1.Seroconversion of serological test for syphilis in infants born to mothers with syphilis serofast reaction

Objective To explore the influence of anti-syphilis treatment to pregnant women with lower titer seroresistance on infantile serum,so as to provide the guidance for clinical diagnosis and treatment.Methods Totally 134 cases of pregnant women with lower titer syphilis serofast reaction were divided into treatment group (75 cases) and untreated group(59 cases) according to whether received anti-syphilis treatment during their pregnancy.The change of syphilis serology toluidine red unheated serum test (TRUST) and Treponema pallidum particle assay (TPPA) for the two groups mothers and babies were compared.Results (1) The first detection of TRUST titers between the two groups of pregnant women did not show statistically significant difference (x2 =0.520,P > 0.05).(2) Among the 134 neonates,20 cases(14.9%) were negetive for both TRUST and TPPA,23 cases (17.2%) were Treponema pallidum particle assay(TPPA) positive only,91 cases (67.9%) were positive for both TRUST and TPPA and showed lower or equivalent TRUST titers compared to their mothers,without significant differences between the two groups(x2 =0.892,P > 0.05).(3)In the two groups of babies with the same TRUST titers,the seroreversion time of TRUST showed no significant differences(P =0.229,0.309,1.000).The negative time of TRUST in infants with neonatal higher titer was later than those with neonatal lower titer in the two groups (all P < 0.05).The infant with TRUST + showed longer duration than those with neonatal TRUST-in TPPA seroreversion(all P < 0.05).The seroreversion time of TPPA in infants with neonatal TRUST titer of 1:4 in untreated group was later than that in treatment group[(14.1 ±1.4)months vs.(12.5 ±1.1)months,t =2.900,P =0.010].Conclusion The treatment for mothers with lower titer seroresistance in pregnant period had no influence in the positive rate of TRUST and TPPA in the neonates and seroreversion time of TRUST in infant.It may have certain effect to shorten the seroreversion time of TPPA in infant with high TRUST titer by the treatment.
2.Safty research of intraoperative intraperitoneal chemotherapy with raltitrexed for advanced colorectal cancer

Objective To investigate the intraoperative intraperitoneal chemotherapy security with raltitrexed in advanced colorectal cancer surgical operation.Methods Sixty patients with colorectal cancer undergone surgery were randomly divided into trial group (n =30) and control group (n =30) according to the random number table method.The trial group was given surgical operation plus with intraperitoneal chemotherapy with raltitrexed.The control group was given surgical operation plus with intraperitoneal saline perfusion.Theroutine blood test,liver and kidney functions,toxic side effects and complications in two groups before and after surgery were investigated.Results The white blood cells in trial group before and after surgery was (6.36 ± 2.63) × 109/L vs.(8.20 ± 2.08) × 109/L,with statistically significant difference (t =3.06,P <0.05).The ratio of absolute neutrophil count in trial group before and after surgery was 65.17% ± 10.36% vs.72.21% ± 10.53% (t =3.22,P < 0.05).The platelets in trial group before and after surgery was (261.03 ±84.74) × 109/L vs.(228.47 ± 58.69) × 109/L (t =2.07,P < 0.05).The white blood cells,the ratio of absolute neutrophil count and the platelets after surgery had no statistically significant differeuces between the two groups (P >0.05).The trial group had higher 1,2 level vomiting (60.00% vs.23.33%;x2 =8.30,P < 0.05),and nausea (30.00% vs.6.67%;x2 =5.46,P < 0.05) incidence rates,but there was no statistically significant difference in other toxic side effects (P > 0.05).The major complications post operation included intestinal obstruction,incision infection,abdominal cavity bleeding,and anastomotic fistula.There were equivalent complications in two groups (6.67% vs.3.33%,x2 =0.35,P >0.05;10.00% vs.6.67%,x2 =0.22,P>0.05;0 vs.0;3.33% vs.0,P>0.05).Conclusion For patients with advanced colorectal cancer,intraoperative intraperitoneal chemotherapy with raltitrexed is safe and feasible,and the adverse reactions can be tolerated without increasing postoperative complications.
3.Pathogenic detection of infectious diarrhea in patients in a district of Beijing from 2011 to 2013

Objective To analyze the distribution and pulsed-field gel electrophoresis (PFGE)of pathogenic bacteria causing infectious diarrhea in a district of Beijing from 2011 to 2013,and provide basis for tracing infection sources.Methods A total of 1 179 stool specimens of infectious diarrhea from patients in a diarrhea outpatient department from January 2011 to December 2013 were collected,all isolated pathogens were identified by serotyping and PFGE analysis.Results 330 enteric pathogens were isolated from 1 179 specimens,the top 4 bacteria were Shi-gella spp .(28.18%,n=93),Salmonella spp .(20.91 %,n=69),Vibrio parahaemolyticus (13.33%,n =44),and diarrheagenic Escherichia coli (3.33%,n = 11 ).18 Shigella sonnei isolates were identified as 8 PFGE patterns, clustering similarity was close to 88%;69 Salmonella spp .strains belonged to 18 serotypes and 41 PFGE patterns, Salmonella senftenberg and Salmonella enteritidis had dominant patterns;no dominant PFGE patterns were obviously identified among 23 strains ofVibrio parahaemolyticus .Conclusion The serotypes and PFGE patterns of pathogenic bacteria in infectious diarrhea in past three years showed a wide distribution characteristics,the dominant PFGE patterns of Salmonella spp .and Shigella spp .need to be paid more attention,and outbreak of infectious diarrhea caused by Salmonella spp .and Shigella spp .should be alerted.
4.Association study between WNK1 gene polymorphisms and ischemic stroke in Uygur

Objective To explore the association between single nucleotide polymorphisms (SNPs) in WNK1 gene and ischemic stroke in Uygur population. Methods Ten tagging single nucleotide polymorphisms (tSNPs) of the WNK1 gene in 295 ischemic stroke patients and 318 control subjects were genotyped,and the association between these tSNPs and ischemic stroke were conducted.The 10 tSNPs were rs3858703, rs11611246, rs7305065, rs1990021, rs34408667, rs12309274, rs1012729, rs956868,rs12828016 and rs953361. They were determined by the Multiplex SNaPshot platform. All data were analyzed using t-test,x2-test and Logistic regression. Linkage disequilibrium and Haplotype were analyzed by Haploview software.Results There were no significant differences between cases (25.6%) and controls(30.0% ) of the 10 tSNPs in WNK1 gene.When the samples were further stratified according to gender,rs11611246 T allele was found to be associated with a reduced risk of ischemic stroke,with a per-allele OR of 0.448(95% CI 0.269-0.746,P =0.002) in female cases than in female controls. The significance remained after adjustment for the covariates of age,and for the covariates of age,BMI,cigarette smoking,alcohol drinking,hypertension,diabetes and hyperlipidemia.In addition,no association between the other 9 tSNPs and ischemic stroke were found in Uygur subjects.Conclusion The study reports a new genetic variant,rs11611246,located in the fourth intron of the WNK1 gene,decreasing the risk of ischemic stroke in Uygur population.The T allele might be the protecting factor of ischemic stroke in female Uygur.
5.Multi-residue determination of 11 pesticides in Shengmaiyin prescription by gas chromatography.

OBJECTIVETo determine the pesticide residues in Shengmaiyin.
METHODThe residues were simultaneously identified and quantified by capillary gas chromatography. The capillary column was OV-1701(0. 32 mm x 30 m, 0.5 microm), splitless injection and Agilent 63Ni electron capture detector ECD were adopted. The carrier gas were high-pure nitrogen, the flow rate were 60 mL x min(-1), the injector temperature was 260 degrees C, and the detector temperature was 300 degrees C.
RESULTIt showed a good linear relationship between peak area and pesticide concentration for 11 kinds of pesticides in the detection concentration range. All correlation coefficients were higher than 0.996 2. The average recovery was 83.1% -114.7%.
CONCLUSIONA simple and efficient monitoring method of pesticide residues is established which provides the foundation for the safe use and analysis of Shengmaiyin.

6.Serum vitamin D levels of atopic dermatitis in adolescents and its clinical significance

Objective:To investigate the relationship between the serum level of vitamin D and severity of atopic dermatitis (AD) in adolescents.Methods:From March 2016 to February 2017, 102 adolescents with AD(AD group) and 100 healthy adolescents(control group) were selected.The distribution and serum levels of vitamin D in the two groups were compared.According to SCORing atopic dermatitis (SCORAD) score, AD patients were divided into mild group, moderate group and severe group.The levels of 25-(OH)D 3, total IgE (TIgE) and peripheral blood eosinophil cells percentage(EOS%) were detected in the three groups. Results:There was no statistically significant difference in the proportion of patients with vitamin D deficiency, insufficiency and sufficiency between the AD group and control group(χ 2=2.718, P=0.257). The serum level of 25(OH)D 3 in the AD group was lower than that in the control group[(18.43±3.73)μg/L vs.(19.47±3.27)μg/L, t=2.112, P=0.036]. The mean values of vitamin D deficiency, insufficiency and sufficiency in the AD group were (11.11±1.65)μg/L, (17.49±1.69)μg/L, (21.73±1.17)μg/L, respectively, which in the control group were (12.86±1.47)μg/L, (17.55±1.34)μg/L, (22.05±1.32)μg/L, respectively.The level of vitamin D deficiency in the AD group was significantly lower than that in the control group ( t=2.588, P=0.017), but there were no statistically significant differences in vitamin D insufficiency and sufficiency between the AD group and control group.In 102 AD patients, 40 cases were mild, 29 cases were moderate and 33 cases were severe.The levels of 25-(OH)D 3, TIgE and peripheral blood EOS% had statistically significant differences among the three groups ( F=6.315, 35.813, 31.285, all P<0.01). The 25-(OH)D 3 level of the severe group was lower than that of the mild group( t=3.64, P=0.097). However, there was no statistically significant difference between the severe and moderate groups and moderate and mild groups ( P>0.05). Regarding the TIgE level and EOS%, severe group>moderate group>mild group( t=8.318, 8.788, all P<0.01; t=4.322, 4.784, all P<0.01). The value of 25-(OH)D 3 was negatively correlated with the TIgE level( r=-302, P<0.01) and EOS%( r=-508, P<0.01) in the AD group. Conclusion:Vitamin D deficiency or insufficiency exists in adolescents with AD.Low level of vitamin D is correlated with high TIgE level and EOS%.The severity of AD is closely correlated with increased serum levels of TlgE and EOS%, as well as decreased serum levels of vitamin D.
7. Clipping for large paraclinoid internal carotid artery aneurysms by non-fluoroscopic occlusion via a micro-bone window frontolateral approach

Objective:
To evaluate the clinical value and outcomes of technical improvement of hybrid operatical clipping for large paraclinoid internal carotid artery aneurysms.
Methods:
A review was conducted on 18 cases of large paraclinoid internal carotid artery aneurysm which were clipped by balloon non-fluoroscopic occlusion of the parent artery via a micro-bone window frontolateral approach in hybrid operating room at Neurosurgery Department of Tianjin Medical University General Hospital from June 2014 to December 2017. There were 8 males and 10 females with age of (63±4) years. There were 6 cases of unruptured aneurysm and 12 cases of ruptured aneurysm of subarachnoid hemorrhage (6 cases of grade Ⅱ, 4 cases of grade Ⅲ and 2 cases of grade Ⅳ in Hunt-Hess classification). Frontolateral approach incision (average length of about 5 cm) and bone window about 3 cm×3 cm were performed. No incision of the neck was needed to expose the internal carotid artery for temporary occlusion. In the operation, the balloon was slowly pushed to the preset position of the internal carotid artery under non-fluoroscopy. The balloon was expanded to block the blood flow of internal carotid artery. Then aneurysm was clipped. The balloon was loosened and retraced to the guiding catheter after clipping. The clipping condition was examined by cerebral angiography. If there was residual aneurysm neck or stenosis of the parent artery, the balloon was pushed under non-fluoroscopy again to temporary occlusion and the clip was adjusted until the aneurysm neck was clamped satisfactorily.
Results:
Eighteen aneurysms were successfully clipped in hybrid operating room. Fourteen aneurysms showed complete occlusion of the aneurysm neck and no stenosis of the parent artery. Four cases showed residual aneurysm neck after clipping by intraoperative angiography, then aneurysms were clipped satisfy by adjusting the aneurysm clip. The patients were followed up for 3 months to 1 year. Ten patients recovered well (modifed Rankin score (mRS): 0), and 3 patients had no obvious disability (mRS: 1). Two patients with Hunt-Hess grade Ⅲ were slightly disabled (mRS: 2). 1 patients with Hunt-Hess grade Ⅲ were moderately disabled (mRS: 3). 1 patients with Hunt-Hess grade Ⅳ were severely disabled (mRS: 4). One elderly patients with Hunt-Hess grade Ⅳ were seriously disabled (mRS: 5).

8.The Effects of Nonylphenol on Calcium Signaling and Cell Proliferation in H9c2 Cell and Its Mechanism

Objective To observe the effects of nonylphenol(NP)on the intracellular calcium concentration changes and cell proliferation,and the involvement of GPR30 receptor in H9c2 cell. Methods The intracellular calcium concentration changes were recorded by using intracellular calcium determination method and cell proliferation was observed by MTT method in H9c2 cell. Results NP(1×10-10 mol/L)increased the intra?cellular calcium concentration changing amplitude and promoted the proliferation of H9c2 cells,while NP(1×10-6 mol/L)decreased intracellular calcium concentration changing amplitude and suppressed cell proliferation. G15 could block the promoting effect of 1×10-10 mol/L NP on the intracel?lular calcium concentration and cell proliferation,but could not block the inhibition of 1×10-6 mol/L NP on the intracellular calcium increase and cell proliferation. Conclusion The results indicate that NP affect rapid calcium signal changes and cell proliferation in non?monotonic dose dependent manner,and its mechanism may be due to the different involvement of GPR30 receptor in different concentrations.
9.Impact of natural selection on the polymorphism of HLA-G 3'UTR among five ethnic Chinese populations.

OBJECTIVETo assess the impact of natural selection and genetic background on the polymorphisms of HLA-G 3-untranslated regions (UTR) among five ethnic Chinese populations.
METHODSPCR and DNA sequencing were used to determine the polymorphisms among 432 individuals from the five ethnic populations. Their genetic background was determined by genotyping of 10 short tandem repeats (STRs).
RESULTSEight variations were identified among Gelao, Mongolian and Kirgiz populations, while only 7 were found in Shui and Dai people. For all 3 southern populations (Gelao, Shui, and Dai), the observed heterozygosites (Ho) was higher than expected heterozygosities (He). But this was reversed for the 2 northern populations (Mongolian and Kirgiz). The Ho and He of the 10 neutral STRs were in random distribution. Ewens-Watterson testing based on haplotypes of the HLA-G 3'UTR has suggested that a natural selection had occurred in the region where Dai and Shui had inhabited, but not in the northern region where Mongolian and Kirgiz population inhabited. Polygenetic trees based on the HLA and STRs were also different.
CONCLUSIONThe HLA-G 3'UTR of Dai and Shui people who lived in southern China may have subjected to a selection pressure. Based on current knowledge, this pressure may have been driven by a pathogenic selection.

10.CD301b+ macrophage: the new booster for activating bone regeneration in periodontitis treatment.

Periodontal bone regeneration is a major challenge in the treatment of periodontitis. Currently the main obstacle is the difficulty of restoring the regenerative vitality of periodontal osteoblast lineages suppressed by inflammation, via conventional treatment. CD301b+ macrophages were recently identified as a subpopulation that is characteristic of a regenerative environment, but their role in periodontal bone repair has not been reported. The current study indicates that CD301b+ macrophages may be a constituent component of periodontal bone repair, and that they are devoted to bone formation in the resolving phase of periodontitis. Transcriptome sequencing suggested that CD301b+ macrophages could positively regulate osteogenesis-related processes. In vitro, CD301b+ macrophages could be induced by interleukin 4 (IL-4) unless proinflammatory cytokines such as interleukin 1β (IL-1β) and tumor necrosis factor α (TNF-α) were present. Mechanistically, CD301b+ macrophages promoted osteoblast differentiation via insulin-like growth factor 1 (IGF-1)/thymoma viral proto-oncogene 1 (Akt)/mammalian target of rapamycin (mTOR) signaling. An osteogenic inducible nano-capsule (OINC) consisting of a gold nanocage loaded with IL-4 as the "core" and mouse neutrophil membrane as the "shell" was designed. When injected into periodontal tissue, OINCs first absorbed proinflammatory cytokines in inflamed periodontal tissue, then released IL-4 controlled by far-red irradiation. These events collectively promoted CD301b+ macrophage enrichment, which further boosted periodontal bone regeneration. The current study highlights the osteoinductive role of CD301b+ macrophages, and suggests a CD301b+ macrophage-targeted induction strategy based on biomimetic nano-capsules for improved therapeutic efficacy, which may also provide a potential therapeutic target and strategy for other inflammatory bone diseases.
