1.Treatment outcome of mucoepidermoid carcinoma in 128 cases

Objectives: To study the treatment outcome of mucoepidermoid carcinoma(MEC).Methods:128 cases of MEC with definite pathologic diagnosis treated in our hospital during the past 15 years were investigated. The relationship between the effects of the therapy and the prognosis was analysed.Result:Treatment with surgical operation only was used in 99 cases,radiotherapy after operation was applied in 26 cases and follow-up was carried out in 76 cases. 5-year survival rate was 90.7%.5 patients who died within 5 years after operation were all with the diagnosis of MEC in clinical stage Ⅳand poor differentiaton.Conclusion:The local extensive excision for the well differentiated MEC,the radiotherapy after operation for poorly differentiated or that of clinical stage Ⅳ,especially in maxilla,may increase 5-year survival rate.Since there were cases of recurrences of MEC 13 years after surgical treatment,long-term follow-up is necessary.
2.The effects of rosuvastatin on the liver and kidney function and muscle pathology in rats

ObjectiveTo study the liver and kidney function and muscle effects of rosuvastatin in rats.MethodsIn this study,twenty healthy male rats (9-month-old) were randomly divided into two groups:normal control group and rosuvastatin treatment group.The first treated group was given conventional feed; The second treated group was given conventional feed and oral rosuvastatin [5mg/( kg · d)] of 12 weeks.The level of liver and kidney function and creatine kinase,histological changes of liver and muscle were examined before and after the treatment.ResultsOur findings demonstrate that the level of liver and kidney function and creatine kinase was no significant difference among the two experimental groups,also no difference before and after treatment( P >0.05).At the same time,liver and muscle showed no abnormal pathology.Conclusion These findings collectively indicate that rosuvastatin shows no significant side effects in the liver and kidney function and muscle pathology compared with control group.
3.Role of ageing on rat endothelial dysfunction

Objective Alterations in vascular structure and function and impairment of associated endothelial cells with aging are one of the independent risk factors in the process of atherosclerotic vascular diseases.The study of age-related endothelial dysfunction plays a critical role in preventing as well as treating heart and cerebral vascular diseases in the elderly.Methods The healthy male rats were divided into the young group (3-month-old),the adult group (9-month-old) and the middle-aged group (15-monthold).The indicators associated with vascular endothelial function,including plasma NO,eNOS,iNOS,ET-1 and aortic NO,eNOS,iNOS,were quantified to evaluate the endothelium-dependent and non-endothelium-dependent vasodilating function of isolated aortic rings.These morphological changes of rat aorta helped to find out the group in which endothelial function changed in the early stage.Results Plasma NO,eNOS,iNOS and aortic NOS activity in the young group,the adult group and the middle-aged group gradually decreased with increasing age (P < 0.05).NO,eNOS and iNOS respectively displayed a negative correlation to age.ET-1 gradually increased with aging (P < 0.05) and showed a predominantly positive correlation to age.Acetylcholine-induced greatest vasodilation of aortic rings lessened with increasing age (P < 0.05).Vasodilation in each group reacted generally consistently with nitroprusside sodium (P >0.05).Aortic morphology,including intima media thickness (T),diameter (D) and intima media thickness / inner diameter (T / D),gradually enhanced with aging (P < 0.05).T,D was positively correlated with age.NO,eNOS,iNOS,ET-1 of the adult group showed maximum slope (P <0.05 respectively) according to group regression analysis,suggesting that vascular endothelial function changed in this group.Meanwhile,T and D of the middle-aged group showed maximum slope (P < 0.05 respectively),suggesting that age-related vascular structural changes predominantly occurred in this period and vascular function changed earlier than the structure.Conclusion Our study demonstrated vascular endothelial dysfunction with aging.Early evident changes of endothelial function occurred in the adult group,which impaired endothelium-dependent vasodilating function.Vascular function changed earlier than the structure.

BACKGROUND:Bone mesenchymal stem cells(BMSCs) can accelerate vascularization of myocardial cell,relieve myocardial remodeling,and improve heart function.However,whether its paracrine action can facilitate the differentiation of CD117+ cardiac stem cells(CSCs) by effecting transforming growth factor-? Ⅲ(TGF-? Ⅲ) receptor expression is still unknown.OBJECTIVE:To study the influence of BMSCs on the expression of TGF-? Ⅲ receptor in the process of differentiation of CD117+ CSCs.DESIGN,TIME AND SETTING:In vitro cytology contract observation.The experiment was performed at the Xinhua Hospital from February 2008 to February 2009.MATERIALS:Neonatal rats,weighing 5-8 g,were used to prepare CSCs.Sprague Dawley rats,4 weeks old,weighing 200-250 g,were prepared for BMSCs.METHODS:CD117+CSCs were obtained from neonatal rats by explant cultures and magnetic bead cell sorting.Immunohistochemical experiment was performed to identify specificity of CSCs.CD117+CSCs suspension were synchronization without serum for 24 hours,washed by PBS for 3 times,and then cultured with DMEM/F12,followed by inoculation into 6-well plate.The cells were divided into the control and co-culture groups.In the control group,CSCs were induced differentiation by cardiac spheres,and Transwell chamber were used in the co-culture group.MAIN OUTCOME MEASURES:The growth of CSCs was observed by inverted microscope.The changes of cardiac Troponin T(cTnT),connexin43,TGF-? Ⅲ receptor,smad2 and phospho-smad2 expressions were detected by western blot at days 1,3,5 and 7 after culture.RESULTS:①CSCs isolated by explants cultures and MACS expressed CD117,which grew in disperse with smaller body.② The expression of cTnT and connexin43 were significantly higher in the co-culture group than that of the control group at days 5 and 7(P
5.Detection of Escherichia coli O157:H7 by multiplex PCR-DHPLC

Objective To develop the PCR-denaturing high performance liquid chromatography (DHPLC) for detection of E. Coli O157: H7. Methods The virulence genes of Shiga-like toxin(SLT) and rfbE were specifically amplified by 2 sets of primers. The target gene fragments of the PCR assay were 224 bp and 499 bp, respectively. Results Analysis of 37 strains demonstrated that this PCR system was specific. The detection limit of the PCR was 4 CFU/ml. Conclusion These results indicated that the multiplex PCR-DHPLC assay can be used for specific and sensitive detection of E. Coli O157:H7.

Objective To investigate the prevalence of hyperuricemia and its relationship with hy-perlipaemia and hyperglycemia in the elderly .Methods We chose the elderly people whose age were over 60 years and who had routine physical examine during August and September in 2009 , the number of them is 1321.Hyperuricemia is defined as the level of serum uric acid (UA) >416 mmol/L.Hyperlipaemia is defined as the level of serum total cholesterol ( TC)≥5.18 mmol/L; triglyceride ( TG)≥1.70 mmol/L;high-density lipoprotein ( HDL) <1.04 mmol/L and low-density lipoprotein ( LDL)≥3.37 mmol/L.Hy-perglycemia is defined as the level of fasting blood glucose ( FBG)≥6.1 mmol/L.Results There were 344 patients with hyperuricemia accounting for 26.04%.With increasing age , the propotion of the patients with hyperuricemia and the mean levels of UA , TC, TG and LDL respectively showed gradually rising ( P<0.05 respectively).The incidence of hyperlipaemia and hyperglycemia was higher in the hyperuricemia group than those in the controls ( P <0.05 respectively ) .Hyperuricemia in the elderly displayed a positive correlation to TC, TG, LDL and FBG( r =0.9954,0.9805,0.9715,0.9682, P <0.05).Conclusion Hyperuricemia , related with hyperlipaemia as well as hyperglycemia , is common in the elderly and should be paid more attention .
7.The correlation study between FGFR2 gene polymorphisms and breast cancer in Qinghai Tibetan areas

Objective To explore the relationship between the fibroblast growth factor 2 (FGFR2) gene polymorphism (rs 2981582 ,rs 1219648 ,rs 2420946) and the breast cancer risk in Tibetan population ,Qinghai province .Methods This is a case con‐trol study .Peripheral blood samples from 210 breast cancer patients and 230 healthy women in Qinghai area were collected .DNA samples were extracted from peripheral blood cells .FGFR2 gene polymorphism (rs 2981582 ,rs 1219648 ,rs 2420946) were typed by Taqman‐MGB probe based on PCR and DNA sequencing ,then analyzed its correlation with breast cancer in Tibetan population , Qinghai province .Results The genotype frequencies of rs 2981582 CC ,CT and TT were 40 .48% ,39 .05% and 20 .47% among the breast cancer patients while 36 .09% ,48 .69% and 15 .22% among the controls .The genotype frequencies of rs 1219648 GG ,AG and AA were 24 .76% ,26 .19 % and 49 .05% among the patients while 23 .91% ,47 .39% and 28 .70% among the controls .The genotype frequencies of rs 2420946 CC ,CT and TT were 29 .05% ,45 .24% and 25 .71% among the patients while 30 .87% , 51 .74% and 17 .39% among the controls .The genotype frequencies of all genetic loci had no significant difference between rs 2981582 and rs 2420946 (P>0 .05) .But the genotype frequencies of rs 1219648 AA have statistical sense (P< 0 .05) ,compared with GG ,the incidence of breast cancer was remarkably increased with AA [OR=1 .65 ,95% CI= (1 .01 ,2 .69)] .Conclusion This study shows that FGFR2 rs1219648 AA is related to breast cancer risk among Tibetan population .
8.Clinical observation of conventional antipsychotics combined with agomelatine in schizophrenia patients with obsessive-compulsive symptoms

Objective To examine the efficacy, safety, economic benefits and social function of conventional antipsychotics combined with agomelatine in the treatment of schizophrenia patients with obsessive-compulsive symptoms. Methods Eighty schizophrenic patients with obsessive-compulsive symptoms were randomly divided into two groups. The study group was treated with conventional antipsychotic drugs combined with agomelatin, and the control group combined with clomipramine for 8 weeks. Positive and negative symptom scale (PANSS) and Yale-Brown obsessive-compulsion scale (Y-BCOS) were used to evaluate the symptoms at before treatment, at discharge and one month after discharge. Social disability screening scale (SDSS), treatment emergent symptom scale (TESS) and the economic benefits was used to evaluate the social function, treatment efficacy and economic status. Results Before treatment, there was no significant difference in the scores of each scale between the two groups (P>0.05). There was no significant difference in PANSS scores between the two groups at all time points (P>0.05). The difference between Y-BCOS score and pre-treatment value was higher in the study group than in the control group (P<0.05). The difference between SDSS score and pre-treatment value was higher in the study group than in the control group (P<0.05). The TMRs were lower in the study group than in the control group at one month after discharge (P<0.05). The TESS score was lower in the study group than in the control group at discharge and one month after discharge. The cost and income were higher and the cost-effect ratio was lower in the study group than in the control group at one month after discharge (P<0.05). PANSS and Y-BCOS were positively correlated with TMR, SDSS and CER (P<0.05), but not with TESS at one month after discharge (P>0.05). TESS was positively correlated with TMR, SDSS and CER (P<0.05). Conclusion The treatment of conventional antipsychotics combined with agomelatine in schizophrenia patients with obsessive-compulsive symptoms are safe and effective. Patients can achieve better social function, good economic and social benefits.
9.Research and development of correlation between myeloperoxidaseand colorectal cancer

Colorectal cancer is one of the most common malignant tumors. Myeloperoxidase (MPO), as an oxidase in neutrophils lysosomes, plays an important role in activating carcinogenic gene intermediates and enhancing exogenous carcinogenesis. It can stimulate oxidative stress reaction in vivo by producing hypochlorite, reactive oxygen species and other oxidants, and induce gene instability factors such as DNA damage, mutation, mismatch repair and so on, which leads to the occurrence and development of colorectal cancer. MPO single nucleotide polymorphism (SNP) is related to the genetic susceptibility of cancer. Mpors 2333227-463G>A can reduce the risk of colorectal cancer. Therefore, the research on MPO, MPO-463G>A will provide new ideas and strong evidence for early detection, prevention, disease assessment and targeted treatment of colorectal cancer.
10.The diagnostic value of KRAS gene mutation in appendiceal adenocarcinoma

Objective:To analyze the mutation characteristics of Kirsten rat sarcoma virus oncogene homology (KRAS) gene in patients with appendiceal adenocarcinoma and its relationship with the activity of Ras Raf Mitogen activated protein kinase/extracellular regulated protein kinase (MAPK/ERK) signaling pathway.Methods:A total of 41 patients with appendiceal adenocarcinoma who were treated in the Lishui Central Hospital from January 2014 to January 2020 were selected as the observation group, and 50 patients with Appendicitis who were operated at the same time were randomly selected as the control group. Clinical and follow-up data were collected, and the mutation of the KRAS gene in the patient′s tissue was measured using the snapshot method. The expression of key proteins in the MAPK/ERK signaling pathway in cancer tissue was measured using Western blotting (WB) assay. We compared the clinical characteristics and prognosis of patients with KRAS mutation and non KRAS mutation appendiceal adenocarcinoma.Results:The KRAS gene mutation rate in the observation group was higher than that in the control group (41.5% vs 10.0%), and the expression levels of p-ARAF/ARAF, p-MEK1/MEK1, and p-ERK1/ERK1 proteins were also higher than those in the control group. The differences between the groups were statistically significant (all P<0.05). The protein expression levels of p-ARAF/ARAF, p-MEK1/MEK1, p-ERK1/ERK1 in KRAS mutation patients in the observation group were significantly higher than those in non KRAS mutation patients. The proportion of stage IV, positive rates of carcinoembryonic antigen (CEA), carbohydrate antigen (CA)199 and CA125 in KRAS mutation patients were higher than those in non KRAS mutation patients, and the survival time and progression free survival time were shorter than those in non KRAS mutation patients, with statistical significance (all P<0.05). Conclusions:The mutation rate of KRAS in appendix adenocarcinoma is high, and the activation of MAPK/ERK signaling pathway caused by KRAS mutation may play a role in the pathogenesis of appendix adenocarcinoma, which has the value of in-depth research.
