Acta Anatomica Sinica 1953;0(01):-
After partial decortication of the parietal cortex of adult rats, division of neurons and glial cells as well as proliferation of macrophages, endothelial cells, pericytes, around the damaged cortical area were observed. The criteria for differentiating mitotic neurons from other cell components especially neuroglia are: (1) the cell body of a dividing neuron especially the pyramidal type is considerably larger than glial cells, and is slightly larger than the adjacent resting neurons of similar type; (2) a dividing neuron has characteristic cell processes, identifiable as axons and apical dendrites, a morphological feature of pyramidal cells; and (3) the dividing neurons contain Nissl granules. The neurons undergo mitosis were located around the lesion area, 37~500 ?m from its border of the lesion. The cortical lamina No. V has more mitotic neurons than any other layers; the explanation for which has been discussed.

Journal of China Pharmaceutical University 2019;50(4):389-396
Organelles have their special functions, they interact with each other and coordinate a series of important physiological functions at the same time. Organelle interaction occurs at membrane contact sites(MCSs), where the membranous organelle endoplasmic reticulum is the core, and specific tethered proteins at the membrane contact site bind to the organelle membrane and various protein complexes work together to perform specific functions, such as lipid transport, Ca2+ transfer, etc. This review studies on the structure and function of membrane contact sites and their key roles in organelle interactions, focusing on the connection between the endoplasmic reticulum and plasma membrane, mitochondria and Golgi, as well as the association between the key proteins at membrane contact sites and the occurrence and development of various diseases.

Journal of Xi'an Jiaotong University(Medical Sciences) 1982;0(04):-
Exon 3 termination mutation of phenylalanine hydroxylase (PAH) gene, the only identified one causing classical phenylketonuria (PKU) in Chinese, was detected in fourteen PKU children from Xi'an. The genomic DNA from these patients was amplified by polymerase chain reaction(PCR) and dot hybridied with specific oligonucleotide probes. This mutation is not present in any of these affected children, which indicates that phenylketonuria in Chinese may be caused by other mutations in phenylalanine hydroxylase locus. PCR amplification combining with oligonucleotide dot hybridization is technically feasible for prenatal diagnosis and carrier screening for PKU.

Objective To study the status of mouse fetal transgenic cells existed in the maternal blood in peri-natal period and provide the information for gene diagnosis using fetal cells circulated in the maternal blood. Methods Mating the female normal mice with male transgenic mice integrated with GFP reporter gene driven by ?-globin promoter and HS2-HS3 elements of LCR. Around the offspring were born, maternal blood was collected and GFP expression level was determined with FACS analysis. Meanwhile, DNA extracted from the tails of the mothers and their offspring as well as the maternal blood were analyzed by PCR.Results GFP positive fetal cells were not found in maternal blood before offspring were born. 1-3 weeks later, GFP positive fetal cells were detected and this population in maternal peripheral blood reached the peak. 4-5 weeks later, they disappeared gradually. PCR results showed no GFP positive band in the mothers. However, a positive fragment of GFP gene was amplified in maternal blood samples after their offspring were born. This result was in accordance with FACS analysis. Conclusion Transgene cells may be useful markers for the study of status of fetal cells existed in the maternal circulating blood. The results would be beneficial for the gene diagnosis using maternal blood as an alternative resource of fetal cells.

Objective To establish axenic cultivation of Pneumocystis carinii (P.c). Methods The organisms of P.c were isolated from the bronchoalveolar lavage fluid (BALF) of the rats with Pneumocystis carinii pneumonia (PCP) and cultured in a medium which was based on IMDM (GIBCO) supplemented with S-adenosyl-L-methionine, putrescine, N-acetyl glucosamine, putrescine, L-cysteine and L-glutamine, and newborn calf serum. The organisms cultured in the system were identified by observing the morphology of cysts in smears stained with Gomori's methenamine silver nitrate stain (GMS). Ultrastructure of the cysts/trophozoites was examined by transmission electron microscopy. The sequences of mitochondria] large ribosomal DNA subunit of the cutured organisms were compared with the Pneumocysti carinii f.sp. ratti variant isolate (GenBank No U20173) and Pneumocystis carinii f.sp.hominis (GenBank No M58605). Results Five isolates of P.carinii received from BALF of 8 rats with PCP were cultured axenically and continuously in the system. The cultured organisms could be stored in frozen condition and used to reinitiate culture, and were amplified by 19-22 times within 72 h. The morphology, ultrastructure and gene sequencing of the cultured organisms confirmed that the isolated organisms were P.carinii. Conclusion Five continuously and axenicly cultured isolates of P.carinii have been received.

Journal of Preventive Medicine 2023;35(2):116-120
Objective:
To examine the associations of sleep with overweight/obesity and central obesity in adults, so as to provide insights into improving sleep quality and preventing obesity in adults.
Methods:
Demographics, height, body weight, waist circumstance and sleep status were collected from the Hubei Provincial Surveillance Program for Adult Chronic Diseases and Their Risk Factors in 2020. Subjects' sleep condiction, overweight/obesity and central obesity were descriptively analyzed. The associations of sleep with overweight/obesity and central obesity were examined using a multivariable logistic regression model.
Results:
A total of 17 789 participants were recruited, with an average age of (56.21±13.05) years, 61.50% women, and mean duration of (7.18±1.56) h/d. There were 7 019 participants with snoring/asphyxia/suffocation (39.46%), 6 108 participants with sleep difficulty (34.34%), 8 064 participants with night waking at least twice (45.33%), 268 participants taking hypnotics (1.51%), and 6 267 participants with early morning awakening and difficulty in sleep again (35.23%), and there were 8 960 participants with overweight/obesity (50.37%) and 6 148 participants with central obesity (34.56%). Multivariable logistic regression analysis showed that sleep duration of <7 h/d (OR=1.081, 95%CI: 1.007-1.159), snoring/asphyxia/suffocation (OR=2.367, 95%CI: 2.222-2.521), and night waking at least twice (OR=1.106, 95%CI: 1.028-1.191) significantly correlated with overweight/obesity, and sleep duration of >8 h/d (OR=0.834, 95%CI: 0.761-0.913), snoring/asphyxia/suffocation (OR=2.153, 95%CI: 2.019-2.297), and night waking at least twice (OR=1.193, 95%CI: 1.105-1.288) were statistically associated with central obesity.
Conclusion
Sleep duration, snoring/asphyxia/suffocation and night waking are associated with overweight/obesity and central obesity.
