Lymphocyte chemiluminescence (Ly-CL) is one of the early events involved in the activation of lymphocytes. This CL is thought to be result ed from the reactive oxygen species (O_2, H_2 O_2, OH, 'O_2) produced by the lymphocytes, the consequent oxidation of luminol and emission of light. In this paper we investigated the experimental condition for ConA-induced human peripheral blood lymphocyte (PBL) CL by using orthogonal design. The effects of new born calf serum (NCS), bovine serum albumin (BSA), temperature and time of cell storage on CL were also studied. The results show that the optimal condition of Ly-CL assay was 1.0 ml PBL suspension (1.4 ? 10~6 cell/ml ), 0.2 ml Luminol solution ( 7 ? 10~(-4)M) and 0.2 ml ConA ( 700 ?g /ml ). The isolated PBL were suspended in phenol red-free Hanks solution containing 0.1% BSA and can be kept for 2 hr at 4℃ before being used without adversely affecting cell viability and CL.

ObjectiveTo analyze the expression and significance of Ets－ 1 (E26 transformation－specific) in chondrosareoma and osteochondroma of the jaws.MethodsEts－ 1 expression was de-tected by immunohistochemistry in 20 cases of chondrosarcoma and 8 cases of osteochondroma of thejaws.Results12 of the 20 cases of chondrosarcoma (t50%) showed an Ets－ 1 positive reastion. Thepositive rates of grade Ⅱ and grade Ⅲ were significantly higher than that of grade Ⅰ ( P < 0.05 ). Re-current cases showed a higher Ets － 1 positive rate than primary ones ( P < 0.05 ). Ets － 1 was presentin 12.5 % (1/8) osteochondroma. The positive rate was significantly different between chondrosarcomaand osteochondrorna ( P < 0.05 ).ConclusionThe results indicate that Ets－ 1 is overexpressed inchondrosarcoma of the jaws and correlated with its pathological grade and recurrence.

ObjectiveTo establish the human periodontal ligament fibroblasts(HPDLFs)that express BMP2 and observe their biologicl characterization. MethodsA phagemid expression vector for BMP2 (pBK-B2) was transfected into HPDLFs by using LipofectAMINE. The BMP2 expression was determined by the immunohistochemical ABC method. The alkaline phosphatase (ALP) activity, osteocalcin (OC) production and capacity of mineralization were measured in the transfected cells. ResultsBMP2 protein was expressed in HPDLFs after gene transfection. The BMP2 gene transfected cells showed prominently elevated ALP activity, OC production and increase in mineralized nodules. ConclusionThe results indicate that BMP2 is expressed in HPDLFs and is involved in inducing differ- entiation of HPDLFs into osteoblast-like cells.

Although Chinese government has input a lot of financial and administrative resources to resolve the problem of “swill-cooked dirty oil”, its policy effect is not so ideal.Meanwhile , a series of challenges such as defec-tive institutional design of kitchen waste recycling and weak regulatory capacity.This paper argues that its overlook on technological innovation and industrial policy have partially led to its policy failure on this issue .Also, this paper summarizes impact factors from the perspectives of technological innovation , industrial policy and government regula-tion.A series of feasible policies include promoting business technological innovation capacity , offering financial sup-ports to industrial development , establishing a long-term plan on kitchen waste recycling and quota system for biodiesel using.Finally, it argues Chinese government should combine its regulatory policy with its technological innovation and industrial policy to achieve its dual policy goals between food safety governance and circular economy development .

Purpose To explore the role of histone H2B monoubiquitination in primary colonic carcinoma and clinicopathological pa-rameters of colonic carcinoma. Methods To detecte the expression of histone H2B monoubiquitination in 116 cases of primary colonic carcinoma and 15 cases of normal colonic mucosal tissue by avidin-biotin-peroxidase complex ( ABC)-immunohistochemistry. Results The level of histone H2B monoubiquitination was significantly reduced in poorly differentiated colonic carcinoma 19. 4% (6/31) compared with that of well-differentiated colonic carcinoma 49. 4%(42/85), moderately differentiated colonic carcinoma 49. 4% (42/85) and normal colonic mucosal tissue 86. 7% (13/15) (P<0. 05). The expression of histone H2B monoubiquitination was closely correlated with tumor differentiation, Dukes stage, TNM stage and lymph node metastasis (P<0. 05), but not depended on sex or age (P>0. 05). Conclusion Histone H2B monoubiquitination is obviously associated with the progression of primary colonic carcinoma. Although its specific mechanism still remains unclear, histone H2B monoubiquitination could be a novel potential molecular marker for early diagnosis, clinic treatment and prognosis evaluation.

Objective To evaluate the clinical value of fractional exhaled nitric oxide (FeNO) level in asthmatic children to predict bronchial hyperresponsiveness by analyzing the correlation between fraction of exhaled nitric oxide and bronchial provocation test.Methods One hundred and fourteen asthma outpatients of Shengjing Hospital were enrolled, FeNO levels, spirometry and bronchial provocation test were measured.Results In the bronchial provocation test,there were 33 positive and 81 negative cases.The positive group had a significantly higher FeNO levels than the negative ones(19.0 × 10-9 vs.16.0 × 10-9 ,p =0.000).By the ROC curve, the best FeNO cut-off value to predict bronchial hyperresponsiveness was 38.5 × 10-9 with high specificity (92.6％) but relatively low sensitivity (36.4％).There was no relationship between methacholine provocative dose causing a 20％ fall in FEV1 (PD20-FEV1) and the level of FeNO.Conclusion FeNO level has important predicting value for bronchial hyperreactivity in children with asthma.The level of FeNO ＞ 38.5 × 10-9 has high predictive value in asthmatic children with bronchial hyperreactivity.

Objective:To investigate the effects of static ultrasound elastography on the elastic modulus of normal breast tissue.Methods: (1)Fifteen cases of normal breasts was tested by static ultrasound elastography at the same locations. Intercepted two pictures each time with one stress-free and the other compressed the maximum amount. For these two cases we measured the breast thickness and calculated the minimum stretch ratios(λ). (2)Fifty breasts with no breast nodules were prospectively enrolled in the study. Intercepted two pictures each time with one stress-free and the other compressed to the minimum stretch ratios(λ). For these two cases we measured the Elastic modulus (E0, Em, kPa). Calculate the elastic modulus change ratio (Rm/0).Results: (1)The minimum stretch ratio caused by static ultrasound elastography was 0.74±0.07. (2)The elastic modulus change ratio of static ultrasound elastography was 4.38±1.72. Conclusion: The normal operation of static ultrasound elastography caused 74 percent deformation in normal breast tissue. There was 4.38 times elastic modular increasing caused by 74 percent deformation in normal breast tissue.
