1.Clinical observation of postoperative chemotherapy combined with tianzhicao capsule in breast cancer

Objective To observe attenuation and synergy of postoperative chemotherapy combined with Tianzhicao capsule in patients who suffered from breast cancer. Methods 66 cases after radical mastectomy were randomly divided into treatment group 31 cases and 35 cases of control group. The patients of control group were treated with chemotherapies of CAF and TA. Treatment group patients were given Tianzhical capsule orally at the same time. Nausea and vomitting,the arrest of bone marrow before and after treatment were observed. The grade of life quality about every patient was evaluated. Results Myelosuppression and the occurrence of nausea and vomitting in treatment group was obviously lower than control group (P < 0.05). The proportion of improvement and stabilization for the quality of life in treatment group was markedly higher than control group (P < 0.05). Conclusion Traditional Chinese drug Tianzhicao capsule had attenuation and synergy to postoperative chemotherapy in breast cancer.
2.S phase kinase-associated protein 2 regulates rat mesangial cells proliferation

Objective To explore whether the change of S phase kinase-associated protein 2 (Skp2) expression could regulate mesangial cell proliferation. Methods Skp2 siRNA and control siRNA, pIRES-GFP-Skp2 plasmid and pIRES-GFP plasmid were designed and synthesized. Cell transfection was performed by Lipofectamine 2000. Skp2 mRNA and protein levels were detected by semiquantitative PCR and Western blotting respectively. Primary culture rat mesangial cells were divided into 6 groups: 0%FCS, 20%FCS, 10%FCS+pIRES-GFP plasmid, 10%FCS+pIRES-GFP-Skp2 plasmid, 20%FCS+control siRNA, 20%FCS+Skp2 siRNA. Cell number was detected by MTT. S phase entry was measured by BrdU labeling. Cell cycle profile was determined by flow cytometric analysis. Results Skp2 mRNA level was significantly down-regulated by Skp2 siRNA compared to control siRNA. Skp2 protein level increased after pIRES-GFP-Skp2plasmid transfection compared to pIRES-GFP plasmid. MTT, BrdU labeling and cell cycle profile demonstrated that cell number (A: 0.419±0.088 vs 0.305±0.036, P<0.01) and S-phase cells (BrdU labeling positive cell: 0.21±0.04 vs 0.15±0.03, P<0.01;S-phase cell number:20.18±0.64vs 14.33±0.37, P<0.01) obviously increased after Skp2 plasmid transfection, while decreased after Skp2 siRNA transfection compared to control groups (A: 0.328±0.069 vs 0.482±0.133, P<0.01;BrdU labeling positive cell: 0.17±0.01 vs 0.24±0.00, P<0.01; S-phase cell number: 16.52±0.75vs 23.81 ±1.25, P<0.01). Conclusion Over-expression of Skp2 stimulates mesangial cell proliferation while down-regulated expression of Skp2 inhibits mesangial cell proliferation.
3.Predictive value of donor's glomerular filtration rate to recipient's weight ratio in early clinical outcome in living-related donor transplantation

Objective To investigate whether the donor's glomerular filtration rate (GFR) to recipient's weight ratio (Dg/Rw) is a useful tool to predict early clinical outcome in living-related do-nor transplantation.Methods A total number of 108 living donor transplant recipients in the Chinese Military 309th Hospital from Jan.2014 to July 2015 were enrolled in this study.The patients who had multi-organ transplantation or developed grafts rejection,delayed graft function,hydronephrosis or renal vascular stenosis were excluded.The 90 qualified recipients were divided into G1 group (Dg/Rw ≤0.81),G2 group (Dg/Rw 0.81~1.11),and G3 group (Dg/Rw≥1.12).We respectively analyzed the relationship between recipient's serum creatinine Scr and Dg/Rw at 3-,7-,30-day and 1 year after transplantation.Results Scr at 3-,7-,30-day and 1 year after transplantation had linear correlation with Dg/Rw.As compared with G1 and G2 groups,Scr level was significantly reduced in G3 group at different time points (P<0.05).Conclusion Dg/Rw has a negative relationship with Scr level after renal transplantation.Pre-transplant Dg/Rw is a potential index to predict the early clinical outcome in living-related donor transplantation.
4.Correlation between de nove anti-endothelial cell antibodies and postoperative adverse events after renal transplantation

Objective To investigate the correlation between de nove anti-endothelial cell antibodies (AECA) and adverse events after renal transplantation and the effect of transplanted renal function within six months after operation.Methods The expression of AECA in serum of 85 renal transplant recipients was detected by indirect immunofluorescence assay (ⅡF) before and 1 day,3 days,7 days,15 days and 1 month after renal transplantation.The patients were divided into double positive group (AECA positive before and after surgery),single positive group (postoperative de nove AECA positive) and negative group (preoperative and postoperative AECA negative) according to AECA test results.The occurrence of adverse events in each group such as DGF,rejection,infection and so on,as well as the serum creatinine levels were recorded at each time point within six months.Results Of the 85 recipients,29 were positive for AECA,including 19 in the double positive group,10 in the single positive group,and 56 in the negative group.The incidence of rejection in single positive group (30%) was higher than that in the rest two groups (5.3% for double positive group,and 17.9% for negative group),but there was no statistically significant difference (P =0.21).The incidence of DGF in the single positive group,double positive group and negative group was 70.0%,26.3 % and 32.1 % respectively.The positive rate of the single positive group was significantly higher than that of the rest two groups (P =0.04),and the duration of DGF was significantly longer in the single positive group than that of the rest two groups (P<0.01).The incidence of infection in the single positive group,double positive group and negative group was 20.0%,21.1% and 8.9%respectively,and there was no significant difference among the three groups (P =0.31).As compared with the double positive group and the negative group,the serum creatinine level was significantly increased in the single positive group at 1st week,1st month,3rd month and 6th month after operation (P =0.02,P =0.04,P =0.04 and P =0.02 respectively).Conclusion Postoperative AECA can increase the risk of DGF,prolong the duration of DGF,and affect the recovery of renal function within 6 months after renal transplantation.
5.The expression and clinical significance of Bax and PHF20 in laryngeal squamous cell carcinoma.

OBJECTIVE:
To investigate the expression of Bax and PHF20 in laryngeal squamous cell carcinoma (LSCC)and to discuss their relevance and the roles in carcinogenesis and development in LSCC.
METHOD:
The expressions of Bax and PHF20 in the LSCC tissues and normal mucosa tissues adjacent to carcinoma were detected by SP immunohistochemistry assay. The relationship between the expressions of Bax and PHF20 and the clinicopathological characteristics including clinical stage, pathological type, histological grade and lymph node metastasis in LSCC were analyzed according to the clinical data.
RESULT:
(1) The expressions of Bax and PHF20 were both significantly lower in the LSCC tissue than that in the normal laryngeal tissue (P < 0.01). (2) In clinical stage grouping, there were no statistical differences of the quantity and positive rate of Bax and PHF20 expressions among supraglottic, glottic and subglottic LSCC (P > 0.05). In histological differentiation grouping, the quantity and positive rate of Bax and PHF20 expressions decreased significantly in poorly differentiated LSCC compared with the well and moderately differentiated LSCCs (P < 0.01, P < 0.05, respectively). In T stage grouping, the quantity and positive rate of Bax and PHF20 expressions were both significantly higher in T1 + T2 compared with T3 +T4 (both P < 0.01). In addition, the quantity and positive rate of Bax and PHF20 expressions were both significantly higher in LSCC with lymph node metastasis compared to that without lymph node metastasis (both P < 0.01).
CONCLUSION
The lack of Bax and PHF20 might contribute to the carcinogenesis and development in LSCC. The positive expression of Bax and PHF20 maybe relative to T term degree, differentiation degree and lymphamatic metastasis of LSCC.

7.Frictional properties of human skin at different ages, gender and anatomical regions.

A reciprocal sliding skin friction testing apparatus was performed to investigate human skin frictional properties of different physiological factors such as different ages, gender and anatomical regions. Student's t-test for non-paired data and one-way analysis of variance (ANOVA) were used in data analysis. Fisher LSD was used for multiple comparison on the statistical significance of the coefficients. The results showed that the skin frictional properties at the different anatomical regions were strongly dependent on the age. The differences of friction coefficients at the different anatomical regions became negligible with the increase of age. The skin frictional properties of different gender at the same anatomical region and age exhibited no significant differences. The friction coefficients of different ages at the same anatomical regions showed significant differences.

8.Mutation analysis and prenatal diagnosis for a family affected with congenital factor VII deficiency.

OBJECTIVETo provide mutation analysis and prenatal diagnosis for a family affected with congenital factor VII(FVII) deficiency.
METHODSDNA was extracted from peripheral blood samples from the proband and his parents. All exons and flanking sequence of the FVII gene were amplified with PCR and subjected to direct sequencing. Prenatal diagnosis was performed by amniocentesis.
RESULTSA homozygous mutation (NM_000131.3) c.572-1G>A was identified in the proband. Both parents of the fetus were carriers of the mutation.
CONCLUSIONA method for molecular diagnosis of congenital factor VII deficiency was established and successfully applied for an affected family.

9.Effects of frictional properties on traumas of rabbit skin.

Simulative experiments on skin traumas between prosthetic socket materials and residual limb skin were investigated by using the means of tribology, histology and animal experiment. Healthy adult rabbits were used as animal model and their denuded back skin was selected as experimental position to simulate residual limb skin. The effects of different normal load and reciprocal sliding frequency on rabbit skin trauma grade were investigated by using a reciprocal sliding skin friction testing apparatus to simulate prosthesis gait. The traumatic subcutaneous tissue slice was stained with hematoxylineosin and the morphology was observed under the optical microscope. The scab thickness of traumatic skin was measured under x 10 object lens. The inflammatory cells were counted in a given visual field under x 20 object lens. The statistical significance analysis of scab thickness and inflammatory cells were carried out to assess the effect of different frictional conditions on skin pathological traumas. The results showed: the greater normal load and higher reciprocal sliding frequency applied on the rabbit skin, the more serious injury to skin and more inflammatory cells in the subcutaneous tissue at the same time. The findings provide a theoretical basis of comfortized prosthesis design and gait analysis for the amputee.

10.Relationship between abnormal glycosylation of serum IgA1 and oxidative stress in patients with primary IgA nephropathy

Objective To study the relationship between abnormal glycosylation of serum IgA1 and oxidative stress in patients with primary IgA nephropathy (IgAN).Methods Fifty-two primary IgAN patients diagnosed by renal biopsy from January 2012 to December 2013 were selected.In the patients,Lee gradeⅠ-Ⅲand estimated glomerular filtration rate(eGFR)≥60 ml/min 41 cases(IgAN-A group),Lee grade≥Ⅲand eGFR<60 ml/min 11 cases(IgAN-B group).Sixty-nine health examination volunteers were selected as control group.The serum galactose-deficient IgA1(Gd-IgA1)was detected by enzyme-linked immunosorbent assay for vicia lectin, the advanced oxidation protein products (AOPPs) was detected by liquid chromatography.Results The urinary protein,urea nitrogen,creatinine and uric acid in IgAN-B group were significantly higher than those in IgAN-A group:(3 613 ± 2 247)mg/d vs.(1 457 ± 1 342)mg/d,(8.30 ± 2.92)mmol/L vs.(5.46 ± 1.55)mmol/L,(155.09 ± 57.97)μmol/L vs.(77.50 ± 22.55)μmol/L and(466.73 ± 120.97)μmol/L vs.(361.70 ± 94.40)μmol/L,the albumin and eGFR were significantly lower than those in IgAN-A group:(36.45 ± 4.71)g/L vs.(39.37 ± 3.97)g/L and (50.71 ± 26.50) ml/min vs.(94.67 ± 23.48) ml/min, there were statistical differences (P<0.05).There were no statistical differences in IgG, IgA, IgM, C3and C4between IgAN-A group and IgAN-B group(P>0.05).The serum Gd-IgA1 and AOPPs in IgAN group were significantly higher than those in control group:0.44 ± 0.17 vs.0.15 ± 0.06 and(120.3 ± 48.5)μmol/L vs.(45.7 ± 19.2)μmol/L,there were statistical differences (P<0.05).The serum Gd- IgA1 and AOPPs in IgAN- B group were significantly higher than those in IgAN-A group:0.59 ± 0.23 vs.0.35 ± 0.12 and(159.4 ± 50.2)μmol/L vs.(90.9 ± 41.7) μmol/L, there were statistical differences (P<0.05).There was a positive correlation between Gd-IgA1 and AOPPs in IgAN group (r = 0.603, P<0.05).Conclusions Gd-IgA1 may up-regulate oxidative stress.The serum levels of Gd-IgA1 and AOPPs are related to histopathological change of IgAN, which suggests that Gd-IgA1 combined with AOPPs may become a new biomarker for prognosis of IgAN.
