adverse reactions.

Esophageal squamous-cell carcinoma (ESCC) is one of the most lethal malignancies in the world and occurs at particularly higher frequency in China. While several genome-wide association studies (GWAS) of germline variants and whole-genome or whole-exome sequencing studies of somatic mutations in ESCC have been published, there is no comprehensive database publically available for this cancer. Here, we developed the Chinese Cancer Genomic Database-Esophageal Squamous Cell Carcinoma (CCGD-ESCC) database, which contains the associations of 69,593 single nucleotide polymorphisms (SNPs) with ESCC risk in 2022 cases and 2039 controls, survival time of 1006 ESCC patients (survival GWAS) and gene expression (expression quantitative trait loci, eQTL) in 94 ESCC patients. Moreover, this database also provides the associations between 8833 somatic mutations and survival time in 675 ESCC patients. Our user-friendly database is a resource useful for biologists and oncologists not only in identifying the associations of genetic variants or somatic mutations with the development and progression of ESCC but also in studying the underlying mechanisms for tumorigenesis of the cancer. Based on domestic clinical practice and experimental data and referred to international experience and viewpoints, the expert group forms a consensus on the clinical application of 177Lu-PSMA radio-ligand therapy in prostate cancer to guide clinical practice.
