1.Pharmacokinetics of sinomenine in rabbits

Objective:To establish a HPLC method for measurement of serum concentration of sinomemine in rabbits and explore it’s pharmacokinetics. Methods:6 rabbits were intragastricly administrated sinomenine with the dosage of 45mg/kg,2 ml blood were collected at 0.25h、0.5h、1.0h、1.5h、2.0h、3.0h、4.0h、6.0h、8.0h、10.0h and 12.0h after oral administration.concentrations of drug were determined by HPLC,datas were calculated with 3P97 software and pharmacokinetics was explored. Results:T1/2(Ka) was(0.31?0.26)h,T1/2(Ke) was(3.15?0.82)h,Tmax was(1.05?0.53)h,Cmax was(15.09?4.15)?g/ml,AUC0→∞ was(84.27?13.02)(?g?h)/L,AUC0→T was(78.29?13.46)(?g?h)/L. Conclusion:absorption and elimination of Sinomenine were quickly and Tmax is short,concentration of drug is high in rabbits after intragastric administration.
2.Expression and Significance of Duodenal Eosinophils in Patients with Functional Dyspepsia

Functional dyspepsia( FD)is a group of clinical syndromes presented as upper abdominal pain,epigastric burning sensation,postprandial fullness,satiety and other symptoms,but lack of organic disease causing these symptoms. Eosinophils( EOS)in gastrointestinal tract play an important role in preventing pathogen invasion and maintaining intestinal epithelial homeostasis. In recent years,many studies have shown a significant increase of duodenal EOS in patients with FD. This article reviewed the expression and significance of duodenal EOS in patients with FD.

OBJECTIVE:To provide feasible basis for establishment of new model of pharmaceutical care in community.METHODS:Pharmaceutical care in time sequence of before or during visiting hospital,after medication were surveyed and analyzed,according to the pharmacy practice of the community medical service stations which belonged to our hospital.RESULTS & CONCLUSIONS:New model was established through pharmaceutical propaganda,medication counseling,consummate patients' information,monitoring of medication process,communication of pharmacists and patients,data analysis,etc.The established model of pharmaceutical care in community is feasible and of high performance.
4.The effect of yemazhui on concentration of levofloxacin in rinse solution of mice tracheae

Objective:To study the effect of yemazhui on concentration of levofloxacin in rinse solution of mice tracheae. Methods:50 mice were randomly divided into blank group、anwhlor group、yemazhui large-dose group、yemazhui middle-dose group, yemazhui low-dose group, each group had 10 mice. After administration of liquor natrii chloridi isotonicus (NS) 30 ml/10kg, amchlor1.00g/kg、yemazhui large dose 64.45g (curde drug)/kg、yemazhui middle dose 32.73g (curde drug)/kg, yemazhui low dose16.36g(curde drug)/kg, mice were intraperitoneal injected levofloxaein at dosage of 0.07g/kg 2hours after administration. The concentration of levofloxacin in excretion of mice tracheae was detected by HPLC. Results:Comparion with blank group, concentration of levofloxacin in excretion of mice tracheae of yemazhui large-dose group、yemazhui middle-dose group、yemazhui low-dose group was 4.58 times、3.18 times、1.86 times the amout of that. Conclusion:Yemazhui can enhance concentration of lwvofloxacin in rinse solution of mice tracheae.
5.Hypofractionated radiotherapy and tumor immunity—new concepts and new combination

Modern immunology has established that tumor immune escape is associated with hidden or missing tumor-specific antigens and tumor-associated antigens,as well as immune suppressors that are released from tumor cells to inhibit the immune cytotoxicity and antigen-presenting cells (APCs).The changes in tumor microenvironment have an impact on tumor immunity and treatment outcomes.The immune effects finally depend on activation and inhibition of T cell receptors and other co-regulated receptors (CD28,CD80/CD86,and CTLA-4) in spite of the existence of APCs and cytotoxic T lymphocytes in tumor microenvironment.Recent studies have revealed that radiotherapy induced not only DNA damage but also immunogenesis in tumor cells.Both conventionally fractionated radiotherapy and hypofractionated radiotherapy can induce immunogenesis in tumor cells.Immunogenic regulation makes many tumor antigens expressed in cells exposed to irradiation,which induces immune recognition and cytotoxicity;cell content (DNA,HMGB1,etc.) released from dead immunogenic cells can trigger immune effects and in situ tumor vaccination,which further induce an abscopal effect of radiotherapy.A lot of anti-tumor immunotherapy fails to achieve satisfactory treatment outcomes.Therefore,how to combine radiotherapy,especially stereotactic body radiotherapy,with anti-tumor immunotherapy has recently become a new challenge for researchers.

Objective To study the effects of the integrity of the tensor tympani muscle on the postoperative hearing recovery and the morphology of tympanic membrane,to provide the reference for the handling of the tensor tympani muscle during the operation.Methods A total of 145 cases of chronic tympanitis were grouped by the integ-rity of the tensor tympani muscle and the implanted types of biological auditory ossicles,the clinical data were ana-lyzed retrospectively.There were 80 cases in the tensor tympani muscle intact group,including 45 cases of PORP group and 35 TORP group.The tensor tympani muscle rupture group of 65 cases consisted of 34 cases of PORP group,and 31 cases of TORP group.The postoperative recovery conditions of tympanic membrane morphology, hearing threshold Ac and air-bone gap(ABG)between the tensor tympani muscle intact group and the tensor tym-pani muscle rupture group were compared 1 month or 3 months,and 6 months after the operation.ResuIts One month after the operation,there was no significant difference in tympanic membrane morphology between the two groups.Three months after the operation,the tensor tympani muscle intact group had a higher ratio about the loca-tion and shape of tympanic membrane ,closer to the normal than the tensor tympani muscle rupture group in which the ratio in the intact group was 81.25% (65/80),while in the rupture group it was 52.31% (34/65 ).After 6 months,with the operation of PORP,the tensor tympani muscle intact group of AC value was 27.48±10.02 dB HL, and ABG value was 13.57±6.36 dB,while the rupture group of AC value was 32.36±9.34 dB HL,and ABG value was 25.73±7.44 dB.With the operation of TORP,the tensor tympani muscle intact group of AC value was 28.76± 7.14 dB HL,and ABG value was 21.02±5.48 dB,while the rupture group of AC value was 39.93 ±5.12 dB HL, and ABG value was 31.41±6.25 dB.The postoperative recovery condition of the tensor tympani muscle intact group was better than those of in the rupture group.ConcIusion The tensor tympani muscle can maintain the morphology and location of the postoperative tympanic membrane.The integrity of the tensor tympani muscle may has a positive effect on the improvement of the postoperative hearing.
8.Construction a shuttle vector containing AaCPR100A epidermal protein gene of Aedes aegypti with two spore-producing-dependent promoters in Bacillus thuringiensis by in-fusion cloning

Abstract: Objective To construct a shuttle vector pHT315-AaCPR100A with two spore-producing-dependent promoters and the target gene AaCPR100A in Escherichia coli-Bacillus thuringiensis. Methods The forward promoter of Cry3A, named Pro-1 (+), was amplified by PCR using pSVP27A plasmid as the template, and the target gene AaCPR100A was amplified using Aedes aegypti RNA reverse conversion cDNA as the template. The plasmid pHT315 was linearized by digestion with Hind Ⅲ and Sal Ⅰ. The forward promoter and the target gene were inserted into the linearized vector pHT315 successively by in-fusion cloning according to the transcription direction. The synthesized plasmid containing the Cry3A reverse promoter sequence was used as the template, and the Pro-1 (-) reverse promoter was amplified by PCR. The intermediate vector containing the forward promoter and the target gene was linearized by EcoR I restriction enzyme, and the reverse promoter was inserted downstream of the target gene by in-fusion cloning in the direction of transcription. Results By agarose gel electrophoresis, the forward promoter, target gene AaCPR100A and reverse promoter bands were clear and of good quality, which could be used for in-fusion cloning experiments. The two spore-producing-dependent promoters and target gene fragments were connected by In-fusion cloning. The recombinant vector pHT315-AaCPR100A was verified by PCR. The forward promoter, target gene fragment and reverse promoter were successfully amplified in the recombinant vector. Nucleotide sequencing verified that the sequencing results of the bidirectional promoter sequence and the target gene sequence were basically consistent with the sequence alignment results, which met the requirements of the construction of vector elements and proved that the recombinant vector was successfully constructed. Conclusions Based on the above results, this study proves that the recombinant shuttle vector with two spore-producing-dependent promoters can be successfully constructed by in-fusion cloning technology, laying the foundation for the construction of engineered Bacillus thuringiensis expressing dsRNA of AaCPR100A.
9.Alterations and Significance of Duodenal Eosinophils and Chromogranin A-positive Cells in Patients With Functional Dyspepsia

Background:Epidemiological studies suggest considerable overlap between functional dyspepsia (FD) and irritable bowel syndrome (IBS).Increased duodenal eosinophil counts have been observed in patients with FD,and the density of chromogranin A (CgA)-positive cells was reduced in duodenum in patients with IBS.Aims:To investigate the alterations and significance of duodenal eosinophils and CgA-positive cells in patients with FD and FD-diarrhea-predominant IBS (IBS-D) overlap.Methods:Outpatients with FD and FD-IBS-D overlap who fulfilled Rome Ⅲ criteria from Aug.2014 to Mar.2015 at the First Affiliated Hospital of Fujian Medical University were enrolled.Biopsy specimens were collected from the duodenal bulb for HE staining and CgA immunohistochemical staining.Eosinophils and CgA-positive cells were counted,and their associations with clinical symptoms were analyzed.Results:Twenty-eight FD patients and twelve FD-IBS-D overlap patients were enrolled in this study.Duodenal bulb eosinophil counts were significantly higher in FD-IBS-D overlap patients than in FD patients [(43.83 ± 7.34)/5HPF vs.(22.90 ± 8.93)/5HPF,P < 0.05],while the density of duodenal bulb CgA-positive cells were significantly lower in FD-IBS-D overlap patients than in FD patients [(13.03 ±5.87)/HPF vs.(19.85 ± 9.18)/HPF,P < 0.05].Spearman correlation coefficient analysis revealed that duodenal eosinophil counts were positively correlated with lower gastrointestinal symptoms such as abdominal pain/discomfort and diarrhea,whereas no correlations were found between density of duodenal CgA-positive cells and both upper and lower gastrointestinal symptoms.Conclusions:Duodenal bulb eosinophil counts are increased and CgA-positive cells are reduced in FD-IBS-D overlap patients than in non-overlap FD patients,which may implicate the difference in clinical symptoms between these two functional gastrointestinal disorders.
10.Spectrum and case fatality of inpatients with malignant tumors from 1995 to 2014 in Shenzhen city

Objective To analyze the epidemiological features,spectrum and case fatality of malignant tumor patients in Shenzhen city,to provide evidence for the development of prevention and treatment strategies on malignant tumor in Shenzhen.Methods All the hospitalized malignant tumor patients including deaths,were monitored from 1995 to 2014 in Shenzhen,and data was analyzed by SPSS 20.0 software.Results There were 160 988 inpatients of malignant tumors between 1995 and 2014 in Shenzhen.The top three hospitalized tumors were lung (13.64%),liver (11.13%) and breast (7.86%) cancers.Numbers of the malignant tumor inpatients had been rapidly increasing during the past 20 years,12.3 times in 2014 higher than in 1995.The total number of deaths due to malignant tumors was 19 460.Deaths of the top three malignant tumors were lung (24.40%),liver (19.84%) and colorectal (8.63%) cancers and the number of deaths was increasing,12.5 times higher in 2014 than in 1995.The overall case fatality rate was 12.09%.The annual percent change (APC) of malignant tumors case fatality rate was 9.7%(95%CI:2.0%-18.0%),during 1995-2003,with an increasing trend (t=2.72,P<0.05).The APC of case fatality rate during 2003-2014 was-3.4%(95%CI:-7.6%-1.1%),but the decreasing trend (t=-1.63,P>0.05) was not statistically significant.The top three major malignant tumors related to case fatality rate were lung cancer (21.62%),liver cancer (21.39%),and esophageal cancer (16.50%).The case fatality rates of leukemia and liver cancer had decreased during the past 20 years.The case fatality rates of cancers in lung,esophagus,stomach,breast,colorectal and nasopharyngeal,had all increased.The number of male patients was significantly exceeding the females (x2=41.691,P<0.01),with sex ratio as 1.65:1.From age 35 and on,the number of deaths due to malignant tumors increased significantly,with the peak after 60 years of age.Conclusions The number of malignant tumor inpatients had an annual increase as well as the case fatality rate.Cancers in lung,liver appeared the leading causes of death among the malignant tumor patients,with elderly in particular.Strategies related to the prevention and treatment of cancers in lung,liver should be strengthened.
