1.The Establishment of Autoimmune Premature Ovarian Failure (POF) Model Using B6AF1 Female Mice

This study aimed at exploring the modeling and identification method of autoimmune POF by the ZP3 polypeptide with the provision of scientific basis for the prevention and treatment of POF.B6AF1 female mice,the hybridization of A/J male mice and C57BL/6 female mice,was immunized by zona pellucida polypeptide fragments.Then,the mice estrous cycle was detected by vaginal exfoliated cells Pasteur's staining.After the 14-day modeling,the mice model was identified on the 7th and 14th day,respectively.Ovarian tissue morphology and anti-zona pellucida antibodies immunofluorescence change were detected by H&E staining and ZP3 immunofluorescence staining.Serum hormone levels of E2 and FSH in the mice were tested by ELISA,with the combination of estrous cycle,ovarian tissue morphology and anti-zona pellucida antibodies immunofluorescence change to judge the success of the POF mice model.It was found that ZP3 polypeptide immunized mice appeared the disorder of estrous cycle after 14 days,so did the marked inflammation observed by ovarian biopsy,a clear transparent zone by ZP3 immunofluorescence staining,and the decrease of serum E2 declined and the increase of serum FSH by ELISA.However,no significant change was found on the 7th day in the modeling.In conclusion,ZP3 polypeptide induced autoimmune POF mice model was established successfully,featuring high incidence and simple method and laying a foundation for the further study of autoimmune POF.
2.Evaluation of binocular minimum thickness of cornea in Chinese myopic population

Background Central corneal thickness (CCT)is an important parameter to evaluate corneal healthy status and is crucial for surgical planning.However,clinical study found that the center of cornea does not correspond to the thinnest point of cornea.Thus,it is essential to characterize the minimum corneal thickness(MCT) and its location.Objective Present study was to determine the thickness and location of MCT and its relationship to the fellow eye using Pentacam High Resolution technique.Methods The 564 eyes from 282 Chinese myopic patients were reviewed in this study.The CCT,MCT,pupillary central corneal thickness(PCT) and x-y coordinate of thinnest point were bilaterally measured.Written informed consent was obtained prior to the ocular biomedical measurement.Results CCT was (540.07±31.78) μm in the right eyes and (539.24±31.06) μm in the lefi eyes; PCT was (540.25±30.75)μm in the right eyes and (539.48±31.00)μm in the left eyes.MCT was (537.87± 31.91)μm in the right eyes and (536.35±31.24)μm in the left eyes,showing significant differences in all the parameters between the right eyes and left eyes expect for PCT(CCT:P=0.046;PCT:P=0.065 ;MCT:P=0.000).The C CT,PCT and M CT were significantly correlated between the right eyes and left eyes (r =0.97,0.97,0.98,P< 0.01).Bland-Altman plot showed a good consistence between the both eyes.The mean distance from the center was (0.50±0.21) mm in the right eyes and (0.56±0.22)mm in the left eyes,showing a significant difference (P =0.000).The difference between CCT and MCT was approximately (2.20±1.74)μm in the right eyes and (2.88±1.75) μm in the left eyes.The location of MCT in both the right eyes and left eyes tended to symmetry along the vertical midline.The distance between the R (x,y) to transformed L (x,y) was (0.29 ± 0.17)mm and the angular distance was (28.28±28.21)degree.Conclusions This study offers a range of MCT and its location in Chinese myopic patients.The difference exists between the CCT and MCT in bilateral eyes.The location of the thinnest point tends to be symmetrical along the vertical midline between the both eyes.The changes of these parameters may be helpful for the diagnosis of some corneal diseases.

4.Effects of Virtual Reality Rehabilitation on Balance for Patients with Parkinson's Disease

Objective To investigate the effects of a training program based on virtual reality on static and dynamic balance perfor-mance in patients with Parkinson's disease (PD). Methods From June, 2014 to June, 2016, 46 patients with PD were randomly divided into control group (n=23) and experimental group (n=23). The control group received routine balance training, while the experimental group re-ceived balance training of virtual reality, for six weeks. They were assessed with Unified Parkinson's Disease Rating Scale part 3 (UP-DRS3), Berg Balance Scale (BBS), TimedUp and GoTest (TUGT) and Hamilton Depression Scale (HAMD) before and after training. The envelope area, anteroposterior standard deviation (AP-SD), mediolateral standard deviation (ML-SD) of centre of pressure (COP) were also measured with posturography. Results The scores of BBS, TUGT and HAMD improved in both groups after training (t>2.657, P<0.05), and improved more in the experimental group than in the control group (t>2.426, P<0.05). The score of UPDRS3 and the parameters of pos-turography improved in the experimental group (t>2.626, P<0.05), and improved more than that in the control group (t>2.112, P<0.05). Con-clusion Virtual reality rehabilitation is more effective than routine balance training on the static and dynamic balance function in patients with PD, and may release their depression.
5.Pinaverium bromide combined with bifid triple viable and doxepin therapy in diarrhea-dominant irritable bowel syndrome

Objective To evaluate the efficacy of stomach intestine power regulator, intestinal microecology preparation and tricyclic antidepressant treatment in irritable bowel syndrome (IBS), and to investigate its pathological mechanism. Methods From November 2006 to November 2010, 103 patients with diarrhea-dominant IBS (D-IBS), who fulfilled the Rome Ⅱ criteria and were excluded from organic disease by entewscope were divided into pinaverium bromide group (26 cases), pinaverium bromide + bifid triple viable group (28 cases), pinaverium bromide + doxepin group (25 cases) and pinaverium bromide +bifid triple viable + doxepin group(24 cases ) by random digits table. The symptom grade, intestinal flora and SCL-90 was tested before treatment and 4 weeks after treatment. Results The total effective rate of pinaverium bromide + bifid triple viable + doxepin group was 83.33%(20/24), significant higher than that in pinaverium bromide group [65.38%(17/26)], pinaverium bromide + bifid triple viable group [71.43%(20/28)], pinaverium bromide + doxepin group [68.00% ( 17/25 )] (P < 0.05 ). Five kinds of intestinal flora and psychiatric symptoms were improved in the four groups, and those in pinaverium bromide + bifid triple viable + doxepin group improved significantly. Conclusions To interfere the correlation factor of IBS can have better efficacy. There is a close relation between brain and gut in patients with IBS, which may be involved in the pathogenesis of IBS.
6.Therapeutic efficacy of 18 α-Diammonium glycyrrhizinate phosphatidylcholine complex in patients with chronic virus hepatitis

Objective To investigate the therapeutic effieacy and safety of 18 α-Diammonium glycyrrhizinate phosphatidylcholine complex (DGPC) in patients with chronic hepatitis B and or C with elevated aminotransferase. Methods 55 patients with chronic hepatitis B and or C, with serum alanine aminotransferase (ALT) of 2 to 10 times the upper limit of normal were randomly assigned to receive DGPC or Diammonium glyeyrrhizinate (DG) for 12 weeks. Then they were followed up for an additional 4 weeks. From week 1 to 10, DGPC or DG was given as 150 nag,three times a day (TID). At the 11th week,the drug was given as 100 mg,TID. Then 50 mg,TID for the 12th week. Results ALT was markedly decreased after receiving DGPC 4,8,12 weeks (P=0.00). ALT normalization rate at the end of therapy was similar (38.5% vs 34.5% ,P =0.76). Drug-related adverse events were similar. Conclusion DGPC can rapidly and safely decrease aminotransferase in patients with chronic viurs hepatitis.

8.Related Factors of Hyperhomocysteinemia among Non-hypertensive People Aged between 40-70 Years in Tianjin

Objective To investigate the concentration of homocysteine (Hcy) and related factors of hyperhomocyste-inemia (hHcy,Hcy≥15μmol/L) among non-hypertensive people aged between 40-70 in Tianjin. Methods Non-hyperten-sive community residents aged 40-70 years were enrolled randomly from May 2011 to December 2012 in six districts in Tian-jin. Plasma Hcy was assessed by enzyme cycling method. Factors related to hHcy were analyzed in multivariate logistic re-gression models. Results Our study included 874 participants (mean age is 57 ± 6 years, 25.5%of all are males) and the con-centration of Hcy was 12.0 μmol/L. The OR (odds ratio)(95%CI; P)for hHcy were 1.048(1.015,1.083; P=0.004), 4.191 (2.359,7.448;P<0.001), 1.280(0.896,1.829;P=0.175), 0.460(0.259,0.816;P=0.008)respectively for age, male, smoking, exercise, and the odd ratio for hHcy were 0.290(0.179, 0.469;P<0.001), 0.168(0.092,0.309;P<0.001)for consumption of vegetable and fruits 250-500 g/d and>500g/d, compared with<250 g/d. Conclusion Male and age were adverse factors for hHcy, consumption of vegetable and fruits, and exercise were positive factors.
9.Evaluation of the immunogenicity of a rBCG expressing GM-CSF and LMP2A

Objective To identify the expression of a fusion gene GCA formed from GM-CSF gene and LMP2A gene of Epstein-Barr virus (EBV) in a recombinant BCG (rBCG) and to study its immunoge-nicity.Methods The rBCG was constructed to express the fusion gene GCA and the expressed products were detected by Western blot assay .ELISA was performed to measure specific antibody titers in serum sam-ples from mice immunized with rBCG .Lactate dehydrogenase assay was used to analyze the cellular immuni-ty of mice.A mouse model of EBV-positive gastric carcinoma was established to evaluate the therapeutic effects of rBCG.Results The target proteins of GM-CSF and LMP2A were successfully expressed in rBCG . The specific antibodies were detected in rBCG immunized mice as indicated by ELISA .The maximum anti-body titer reached 1 ∶27 900 [(326.5±7.8) pg/ml] as injection with rBCG 5×108/mouse.The rBCG in-duced cytotoxicity of cytotoxic lymphocytes (CTLs) to EBV-positive gastric carcinoma cells (GT39) (with a killing rate of 89.6%±6.8%) was significantly higher than that of control group (P<0.05) The sizes of tumor in PBS control group [(1964.0±548.7) mm3] and BCG group [(1268.65±72.4) mm3] were big-ger than those in rBCG group [(168.64±78.80) mm3].Conclusion The rBCG expressing GM-CSF and LMP2A fusion gene was successfully constructed .The rBCG could induce humoral and cellular immune re-sponses in mice and inhibit the growth of tumor .
10.Effect of ultrasound in repair of rabbit articular cartilage injury

Objective To observe the effect of low intensity pulsed ultrasound (LIPU) in repair of the articular cartilage injury in rabbits. Methods Osteochondral defects in the cartilage surfaces of femoral intercondylar fossae were made in 33 healthy adult New Zealand White rabbits, of which the knee lesions of the left hind legs were treated with LIPU (LIPU group) while that of the right ones used as control group. All animals in two groups were sacrificed respectively at the 2nd, 4th and 8th weeks postoperatively for gross appearance observation and histological examination. Results The ultrasound-treated tissues became more mature with in advance repair or regeneration. The repaired tissues in the LIPU group were near to normal hyaline cartilage tissues at the 8th week postoperatively, while the repaired tissues in the control group were weak, near to fibrocartilaginous tissues. Conclusion LIPU can promote repair of articular cartilage injury in rabbits.
