1.THE NURITIVE STUDY ON THE AMINO ACID MIXTURE SOLUTION FROM BEAN AND MAIZE EMBRYO DREGS BY MICROBIAL DEGRDATION
Acta Nutrimenta Sinica 1956;0(04):-
The amino acid mixture solution was prepared from bean and maize embryo dregs by microbial degradation. The solution contains eighteen kinds of amino acid. The eight kinds of essential amino acid was 41.5 per cent of the total amino acid mixture solution and the resultant ratio of amino acid, produce artio, utilization ratio of raw material protein and resultant ratio of amino acid nitrogen were 40, 18, 87.2 and 52.6 per cent respectively. The experimental animals were given basic food and amino acid mixture solution as a substitute for drinking water. The experiment was divided into four groups i.e. A (100% instead of the water), B(50%), C (33%), D (100% tap water). The growth rate, swimming time and antianoxic survival time of all test animals were investigated. The result showed that amino acid mixture solution from bean and maize embryo dregs might play a benefitial roles in animal growth rate, antifatigue, maintaning strength of anti-anoxia and increasing stamina.
2.Expression and antitumor activity of fusion protein RGD-TRAIL in Pichia pastoris.
Acta Pharmaceutica Sinica 2015;50(5):552-9
To compare the activity of RGD-TRAIL in different expression systems, RGD-TRAIL in both Escherichia coli (E.coli) and Pichia pastoris was constructed and expressed. In vitro activity of RGD-TRAIL from Pichia pastoris expression system was also analyzed. Genetic engineering techniques were used to construct recombinant plasmid pET30-rgd-trail and pHBM-rgd-trail. The recombinant protein RGD-TRAIL was purified with Ni ion affinity chromatography after induction. MTT assay, ELISA, scratch wound healing, transwell migration assay and Hoechst 33342 staining were performed to detect the effects of RGD-TRAIL on proliferation, binding activity, migration and apoptosis. The expression of apoptosis-associated proteins was detected by Western blotting. Recombinant protein RGD-TRAIL was successfully expressed in a form of inclusion body in E.coli, while expressed secretorily in Pichia pastoris. It possessed more potent cytotoxicity than RGD-TRAIL in E.coli by MTT assay. The RGD-TRAIL expressed by Pichia pastoris showed powerful binding affinity with cancer cells expressing α(v), DR4, DR5 and highly potent cytotoxicity through inducing apoptosis of cancer cells. Nuclear fragmentation was examined by Hoechst 33342 staining. Cleaved PARP and caspase-3 were also detected after incubation with RGD-TRAIL. Additionally, RGD-TRAIL inhibited migration significantly in A549 and HT1080 cells. The results demonstrate that Pichia pastoris expression system is more suitable for the recombinant protein RGD-TRAIL. Its binding affinity and antitumor activity might make RGD-TRAIL a promising candidate for cancer therapy.

90 bpm and nicardipine 10?g?kg-1 when MAP increased by 20% of the baseline value. Radial artery was cannulated for continuous BP monitoring and blood sampling. MAP, ECG, HR, SpO2 , PET CO2 and body temperature were continuously monitored during anesthesia. Blood samples were taken before induction of anesthesia (T1), when dura was cut (T2), when aneurysm was clipped (T3 ) and 30 min after clipping (T4 ) for determination of plasma concentrations of AT- Ⅱ , ET and CGRP by radioimmuno-assay.Results Plasma concentration of AT- Ⅱ and HR did not change significantly through out the study. Plasma CGRP concentration and MAP decreased significantly during operation at T2-4 as compared to the baseline values (T1 ) . Plasma ET concentration increased significantly at clipping (T3 ) as compared to that at T2. Conclusion The action of vaso-constrictors (AT- Ⅱ , ET) predominates over that of vaso-dilator during operation. It is important to prevent acute cerebral vasospasm during intracranial aneurysm clipping.
