1.Prophylactic efficacy of levetiracetam, valproate or phenobarbital on febrile convulsions in rats.

OBJECTIVETo study and compare the prophylatic efficacy of levetiracetam, valproate and phenobarbital on febrile convulsions in rats.
METHODSSixty Wistar rats were randomly administered with levetiracetam (200 mg/kg), valproate (250 mg/kg), phenobarbital (30 mg/kg) or normal saline (8 ml/kg) for 5 days. Five days later, febrile convulsions were induced by hyperthermal bath (45 Celcius degree). The latency, duration and the severity of seizures were observed.
RESULTSIn all the three drug-treated groups, the latency was significantly prolonged, and the duration and the severity of seizures were notably reduced compared with the saline group (P<0.05 or 0.01). The phenobarbital group had the shortest duration of seizures and the least severe seizures among the three drug-treated groups. There were no significant differences between the levetiracetam and valproate groups.
CONCLUSIONSContinuous administration of levetiracetam, valproate or phenobarbital is effective in preventing recurrent febrile convulsions in rats. Phenobarbital appears to be more effective than levetiracetam and valproate. There were no significant differences in the prophylactic efficacy between levetiracetam and valproate.

2.Dendrobium officinale stereoscopic cultivation method.

The study is aimed to make the most of available space of Dendrobium officinale cultivation facility, reveal the yield and functional components variation of stereoscopic cultivated D. officinale, and improve quality, yield and efficiency. The agronomic traits and yield variation of stereoscopic cultivated D. officinale were studied by operating field experiment. The content of polysaccharide and extractum were determined by using phenol-sulfuric acid method and 2010 edition of "Chinese Pharmacopoeia" Appendix X A. The results showed that the land utilization of stereoscopic cultivated D. officinale increased 2.74 times, the stems, leaves and their total fresh or dry weight in unit area of stereoscopic cultivated D. officinale were all heavier than those of the ground cultivated ones. There was no significant difference in polysaccharide content between stereoscopic cultivation and ground cultivation. But the extractum content and total content of polysaccharide and extractum were significantly higher than those of the ground cultivated ones. In additional, the polysaccharide content and total content of polysaccharide and extractum from the top two levels of stereoscopic culture matrix were significantly higher than that of the ones from the other levels and ground cultivation. Steroscopic cultivation can effectively improves the utilization of space and yield, while the total content of polysaccharides and extractum were significantly higher than that of the ground cultivated ones. The significant difference in Dendrobium polysaccharides among the plants from different height of stereo- scopic culture matrix may be associated with light factor.

Eleven compounds were isolated from the culture of Streptomyces sp. CPCC 202950 by a combination of various chromatographic techniques including column chromatography over macroporous resin HP-20, MCI, and reversed-phase HPLC. Their structures were identified as 1H-pyrrole-2-carboxamide(1),5'-deoxy-5'-methylthioinosine(2), vanillamide(3), trans-3-methylthioacrylamide(4), 1,2,3,4-Tetraydro-1H-pyrido[3,4-b]indole-3-carboxylic acid(5), cyclo(L-pro-L-tyr) (6), N-[2-(4-hydroxyphenyl)]ethylacetamide(7), benzamide (8), cyclo ('L-leucyl-trans-4-hydroxy-L-proline)(9), cyclo-(Phe-Gly) (10), and tryptophan (11). Among them, compounds 1 and 2 were new natural products. In the preliminary assays, none of the compounds exhibited obvious inhibition of HIV-1 protease activity (IC50 > 10 micromol x L(-1)).

4.Analysis of monitoring results of Chinese iodized salt surveillance in 2010

Objective To understand the situation of iodized salt consumption at the household level and non-iodized salt distribution in those areas with low iodized salt coverage.Methods In 2010,iodized salt was monitored in 31 provinces and Xinjiang Production and Construction Corps in accordance with the Monitoring Program of the National Iodine Deficiency Disorders (Trial) (hereinafter referred to as the Program) requirements.Under the jurisdiction of counties (cities,districts,banners) with more than 9 townships (towns,street offices),based on the location of east,west,south,north and center,9 townships (town,district offices) were selected using simple random sampling method; 4 administrative villages (neighborhoods) were selected in each township (town,district office); and 8 residents in each administrative village (neighborhood) were selected.Under the jurisdiction of counties (cities,districts,banners) with less than 9 townships (towns,street offices),based on the location of east,west,south,north and center,1 township(town,district office) was selected using simple random sampling method; 4 administrative villages(neighborhoods) were selected in each township(town,district office);and 15 residents in each administrative village(neighborhood) were selected.Iodized salt coverage rate,qualification rate of iodized salt and consumption rate of qualified iodized salt were calculated in various provinces.The salt samples were tested by semi-quantitative method on the spot and then tested with quantitative method in laboratories.The standard of qualified iodized salt was set as 20-50 mg/kg and that of non-iodized salt was set as < 5 mg/kg (GB/T 13025.7-1999).Results In 2010,a total of 2862 counties(districts,cities and banners) and 14 divisions of Xinjiang Production and Construction Corps,reported the monitoring results,and the monitoring coverage rate was 99.79%(2876/2882).A total of 826 696 copies of edible salt samples were tested,the coverage rate of iodized salt was 98.63%,the consumption rate of qualified iodized salt was 97.95%,and the coverage rate of qualified iodized salt was 96.63%.At province level,only in Tibet iodized salt coverage rate was < 90%.At county level,2755 counties qualified iodized salt coverage rate was ≥90%,and 33 counties iodized salt coverage rate was < 80%.The counties with qualified iodized salt coverage rate of 90% or more accounted for 96.63%(2785/2882) of the total counties.Conclusions The counties where non-iodized salt coverage is higher than 20% mainly distributed in the western or coastal areas and adjacent areas with higher iodine.These areas need policy and funding support from governments at all levels to reducc the gap between these areas and other areas.
5.Protective effect and mechanism of astragaloside IV on oxidative stress injury of mesangial cells.

OBJECTIVETo study the protective effect of astragaloside IV (AS IV) on H2O2 induced human mesangial cells (HMC), and further explore its molecular mechanism.
METHODThe cultured mesangial cells were divided into 5 groups: the normal control group, the H2O2 model group, the AS IV (12.5, 100 nmol x L(-1)) group and the Tempol (1 x 10(5) nmol x L(-1)) group. The MTT method was used to observe cell viability. Hoechst 33258 staining was used to observe the HMC apoptosis and DHE staining was used to detect the generation of reactive oxygen species (ROS). The flow cytometry was used to detect the changes in cell cycle. Western blot was used to detect the expression of Cyclin D1, CyclinA, p38, and T-p38.
RESULTH2O2 (1 x 10(5), 2 x 10(5), 3 x 10(5), and 4 x 10(5) nmol x L(-1)) could induce HMC oxidative stress injury, with significant decrease in the cell survival rate. AS IV (100 nmol x L(-1)) could significantly inhibit HMC oxidative stress injury induced by H2O2 (3 x 10(5) nmol x L(-1)), increase the survival rate of HMC cells, inhibit cell apoptosis, and decrease intracellular ROS production. AS IV could also increase the expression of Cyclin D1, recover normal cell proliferation, and decrease the expression of p38.
CONCLUSIONAS IV can protect H2O2 induced oxidative stress injury in mesangial cells. Its mechanisms may be related to inhibiting the p38/MAPK signaling pathway, increasing the expression of Cyclin D1 and decreasing the intracellular ROS oxidative stress injury.

6.Effect of astragalus membranaceus on the proliferation, osteogenic capacity and structure of periodontal ligament cells in vitro.

OBJECTIVETo investigate the effect of Astragalus membranaceus (APS) on the proliferation, osteogenic capacity and structure of periodontal ligament cells (PDLCs) in vitro.
METHODSPDLCs were cultured in vitro with APS of 0.08, 0.1, 0.2, 0.4 mg x mL(-1). Methyl thiazolyl tetrazolium (MTr), alkaline phosphatase (ALP) and cell structure were detected to determine the proliferation and differentiation of PDLCs proliferation and differentiation.
RESULTSWhen the APS was 0.2 mg x mL(-1), the absorbance of MTT and ALP exhibit significantly increased as compared to the control (P < 0.05). The cells cultured in vitro with APS of 0.2 mg x mL(-1) had the normal structure.
CONCLUSIONAPS with proper concentration in short-term culture may promote the proliferation and differentiation of PDLCs.

7.The expression of vascular endothelial growth factor-C in oral squamous cell carcinoma and its associations with angiogenesis, lymphangiogenesis and lymph node metastasis.

OBJECTIVETo study the expression of vascular endothelial growth factor-C (VEGF-C) in oral squamous cell carcinoma (OSCC) and its relation to angiogenesis, lymphangiogenesis, as well as lymph node metastasis.
METHODSSixty-seven archival specimens from patients with oral squamous cell carcinoma were investigated, whose clinicopathologic data were completely conserved. Immunohistochemical staining was performed to detect the expression of VEGF-C, microvessel density (MVD), lymphatic vessel density (LVD). The correlations between VEGF-C expression and MVD, LVD, as well as other clinicopathological features were measured.
RESULTSAlthough no correlation between VEGF-C expression and tumor location, histological grade, or gender of the patients was observed (P > 0.05), OSCC patients with more advanced clinical stages and lymph node metastasis were prone to have high expression of VEGF-C (P = 0.015 and P < 0.001, respectively). Cases with high-expression of VEGF-C also showed significantly more often higher LVD (P = 0.001) but not MVD (P = 0.125). In addition, cases with lymph node involvement presented higher LVD than other cases (P = 0.026).
CONCLUSIONVEGF-C may promote lymph node metastasis by inducing lymphangiogenesis in OSCC.

8.99Tcm-octreotide somatostatin receptor imaging in the diagnosis of lung cancer

Objective To explore the efficacy of 99Tcm-octrotide somatostatin receptor imaging in the diagnosis of lung cancer. Methods Thirty consecutive patients with pulmonary neoplasms were studied. All lesions detected by scintigraphy were confirmed by histopathology and/or by other imaging modalities. Tumor to non-tumor tissue ratios (T/N) was calculated for semiquantitative analysis. The statistical analysis was carried out using Student t test. Results The sensitivity, specificity and accuracy in differentiating malignant from benign lesion was 95.7%, 71.4% and 90.0%, respectively. Positive image finding was obtained in 24 of 30 patients, and 2 of those was actually false positive on 99Tcm-octrotide imaging. Six patients had negative 99Tcm-octrotide imaging, and one of these was false negative. Forty-six lesions were detected by nuclear imaging, and 3 of these 46 lesions had not been revealed by CT or ultrasound examination. T/N ratio was 2.78±1.07 for SCLC and 1.75±0.31 for NSCLC, respectively(t=3.82,P< 0.05). Conclusion 99Tcm-octreotide somatostatin receptor image can display the distribution of somatostatin receptor in lung cancer. The octreotide uptake rate in small cell lung cancer is significantly higher than in non-small lung cancer, therefore, 99Tca-octreotide image may be more helpful in detecting small cell lung cancer. It can be used as a supplementary diagnostic method to other conventional method.

10.A study on the prevalence rates of human immunodeficiency virus, hepatitis B virus and hepatitis C virus infections in intravenous drug users.

OBJECTIVETo study HIV, HBV and HCV infections in intravenous drug users.
METHODS2025 blood samples from intravenous drug users were collected from Sichuan, Hunan, Guangxi and Xinjiang regions, and tested for anti-HIV, anti-HCV, HBsAg using enzyme-linked immuno-sobent assays (ELISAs).
RESULTSThe positive rates of anti-HIV,anti-HCV and HBsAg were14.7%-30.4%, 60.7%-85.5% and 6.6%-22.4% in the intravenous drug users, respectively. The co-infection rates of HIV/HBV, HIV/HCV, HCV/HBV and HIV/HCV/HBV were 0%-0.4%, 11.6%-27.2%, 2.3%-14.3% and 1.6%-4.8% respectively in this population.
CONCLUSIONThe infection rates of HIV, HBV and HCV were higher in the intravenous drug users than that in general populations in the same regions, and HIV/HCV co-infection appeared most frequent in this population.
