Circular dichroism (CD) is an useful technique for monitoring DNA conformation changes resulting from changes in environmental conditions, such as temperature, ionic strength, and pH, and also for the study of the interaction between DNA and ligands (including small molecules and proteins). CD spectroscopy of DNA arises from the asymmetric backbone sugars and by the helical structures often adopted by nucleic acids. By the interpretation of induced circular dichroism (ICD) of ligand signals resulting from the coupling of electric transition moments of the ligand, DNA bases within the asymmetric DNA environment, ligand-DNA interactions, as well as the DNA-binding mode can be assessed. A number of important conclusions have been reported that related to the observed ICD signals resulting from the interactions between intercalators and groove binders with DNA. If short oligonucleotide sequences are used in the study, sequences-specific of binding also can be deduced. CD determination requires smaller amounts of sample, and not limited by the molecular weight or size and can be performed rapidly; though CD is of low resolution, but it's a complement to NMR and X-ray diffraction methods. This review will introduce the characters of the CD spectra of DNA, and its application to the studies of DNA with small molecules; some progress of the studies in our laboratory will also be discussed. CD is expected to be used as a screening method in seeking more DNA-targeted drugs, such as, antineoplastic, antimicrobial and antiviral drugs.

Objective To evaluate the clinical features and guanosine triphosphate cyclohydrolase 1 (GCH-1) gene mutation in a family with dopa-responsive dystonia (DRD).Methods The clinical features of this family were collected and their peripheral blood samples were screened for mutation in GCH-1 gene using PCR and DNA direct sequencing. Results The clinical features among each patient in this family were different. But all affected family members had quite a good response to levodopa treatment without significant adverse reactions.DNA test showed an AT deletion mutation at point of 631-632 in the 6th exon of GCH-1 gene in 5 affected members and 1 asymptomatic immediate family member. Conclusions Clinical heterogeneity is an important characteristic of DRD and clinical symptoms vary intra-families.Same gene type may cause different phenotype and not all carriers are patients. The deletion mutation at point of 631-632 in the 6th exon of GCH-1 gene should be considered as a pathogenic mutation for DRD.

Objective To improve the recognition and MRI diagnosis for intraspinal epidermoid cyst. Methods MRI appearances of 20 cases with histologically confirmed intraspinal epidermoid cyst in the spinal canal were analyzed. Results The lesions were ovoid in 15 cases. The lesions were low signal intensity on T1WI and high signal intensity on T2WI with smooth border, there were no enhancement on contrast-enhanced scan,5 lesions appeared as irregular shape.3 cases showed circular enhancement. Conclusion MRI is of important value in diagnosis of localization and quality of intraspinal epidermoid cyst.

Chinese Acupuncture & Moxibustion 2015;35(12):1309-1314
By bibliometric method, acupuncture-moxibustion literature from 2003 to 2013 included by SCIE and GoPubMed was analyzed. The number of annually published literature, distribution of country, agency and area, author, journal, subject category, high-frequency words and H index were analyzed to explore the current situation and development trend of acupuncture and moxibustion.

Objective To observe on the variance of serum IL-18 level and lymphocyte subpopulations in the patients with obese diabetic(T2DM).Methods The diabetic obese group contained 31 cases, the non-diabetic obese group contained 33 cases, healthy normal body weight group contained 23 cases, the blood glucose(FPG),insulin (FPI),blood total cholesterol(TC),triglyceride(TG),IL-18 and the cell population of lymphocyte subpopulations CD3+and CD4+、CD8+ in peripheral blood were determined, the insulin sensitivity index were calcnlated. Results Compared with the healthy normal body weight group, the blood fat,FPG,FPI,IL-18 of T2DM patients significantly increased, there were significant statistical difference(P < 0.05 or P < 0.01).The ISI significantly decreased(P < 0.01).Compared with the non-diabetic obese group, the blood fat and IL-18 of T2DM patients significantly increased (P<0.05).Compared with the diabetic obese group, the subpopulations cell population of CD3+、CD4+、CD8+ in the non-diabetic obese group significantly decreased, the ratio of CD4+/CD8+ significantly increased, there were significant statistical difference(P<0.05 or P<0.01)in the two groups comparison.Compared with the non-diabetic obese CD4+/CD8+ significantly increased(P<0.05).Conclusion T2DM patients should prevent the hyperinsulinemia, control blood sugar, reduce insulin resistance, sustain normal lipid metabolism, boost immune function.

Objective To investigate the effects of a training program based on virtual reality on static and dynamic balance perfor-mance in patients with Parkinson's disease (PD). Methods From June, 2014 to June, 2016, 46 patients with PD were randomly divided into control group (n=23) and experimental group (n=23). The control group received routine balance training, while the experimental group re-ceived balance training of virtual reality, for six weeks. They were assessed with Unified Parkinson's Disease Rating Scale part 3 (UP-DRS3), Berg Balance Scale (BBS), TimedUp and GoTest (TUGT) and Hamilton Depression Scale (HAMD) before and after training. The envelope area, anteroposterior standard deviation (AP-SD), mediolateral standard deviation (ML-SD) of centre of pressure (COP) were also measured with posturography. Results The scores of BBS, TUGT and HAMD improved in both groups after training (t>2.657, P<0.05), and improved more in the experimental group than in the control group (t>2.426, P<0.05). The score of UPDRS3 and the parameters of pos-turography improved in the experimental group (t>2.626, P<0.05), and improved more than that in the control group (t>2.112, P<0.05). Con-clusion Virtual reality rehabilitation is more effective than routine balance training on the static and dynamic balance function in patients with PD, and may release their depression.

OBJECTIVE: To establish an HPLC method for the content determination of prostaglandin A1 and prostaglandin B1 in Alprostadil for injection.METHODS: The determination was performed on Alltech Alltima C18 column with mobile phase consisted of phosphate puffer(pH=6.3)-acetonitrile-methanol(70 ∶ 25 ∶ 5) at a flow rate of 1.5 mL? min-1.The detection wavelength was set at 196 nm.The column temperature was set at room temperature and the injection volume was 20 ?L.RESULTS: The prostaglandin A1 and prostaglandin B1 were well separated from main component and other impurities. The linear range of prostaglandin A1 and prostaglandin B1 were 0.175~19.00 ?g?mL-1(r=0.999 7) and 0.23~19.90 ?g?mL-1(r=0.999 2).The contents of prostaglandin A1 in 3 batches of samples were 4.7%,4.9% and 4.3%,and the contents of prostaglandin B1 in 3 batches of samples were 0.6%,0.8% and 0.5% respectively.CONCLUSIONS: This method is proved to be simple, specific and suitable for the content determination of related substances in Alprostadil for injection.
