1.The effect of nitriglycerin on ET/NO and TXA2/PGI2 in severe acute pancreatitis rats

Objective To explore the effect of nitroglycerin on ET/NO, TXA2/PGI2 and pancreas pathomorphology changes in severe acute pancreatitis (SAP) rats. Methods Sixty SD rats were random divided into five groups, including control group (A group, n = 12) and experimental group(B,C,D and E group, n = 12). The SAP was induced by injection of 5% sodium taurocholate through retrograde common biliopancreatic ducts via duodenal papilla with epidural catheter. Group C, D and E were intravenously injected with nitroglycerin 0.5μg/kg/min,1μg/kg/min and 2μg/min in 30 min respectively, and group A and B was injected with Sodium Chloride 0.5ml. The indexes of changed pathomorphology and ET/NO, TXB2/6-keto-PGF1a, were determined at the 6th and 12th hour after operations, respectively. Results The specimen data of the 6th and 12th hour displayed that the indexes of changed pathomorphology, ET, ET/NO, TXB2, and TXB2/6-keto-PGF1a of the group C,D and E degraded respectively, compared to group B(P < 0.05). Conclusion The nitroglycerin could degrade ET, ET/NO,TXA2 and TXA2/PGI2, improve the microcirculation of pancreas, and delay the pathological inflammation change in SAP rats.

BACKGROUND:High-dose antibiotics for bone infection have many adverse reactions, and its outcomes are not perfect. Thus, to explore a degradable material as a vector to prevent bone infection is valuable. OBJECTIVE:To study drug release characteristics of gatifloxa-poly sebacic anhydride local control ed release system in vivo. METHODS:A 3 mm × 6 mm bone window was made at right knee joint of New Zealand rabbits. The gatifloxacin-poly sebacic anhydride sustained release preparation was implanted. Heart blood, bone tissue and myeloid tissue specimens were obtained at 1, 2, 3, 6, 9, 12, 15, 18, 25 and 30 days after surgery. High-performance liquid chromatography was utilized to determine gatifloxacin concentration. Scanning electron microscope was employed to observe the structural changes before and after implantation of gatifloxacin-poly sebacic anhydride sustained release preparation. RESULTS AND CONCLUSION:After implantation of gatifloxacin-poly sebacic anhydride sustained release preparation, drug concentration gradual y decreased in the myeloid tissue, peaked at 1 day, stabilized at 3-15 days, gradual y reduced at 15-30 days. However, the drug concentration was stil higher than the minimal inhibitory concentration 0.1 mg/L against Staphylococcus aureus at 30 days. The peak of drug concentration in the bone tissue occurred at 3 days, and stabilized at other days, which was higher than 0.1 mg/L. At the same time point, drug concentration in the blood specimen was lower than that in the myeloid tissue and bone tissue. The degradation of gatifloxacin-poly sebacic anhydride sustained release preparation was surface erosion, and the shape of the degradation residue is smal globular. The change of the internal structure of gatifloxacin-poly sebacic anhydride sustained release preparation was not found. In the drug release procedure, gatifloxacin-poly sebacic anhydride sustained release preparation did not show disintegration or fragmentation. These results indicated that
gatifloxacin-poly sebacic anhydride local sustained release preparation has good abilities of drug load and drug release.
4.Relationship between serum retinol-binding protein 4 and non-alcoholic fatty liver disease in schizophrenia

Objective To investigate the serum retinol-binding protein 4 (RBP4) level in olanzapinetreated schizophrenia with non-alcoholic fatty liver disease (NAFLD) via a case-control study and to explore its relationship with NAFLD.Methods Schizophrenia receiving olanzapine treatment,with or without NAFLD were enrolled as cases (n=60) or controls (n=60).The serum retinol-binding protein 4,metabolic syndrome component including body mass index (BMI),waist-to-hip ratio (WHR),fasting plasma glucose (FPG),total cholesterol (TG),total glycerin (TC) and blood pressure were assayed.TyG index,CT value ratio of liver and spleen were used for insulin resistance or NAFLD diagnosis and severity assessment respectively.Results The serum level of RBP4 was significantly higher in cases than that in controls ((70± 13)mg/L vs (52±12)mg/L ; t =4.943,P<0.01) and the same result was showed after influencing factor adjustment thru covariance analysis (F=16.797,P<0.01).Moreover,cases had significantly higher TyG index,BMI,WHR,FPG,TG and systolic pressure (t=2.383-4.300,P<0.05-0.01).Cases with severe or moderate NAFLD had significantly elevated serum RBP4 level compared with mild cases((79± 16) mg/L,(72±8) mg/L vs (63t9) mg/L,d =16.2,9.9 ; P<0.01,0.05).A significantly negative correlation between RBP4 level and the CT value ratio of liver and spleen (r=-0.244,P<0.05),and a significantly positive correlation between RBP4 level and TyG index,BMI,WHR,TG(r=0.197-0.244,all P<0.05) were observed.The partial coefficient between RBP4 level and CT value of liver and spleen was significant with influence adjusted(r=-0.453,P<0.01).Logistic regression showed that serum RBP4 level(β3=0.105,P<0.01)and TyG index,hyperlipidemia,obesity,central obesity or high blood pressure (β =1.288-9.711,P< 0.05-0.01) were closely associated with NAFLD.Conclusion A heighten serum RBP4 level may be one of NAFLD risk factors in schizophrenia treated with olanzapine.
5.Clinical study on intra-abdominal hypertension secondary to fulminant acute pancreatitis

Objective To study the treatment of intra-abdominal hypertension syndrome (IAHS) secondary to fulminant acute pancreatitis(FAP). MethodsWe retrospectively analyse therapeutic results of 14 IAHS cases secondary to FAP during the period of 1998~2003. ResultsFour out of the 6 cases receiving nonoperative therapy died with motality rate of 67.7% (4/6). Two out of the 8 cases treated by early surgery died. The motality rate was 25% (2/8), which was significantly different from that treated conservatively . ConclusionModerate to severe IAHS cases secondary to fulminant acute pancreatitis should undergo exploration in the early phase of disease to improve the prognosis of FAP.
6.Application of the Animal Energy Metabolism Monitoring System (AEMMS) in Acute Alcoholic Liver Injury with the Treatment of Acupuncture

This study aimed to explore the value of animal energy metabolism monitoring system (AEMMS) to evaluate the energy metabolism of acute alcohol liver injury in rat with the treatment of acupuncture.Tirty rats were randomly divided into the control group,the model group and the EA group.The model group and the EA group were gavaged with 50% ethanol for the establishment of the animal model of acute alcoholic liver injury.EA was given by needling Zusanli (ST-36) 30 min after intragastic administration every day in the EA group.The treatment of acupuncture continued 3 days.In addition,the RER,heat,feeding and drinking were continuously recorded for 96 h in each group at the same time by AEMMS.As a result,the RER,feeding and drinking indices significantly decreased in rats with ALI with an increase in the heat index.There was an up-going tendency of the RER,feeding and drinking indices,and a declined tendency of the heat index in the model rats after EA treatment.It was concluded that AEMMS presented a practical value in the study of the energy metabolism of rats with ALI with the treatment of acupuncture.
7.Total mesopancreas excision in radical resection of pancreatic head carcinoma

Objective To evaluate the concept of artery first and total mesopancreatic excision in radical resection of pancreatic head carcinoma through both anterior and posterior approaches.Method The anterior approach was to identify the superior mesenteric artery (SMA) and the posterior approach to confirm the possibility of negative margin at the origin of SMA,on the posterio-lateral vascular wall of superior mesenteric vein (SMV) and the supposed posterior of the mesopancreas.The resection scope were with the celiac trunk and common hepatic artery as the upper boarder,the SMA as the left boarder,the inferior mesenteric vein (IMA) level as the lower boarder,to achieve a complete mesopancreatic excision,namely the en bloc resection of all the involved nerve,the lymph tissue and vascular tissue along the right side of the axial composed by SMA and celiac trunk.Results Of the 15 patients,11 had radical Whipple procedure,among which 2 had a combining SMV resection and reconstruction.1 case suffered from delayed gastric emptying and 2 cases from bile leakage.There was no mortality.The postoperative pathology reported carcinoma in all 11 cases,with duodenum and low bile duct involved in 4 cases,with the duodenum involved in 6 cases,no surrounding tissue involvement was identified in 1 case.Nerve involvement was found in 7 (7/11),vascular involvement in 10 (10/11),and lymphnode metastasis was (2.5 ± 3.8/12.9 ± 4.9).Conclusions The radical resection of pancreatic head carcinoma using the concept of artery first and the total mesopancreatic excision is helpful for an early evaluation of the possibility of radical resection and guarantees negative margins.
8.Acute pulmonary injury caused by echinococcus granulosus cyst fluid

AIM: to establish an animals model of pulmonary embolism caused by echinococcus granulosus (E.g)cyst fluid. METHODS: Cyst fluid were isolated from sheep liver . Twenty-one rabbits were randomized into 3 groups: group Ⅰ(saline group), group Ⅱ(clear cyst fluid group), group Ⅲ (sand-contained cyst fluid group). Operation was performed on each animal to place a femoral artery catheter and a femoral vein catheter. Saline, clear cyst fluid or sand-contained cyst fluid were given to rabbits at a dose of 3 mL/kg body weigh . Observations were then made at 10, 30, 60 minutes to determine the changes of MAP, blood gas and vaso-active substance. Then ECT scanning image was obtained .After observation, all animals were killed and the lungs were harvested for histological examination by light microscopy. 9.Comparison of gene frequencies of short tandem repeats loci on Y Chromosome between offenders with initiative-aggressive behavior and impulsive-aggressive behavior

Objective To explore the distributional differences of the gene frequencies of 22 short tandem repeats loci on Y Chromosome(Y?STRs) between offenders with Initiative?aggressive behavior and impulsive?aggressive behavior,and to probe into the genetic factors of initiative?aggressive behavior and im?pulsive?aggressive behavior. Methods Biological samples of 271 offenders with initiative?aggressive behav?ior and 271 offenders with impulsive?aggressive behavior were collected and PCR compound amplification was carried out with the aid of PowerPlex Y23 System. Then the PCR products were subjected to electrophoresis and gene detection with AB3500xL gene analysis system so as to calculate and compare the alleles and haplo?types of 22 Y?STRs gene frequency in the two groups. Results The distribution of allele frequency were sig?nificantly difference in locus DYS437(P=0.022) between two groups,not in the other 21 Y?STRs loci( all P>0.05) . Univarite analysis showed significant differences at allelle 14 in locus DYS437 between both groups ( initiative?aggressive behavior group:69. 37%;impulsive?aggressive behavior group:58. 67%; P=0. 009 ) . Conclusion Loci DYS437 may be associated with aggressive behavior. In the group of aggressive behavior, allelle 14 on locus DYS437 may be the susceptible factor of initiative?aggressive behavior and the resistant factor of impulsive?aggressive.
10.Comorbidity of schizophrenia and depression disorder based upon differential expression of microRNA

Objective To investigate the differential expression of microRNA (miRNA) in schizophrenia (SZ) patients,and explore the comorbidity of SZ and depression disorder based upon miRNA expression.Methods Affymetrix array analysis was used to investigate the differentially expressed miRNA in SZ patients firstly,and then quantitative real-time polymerase chain reaction (qRT-PCR) was further carried out to confirm the selected miRNA in peripheral blood mononuclear cells of 40 SZ patients,whom were administered by Positive and Negative Syndrome Scale (PANSS) and the selected miRNAs in depression disorder patients has also been confirmed by Affymetrix array analysis and qRT-PCR in our previous studies.Results Affymetrix array analysis indicated that there existed 33 miRNAs which differentially expressed (32 up-regulated and 1 down-regulated) compared with normal controls.qRT-PCR results suggested that the expression of 8 miRNAs (miR-1273d,miR-1303,miR-3064-5p,miR3131,miR-3687,miR-4428,miR-4725-3p and miR-5096) were significantly up-regulated in SZ;the miRNA differentially expressed in depression disorder patients also had differential expression in SZ patients (P<0.05).There were significant correlation between the miRNAs differentially expressed in depression disorder patients and in SZ patients (P<0.01).MiR-1972 differentially expressed in depression disorder patients had significant positive correlation with the positive symptoms of PANSS (P<0.05),and miR-26b was positively correlated with composite factor (P<0.05).Conclusion Comorbidity of SZ and depression disorder is observed not only on the clinical symptoms,but on the molecular genetic basis.
