Objective To establish a RP-HPLC method for determination of aconitine in Shenhailong capsules.Methods Zorbax SB-C18column(4.6 mm?250 mm,5 ?m)was used,with(methanol∶water∶triethyl amine)60∶40∶0.1,as the mobile phase.The flow rate was 1.0mL?min-1and the column temperature was at 30℃.The detection wavelength was 240nm.Results Aconitine concentration presented a good linear range of 0.04?10-3～4?10-3g?L-1(r = 0.9999).with the average recovery of 98.4%,and RSD :1.0%.ConclusionThe method was accurate,nice in reproducibility and with few interference.It could be used for quality control of Shenhailong capsules.

Objective To know the sleeping quality of clinical nurses in Urumqi, and analyze the influencing factors, put forward the corresponding countermeasures for different factors to improves the sleeping quality. Methods Randomly selected 407 nurses in clinical departments as the research objects in Urumqi top three hospitals, using the Pittsburgh sleep quality index scale (PSQI) and general situation questionnaire to investigate, and then analyzed the results of the survey. Results The incidence rate of sleeping disorders in emergency nurses was the highest (80.00%), followed by pediatric, intensive department, internal medicine, surgical department, operating room, outpatient and others was the lowest (39.02%). Gender (t=-2.015, P =0.045), age (F=8.509, P =0.000), establishment(F=3.609, P =0.013) were the influence factors of PSQI scores. Only the difference of the scores of hypnotic drugs between different ethnic groups was significant, P<0.05, while the others had no significant difference, P>0.05. Conclusions Poor sleeping quality of nurses existed in every department, its sleeping quality status, department, gender, age and establishment have certain correlation, interventions should be strengthened, to improve the overall level of sleeping.

Assembling an adapted smoothing method and a classifier of wavelet transform combined support vector machine ( SVM) , a Raman spectrum recognition approach was built for low signal noise ratio situation. Firstly, spectra data were denoised by the adapted smoothing method. The smoothing window was adapted to the signal noise ratio, which would effectively remove noise with the intensity of the signal well remained. Secondly, the wavelet transform was used for dimension reduction of the data. The decomposition level of wavelet transform was optimized according to the best classification result of the training set. Lastly, SVM was used for classification. Cross Validation ( CV ) was applied to obtain the optimized parameters of SVM. Conditions for the effective parameters were searched considering the relation between the cross_validation result and the classification accuracy. Combined with the surface enhanced Raman scattering ( SERS ) technology , the developed spectrum recognition approach was used for qualitative analysis of methamphetamine ( MAMP ) and 3, 4_methylenedioxymethamphetamine ( MDMA ) in peopleˊs urine, where the detecting accuracy is above 95. 0%. The uniform Au nanorods (NRs) SERS substrate synthetized by the Hefei Institute of Intelligent Machines of Chinese Academy of Sciences was used for the experiment. Raman spectra were acquired using an Inspector Raman ( DeltaNu) spectrometer, with the excitation wavelength of 785 nm and the integrate time of 5 seconds.

Objective To know the influence of preoperative respiratory function exercise on the postoperative pulmonary function among patients with lung cancer. Methods Divided 80 patients with lung cancer into the observation group and the control group, there were 40 cases in the each group. Routine perioperative nursing cares was used in the control group, while a two weeks respiratory function exercise was used in the observation group. Compared the incidence rate of complications be-tween the two groups after the intervention, some indexes which can indicate the respiratory function were compared before and after the exercise in the observation group. Results The indexes such as PaO_2, SpO_2, MVV, TCL and FEV in the observation group after the respiratory function exercise were better than before significantly. The incidence rate of complications in the control group was more than in the observa-tion group. Conclusions Strengthen respiratory function exercise can effective improve the respiratory func-tion for patients with lung cancer when cured by thoractomy, which can promote patients' compliance, and then avoid certain postoperative complications.

Aim To investigate the effect and mechanism of valdecoxib on the apoptosis of human esophageal cancer cells.Methods Flow cytometry was used to observe the effect of valdecoxib on apoptosis and the cell cycle distribution of Eca109 cells.Transmission electron microscope was further used to detect the cell apoptosis.The content of LDH was examined using LDH kit.The expressions of p-p38MAPK,Fas and FasL protein were detected using flow cytometry.Results Valdecoxib of 25～400 ?mol?L~(-1) significantly induced the apoptosis of Eca109 cell line,and the rate of apoptosis was increased from(2.95?0.83)% to(48.46?0.73)%,50～400 ?mol?L~(-1) valdecoxib also decreased the proliferation index and the proportion of cells in the S phase,increased the proportion of cells in the G_0/G_1 phase,but had no effect on the proportion of cells in the G_2/M phase.Compared with those in Eca109 cells cultured in the medium with solvent,the expression of p-p38MAPK,Fas and FasL was higher in the Eca109 cells exposed to valdecoxib in a dose-dependent manner in 72 h.Conclusion Valdecoxib can induce apoptosis of Eca109 cell line partly by up-regulating the expression of p-p38MAPK/Fas/FasL.

Questionnaire-based survey, physical examination, and blood testing were conducted according to cluster random samplings in Kazakh residents in Xinjiang.2 760 samples were collected to analyze the association of different strata of waist circumference and clustering of metabolic syndrome (MS) components.Accoding to International Diabetes Federation standard, the prevalence of ≥1and ≥2 components of MS showed increasing trend with the increase of waist circunference, and odds ratio of clustering of MS components also increased significantly.The distance of receiver operating characteristic curve was the shortest and the prevalence of MS was 22.1% ;22.4% in men, and 21.9% in women;when the waist circumference was ≥91 cm for men, and ≥88 cm for women.

Objective To investigate the association of lipoprotein lipase gene Hind Ⅲ and S447X polymorphisms with metabolic syndrome. Methods PCR-RFLP was used to detect lipoprotein lipase Hind Ⅲ and S447X genotypes in 401 subjects(including 201 controls, 200 metabolic syndrome patients). Results ( 1 ) The levels of waist circumference ( WC ) , hip circumference ( HC ) , waist-to-hip ratio ( WHR ) , body mass index (BMI), systolic blood pressure (SBP), diastolic blood pressure ( DBP) , total cholesterol ( TC) , triglyceride (TG), high-density lipoprotein-cholesterol (HDL-C), low-density lipoprotein-cholesterol (LDL-C), and fasting plasma glucose (FPG) were significantly different between metabolic syndrome group and control group (all P< 0.05). (2)The frequencies of H+H+ genotype,H+allele,SS genotype, and S allele for metabolic syndrome were all significantly higher than those for controls( H+H+ genotype:66. 5% vs 54.2% ,P=0.012; H+ allele:78.0% vs 71.4%, P=0.031;SS genotype:89.5% vs 77. 1% , P = 0.001; Sallel:94.5% vs 87. 56% , P = 0.001). (3) The levels of WC, HC, WHR, BMI, SBP, DBP, TG, LDL-C, and FPG in H + H-/H-H- genotype were significantly lower than those in H+H+ genotype, HDL-C was significantly higher than that in H+H+ genotype ( all P<0. 05). The levels of WC, HC, WHR, BMI, SBP, DBP, TC, TG, and FPG in SX/XX genotype were significantly lower than those in SS genotype, HDL-C was significantly higher than that in SS genotype ( all P< 0.05). (4)Multi-way logistic regression analysis suggested that risk factors for metabolic syndrome were smoking, drinking, and SS genotype (OR value was 4.289,2.268, and 2. 597, respectively ). (5) Result of interaction analysis among different factors indicated that the risk for metabolic syndrome in smoker with SS genotype was 3. 996 times of non-smokers with SX/XX genotype. Conclusions The lipoprotein lipase gene S447X polymorphism is associated with metabolic syndrome risk in Kazakh, and SS genotype and S allele may serve as genetic risk factors of metabolic syndrome, H + H-/H-H- and SX/XX genotypes yield beneficial effect for lipid and blood pressure. SS genotype and smoking may exist additive effect.

Objective To investigate the effect and possible mechanism of arsenic trioxid(As2O3)on proliferation of RSC-364 synoviocyte lines stimulated with TNF-?.Methods RSC-364 synoviocytes were cultured with standard medium as control group or medium supplemented with 10 ?g/LTNF-? and different concentrations of As2O3 respectively. MTT assay were carried out to study cell proliferation. Proliferation index (PI) and cell cycle were detected by flow cytometry (FCM). RT-PCR was used to detect the mRNA expression of High mobility group box chromosomal protein (HMGB)1. HMGB-1 and proliferation cell nuclear antigen (PCNA) proteins were detected by immunocytochemistry and FCM. Objective To investigate the effect and possible mechanisms of high mobility group box (HMGB) 1 on the proliferation of RSC-364 synoviocytes. Methods ① RSC-364 cells stimulated by 10 μg/L TNF-α and cells of the normal control groups were collected at 6, 12, 24 h respectively in vitro. HMGB1mRNA and protein was detected by reverse transcription polymerase chain reaction (RT-PCR) and immunocytochemistry (ICC); ②RSC-364 cells induced by 10 μg/L HMGB1 were collected in 6, 12, 24 h respectively, so did normal control group cells in vitro. The expression of signal transducer and activator of transcription (STAT) mRNA 1 was detected by RT-PCR. The expression of STATland SOCSI proteins were detected by ICC and flow cytometry analysis (FCM). The expression of PCNA was detected by ICC. Results ① Compared with the control group, TNF-α markedly up-regulated HMGBI mRNA at 6, 12, 24 h respectively [0.86, 0.92, 1.06 vs 0.70, P<0.01 ], as well as protein expression level. Positive signal of HMGB1 proteins was not only expressed in nuclear but also in cytoplasm after stimulation. ② Compared with normal group, HMGBI increased the expression of P-STAT1 mRNA and protein at 6, 12, 24 h respectively [0.30, 0.69, 1.05 vs 0.24, P<0.01 ] and [1.34±0.09，1.55±0.16，1.74±0.13 vs 1.00±0.15，P<0.01]. The expression of SOCSI protein increased significantly in HMGB1 group at 6 and 12 hours ( 1.43±0.10 vs 1.58±0.05), but it decreased at 24 hours (1.24±0.15). ③The expression of p-STATI protein was negatively correlated with that of SOCS1 protein. Conclusion HMGB1 appears to be an important mediator in the proliferation of RSC-364 cells, partly by up-regulating the expression and aetivity of p-STAT1.
