1.Relationship between Complications and Neurologic Subtype or Gross Motor Function in Preterm Infants with Cerebral Palsy

Objective To investigate the distribution of complications and the relationship with neurologic subtype and gross motor function in preterm infants with cerebral palsy (CP). Methods The type, grade of Gross Motor Function Classification System (GMFCS), intelligence,speech, ophthalmologic consultation, brainstem auditory evoked potential and electroencephalogram of 135 preterm infants with CP were reviewed. Results There were 284 complications in total, (2.10±1.33) per child, and was significantly different among various types of CP (F=5.50, P<0.001). The incidence of mental retardation and speech disorder was significant different among various types (P<0.05). The incidence of mental retardation, speech disorder, visual impairment and epilepsy increased significantly (P<0.05) in spastic quadriplegia infants,compared with those with diplegia and hemiplegia. The incidence of mental retardation, speech disorder, visual impairment and musculoskeletal disorder was significantly different (P<0.05) among various grades of GMFCS. The frequency of complications was more in children unable to walk (GMFCS Ⅳ~Ⅴ) than able to walk (GMFCS Ⅰ~Ⅲ) for children over 2 years old (t=70.05, P<0.001). Conclusion The incidence of mental retardation, speech disorder, visual and hearing impairment, secondary musculoskeletal disorder and the multiple disorders are related with neurologic subtype and/or the grade of GMFCS.
2.A MORPHOLOGICAL STUDY OF THE EFFECT OF ACUTE EXPERIMENTAL ISCHEMIA ON THE PYRAMIDAL CELLS OF CANINE CEREBRAL CORTEX

In the present study, 10 healthy adult dogs were used,8 for the experimental group and 2 for the normal group. We observed the histochemical and ultrastrac- tural changes of the enzymes in the pyramidal cells in the cerebral cortex under the condition of acute ischemia. The result demonstrated that the activity of LDH, AcP and AchE was increased, while the activity of SDH, MAO, ATP ase was decreased. The ultrastructural chnges showed that in the acute ischemia gromp, there were enlargement perinucleus spaces of neurone, brisement, disappearance and vasicularization of the mitochondrial crista. This indicated that acute ischemia had an obvious effect on the histochcmistry of enzymes and the ultra- structure of the neurone in the cerebral cortex.
3.Effect of deproteinised calf blood extract on experimental gastric ulcer in rats

AIM To observe the effect of deprot einised calf blood extract(DCBE) on experimental gastric ulcer in rats. METHODS Ulcer model of SD rats induced by acetic acid, half male and female, were divided into three group s randomly, the control group and the two treatment group DCBE (30 mg?kg -1 or 60 mg?kg -1 ). Following the two and four weeks of experiment, the gastric acid degr ee, activity of pepsin, area of ulcer, the number of tela leukocyte and activity of myeloperoxidase (MPO) were observed. RESULTS Gastric acid deg ree and activity of pepsin showed no significant difference between treatment gr oup and control group, either in DCBE 30 mg?kg -1 or in 60 mg?kg -1 , but area of ulcer, the number of tela leukocyte and the activity of myeloperoxi dase (MPO) showed a significant difference between treatment group and control group and the e ffects of 60 mg?kg -1 group were significantly decreased as compared with that of 30 mg?kg -1 group. CONCLUSION DCBE can significantl y promote tissue healing of gastric ulcer induced by acetic acid in SD rats.
4.Immunohistochemical analysis of the expression of Hes1 in adult mouse brain

BACKGROUND:In recent years,it has been shown in studies that regulation of the expression of Has1 is very important to maintain neural stem cells and regulate its differentiation during the development of embryonic nervous system.Moreover,upregulation of Hes1 expression is required for quiescent adult flbroblasts resuming proliferation.This indicates that there is a close relationship between Hes1 and some potential stem cells in adult.Therefore,the study of Hes1 expression in neural stem cells and its relationship with adult neurogenasis is put on the agenda.However,the expression of Hes1 in adult brain is still not clear.OBJECTIVE:To observe the expression of Hea1 in adult mouse brain and the cell types of Has1 positive cells.METHODS:Totally 12 adult male C57BL/6 mice were randomly evenly divided into two groups by using the random number table.For the first group,the mouse brain was removed directly and immunohistochemical staining was used to detect the expression of Has1 in the adult mouse brain.For the second group,Brdu was injected intraperitoneally to every mouse with a dosage of 200 mg/kg,once a day,for 3 days.The mouse brain was removed at 4 days and then double staining was carded out to observe and analysis the cell types of Hes1 positive cells in hippocampus.RESULTS AND CONCLUSION:Hes1 was expressed in all the observed anatomical site where neural cells exist,Brdu positive cells nearly all expressed Hea1,NeuN positive cells all expressed Has1,whereas GFAP positive cells did not express Hes1 completely.Hes1 was expressed in all neurons and neural stem cells,and glial cells did not express Hes1.
5.Effect of overexpression of DNA methyltransferase 3B4 gene on proliferation of 293A cells

Background and purpose:DNMT3B has nearly 40 known splice variants expressed in a tissue-and disease-speciifc manner, but the roles of these splice variants in the cell are still unclear. The aim of this study was to investigate the effects of overexpression of DNA methyltransferase 3B4 (DNMT3B4) gene on proliferation of human embryo kidney 293A cells. Methods:293A cells were transfected with plasmid pCMV-DNMT3B4 or pCMV-2B and then treated with G418 to get the stable cell line. The stable cell lines were determined for proliferation level by MTT method, and for cell cycle distribution by lfow cytometry. The expression of p21 was detected by real-time PCR and Western blot. The methylation status of p21 gene promoter was detected by methylation-speciifc PCR (MS-PCR). Results:The absorbance value in DNMT3B4-1 and DNMT3B4-2 clone were (58.92±3.47)%and (68.82±5.64)%as compared to 293A-vector cells using MTT method. DNMT3B4 overexpression signiifcantly decreased cell proliferation (P<0.05). S phase fraction of 293A-vector cells was (40.44±0.91)%. While in DNMT3B4-1 and DNMT3B4-2 clone cells, the S phase fraction was (35.88±2.00)%and (37.00±1.79)%respectively. Overexpression of DNMT3B4 could significantly decrease S phase fraction (P<0.05). The expression of p21 in DNMT3B4 overexpressed cells was increased, but the methylation status of p21 gene promoter was unchanged.Conclusion:Overexpression of DNMT3B4 can inhibit 293A cell proliferation and can facilitate p21 expression.
6.Alterations of Treg cells, Th17 cells and some related cytokines in peripheral blood of patients with hand-foot-mouth disease

Objective To investigate the alterations of Treg cells , Th17 cells and related cyto-kines in peripheral blood of patients during the early stage of hand-foot-mouth disease ( HFMD) .Methods Flow cytometry was performed to analyze the percentages of Treg cells ( CD4+CD25+Foxp3 T cells) and Th17 cells ( CD3+CD8-IL-17+T cells) in peripheral blood samples collected from 49 patients with severe HFMD , 26 patients with common HFMD and 30 healthy children.The levels of IL-6, IL-10, IL-17, IL-23 and TGF-β1 in serum samples were measured by ELISA .Results The percentages of Treg cells , ratios of Treg/Th17 cells, serum levels of TGF-β1 and IL-10 in patients with HFMD were significantly decreased as compared with those of control group (F=5.580, 6.205, 0.000, 0.014, respectively, P<0.05).Patients with se-vere HFMD showed a significantly increased Th17 cells (F=3.189 P<0.05) and a tendency of enhanced IL-17 expression , but no significant differences with the levels of IL-17 were observed .No significant differ-ences with the expression of IL-23 in the patients among each group were detected (P>0.05).The levels of IL-6 in serum samples from severe disease group were obviously increased as compared with those of common HFMD group and control group (F=7.318, P<0.05).Conclusion The results of this study demonstrated that the levels of Treg , Th17 cells and some related cytokines were varied in peripheral blood of patients dur-ing the early stage of HFMD .Inflammatory responses were enhanced to promote anti-virus activities by sup-pressing Treg cells and stimulating Th17 cells.
7.Evaluation of fetal cardiac function by brain natriuretic peptide and velocity vector imaging in congenital heart disease

Objective To investigate the clinical value of evaluation of fetal cardiac function in congenital heart disease by brain natriuretic peptide (BNP) and velocity vector imaging (VVI). Methods Fetuses who came from Shenzhen Maternity & Child healthcare Hospital were divided into the congenital heart disease group and the control group. At the same time we collected amniotic fluid and assayed BNP concentration. Using the VVI software, the velocity, strain and strain rate of the global and segmental of the left ventricle were measured. Comparison and correlation were made between the two groups. Results There was significantly difference of BNP concentrations in amniotic fluid between two groups. The gestational age had significant positive correlation with BNP concentrations in disease group. The comparison of global velocity, strain and strain rate of left ventricle between the two groups showed significant differences. All of the left ventricular dynamic parameters in disease group were lower than those of the control group. Conclusions Compared with the control group, the disease group had a high level of BNP in amniotic fluid and a lower level of dynamic parameters of left ventricular. There was a positive correlation between BNP concentration and gestational age in disease group. So we can conclude that theBNP concentration can be a biological parameter for evaluating the latent impairments of fetal cardiac function.
8.Longitudinal study of early neural development in premature infants with different gestational age and birth weight

Objective To investigate the trend of early neural development in premature infants. Methods At the age of 12 months and 24 months, Bayley Scales of Infant Development were used to assess the mental development index (MDI) and the psychomotor development index (PDI) in preterm (corrected age) and full-term infants. Results At 12 months, there was no significant difference in corrected age PDI scores among different gestational age groups (<32 , 32–33+6 and 34–36+6 weeks) (P=0.820). The actual age MDI and PDI scores of full-term infants and premature infants in 34~36+6 weeks group were significantly higher than those of premature infants in <32 and 32-33+6 weeks groups, and the PDI score of full-term infants was significantly higher than that of premature infants in 34-36+6 weeks group (P<0.05). There was no significant difference in actual age PDI scores among different birth weight groups (P=0.166). The actual age MDI and PDI of full-term infants and premature infants in birth weight≥2500 g group were significantly higher than those of premature infants in <1500 g, 1500~1999 g and 2000~2499 g groups (P<0.05). At 24 months, the actual age MDI scores of full-term infants were significantly higher than those of premature infants in different gestational age and birth weight groups (P<0.05). The actual age MDI curve of premature infants in birth weight <1500g group showed a downward trend, while the actual age PDI curve showed a significant upward trend. Conclusion The neurodevelopment of preterm infants at the corrected age of 12 and 24 months reaches the level of full-term infants.
9.Adipose-derived mesenchymal stem cells from hepatitis B virus infection patients differentiate into hepatocyte-like cellsin vitro

BACKGROUND: It has been reported that human adipose-derived mesenchymal stem cells can be induced to differentiate into hepatocyte-like cells with the function of albumin synthesis and urea secretion in vitro.OBJECTIVE: To investigate the potential of adipose-derived mesenchymal stem cells differentiating into hepatocyte-like cells in vitro.METHODS: Adipose-derived mesenchymal stem cells were isolated from the subcutaneous fat of hepatitis B virus infection patients by collagenase digestion and adherent method. Adipose-derived mesenchymal stem cells were induced by three-phase induction method and observed morphologically. The expression levels of alpha-fetoprotein, albumin and cytokeratin 18 were detected by immunohistochemical staining and glycogen synthesis function was detected by glycogen staining method.RESULTS AND CONCLUSION: Most of adipose-derived mesenchymal stem cells induced by three-phase induction method were differentiated into hepatocyte-like cells with polygonal morphology. Immunohistochemistry staining results showed that hepatocyte-like cells expressed alpha-fetoprotein, albumin and cytokeratin 18, and the expression levels of albumin and cytokeratin 18 increased with the culture time. The induced cells had the function of glycogen synthesis and were positive for periodic acid Schiff staining. These results showed that the subcutaneous adipose-derived mesenchymal stem cells could be induced into functional hepatocyte-like cells in hepatitis B virus infection patients.
10.The detection of c-myc,Bcl-2 protein and DNA ploidy in invasive breast carcinoma and its significance

Objective To investigate the clinical significance of c-myc、Bcl-2 protein expression, DNA ploidy and their relationship in breast cancer. MethodsWT5”BZ The expression of c-myc、Bcl-2 protein in 146 breast tumor tissues was examined by using immunohistochemical methods(S-P)and DNA ploidy in 72 cases of breast carcinoma by flow cytometry. ResultsKG1 Nuclear expression of c-myc protein was detected in 9 6% of tumors, and it was related to the DNA aneuploidy and prognosis. Cytoplasmic expression of c-myc protein was present in 91 8% of the carcinomas. Moderate to strong c-myc protein expression in cytoplasmic was associated with the positive status of ER. Bcl-2 protein was positive in 78 8% of cancers. Overexpression of Bcl-2 protein was associated with ER(+) status and the lack of axillary lymph node metastasis. DNA aneuploidy was found in 51 4% of 72 cancers, it was related to nuclear expression of c-myc protein and prognosis.WT5”HZConclusion Nuclear expression of c-myc protein and DNA ploidy are important prognostic factors.
