1. Breeding of Erigeron breviscapus triploid and observation of biological properties

Objective: To breed Erigeron breviscapus triploid and evaluate the important biological properties of triploid Erigeron breviscapus. Methods: Tetraploid doubled by colchicine was hybridized with diploid cultivar to get the E. breviscapus triploid; The ploidy of E. breviscapus was identified by the chromosome number. The morphological charcteristics of E. breviscapus triploid in the field were observed and scutellarin was tested by HPLC. Results: Through hybirdization of diploid cultivar and tetraploid, E. breviscapus triploid (2n=3x=27) was obtained successfully. Compared with its parents, the plant height, leaf length and width, number of petals, stoma size of E. breviscapus diploids are larger than those of E. breviscapus diploids, but less than those of E. breviscapus tetraploids. However, leaf number and flower diameter of E. breviscapus triploids are better than those of the parents. The plant of E. breviscapus triploids grew vigorously, and the content of scutellarin (1.38%) is lower than that of E. breviscapus tetraploids (1.51%) and higher than that of E. breviscapus diploids (1.22%). However, the yield of E. breviscapus triploid is obvious higher than that of E. breviscapus diploid with 2.14 times and E. breviscapus tetraploid with 1.84 times. Conclusion: The content of scutellarin in E. breviscapus triploid bred by hybridizing E. breviscapus diploid and E. breviscapus tetraploid is lower than that of E. breviscapus tetraploid, while is much higher than that of E. breviscapus diploid. E. breviscapus triploid is an ideal and important one with the development and utilization value based on the characteristics mentioned above.

4.The effect of Xuebijing injection for severe acute pancreatitis:a Meta analysis

ObjectiveTo assess the efficacy and safety of Xuebijing injection for the treatment of severe acute pancreatitis (SAP).Methods An extensive search of related literatures from the Cochrane Library, EMBASE, China Biology Medicine (CBM), CNKI, VIP and Wanfang data up to March 2014 was performed. Randomized controlled trials (RCTs) regarding Xuebijing injection for the treatment of SAP were collected regardless of languages. Jadad scale was taken for quality evaluation of the included studies by two researchers. The patients in control group were given conventional treatment, and those of the Xuebijing group were given Xuebijing injection on the top of conventional treatment. The Cochrane Collaboration RevMan 5.2 software was used for data analysis regarding the effect of Xuebijing injection on the mortality, incidence of complication, effective rate, the length of stay in hospital, and the safety of the drug in patients with SAP.Results A total of 15 published reports meeting the inclusion criteria were enrolled. The methodological quality of the trials was low. Meta analysis showed that the mortality in Xuebijing group was significantly lower [odds ratio (OR) = 0.37, 95% confidence interval (95%CI) =0.17 - 0.77,P = 0.008], and the incidence of complication was also significantly decreased (OR = 0.26, 95%CI =0.14 - 0.45,P< 0.000 01) as compared with those of control group. The effective rate in Xuebijing group was significantly higher than that of the control group [relative risk (RR) = 0.85, 95%CI = 0.80-0.91,P< 0.000 01]. The length of stay in hospital in Xuebijing group was significantly shorter than that of the control group [mean difference (MD) = -5.28, 95%CI = -6.69 to -3.86,P< 0.000 01]. Adverse reactions of Xuebijing injection were reported in 2 studies. The adverse reaction in one study was headache and nausea, which were relieved by adjusting the speed of intravenous infusion, and mild rash was reported in another case, and it disappeared after the withdrawal of Xuebijing. Conclusions The currently available evidence shows that Xuebijing injection may have some therapeutic effect on SAP. Because of the low methodological quality of the included trials, multi-center and high-quality RCTs with large sample sizes are needed to provide stronger evidence.
5.Effect of propofol on the apoptosis of PC12 cell induced by glutamic acid

Objective To explore the effect of propofol on the apoptosis of PC12 cell induced by glutamic acid.Methods PC12 cells were in-duced 10 mmol · L-1 glutamate for 48 h, and were divided into model group , propofol low , medium and high dose groups.The normal cells were used as control group.Control group and model group were received culture medium without any drugs for 48 h.Propofol low , medium and high dose groups were given 12.5 , 25.0 , 50.0 μmol · L-1 propofol for 48 h.The viability of PC12 cell was measured by MTT assay.PC12 cell apoptosis was measured by flow cytometry.The activity of Caspase -3 was determined by spectrophotometric method.The expression of FBJ osteosarcoma oncogene ( c -fos ) and early growth response protein 1 ( Egr -1 ) were detected by Realtime -PCR and western blot.Results Compared with the normal group , the cell apoptosis rate , the activity of Caspase -3 , the expression of c -fos mRNA and protein increased , and the cell viability , the expression of Egr -1 mRNA and protein decreased in the model group ( P<0.01 ).Compared with the model group, the acitivity of Caspase -3, the cell apoptosis rate , the activity of Caspase -3, the expression of c-fos mRNA and protein decreased , and the cell viability , the expression of Egr-1 mRNA and protein increased in propofol low -dose , medium -dose and high -dose group ( P <0.05 ).Conclusion Propofol suppressed the apoptosis of PC 12 cell induced by glutamic acid , which was related with the expression of c-fos and Egr-1.
6.The expression and significance of structural proteins, VEGF and Ang-1 in cavernous venous malformations of the body surface.

OBJECTIVETo study the expression and significance of structural proteins, VEGF and Ang-1 in cavernous venous malformations of the body surface.
METHODSTissue samples came from 25 cases of cavernous venous malformations, 12 cases of normal moderate veins and 12 cases of normal small veins. Envision immunohistochemical stain was used to investigate the expression of IV collagen, fibronectin, laminin, VEGF and Ang-1. The results were analyzed semi-quantitatively.
RESULTSThe distribution of structural proteins in cavernous venous malformations is similar to moderate and small veins, but the expression in venous malformations is less obviously. VEGF expression in cavernous venous malformations and small veins is stronger obviously than moderate veins. Ang-1 expression in small veins is stronger remarkably than cavernous venous malformations and moderate veins.
CONCLUSIONThe abnormal expression of structural proteins may be an important factor in etiopathology and progress of cavernous venous malformations. There is disturbance of blood vessel remodelling in the sinusoid of cavernous venous malformations, with which the less expression of Ang-1 may be related.

7.Expression of SOX4 gene and early recurrence of hepatocellular carcinoma: their relationship and the clinical significance.

OBJECTIVET To explore the relationship between the expression of SOX4 gene and early recurrence of hepatocellular carcinoma (HCC) after curative resection.
METHODSSOX4 expression was detected immunohistochemically in 60 HCC patients including 30 with and 30 without early recurrence after curative resection, with 30 normal liver specimens as the control.
RESULTSThe expression of SOX4 was significantly higher in HCC than in normal liver (41.7% vs 16.7%, P<0.05), and in HCC tissues, the expression was significantly higher in early recurrent HCC after curative resection than in HCC without early recurrence (56.7% vs 26.7%, P<0.05). SOX4 expression was inversely correlated to the patients' gender, age, tumor size, HBsAg, and Edmonson grade (P<0.05).
CONCLUSIONSOX4 is closely associated with early recurrence of HCC after curative resection, and its overexpression may contribute to early recurrence of HCC. SOX4 may serve as a new molecular indicator for evaluating the prognosis of HCC.

8.A clinical pathological study on cavernous venous malformation of the body surface.

OBJECTIVETo investigate the clinical pathology of cavernous venous malformations of the body surface.
METHODSTissue samples of cavernous venous malformations from 42 cases were stained with hematoxylin and eosin to observe the pathologic structure. The clinical manifestations and case history were summarized accordingly.
RESULTSThere was no distribution difference of the malformation in sex and body sides, but with obvious difference in anatomic sites. The malformation occurred most frequently at the head and neck, more frequently at extremities and least frequently at the trunk. According to pathologic structure, cavernous venous malformations of the body surface can be divided into three types: the cellular, the canaliform and the mixed.
CONCLUSIONThe cause of distribution difference in anatomic sites remains unclear. Internal hemorrhage and infection may account for the increased growth and ache of the lesion. The different pathologic structure of the malformation may cause different clinical manifestations.

9.Study on the association of cholesteryl ester transfer protein gene mutations with the susceptibility to coronary atherosclerotic heart disease.

OBJECTIVETo determine the frequencies of 4 mutations of cholesteryl ester transfer protein (CETP) gene in Chinese population and to investigate the association of the mutations with lipid metabolism and the susceptibility to coronary atherosclerotic heart disease (CHD).
METHODSThe target fragments of CETP gene were amplified and analyzed by PCR-restriction fragment length polymorphism technique in 209 unrelated control individuals and 203 CHD patients. The test for Hardy-Weinberg equilibrium was performed using HWE program and statistical analysis was implemented in statistical package SPSS.
RESULTSIVS14A and 451Q mutant genes were not found in either control group or patient group. The frequencies of 405V mutant allele were 0.443 and 0.413 in controls and patients, respectively, while 442G mutant gene frequencies were 0.007 and 0.025, respectively. The observed allele frequencies of I405V and D442G mutation were in accord with Hardy-Weinberg equilibrium. The frequency of 442G mutant gene in patients was significantly higher than that in controls (P=0.043). Compared with the CHD patients without D442G mutation, the 442G heterozygous CHD patients exhibited a significant increase in plasma TC and LDL-C concentration (P=0.017; P=0.041).
CONCLUSIONIVS14A and 451Q mutants of CETP gene were rare in Chinese population and 442G mutant gene was possibly one of the susceptibility factors to CHD in Chinese.

10.Relationship between expression of multidrug-resistant genes in nasopharyngeal carcinoma tissue and sensitivity to chemotherapy.

OBJECTIVETo study the relationship between multidrug-resistant (MDR) expression in nasopharyngeal carcinoma (NPC) and its sensitivity to chemotherapy.
METHODSThe specimens of 23 NPC cases were studied by immunohistochemistry with monoclonal antibody of P-glycoprotein (P-gp), multidrug resistance relation protein (MRP), lung-resistance related protein (LRP), topoisomerase II (Topo II), thymidylate synthase (TS), glutathione-S-transferase (GST-pi). Among them, 20 specimens were taken from primary NPC lesion which were treated with two course of cisplatin (DDP) and 5-fluorouracil (5-FU), 3 specimens were taken from cervical lymph-node of recurrent NPC patients who were treated by radical dissection.
RESULTSVarious MDR parameters were expressed differently in 22 cases except for 1 clear cell carcinoma case. The difference was statistically significant (P < 0.05). However, there were no significant difference of MDR expression either among various carcinoma pathomorphology cell groups or among different clinical stage groups. Expression of LRP and TS were found in 10 and 14 cases respectively and the chemotherapy responders rates were 20% (2/10) and 28.5% (4/14) respectively. While the chemotherapy responders rates were 70% (7/10) and 5/6 in cases without expression. There was significant difference (P < 0.001, and P < 0.05).
CONCLUSIONThe NPC patients with LRP and TS expression may be less sensitive to chemotherapy with DDP + 5-FU.
