Chinese Pharmacological Bulletin 1987;0(03):-
The influences of the N -Isosuccinyl - dichloro - cinnamoylamide (7903) on the NA and 5 -HT systems in the brain were studied. Acute administration of 7903 could significantly enhance the level of NA and 5 -HT in mice brain and cortex of rats, and the level of 5 -HT in the hyporthalamus of rats was tending to be enhanced. However, chronic administration of 7903 (two weeks ) could decrease the level of NA and the ra-tio of 5-HIAA/5-HT in the cortex of rats; Chronic administration of 7903 could also decrease the turnover of NA and 5- HT in mice brain, but had no effects on the special binding of ? - receptor to 3-H - DHA and that of 5 -HT receptor to H - ketanserine in the cortex of rats.

Objective:To evaluate the mechanical performance of customized metal prosthesis with tibia stems of varying lengths for tibial bone defects reconstruction. Methods:Morphologically matched postoperative finite element models of bone defect revision were developed, with three gradients (15 mm, 30 mm, and 45 mm) according to the degree of bone defect and were reconstructed with 3D printed tantalum metal prosthesis using three tibial stem lengths (80 mm, 120 mm, and 150 mm), respectively. Conventional static and dynamic (walking gait) loading was performed to analyze the peak tibial stress distribution and accumulated sliding distance at the prosthetic interface, and to assess the effects of the three tibial stems of different lengths on the stability of the customized tibial defect restorations and the internal tibial stress state. Results:The peak accumulated sliding distance of the dynamically loaded morphologically matched restorations ranged from 17.94 to 21.31 mm with static loading, which were 68% to 84.3% higher than those of 10.26 to 11.69 mm with static loading. The peak tibial stresses in the dynamically loaded model were greater than those in the statically loaded model, with an increase of 28%-49.2%, including 132.94-143.88 MPa in the statically loaded model and 170.41-200.14 MPa in the dynamically loaded model. The overall accumulated sliding distance of the tibia prosthetic model gradually decreased from the tibial osteotomy surface, and the accumulated peak sliding distances ranged from 10.26 to 11.69 mm for static loading, and from 17.94 to 21.31 mm for dynamic loading. The bone tissue stresses in the anterolateral and medial-posterior tibia increased gradually from top to bottom, and the maximum stress value in each section was in the posterior medial tibia (the maximum value was 200.14 MPa). The highest bone tissue stress in the lateral tibia was affected by the tibial stem length, which resulted in a different location, and it was the area most affected by stress shielding (maximum value of 170.65 MPa).Conclusion:For stability assessment of morphologically matched tantalum customized prosthesis, physiological gait dynamic loading studies are more reliable than static loading; the choice of tibial stem length depends on a combination of accumulated peak sliding distances and tibial bone stress distribution factors.

Objective To promote the application of the best evidence for supportive care in the management of depression in patients with head and neck cancer, and to improve the quality of clinical care. Methods Follow the three stages of clinical evidence practice of Joanna Briggs Institute (JBI): baseline review, practice change and effect evaluation, the best evidence of supportive nursing in depression management for head and neck cancer patients were retrieved, and 4 quality review indicators were developed. A total of 50 hospitalized head and neck cancer patients and 12 nurses were selected to collect data by means of field observation, interviews and nursing records. The obstacles to the application of evidence were analyzed and a practical scheme was worked out. Results The nurse's implementation rate of the four review indicators increased from 0-90% at the baseline review to 90%-100% after the evidence was applied. Except for Indicator 3 "Indicators nurses introduced the pressure on patients and their families to face cancer", the implementation rates of the other three indicators were significantly improved (P<0.01). After the application of evidence, the incidence of depression was decreased (P<0.05). The score of Relaxation, Sleep and Fatigue Management Knowledge Questionnaire was (18.20±14.09) before the application of evidence, which was lower than that after the application of evidence (80.60±12.84), and the difference was statistically significant (t=-23.14, P<0.01). The nurses' score of Supportive Nursing Knowledge Questionnaire before the application of evidence was (63.83±10.78), which was lower than that after the application of evidence (95.5±3.18), and the difference was statistically significant (t=-9.76,P<0.01).Conclusions The evidence-based application program can standardize nurses' supportive nursing behaviors for patients with depressive symptoms, reduce the severity of depression, improve the self-support knowledge level, and improve the quality of depression management in hospitalized patients with head and neck cancer.

Objective To investigate the changes of renal resident dendritic cells (rDC)during kidney ischemia-reperfusion injury (IRI). Methods C57BL/6J mice models with bilateral renal warm ischemia were established. The kidney tissue was prepared for single cell suspension at 24 h and 48 h after reperfusion. The changes in the percentage of CD45 +cells and CD1 1 c +rDCs were evaluated by flow cytometry. The renal tissues of mice labeled with green fluorescent protein and diphtheria toxin receptor (CD1 1 c +GDTR ) were prepared for single cell suspension. The percentage and phenotype of CD1 1 c +rDCs were analyzed by flow cytometry. CD1 1 c +GDTR mice models with bilateral renal warm ischemia were established. The renal tissue was prepared for single cell suspension at 24 h after reperfusion. CD45 + cells was gathered by magnetic-activated cell separation (MACS ). The expression levels of co-stimulatory molecules on the rDC surface were analyzed by flow cytometry. Results At 24 h after reperfusion, the percentage of CD45 +cells in the kidney of C57BL/6J mice was significantly elevated, and further increased at 48 h after reperfusion. At 24 h after reperfusion, the quantity of CD1 1 c +rDCs was equally increased, whereas the percentage of CD1 1 c +rDCs in CD45 +cells was dramatically declined and restored at 48 h after reperfusion, slightly higher compared with that in the sham group. In healthy CD1 1 c +GDTR mice, the percentage of CD45 +cells in the kidney was lower than 1%,consisting of approximately 40%of CD1 1 c +rDCs,which mainly presented as CD11bintF4/80 -MHCⅡ+. At 24 h after reperfusion, the percentage of CD11c +F4/80 -subset rDC surface co-stimulatory molecules was significantly enhanced, such as CD40,CD80 and CD86. Conclusions Following warm IRI,the percentage and quantity of rDCs,and the expression level of rDC surface co-stimulatory molecule are significantly increased, prompting that renal rDC infiltration is increased and phenotype becomes matured.
