Objective To investigate the effect of astilbin on expression of CTLA-4 in activated T cells of mouse heart transplantation model with acute rejection. Methods Cardiomyocytes of BALB/ c mouse and spleen cells of C57BL/6 mouse were separated. The cardiomyocytes(2?10~5/ml)as irri- tation cells and spleen cells(1?10~6/ml)as responsers were mixed and cultured. The model of mouse heart transplantation with acute rejection in vitro was established.Three groups were set up:control group,Astilbin(15/?g/ml) group,Astilbin+anti-CTLA-4 mAb 9H10 (30?g/ml)group.Apoptosis of T cells was observed by TUNEL.The expression of CTLA-4 in activated T cells was detected by RT-PCR and Western blot. Results Apoptosis indexes of activated T cells in Astilbin group were sig- nificantly higher than those in the control group(73.4%?12.5% vs 35.1%?9.2%,P<0.01). The expression of CTLA-4 in Astilbin group was significantly higher than control group(P<0.01), but there was no apparently difference between control group and Astilbin+CTLA-4 mAb group(P>0.05).Conclusion Astilbin induces apoptosis of activated T cells in heart transplantation, which may be partially related to its enhanced expression of CTLA-4.

Journal of Environment and Health 1989;0(06):-
Objective To study the effects of di-butyltin dilaurate (DBTD) exposure on pregnancy outcome in Wistar rat and evaluate it's effect on sexual development of fetuses. Methods Timed pregnancy rats were treated with corn oil or DBTD (10, 20, 30 mg/kg body weight) from days 12-19 of gestation. On gestational day (GD) 20, dams were sacrificed to investigate the pregnancy outcome. Results There was a downtrend in weight of dams on GD20 as the DBTD exposure dose increased. The weight of dam's utero significantly decreased in 30 mg/kg group. A significant decrease in fetal weights was observed in all DBTD groups, and fetal sizes in 20, 30 mg/kg groups. Exposure to DBTD from GD 12-19 resulted in a distinct increase in normalized anogenital distances in female fetuses, but no effects were seen in male ones. DBTD exposure did not result in external malformations, however, delayed ossification of fetal phalanges was observed in all DBTD treated groups. Dead fetuses and absorbed embryos were observed in 20, 30 mg/kg DBTD groups. Conclusion The results of the present paper show that DBTD exposure has some adverse effects on fetal development and may exert a masculinizing effect on female fetuses.

Objective To study the anti-injury effects of grape seed proanthocyanidin (GSP) on the umbilical vein endothelial cell (HUVEC)injured by hydrogen peroxide(H2O2) and its mechanism. Method Oxidative damage model of endothelial cell was induced by H2O2,and the cells were divided into five groups, including normal groups, oxidative damage group and three (100,50,10 ?g/ml) GSP pretreated groups. MTT assay was used to detect the protection of GSP,Reverse transcription PCR (RT-PCR) was used to detect the protein expression in mRNA level of monocyte chemoattractant protein (MCP-1) and flow cytometry(FCM) was used to detect apoptosis rate and quantitate the expression of intercellular adhesion molecule(ICAM-1)and vascular cell adhesion molecule ( VCAM-1). Results The cell optical density in oxidation damaged group was lower than that in normal control. When pretreated by GSP,the OD value was increased, and was insignificantly different between high GSP group and normal control. In H2O2 damage group, the expression of MCP-1 mRNA and the apoptosis rate were increased, and the numbers of positive cells which express ICAM-1 or VCAM-1 were obviously more than those in pretreated groups respectively.Futhermore, these effects were dose dependent. Conclusion GSP has the protective effect on oxidative damage of HUVEC. The mechanism may be related to stabilization of cell wall and inhibition of inflammatory reaction and apoptosis induced by ICAM-1,VCAM-1 and MCP-1.

This investigation describes a new precise, sensitive and accurate stereoselective RP-HPLC method for determination of the enantiomers of a novel alpha- and beta-receptor blocking agent, 1-[4-(2-methoxyethyl) phenoxy]-3-[[2-(2- methoxyphenoxy) ethyl]amino]-2-propanol (TJ0711), in rat plasma. GITC was used for precolumn derivatization of TJ0711 enantiomers. Enantiomeric resolution was achieved on a Eurospher-100 C18 column (250 mmx4.6 mm ID, 5-mum particle size), with UV detection at 255 nm, and the mobile phase consisted of acetonitrile and water (58:42, v/v) containing 0.02% glacial acetic acid (v/v). Using the chromatographic conditions described, TJ0711 enantiomers were well resolved with mean retention time of 10.2 and 11.5 min, respectively. Linear response (r>0.999) was observed over the range of 0.125-12.5 mug/mL of TJ0711 hydrochloride enantiomers. The mean relative standard deviation (RSD%) of the results of within-day precision was [Symbol: see text] 10%. The proposed method was found to be suitable and accurate for the quantitative determination of TJ0711 enantiomers in rat plasma, and it can be used in pharmacokinetic studies.

Aim To make ethanol from bistortae(EFB)from traditional Chinese medicine bistortae, and investigate the protection on the injury of the human umbiliar vein endothelial cell(ECV304)induced by hydrogen peroxide(H2O2).Methods ECV304 cells were cultured and divided by five experimental groups including normal, injury model induced by H2O2 and three pretreated groups with EFB;flow cytometry(FCM)methods Annexin V/PI staining and PI staining were used to analyze the effect of the medicine on the apoptosis and cell cycle;reverse transcription PCR(RT-PCR)was used to detect the mRNA expression level of apoptosis associated protein caspase-3 in all experiment-al groups. Results FCM Annexin V/PI staining result showed early and advanced stage apoptosis rate was lower when predisposed than that of the damaged group,FCM PI staining showed there were more cells in G0/G1 stage in oxidation damage group, but less in S stage, and hypodiploid peak could be observed,RT-PCR showed that the expression of caspase-3 was weakened when predisposed by EFB than without this disposal(P

Objective To investigate the changes of plasma adrenomedullin(ADM),endothelin-1(ET-1) in infants with severe pneumonia complicated by acute congestive heart failure, and its relationship with heart function. Methods Forty-seven bronchopneumonia patients in their acute phase were divided into three groups:group A1,severe pneumonia complicated by acute congestive heart failure without congenitive heart disease(CHD)(n=15);group B1,severe pneumonia complicated by acute congestive heart failure with CHD(n=12);group C1,mild bronchopneumonia(n=20).A2,B2 and C2 groups were the convalescence groups of A1,B1 and groups C1 respertively.Group D,20 healthy infants were used as control group.Plasma ADM,ET-1 levels of patients in acute phase(pre-treatment)and convalescent phase and controls were measured by specific radioimmunoassary. Results 1.The plasma ADM levels significantly increased in the acute phase of A1,B1 and C1 compared with healthy controls(Pa0.05).3.The plasma ADM,ET-1 levels in A2 and B2 groups significantly decreased compared with those in group A1 and B1(Pa0.05).Conclusions The ADM,ET-1 play very important roles in the pathophysiological processes of pneumonia and congestive heart failure in infants.ET-1 may play an important role in the pathogenesis of severe pneumonia in infants complicated with congestive heart fai-lure.The level of plasma ET-1 is related with the degree of congestive heart failure.
