A quail-origin subtype of the influenza virus was isolated from a human-infecting H7N9 subtype of the avian influenza virus found in a live poultry market and was given the name A/Quail/Hangzhou/1/ 2013 (H9N2). We analyzed the whole genome of this virus and its biologic characteristics. Sequence analyses suggested that the: HA and NS genes belonged to a CK/BJ/1/94-like lineage; NA, NP, PA and PB1 genes belonged to a SH/F/98-like lineage; M and PB2 genes belonged to a G1-like lineage. Analyses of key amino acids showed that the cleavage site in HA protein was PSRSSR ↓ GL, and that the HA protein had a human receptor-binding site with Leu226. Deletion of amino acids 69 - 73 was detected in the stalk of NA protein, the M2 protein had an Asn31 mutation, and the NS1 protein had two mutations at Ser42, Ala149. The intravenous pathogenicity of this virus was 0.36. A study in chickens suggested that all inoculated birds shed the virus from the trachea and cloaca on the third day post-infection (p. i. ) until 11 days. All chickens that had direct contact shed the virus on the second day p. i. until 8 days. Results of virus reisolation suggested that lung and tracheal tissues could shed the virus in 5 days, whereas the other organs could shed the virus in 3 days. These results suggest that this virus strain is H9N2 subtype LPAIV, whose lineage is prevalent in mainland China. This research provides evidence on how to monitor and prevent the H9N2 subtype of the avian influenza virus.

Using universal primer Tyl-copia retrotransposon RT, the conserved reverse transcriptase domain of about 260 bp was amplified by RT-PCR from the Dendrobium officinale which induced by 100 micromol x L(-1) abscisic acid (ABA), indicating these retrotransposons activated by 100 micromol x L(-1) ABA. The amplicons were recovered and cloned, then sequenced and analyzed by related bioinformatics software. Forty-two Ty1-copia like retrotransposon RT transcriptionally activated were obtained with high heterogeneity. The length of these sequences varied from 247 to 266 bp, and was rich in AT and homology ranged from 46.3% to 98.9%. The same to Ty1-copia like retrotransposon RT of genome, different c/s-acting regulatory elements induced by stress conditions and the starting transcription signals, corresponding to CAAT box, TATA box conserved sequences and some other regulatory elements. The c/s-acting regulatory elements induced by stress conditions of reverse transcriptase transcriptionally activated of Tyl-copia retrotransposons were significantly increased than that of Ty1-copia like retrotransposon RT of genome. When being translated into amino acids, fifteen sequences presented stop codon mutation, nineteen sequences presented frameshift mutation, and all sequences presented conserved sequence "SLYGKQ" mutation. Five categories were identified through phylogenic analysis after alignment analyses of their amino acid sequences, and with Ty1-copia like retrotransposon RT of genome having low homology, which indicated that reverse transcriptase transcriptionally activated of Ty1-copia retrotransposons which induced by ABA had Significantly differences with Ty1-copia like retrotransposon RT of genome.

OBJECTIVETo analyze the rules for acupoint selection of acupuncture and moxibustion in domestic clinical treatment of perimenopausal syndrome based on data mining technology in modern times.
METHODSThe relevant literature were retrieved from Chinese Biomedical Literature Database (CBM), China National Knowledge Infrastructure (CNKI) and Wanfang database on this disease treated with clinical acupuncture and moxibustion in China from 1978 to 2013. The database of acupuncture-moxibustion prescription was set up. The relevant regulations of data mining technology were used to analyze the rules for acupoint selection.
RESULTSTotally, 211 papers, 254 acupuncture-moxibustion prescriptions and 130 acupoints were included. The total frequency of acupoints application was 2193 times, with 14 meridians involved. The utilization of the acupoints in the lower limbs and on the back were 33.0% (723/2193) and 23.8% (521/2193) and those of yin and yang meridians were 51.8% (1136/2193) and 44.0% (965/2193), respectively. The utilization of the specific acupoints accounted for 88.7% (1946/2193).
CONCLUSIONIn clinical treatment of perimenopausal syndrome with acupuncture and moxibustion in modern times, the acupoint selection from involved meridians is the basis, associated with multiple methods of acupoint combination; yin and yang meridians are equally important and the specific acupoints are considered particularly critical in application.

?AlM: To observe the effect of Qingguang'an on elastic fiber, MMP-7, TlMP-1 in scarring area of filtration canal after glaucoma surgery through the four Qingguang'an effective groups and Qingguang'an granules, to discuss and compare their mechanism of action on scarring area of filtration canal.?METHODS:Four effective components of Qingguang'an were used in groups D, E, F, G and H after glaucoma surgery, compared with group A ( blank ) , group B (model) and group C ( MMC) to observe the effect of elastic fiber, MMP-7, TlMP-1 in scarring filtration canal.?RESULTS:Compared with the preoperative basic lOP and 2d , 1, 2, 4wk postoperative lOP of groups C, E and H, the lOP of three group rose up slower than other groups, and kept the lowest data at 28d. There was significant difference compared with the rest of A, B, D, F, G groups (P<0. 05). The area and density of elastic fiber in surgery group were significantly different with that of black control group ( P<0. 05 ), but there were no statistical differences between groups C and H, groups C and F, groups H and E (P>0. 05). The difference was statistically significant among other groups (P<0. 01).?CONCLUSlON:The scarring area of filtration canal after glaucoma surgery is the major reason which lead to the failure of surgery. Qingguang'an effective group 2, Qingguang'an granules and MMC could reduced the scar tissue by restrained the elastic fiber, TlMP - 1 and increased the MMP-7. By observing the experimental results that both Qingguang'an effective group 2 and Qingguang'an granules could restrained the scarring area of filtration canal, the effects were unbiased, Qingguang'an granules group is better than effective group 2.

OBJECTIVETo study a new method of simultaneous repair of alveolar cleft and the deformity of the nose and lip.
METHODS42 patients with cleft of alveolar process were included in this study. We filled the cleft of alveolar process with iliac cancellous bone and covered the depression of piriform aperture and the nasal side with iliac cartilage. Simultaneously, we corrected the nasal deformity using V-Y plasty of a mucosa-cartilage compound flap at the nasal vestibule.
RESULTSIn the patients aged 9 approximately 11 years, the mean height of the repaired alveolar process was 86.7%; the mean thickness was 89.6%; the contour and function of the alveolar process was grade I or II. The canine erupted at the place of bone grafting. The depression of the piriform aperture and alae nasi was repaired. The contour of the nares was symmetrical. In the patients over 12 years, the mean height of the alveolar process was 70.1%; the mean thickness was 71.7%; the contour and function of the alveolar process was grade II or III. The deflexion of nasal dorsum and nasal dome was not ameliorated in 2 patients of 18 years old.
CONCLUSIONSThe application of iliac cancellous and chondral bone to simultaneously repair cleft of alveolar process and the nasal deformity is an ideal plastic method. It can recover the height of alveolar process and the continuity of dental arch, ameliorate the nasal deformity, and redress the deflexion of nasal dorsum and nasal end. It can also eliminate the psychological obstacle of the patients at an early stage.

Objective:To study the effects of silencing protein arginine methyltransferase 6 (PRMT6) gene on cell proliferation and migration of gastric cancer cell line MGC-803, and explore its related molecular mechanism. Methods:The expression levels of PRMT6 mRNA and protein in human normal gastric mucosa epithelial cell line GES-1 and human gastric cancer cell lines (AGS, SGC-7901, MGC-803) were detected by quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting. The gastric cancer cell line MGC-803 was divided into silencing control group (NC group), PRMT6 silencing group (si-PRMT6 group), nuclear factor-κB (NF-κB/p65) overexpression group (pcDNA-p65 group), si-PRMT6+ pcDNA-p65 group and PRMT6 silencing and matrix metalloproteinase 9 (MMP9) overexpression group (si-PRMT6+ pcDNA-MMP9 group). Western blotting was used to detect the protein expression levels of PRMT6, NF-κB/p65 and MMP9. CCK-8 kit and Transwell assay were used to measure cell proliferation and migration rates. Results:The results of qRT-PCR showed that the relative expression levels of PRMT6 mRNA in GSE-1, AGS, SGC-7901 and MGC-803 cells were 1.041±0.114, 2.141±0.132, 2.716±0.231, 2.825±0.300, and the difference among the four groups was statistically significant ( F=46.082, P<0.001). Compared with GSE-1 cells, PRMT6 mRNA expression levels were significantly increased in AGS, SGC-7901 and MGC-803 cells (all P<0.001). After 48 hours of silencing PRMT6 in MGC-803 cells, in NC group and si-PRMT6 group, the PRMT6 mRNA expression levels were 0.921±0.110 and 0.303±0.045, the PRMT6 protein expression levels were 1.032±0.105 and 0.289±0.043, the cell proliferation activities were 0.917±0.089 and 0.660±0.069, the cell migration rates were (89.122±5.109)% and (30.831±4.463)%, and the p-p65/p65 protein relative expression ratios were 0.947±0.143 and 0.285±0.023. The relative expression levels of PRMT6 mRNA and protein, cell proliferation activity, cell migration rate, protein relative expression ratio of p-p65/p65 in si-PRMT6 group were significantly lower than those in NC group, with statistically significant differences ( t=9.006, P<0.001; t=11.338, P<0.001; t=3.954, P=0.017; t=14.881, P<0.001; t=7.919, P<0.001). Western blotting results showed that the MMP9 protein relative expression levels in NC group, si-PRMT6 group, pcDNA-p65 group and si-PRMT6+ pcDNA-p65 group were 1.202±0.138, 0.318±0.018, 2.849±0.217 and 1.595±0.194, with a statistically significant difference ( F=127.410, P<0.001). Further pairwise comparison showed that the protein relative expression level of MMP9 in si-PRMT6 group was significantly lower than that in NC group ( P<0.001), while that in si-PRMT6+ pcDNA-p65 group was significantly lower than that in pcDNA-p65 group ( P=0.002). Then, MGC-803 cells were co-transfected with si-PRMT6 and pcDNA-p65 or pcDNA-MMP9 for 48 h. Chinese Journal of Applied Physiology 2012;28(3):268-270
OBJECTIVETo investigate the effects of pH0 on the electrophysiological properties of vascular smooth muscle cells (VSMCs) in brain artery of spontaneously hypertension rats (SHR).
METHODSWe studied the effects and the ion mechanism of pH0 on whole-cell membrane current of VSMCs in brain artery of 200 - 250 g SHR by whole-cell patch clamp recordings.
RESULTS1. Acidic pH0 could inhibit the outward current of VSMCs of brain artery in SHR in a voltage-dependent manner. It induced a more pronounced inhibition of the outward current from 0 to + 60mV; 2. In the presence of 1 mmol/L TEA, the inhibition of acidic pH0 on the outward current of VSMCs of brain artery was inhibited.
CONCLUSIONThe changes of outward current of VSMCs of brain artery in SHR induced by pH0 may be connected with BKCa channel.
