1.Clinical value of dual-source CT in evaluating coronary artery disease

Objective To explore the clinical value of dual-source CT(DSCT) in the diagnosis of coronary artery disease by comparing the result of DSCT and coronary angiography(CAG).Methods From July 2009 to August 2010,36 patients with suspected coronary hear disease underwent both DSCT coronary angiography(DSCTCA) and selective GAG examination.The results of DSCT coronary angiography were compared with CAG which was the gold standard of diagnosing coronary disease.The sensitivity,specificity,positive and negative predictive value,and accuracy of DSCT were evaluated.Results 144 coronary arteries were well evaluated of 36 cases.The sensitivity,specificity,positive and negative predictive value,and accuracy of DSCT in the diagnosis of coronary hear disease were 100.00%,66.67%,93.75%,100.00% and 97.20% respectively by the number of patients;The McNemar test showed no significant difference between two methods( x2 =0.500,P >0.05 ) with a higher diagnosis concordance (Kapp =0.768,P <0.01).By detecting moderate to severe coronary artery stenosis,the sensitivity,specificity,positive and negative predictive value,accuracy were 95.24%,96.08%,90.91%,98.00%,95.83%,respectively.The McNemar test showed no significant difference between two methods(x2 =0.167,P >0.05) with a higher diagnosis concordance ( Kapp =0.899,P < 0.01 ).Conclusion DSCT has a diagnostic accuracy of coronary heart disease close to that CAG,especially in evaluating patients with moderate to severe coronary artery stenosis,it has high compliance and consistency with CAG.DSCT is a noninvasive,safe and reliable radiographic method.
2.Effects of repeated sevoflurane anesthesia on long-term cognitive function in lactating rats

Objective To evaluate the effects of repeated sevoflurane anesthesia on long-term cognitive function in lactating rats.Methods Twenty-four healthy Spragne-Dawley rats,aged 7 days,weighing 14-17 g,were randomly divided into 3 groups (n =8 each):control group (group C),2.6 % sevoflurane group (group S1),and 1.5% sevoflurane group (group S2).At 7,14 and 21 days after birth,2.6% and 1.5% sevoflurane and carrier gas were inhaled for 1 h in groups S1,S2 and C,respectively.Visible plafform trial was carried out on 28 and 29 days after birth,and the swimming speed of the rats was recorded.Place navigation test was performed on 32-36 days after birth,and the escape latency was recorded.Spatial probe test was carried out on 36 days after birth,and the time spent in the platform quadrant,swimming distance and the number of times crossing the platform quadrant was recorded.Results Compared with group C,the escape latency was significantly prolonged in groups S1 and S2 (P < 0.05).The escape latency was significantly longer in group S1 than in group S2 (P <0.05).There were no significant differences between the three groups in the swimming speed,time spent in the platform zone,swimming distance and the number of times crossing the platform quadrant (P > 0.05).Conclusion Repeated inhalation of sevoflurane during lactation can impair the long-term declarative memory,which is concentration-related,while it has no effects on the associative learning in rats.
3.The progress of the research on the relationship between stanniocalcins and cancers

Stanniocalcin (STC) was ifrst found as a calcium- and phosphate-regulating hormone produced in bony ifsh by the corpuscles of Stannius. In mammals, the homolog STC-1 displays a relative high amino acid sequence identity (nearly 50%) with ifsh STC, and STC-2 has a lower identity (nearly 35%) with STC-1 and ifsh STC. Both STC-1 and STC-2 are expressed in a variety of tissues. The functions of STC have not been understood. But some ifndings have been reported on their cellular localization, gene structure, and expression in different physiological and pathological conditions, which will be clues in elucidating the functions of STC in mammals. Moreover, STC-1 and STC-2 are expressed in many tumor cell lines, suggesting other biological functions of STC in mammals other than mineral metabolism.

Objective To explore the relationship between glycosylated hemoglobin and pulse wave velocity in non-diabetic elderly people.Methods Totally 163 subjects were included after exclusion of liver or kidney disfunction,diabetes mellitus or malignant tumor.They were divided into two groups:group A (HbA1c<5.7%) (n=54) and group B (HbA1c 5.7%-6.4%)(n=109).The age,pulse wave velocity,systolic blood pressure,diastolic blood pressure,blood chemistry and hypertension were compared between the two groups.Multi factor analyse was conducted to analyze the relative factors for pulse wave velocity.Results Systolic blood pressure was higher in group B than in group A (P < 0.05).Multi-linear stepwise regression analyses suggested that age,hypertension,glycosylated hemoglobin were positively related to pulse wave velocity in non-diabetic elderly people (β values were 20.382,144.061,93.195,all P<0.10).Conclusions Glycosylated hemoglobin is related to arterial stiffness in non-diabetic elderly people,it can be used as a predictive and control target of cardiovascular and cerebrovascular disease in old people.
6.Analysis of Dyslipidemia in 12908 Subjects in Enterprises in Beijing

Objective To explore dyslipidemia in the people in the part of enterprises in Beijing.Methods Total cholesterol, triglyceride, high-density lipoprotein cholesterol, etc., in blood were detected in the physical examination of 12908 subjects in enterprises in Beijing. Dyslipidemia was analysed for the distribution of gender and the trend of age.Results The findings showed that the occurrence of hypercholesteremia is 21.3% with 20.3% for male vs 23.0% for female; hypertriacylglycerolemia is 30.4% with 36.19% for male vs 20.49% for female; and low level of high-density lipoprotein is 6.5% with 9.4% for male vs 1.6% for female. The trend of dyslipidemia increased with age. The subjects with coronary artery disease and related high risk diseases, who need medicine care, occupied 80.6% of the subjects with dyslipidemia.Conclusion Dyslipidemia increases with age and so regular detection of lipid is necessary especially for the subjects with coronary artery disease and related high risk diseases.
7.MRI findings of the brain after gas explosion and its relationship with post-traumatic stress disorder

Objective To investigate MR findings and dynamic changes of the brain after gas explosion,and to evaluate the relationship between MR findings and post-traumatic stress disorder (PTSD).Methods Forty-nine survivors of a gas explosion (group A) were examined with brain MRI within 1 to 3 days,and serial MR follow-up examinations were also performed.Forty miners not under the ground that day were assigned as group B,and 40 staff working on the ground as group C.The signal intensity values of hippocampus and globus pallidus on T2WI were measured in the three groups and F test was performed by using SPSS 13.0.The relationship between signal intensity values of hippocampus/globns pallidus and PTSD was explored,and the relationship between ADC values of hippocampus and PTSD was also investigated.Results In group A,slight low signal on T1WI and high signal on T2WI were detected on initial MRI in hippocampus (33 cases),globus pallidus (12 cases),cortex (10 cases),and midbrain (2 cases),respectively.On follow-up MRI at 2 months,lesions in hippocampus disappeared (25 cases) or remained slight high signal on T2WI (8 cases),lesions in globus pallidus disappeared (3 cases,5 sides) or showed shrinkage and encephalomalacia (9 cases),cortical lesions resulted in encephalomalacia in 2 cases and returned normal in the others,and lesions in the midbrain showed encephalomatacia.For comparison of T2 signal intensity values in hippocampus and globus pallidus,there was significant difference between group A and group B(P <0.01),and also between group A and group C(P <0.01),but no difference was detected between group B and group C (P>0.05).In group A,the T2 signal intensities of PTSD and non-PTSD were 455±37 and 462±53 in the left hippocarnpus,and 458±36 and 460±43 in the right hippoeampus on 1 to 3 days,and the T2 signal intensities of PTSD and non-PTSD were 438±29 and 424±37 in the left hippocampns,and 442±31 and 430±32 in the right hippocampus at 2 months.The T2 signal intensities of PTSD and non-PTSD were 361 ±35 and 366±63 in the left globus pallidus,and 363 ±41 and 375±62 in the right globus pallidus on 1 to 3 days,and the T2 signal intensities of PTSD and non-PTSD were 341±24 and 337±39 in the left globns pallidus,340±26 and 332±35 in the tight glohus pallidns at 2 months.There was no difference of T2 signal intensity values in hippocampus and globus pallidus between PTSD and non-PTSD( t=0.350,0.826,0.503,0.907,P>0.05).In group A,ADC values of PTSD and nun-PTSD were (8.1±1.1)×10-4 and(8.1 ±0.9)×10-4mm2/s in the left hippocampus,and (8.2±1.0)×10-4 and(8.2±0.8)×10-4mm2/s in the tight hippocampus on 1 to 3 days,ADC values were (8.8±0.7)×10-4 and (9.0±1.0)×10-4mm2/s in the left hippocampus,and (8.5±0.9)×10-4 and (9.3±1.1)×10-4mm2/s in the tight hippocampus at 2 months.ADC values in hippocampns showed no difference between PTSD and non-PTSD(t=0.016,0.081,P>0.05)on initial MRI,but showed significant difference between PTSD and non-PTSD in tight hippocampus (t=7.407,P < 0.05) on follow-up MRI at 2 months,while no difference in left hippocampus (t =0.333,P>0.05) was observed at 2 months.Conclusion Hippocampns and globus pallidus are the most vulnerable structures in gas explosion.The occurrence of PTSD may be related to the injury of fight hippocampus,but not related to the injury of globns pallidus.
8.Microneedle-based interstitial fluid extraction for drug analysis:Advances,challenges,and prospects

Similar to blood,interstitial fluid(ISF)contains exogenous drugs and biomarkers and may therefore substitute blood in drug analysis.However,current ISF extraction techniques require bulky instruments and are both time-consuming and complicated,which has inspired the development of viable alterna-tives such as those relying on skin or tissue puncturing with microneedles.Currently,microneedles are widely employed for transdermal drug delivery and have been successfully used for ISF extraction by different mechanisms to facilitate subsequent analysis.The integration of microneedles with sensors enables in situ ISF analysis and specific compound monitoring,while the integration of monitoring and delivery functions in wearable devices allows real-time dose modification.Herein,we review the progress in drug analysis based on microneedle-assisted ISF extraction and discuss the related future opportunities and challenges.
9.Effect of Cichorium glandulosum Boiss.et Huet.on fecal bile acid profile in obese mice based on targeted metabolomics technique

Objective To investigate the effect of ethanolic extract of Cichorium glandulosum Boiss.et Huet.on fecal bile acid profiles in high-fat-diet-induced obese mice.Methods Twenty-four 6-week-old C57 BL/6 male mice were divided randomly into normal,model,drug-administration,and metformin groups.Mice in the normal group were fed a regular diet and mice in the other three groups were given high-fat diets.The drug-administration group was gavaged with 10 mL/kg ethanol extract of Cichorium glandulosum Boiss.et Huet.daily,and the metformin group was gavaged with 10 mL/kg metformin daily.After 10 weeks,livers were collected to measure hepatic total triglycerides(TG),total cholesterol(TC),low-density lipoprotein-cholesterol(LDL-C),and high-density lipoprotein(HDL)-C.Feces were collected and analyzed.Results Body weight(P<0.0001),liver TG(P<0.05),and TC(P>0.05)were all significantly higher in model mice compared with normal mice,while LDL-C(P>0.05)and HDL-C(P<0.001)were significantly lower,indicating abnormal weight gain and lipid metabolism.Alcoholic extract of Cichorium glandulosum Boiss.et Huet.significantly reduced body weight(P<0.0001),liver TG(P<0.0001),serum TG(P<0.05),TC(P<0.01),and LDL-C(P<0.05)in mice.Methodsological validation showed that the current method could accurately quantify 52 bile acids in feces.Analysis of the concentration of each type of bile acid revealed that alcoholic extract of Cichorium glandulosum Boiss.et Huet.significantly increased the secondary/primary bile acid ratio(P<0.05).Multivariate analysis showed that the bile acid metabolic pattern was significantly altered in all groups.Eight differential bile acids were screened in the drug-administration group relative to the model group using variable importance of projection>1 and P<0.05.A search of the Kyoto Encyclopedia of Genes and Genomes database revealed that the differential bile acids were mainly involved in the secondary bile acid biosynthesis pathway.Correlation analysis showed that four differential bile acids,deoxycholic acid(r,=0.6445,P<0.001),isolithocholic acid(r,=0.5879,P<0.01),3β-deoxycholic acid(r,=0.6649,P<0.001),and ω-rhamnoglutaric acid(rs=0.5387,P<0.01),in feces were strongly positively correlated with body weight.Conclusions Cichorium glandulosum Boiss.et Huet.alcoholic extract may play a role in weight reduction and amelioration of dyslipidemia by modulating secondary bile acid biosynthesis and altering fecal bile acid metabolic profiles.
10.Reuse of recombinant Escherichia coli to produce succinic acid by bioconversion.

The possibility of reusing Escherichia coli cells from the broth for succinic acid production was investigated. Using succinic acid yield and productivity as criterion, we investigated the effects of cell concentration, initial glucose concentration, different neutralizers on the bioconversion. The results revealed that E. coli could convert glucose to succinic acid in a water solution of glucose and a neutralizer. According to the results, the optimal condition was as follows: the cell concentration was 50 (OD600), glucose concentration was 40 g/L and neutralizer was MgCO3. Under the optimum conditions, we carried out the consecutive batch bioconversion in 7 L fermenter. Succinic acid yield reached 91% with the productivity of 3.22 g/(L x h) for the first conversion. For the second conversion, succinic acid yield reached 86% with productivity of 2.04 g/(L x h). Furthermore, we achieved a high mass yield above 83% with the productivity of 1.82 g/(L x h) for the third bioconversion.
