Objective:To identify the function of extracellular soluble vimentin that promotes proliferation, activation and chemotaxis of inflammatory cells. Methods: The proliferation of rat splenocytes stimulated with vimentin was evaluated in vitro. The lymphocyte counts and vimentin-antibody levels of peripheral blood in mice immunized with vimentin were detected in vivo. Peritoneal macrophages were collected and cultured with different concentrations of vimentin to detect the effect on phagocytosing chicken red blood cells. The chemotaxis of NIH3T3 fibroblast towards vimentin was observed in transwell chamber. Results:In vitro vimentin dose dependently promoted the proliferation of splenocytes. The proliferation indexes of primed and naive splenocytes cultured with 16μg/ml vimentin were reached up to ( 196. 0 ± 9. 7 )% and ( 208. 9 ± 4. 6 )% respectively without significant difference. In vivo vimentin significantly enhanced the lymphocytes number(109 L-1)of peripheral blood(5. 74±0. 51 vs. 1. 69±0. 13)and the levels of vimentin specific antibody( OD value 2. 31 ± 0. 06 vs. 0. 19 ± 0. 08 ) that shown no significant difference from immunization with vimentin plus CFA. In vitro vimentin dose dependently stimulated phagocytic ability of macrophages and performed the chemotactic effects on NIH3T3 fibroblasts. Conclusion:Extracellular soluble vimentin promotes the proliferation, activation and chemotaxis of concerned inflammatory cells and possesses the functions as an alarmin.

Objective: To observe the effectiveness and safety of electroacupuncture (EA) plus Luohua Anshen oral liquid for patients with perimenopausal insomnia. Methods: A total of 66 participants who met the inclusion criteria were enrolled in the randomized controlled trial and allocated to a treatment group and a control group at a ratio of 1:1, with 33 cases in each group. Both groups were given Luohua Anshen oral liquid as a basic treatment. The treatment group was additionally given EA every other day, three times a week. Both groups were treated for four weeks and a four-week follow-up was conducted. The scores of Pittsburgh sleep quality index (PSQI), Kupperman index (KI) and traditional Chinese medicine sleep syndrome scale (TCMSSS) were recorded at pre- and post-treatment, and at the follow-up. Meanwhile, adverse effects were monitored and recorded. Results: After four-week treatment, the global scores of PSQI, KI and TCMSSS in both groups declined significantly (all P<0.05), and the decreases in the treatment group were more significant than those in the control group (allP<0.05). The global scores of PSQI, KI and TCMSSS in both groups at the follow-up visit were significantly different from the corresponding baseline (allP<0.05), while insignificantly different from those assessed at post-treatment (allP>0.05). The total effective rate was 93.9% in the treatment group, significantly higher than 72.2% in the control group (P<0.05). No significant adverse event was reported in this trial excepted one patient experienced slight dizziness in the first acupuncture treatment. Conclusion: EA plus Luohua Anshen oral liquid is safe for perimenopausal insomnia with satisfactory short- and long-term effectiveness, and it shows certain advantage compared with using Luohua Anshen oral liquid alone.

Objective To investigate the effect and mechanism (a selective cyclooxygenase-2 inhibitor) on invasive ability of human nasopharyngeal carcinoma (NPC) line CNE-2Z. Methods The proliferation of NPC cells was examined by MTT assay. The invasive and migrating ability of NPC cells was detected with transwell chamber. E-cadherin protein expression was detected by immunocytochemistry and the expressions of Cox-2 and E-cadherin mRNA were analyzed by reverse transcription-polymerase chain reaction (RT-PCR). Results MTT showed that celecoxib inhibited CNE-2Z proliferation in dose-dependent manner, the survival rate of cells treated with 25, 50, 100 μ mol/L celecoxib (-x±s) for 24 h was(94. 75 ±1.34)%, (91.77 ±2. 70)% , (64. 54 ± 1.20)%, respectively, and the survival rate of cells treated for 48 h was ( 88.41±1.28 ) %, ( 78. 84 ± 1.56 ) %, ( 52. 46 ± 2. 25 ) %, respectively, the concentration of 50% inhibition concentration of a substance (IC50) was 100 μmol/L, the difference was statistically significant between different concentration groups in the same time-point ( repectively, F were 462. 204 and 1328. 306, P <0. 01 ). Treated with different concentrations of celecoxib(0, 25, 50 μmol/L) for 24, the cell numbers (-x±s) through PVPF by tumor invasion assay were (263.7 ± 13.5), (185.3 ±8.7) and (144. 0 ± 8. 2), the difference was statistically significant between the experimental and control group (F =102. 089, P <0. 01 ). Immunocytochemistry showed that celecoxib significantly induced the increase of Ecadherin protein expression, also with a dose-dependence in 0 μmol/L, 25 μmol/L, 50 μmol/L group was (21.7 ±2. 6), (28. 7 ±2. 4), (40. 3 ± 1.3), and 50 μmol/L group increased significantly ( F =78. 637,P <0. 01 ). RT-PCR showed that celecoxib reduced the expression of Cox-2 mRNA expression in 25, 50 μmol/L group decreased significantly compared with the control group (respectively, t were 23. 950 and 36. 651, P < 0. 01 ), but it enhanced the expression of E-cadherin mRNA expression in 25, 50 μmol/L group was significantly higher ( respectively, t were 35. 829 and 81. 497, P <0. 01 ). Conclusion Celecoxib can inhibits the invasive ability of NPC cell line CNE-2Z, which possibly relates with the upregulated expression of E-cadherin.
