1. Advantages of robot assisted percutaneous biopsy in the diagnosis of spinal lesions

BACKGROUND: Robot assisted orthopedic surgery has been gradually popularized in clinical practice, and its application fields include spine, joint, trauma and sports medicine, which are precise and minimally invasive. However, there are few reports on the application of robot assisted puncture biopsy in the diagnosis and treatment of spinal lesions. OBJECTIVE: To explore the clinical value of percutaneous biopsy assisted by orthopedic surgical robot in the diagnosis of spinal lesions. METHODS: The clinical data of 38 patients with spinal lesions treated in Sichuan Provincial People’s Hospital from November 2018 to January 2020 were analyzed retrospectively. All patients underwent percutaneous biopsy assisted by TINAVI orthopedic surgery robot. The biopsies were sent to the pathology department for histopathological examination. RESULTS AND CONCLUSION: (1) The successful rate of biopsy was 100% in 38 samples. Of them, 21 cases affected metastases; 4 cases had primary malignant tumors; 5 cases suffered from borderline tumors; 5 cases had benign tumors; and 3 cases experienced non-specific inflammatory reaction. (2) The average operation time was (37.11±5.65) minutes, and no complications occurred in all patients. (3) Robot assisted percutaneous biopsy is a safe, rapid, minimally invasive and accurate method for the diagnosis of spinal lesions.
2.Research and Practice of Vehicle-mounted Mobile CT Based on Emergency Medical Rescue.

OBJECTIVE:
In order to solve the problem of inadequate CT screening ability in emergency medical rescue in remote mountainous areas, high-altitude areas, other public health events and sudden natural disasters, a vehicle-mounted mobile CT suitable for emergency medical rescue is studied.
METHODS:
A vehicle chassis system suitable for long-distance transportation and a cabin system suitable for epidemic prevention and control was designed. A domestic 32-slice CT with small volume, light weight and high stability was selected to design a vehicle-mounted mobile CT suitable for emergency medical rescue.
RESULTS:
The high-performance vehicle-mounted mobile CT can provide rapid support, and provide large-scale screening, emergency medical rescue, a supplement to daily CT scanning in peacetime and wartime.
CONCLUSIONS
The vehicle CT has high stability, fast scanning speed and high social and economic value.
3.Effect of long non-coding RNA Linc01296 on cisplatin resistance of ovarian cancer OVCAR-3 cells and its correlation to clinicopathological characteristics

Objective: To investigate the effect of long non-coding RNA(lncRNA) Linc01296 on cisplatin (DDP) resistance in ovarian cancer cells and the mechanism. Methods: Fifty-three cases of ovarian cancer tissues, 22 cases of borderline ovarian tumor tissues and 17 cases of benign ovarian mass tissues were collected from Department of Gynecology, Sichuan Provincial People's Hospital during October 2017 to October 2018. The differential expression of Linc01296 mRNAin above tissue samples was detected by qPCR, and its relationship with clinicopathological data was analyzed. Human ovarian cancer cell line OVCAR-3 (OVCAR-3 Control) and its DDPresistant cell line (OVCAR-3 DR) were cultured. Lentiviral vectors expressing siRNA that targeting Linc01296 or siRNA control were transfected into OVCAR-3 DR cells respectively, as siLinc01296 group and siControl group. CCK-8 assay was used to detect DDP semi-inhibitory concentration (IC50) and drug resistance index of each group; real-time fluorescence quantitative PCR (qPCR) was used to detect the mRNA expressions of Linc 01296 and tumor stem cell related genes (Oct-4 and Sox-2) in cell lines of OVCAR-3 control, DR, DR siControl and siLinc 01296; and the protein expressions of Oct-4 and Sox-2 in each group were detected by Wb. Results: DDP IC50 and drug resistance index in OVCAR-3 DR group was significantly higher than that in Control group (all P <0.01), and DDP IC50 and drug resistance index in siLinc01296 group was significantly lower than that in DR siControl group (all P <0.01), but significantly higher than that in Control group (all P <0.01). The mRNA and protein expressions of Linc01296, Oct-4 and Sox-2 in DR group were significantly higher than that in Control group (all P <0.01), while those expressions in siLinc01296 group were significantly lower than those in DR siControl group ( P <0.01), and the mRNA and protein expressions of Oct-4 and Sox-2 in siLinc01296 group were higher than that in control group ( P <0.01). The expression of Linc01296 in ovarian cancer tissues was significantly higher than that in borderline ovarian tumor tissues and benign ovarian mass tissues ( P <0.01), which was positively correlated with lymph node metastasis and clinical stage ( P <0.05). Conclusion: Linc01296 can promote the occurrence of DDP resistance via promoting the expression of cancer stem cell markers in ovarian cancer; Linc01296 is highly expressed in ovarian cancer tissues and closely related to disease progression of patients.
4.Discussion on Standardization Management Mode of Medical Equipment Based on Whole Process Quality Control.

This paper introduced the current situation of medical equipment. Based on the current problems such as technology assessment difficulty, weak consciousness on maintenance management, less quality control during clinic use and backward information level, the standardized management based on the whole process quality control was proposed. Combined the whole process quality control system with the construction of quality management information, the management level of the medical equipment was enhanced. The application of standardized management based on the whole process quality control can make the medical equipment work better in the clinic and set a good foundation for the development of hospital.

5. Effect of survivin siRNA on biological behaviour of breast cancer MCF7 cells

Objective: To investigate the expression of survivin in breast cancer cell lines and explore the effect of survivin siRNA on biology behavior of breast cancer cells. Methods: Western blot was performed to detect the expression of survivin in breast cancer cell lines. Eukaryotic expression vector pIRES2-EGFP-Survivin siRNA was constructed and transfected in MCF7 cells with liposome, the efficiency of survivin siRNA was measured by Western blot and RT-PCR. Cell proliferation and apoptosis were detected by CCK8 and cell flow respectively. Cell migration and invasion was measured by transwell assay. Results: Survivin was highly expressed in MCF-7. Green fluorescence was found in MCF-7 cells tranfected with survivin siRNA and control siRNA by inverted fluorescence microscopy, the protein and mRNA level of survivin was significantly lower in cells tranfected with survivin siRNA compared with control group. Compared with control group, interfering the expression of survivin by siRNA significantly decreased the proliferation, migration and invasion of MCF-7 cells, the percentage of apoptosis cells was greatly promoted. Conclusions: Interfering the expression of Survivin can inhibit the cell proliferation, migration and invasion, and promot apoptosis in MCF-7.
6. Netrin-1: The new tumor markers in renal clear cell carcinoma

Objective: To explore the expression of Netrin-1 protein in human renal clear cell carcinoma (RCCC) and the relationships between Netrin-1, pathology and prognosis. Methods: 72 cases of RCCC admitted in our hospital from 2008 June to 2009 June and their adjacent tissues were selected for study. They included 30 cases in stage I-II, 42 cases in stage III-IV; 9 cases in grade I, 9 cases in grade II, 40 cases in grade III and 14 cases in grade IV. All cases were followed up for more than 5 years. Survival analysis lines were made by Kaplan-Meier method and the difference between groups was tested by the Log-rank test. The expression of Netrin-1 protein was detected by immunohistochemistry staining and its clinical significance was analyzed. Results: Renal clear cell carcinoma: 51 cases in high expression of Netrin-1 and 21 cases in low expression, normal tissues: 12 cases in high expression of Netrin-1 and 60 cases in low expression, the difference between the two groups is significant (χ2=42.921, P<0.01). The difference of the expression of Netrin-1in Fuhrman grade and AJCC clinical stage is significant (χ2=8.000, χ2=6.203; P<0.05). The 5-year survival rate in low protein expression group and in high protein expression group was 79% (17/21) and 62% (32/51). The survival curve had different trend, with no significant difference between groups ((χ2=1.360, P=0.245). Conclusions: Netrin-1 protein plays an important role in the development of RCCC. It might be a new specific tumor marker of RCCC, and might become a new target in treatment of RCCC.
7. Reoperation methods and effectiveness of urethral stricture after urethroplasty of hypospadias

Objective: To discuss the reoperation methods of urethral stricture after urethroplasty of hypospadias and their effectiveness. Methods: Between September 2010 and April 2018, 169 patients with urethral stricture after urethroplasty of hypospadias, who underwent ineffective conservative treatments first, were accepted. The age ranged from 1 year and 7 months to 41years with a median age of 5 years and 8 months. The stricture located at the external urethral orifice in 80 cases, internal anastomosis connection in 87 cases, and constructed urethra in 2 cases. The symptoms of urethral stricture occurred at 2 weeks to 52 months after urethroplasty, with a median time of 4.5 months. The patients with external urethral orifice stenosis were treated with urethral meatus augmentation (74 cases) and urethral advancement (6 cases). The patients with internal anastomosis connection stenosis were treated with internal urethrotomy with urethroscopy (10 cases), urethrotomy and one-stage urethroplasty (26 cases), and urethrostomy (51 cases) including 43 cases of two-stage urethroplasty. The patients with constructed urethral stricture were treated with urethrolysis. Results: One hundred and fifty-four patients were followed up 6-86 months with an average of 47 months. The stenosis was relieved in 137 cases, and re-stenosis in 12 cases, urethral fistula in 4 cases, all of which were treated successfully. In addition, 1 case with mild urethral diverticulum did not need to be treated. Conclusion: If it is ineffective for the conservative treatment of urethral stricture after urethroplasty of hypospadias, appropriate surgical treatments could be selected according to the location and length of the stricture, local tissue conditions, complications, and so on.
8. Duckett URETHROPLASTY-URETHROTOMY FOR STAGED HYPOSPADIAS REPAIR

OBJECTIVE: To explore the surgical outcome of Duckett urethroplasty-urethrotomy for staged hypospadias repair. METHODS: Fifty-three patients with hypospadias were treated by 2 stages between August 2013 and September 2014. The age ranged from 10 months to 24 years and 3 months (median, 1 year and 10 months). There were 5 cases of proximal penile type, 2 cases of penoscrotal type, 36 cases of scrotal type, and 10 cases of perineal type. Urethroplasty was performed with tubed transverse preputial island flap only in 27 cases or combined with urethral plate in the other 26 cases, thus a urethrocutaneous fistula was intentionally created; stage II fistula repair was carried out at 1 year after stage I repair. RESULTS: The length of the new urethra ranged from 2 to 8 cm with an average of 3.6 cm. The patients were followed up 5-17 months with an average of 8 months after stage II repair. After stage I repair, urethral fistula was noted at other site in 3 cases, skin necrosis in 1 case, glandular stricture in 2 cases, cicatric curvature in 1 case, and position and morphology of urethral orifice not ideal in 4 cases. After stage II repair, urethral fistula was noted in 2 cases, mild urethral diverticulum in 2 cases, and stricture at temporary repair site in 1 case. HOSE score was 12-16 at 3 months after stage II repair (mean, 14.5). At 3-14 months after stage II repair, the maximum flow rate ranged from 3.9 to 22.7 mL/s with an average of 8.6 mL/s. CONCLUSIONS: Duckett urethroplasty-urethrotomy can be used as staged repair for primary treatment of hypospadias because of high safety, low complication incidence, and satisfactory appearance.
9. Efficacy of percutaneous microwave ablation on low-risk papillary thyroid microcarcinoma and impact on patient's life quality

Objective: To observe the efficacy of percutaneous microwave ablation on low-risk papillary thyroid microcarcinoma (PTMC) and impact on patient's life quality. Methods: Totally 75 pathologically confirmed patients with single low risk PTMC (T1N0M0) were treated with ultrasound-guided percutaneous microwave ablation and followed up at 1, 3, 6, 12 and 24 months respectively after operation. The therapeutic effect and complications were observed, and score the physical component summary (PCS), mental component summary (MCS) and 36-item short form health survey (SF-36) scores were counted. Results: The operation was successfully completed in all 75 patients. Four patients experienced temporary hoarseness during the ablation process and recovered within 2 h, while 15 patients complained of radiation-induced ear root pain, neck pain and other symptoms which immediately relieved after ablation. Twenty-four months after operation, the effective rate of treatment was 100% (75/75), the lesion disappearance rate was 93.33% (70/75), while no local recurrence occurred. The maximum diameter and volume of lesions increased 1 and 3 months after operation but decreased 6, 12 and 24 months after operation (all P<0.05). There was no significant difference of thyroid stimulating hormone (TSH), free triiodothyronine (FT3) nor free thyroxine (FT4) levels among all time points (all P>0.05). PCS scores at different time points before and after operation showed no statistical significant difference (all P>0.05). MCS scores and SF-36 scores of all time points after operation were higher than those before operation (all P<0.05). Conclusion: Percutaneous ablation is safe and effective for treating low risk PTMC, which can improve the patient's life quality.
10.Synergistic Inhibitory Effect of Arsenic Trioxide Combined with Itraconazole on Hedgehog Pathway of Multiple Myeloma NCI-H929 Cells.

OBJECTIVETo investigate the antitumor effect of arsenic trioxide (ATO) combined with itraconazole (ITRA) on human multiple myeloma NCI-H929 cells by synergistically inhibiting Hedgehog (HH) signaling pathway.
METHODSThe inhibitory rate of NCI-H929 cells was assayed by MTT method. Tumor weight, tumor weight inhibition rate, and tumor volume of mouse model with multiple myeloma were examined. The ELISA were appled to detect the M-protein, qPCR and Western blot were used respectively to detect the expression level of Ptch, SMO, Gli and downstream target genes, the survival rate of tumor-bearing mice was analyzed.
RESULTSATO combined with ITRA significantly inhibited NCI-H929 cell proliferation as compared with a single administration. The combination of ATO and ITRA could synergistically inhibit tumor growth and obviously reduced tumor burden, survival time of tumor-bearing mice was significantly prolonged. qPCR and Western blot results confirmed that the ATO combined with ITRA could significantly down-regulated expression of Gli1, leading to significantly decrease of cyclinD1 and BCL-2 expression levels.
CONCLUSIONATO combined with ITRA can more strongly suppress the growth of multiple myeloma NCI-H929 cells, as compared with a single administration. The synergistic effect of ATO and ITRA significantly down-regulates expression of Gli1 in HH signaling pathway, moreover the inhibition of target gene overexpression may be one of two drug mechanisms.
