2.Serum NT-proBNP level and its influencing factors in very old male heart failure patients with preserved ejection fraction

Objective To study the diagnostic value of serum N-terminal probrain natriutetic peptide (NT-proBNP) level and its influencing factors in very old male heart failure (HF) patients with preserved ejection fraction.Methods Two hundred and thirty-six very old male HF patients with preserved ejection fraction admitted to our hospital from February 2008 to July 2016 were divided into HF group (n=92) and HF-free grup (n=144).Their biochemical parameters were tested and analyzed by correlation analysis.Results The serum NT-proBNP level and logNT-proBNP were significantly higher in HF group than in HF-free group (2205.4±2128.4 ng/L vs 457.3± 312.2 ng/L,P=0.000;3.3±0.3 vs 2.5±0.3,P=0.004).Univariate correlation analysis showed that serum NT-proBNP level was positively related with AF,diuretics,LVEF,left atrium diameter,eGFR,creatinine and logcTnI (P<0.05,P<0.01).Multivariate linear regression analysis revealed that AF,LVEF,left atrium diameter,eGFR,and logcTnI were the independent risk factors for NT-proBNP and that NT-proBNP was negatively related with LVEF and eGFR.Conclusion The serum NT-proBNP level in very old male HF patients with preserved ejection fraction can be influenced by a variety of factors.Increasing LVEF and eGFR can reduce their serum NT-proBNP level and improve their outcome.
4.Inhibition of bladder activity by 5-HT1 subtype-selective serotonin receptor agonists in cats with chronic spinal cord injury

Objective To investigate the effect of bladder activity by 5-hydroxytryptamine serotonin receptor agonists with chronic spinal cord injury(SCI). Methods Intact cats and SCI cats were catheterized through the bladder dome for saline-filling cystometry.Dose-response curves for i.v.8-OH-DPAT and GR-46611 were followed in three cases each by WAY-100635.Threshold volume,capacity,residual volume,micturition volume,and arterial pressure were measured. Results Intact cats showed few significant changes in cystometric variable.SCI cats responded to both 8-OH-DPAT and GR-46611 with dose-dependent increases in threshold volume,capacity,and residual volume.Effects of 8-OH-DPAT but not GR-46611 were largely reversed by WAY-100635. Conclusions Both 5-HT_ 1A and 5-HT_ 1B/1D agonists may offer a promising means of reducing bladder hyperactivity and increasing bldder capacity in patients with chronic SCI.
5.Establishment of migraine rheumatism stasis syndrome animal model.

OBJECTIVETo establish the migraine rheumatism stasis syndrome animal model.
METHODThe rat migraine rheumatism stasis syndrome animal model was established through rheumatism stimulation with manual climate box, 5-HT reduction caused by reserpine and local cerebral vasospasm. General vital signs (activity, weight, eye gum, hair, feeding, excrement), head scratch frequency and image collection were observed to analyze the changes in biological signs of stasis syndrome (tongue image RGB), thrombin and serotonin of model rats.
RESULTThe reserpine group and the reserpine plus rheumatism model group showed significant reduction in blood coagulation time, pain threshold and 5-HT content in blood and brain (P < 0.01); the reserpine plus rheumatism model group showed an increase in eye gum and decreases in activity, feeding, with thin sloppy stool. According to the tough RGB values, the control group showed light red toughs, the reserpine group showed dark purple toughs, the reserpine plus rheumatism model group showed gray toughs, with notable differences in tough RGB values in all three group.
CONCLUSIONThe rheumatism stimulation with manual climate box, 5-HT reduction caused by reserpine and local cerebral vasospasm can be used to induce the migraine rheumatism stasis syndrome animal model, but its modeling assessment method and process shall be further improved.

7.Application of 3-dimensional CT reconstruction in the diagnosis of posterior urethral strictures or ankylurethria

Objective To study the diagnostic value of 3-dimensional CT reconstruction in posterior urethral strictures or ankylurethria.Methods Thirty patients with strictures or ankylurethria of posterior urethra caused by pelvic fracture underwent helical CT scan and 3-dimensional reconstruction of the urethral canal as well as radiographic urethrography before and post open urethral reconstruction to observe the urethral anatomy,the length and position of the urethral strictures,the depth of periurethral scar.Results The mean stricture or ankylurethria length measured by radiographic urethrography was 4.0 cm (range from 1.0 cm to 7.0 cm),and the correlation coefficient of stricture or ankylurethria length was 0.92,21 (70%) patients were diagnosed accurately by radiographic urethrography.The mean stricture or ankylurethria length measured by 3-dimensional CT reconstruction was 4.3 cm (range from 1.2 cm to 7.6 cm),and the correlation coefficient of stricture or ankylurethria length was 0.96,there were 28(93%) patients diagnosed accurately by 3-dimensional CT reconstruction.The mean stricture or ankylurethria length measured by open urethral reconstruction was 4.2 cm (range from 1.5 cm to 7.5 cm).Five patients with urethrorectal fistula were also diagnosed accurately by 3-dimensional CT reconstruction rather than by radiographic urethrography.Conclusions 3-dimensional CT reconstruction of the urethral canal can accurately evaluate the urethral anatomy,the length and position of the urethral strictures,as well as the depth of periurethral scar after crush injury and provide useful information for operation that may not be provided by radiographic urethrography.3-dimensional CT reconstruction may become the most valuable means for detecting posterior urethra strictures or ankylurethria with urethrorectal fistula.
8.Expression and anti-tumor characterization of fusion gene GM-CSF and LMP2A of EB virus gene

Objective:To construct GM-CSF and EB virus LMP2A fusion gene recombinant adenovirus and identificate it , using recombinant adenovirus to do immunologic study.Methods:With RT-PCR and Western blot to detect GC 2A gene expression of the recombinant adenovirus , with ELISA to analysis antibodies of vAd-GC2A stimulation in murine body , lactate dehydrogenase assay to detect the mouse cellular immunity effect , thereby the recombinant adenovirus immune effect was initially analysis.T-test method was used to do preliminary analysis and evaluation of the immune effect of recombinant adenovirus .Results:GC2A fragment amplified by PCR, the size was 1 961 bp and it was the expected.The results showed, the recombinant adenovirus vAd-GC2A proteins can be recognized by sera antibody of GM-CSF ( LMP2A); the results of ELISA showed that vAd-GC2A can stimulate the generation of antibodies.EBV-positive tumor cell killing rate (%) by CTL induced recombinant adenovirus was (66.7 ±6.9)%, at the same time the adenovirus type 5 wild strains and PBS were (24.3 ±2.5)%and(32.4 ±3.1)%only(P≤0.05).Conclusion:Fusion gene has been successfully constructed , vAd-GC2A in mice have humoral and cellular immune function.The data suggest that the vAd-GC2A should play a potent role in preventing the positive EB virus tumor.
9.Evaluation of the immunogenicity of a rBCG expressing GM-CSF and LMP2A

Objective To identify the expression of a fusion gene GCA formed from GM-CSF gene and LMP2A gene of Epstein-Barr virus (EBV) in a recombinant BCG (rBCG) and to study its immunoge-nicity.Methods The rBCG was constructed to express the fusion gene GCA and the expressed products were detected by Western blot assay .ELISA was performed to measure specific antibody titers in serum sam-ples from mice immunized with rBCG .Lactate dehydrogenase assay was used to analyze the cellular immuni-ty of mice.A mouse model of EBV-positive gastric carcinoma was established to evaluate the therapeutic effects of rBCG.Results The target proteins of GM-CSF and LMP2A were successfully expressed in rBCG . The specific antibodies were detected in rBCG immunized mice as indicated by ELISA .The maximum anti-body titer reached 1 ∶27 900 [(326.5±7.8) pg/ml] as injection with rBCG 5×108/mouse.The rBCG in-duced cytotoxicity of cytotoxic lymphocytes (CTLs) to EBV-positive gastric carcinoma cells (GT39) (with a killing rate of 89.6%±6.8%) was significantly higher than that of control group (P<0.05) The sizes of tumor in PBS control group [(1964.0±548.7) mm3] and BCG group [(1268.65±72.4) mm3] were big-ger than those in rBCG group [(168.64±78.80) mm3].Conclusion The rBCG expressing GM-CSF and LMP2A fusion gene was successfully constructed .The rBCG could induce humoral and cellular immune re-sponses in mice and inhibit the growth of tumor .
10.Anatomy of Blood Vessels in Amputated Lower Extremity Residual Body after Atherosclerosis Obliteration and Its Clinical Significance

Objective To provide the anatomical basis for detecting distal outflow tract in late atherosclerosis obliteration in lower extremities.Methods Ten lower extremities that were amputated above knees because of late atherosclerosis obliteration were used in this experiment.The blood vessels in the residual bodies were perfused to run blood vessel cast mould to observe the anatomical and pathological change of the popliteal artery,the anterior and posterior tibial arteries and their collateral vessels.The number and distribution of those collateral vessels were also observed.Results The popliteal artery,anterior and posterior tibial arteries were all occluded due to atherosclerosis.However,there were three types of those collateral arteries:① Atheromatous plaque in bole stretched into collateral arteries and led to occlusion.② Obliteration was only observed at the initial segment,with no obstruction at the distal end but extenuated.③ The collateral arteries originated from the bole artery symmetrically,keeping communicative with each other through punctiform interspaces.The last two types were mainly distributed at the inferior segment of popliteal artery,the superior segment of anterior and posterior tibial arteries,forming vascular anastomosing network in the whole cnemis muscle group.Conclusion Un-obstructed collateral arteries in certain places can be still found,though atherosclerosis obliteration is formed in popliteal artery,anterior and posterior tibial arteries in lower extremities.Therefore,it may be possible to construct collateral outflow tracts if endo-membrane stripping operation is performed.
