1.TAB182 promotes DNA homologous recombination repair by stabilizing RPA2 mRNA

Objective:To investigate the regulating molecules and acting mechanism of TAB182 in HR pathway.Methods:TAB182 in human breast cancer MCF-7 cells was knocked down by shRNA strategy, the TAB182 knockdown MCF-7 as the TAB182 knockdown group, and the MCF-7 cell using the shRNA negative control as the TAB182 negative control group. RNA sequencing and qRT-PCR were performed to screen and verify the differentially expressed genes of HR pathway related to TAB182 depression. Western blot was used to detect protein expression. Immunofluorescence staining of nuclear RAD51 and BrdU was used to check the 3′ ssDNA formation by the end resection. The cell cycle arrest and apoptosis were measured by flow cytometry. Cloning formation assay was used to evaluate the sensitivity TAB182-knockdown cells to radiation.Results:Both quantitative RNA sequencing and qRT-PCR assays showed that TAB182-knockdown significantly decreased the mRNA expression of RPA2( t=17.97, P<0.05). Compared with the TAB182 negative control group, the protein level of RPA2, the number of RAD51 foci, and the 3′ ssDNA-binding nuclear protein marker BrdU in TAB182-knockdown cells were significantly reduced. At 4, 8, and 12 h after actinomycin D treatment, the attenuation of RPA2 mRNA in the TAB182-knockdown cells was accelerated ( t=5.37, 3.79, 3.69, P<0.05). Compared with the TAB182 negative control group, the radiosensitivity and radiation-induced apoptosis in the TAB182-knockdown group were increased ( t=3.48, 11.05, P<0.05), and at 24 h after irradiation, the cell cycle block time was prolonged ( t=8.40, P<0.01). Conclusions:TAB182 plays a role in maintaining RPA2 mRNA stability, thereby promoting HR repair. TAB182 knockdown cells are highly sensitive to ionizing radiation.
2.Clinical application of painless oral anesthesia instrument combined with piezosurgery in the orthodontic traction of impacted maxillary anterior teeth

Objective :
To investigate the clinical efficacy of painless oral anesthesia combined with piezosurgery compared with ordinary cassette syringe combined with high speed turbine handpiece in the Fenestration operation of orthodontic traction of impacted maxillary anterior teeth.
Methods :
A total of 128 cases of impacted maxillary anterior teeth were treated with Fenestration operation and orthodontic traction, and the patients were randomly divided into two groups. Group A (64 patients) received painless oral anesthesia combined with piezosurgery. Group B (64 patients) received ordinary cassette syringe combined with high speed turbine handpiece. The cooperation of fenestration and the swelling and pain after operation were compared between the 2 groups.
Results :
The proportion of 0 and Ⅰ in the anesthesia group was 45.3% and 31.3% respectively, and the conventional group 0 and I accounted for 32.8% and 20.3% respectively. The difference between the two groups was statistically significant (Z=-2.676, P < 0.05). The pateints' cooperative degree in group A was better than that in group B (P < 0.05). The ratio of pain and swelling in the anesthesia instrument combined with Piezosurgery group was 81.2%, the conventional group had a grade I ratio of 59.4%, and the anesthesia instrument combined with Piezosurgery group was lighter than the conventional group, The difference was statistically significant (Z=-2.777, P < 0.05); anesthesia combined with Piezosurgery group after surgery accounted for 81.2% of the swelling, the conventional group of pain accounted for 71.9%, There was significant difference between the two groups (Z=-2.097, P < 0.05). Symptoms and signs as swelling and pain degree in group A were relatively minor than those in group B (P < 0.05).
Conclusion
It is worthy clinical promotion to use Painless oral anesthesia combined with piezosurgery in the fenestration and orthodontic traction of impacted maxillary anterior teeth, for its better cooperation and minor post-surgery reaction.
3.Activated coagulation factor Ⅺ(FⅪa)activity in intravenous immunoglobulin preparations from China: An analysis test

【Objective】 To investigate the activity level of coagulation factor Ⅺ(FⅪ) in IVIG products, which would provide support for improving the safety and quality control of IVIG products in China. 【Methods】 A total of 71 batches of IVIG products(half of which were closed to expiry date) from 23 domestic manufacturers (brands) were submitted for inspection and assayed for procoagulant activity including FⅪa, FⅪ and NAPTT activity with a chromogenic assay, coagulation proenzymes levels using one stage clotting assays and non-activated partial thromboplastin time (NAPTT). 【Results】 FⅪa(mIU/mL): 32 lots of IVIGs from 15 manufacturers(brands) possessed activities below 0.1 or the detection limit of the assays, 24 IVIG lots from 12 manufacturers were between 0.1~1(0.39±0.20), the remaining 15 IVIG lots from 6 manufacturers had FⅪa activity greater than 1 up to 47(13.27±15.61). FⅪ(mIU/mL): there were no detectable activities of FⅪ in 58 lots from 21 manufacturers, the other 13 lots from 6 manufacturers had IVIGs between 13~309(69.0±98.1), which included in 15 lots of IVIG whose FⅪa activity were greater than 1mIU/mL. NAPTT(s): The coagulation times for aforementioned 15 IVIG lots were all above 150s in the NAPTT test. The NAPTT ratios varied between 0.67 and 0.94(0.83±0.07), and 7 of 15 IVIGs had NAPTT ratios below or equal to 0.8. 【Conclusion】 There are substantial differences in the FⅪ and FⅪa activities in the IVIG preparations from 23 different manufacturers. Most of them had a lower activities; certain lots from specific manufacturers had relatively higher FⅪa activities. Manufactures should evaluate the manufacturing processes and monitor IVIG preparations for the presence of factor Ⅺ-like activity in case of thrombotic risks.
4.Effects of core muscle training combined with balance cup therapy in patients with chronic nonspecific low back pain

Objective:To explore the effect of core muscle group training combined with balance cup therapy in patients with chronic nonspecific low back pain.Methods:From January 2017 to December 2019, convenience sampling method was used to select 130 patients with chronic nonspecific low back pain in Guangdong Province Foshan Hospital of Traditional Chinese Medicine. According to the random number table, patients were divided into core muscle group and combined treatment group, with 65 cases in each group. The core muscle group was given the Swiss ball to perform core muscle training in the order of sitting, double bridge, knee flexion double bridge, reverse bridge and push-ups. The combined treatment group was given a balance tank based on core muscle training, followed by flash tank, walking tank, and sitting tank treatment. After 4 weeks of intervention, we compared the scores of the Visual Analogue Scale (VAS) , Roland-Morris Disability Questionnaire (RMDQ) , Finger-Floor Distance (FFD) , and static and dynamic muscle endurance time, and the total effective rate of treatment between the two groups of patients.Results:After intervention, the scores of VAS, RMDQ and FFD of combined treatment group were lower than those of core muscle group, and the differences were statistically significant ( P<0.01) . The static and dynamic muscle endurance time of combined treatment group were higher than those of core muscle group, and the differences were statistically significant ( P<0.01) . The total effective rate of combined treatment group was 90.77% (59/65) , which was higher than 76.92% (50/65) of core muscle group, and the difference was also statistically significant ( P<0.05) . Conclusions:Core muscle training combined with balance cup therapy can reduce the degree of pain in patients with chronic nonspecific low back pain, improve waist dysfunction, waist flexibility and muscle endurance, and have good clinical effects.
5.Flipping moxibustion ofmedicine combined with western medication for rheumatoid arthritis with cold dampnesssyndrome.

OBJECTIVETo compare the effects of flipping moxibustion ofmedicine combined with western medication and simple western medication for rheumatoid arthritis with cold dampnesssyndrome.
METHODSEighty patients were randomly assigned into an observation group and a control group, 40 cases in each group. Oral methotrexate (1 time a week) and sulfasalazine (twice a day except the day for methotrexate) were used in the two groups. Patients in the observation group were treated with flipping moxibustion ofmedicine, twice a week. The main acupoints were Shenshu (BL 23), Guanyuan (CV 4), Zusanli (ST 36), Yinlingquan (SP 9), and the matching acupoints were in the meridians related to the disease location. All the treatment was given for continuous 4 weeks. The TCM symptom score, visual analogue scale (VAS) score, blood sedimentation (ESR), rheumatoid factor (RF) and C-reactive protein (CRP) were observed before and after treatment. The effect was evaluated.
RESULTSThe total effect rate in the observation group was 95.0% (38/40), which was better than 77.5% (31/40) in the control group (<0.05). After treatment, the VAS score, TCM symptom score, RF, ESR, CRP levels decreased in the two groups (all<0.05), with better effects in the observation group (all<0.05). The adverse reactions in the observation group were lower than those in the control group.
CONCLUSIONFlipping moxibustion ofmedicine combined with western medication for rheumatoid arthritis with cold dampnesssyndrome are better than simple western medication, which are safer and more effective.
6.Clinical application of cone beam CT in the diagnosis of traumatic anterior teeth before orthodontic treatment

Objective:
To explore the clinical application of cone beam CT in the diagnosis of traumatic anterior teeth before orthodontic treatment in order to reduce the risk of orthodontic treatment failure due to adhesion.
Methods :
48 fixed orthodontic treatment cases with the history of anterior teeth trama were selected. The anterior teeth were examined by CBCT to exclude root and alveolar bone adhesions.
Results :
3 cases of root-alveolar bone adhesion were found, and the upper anterioa teeth of the remaining 45 cases were successfully moved by orthodontic treatment.
Conclusion
CBCT examination is recommended to check the periodontal ligament in patients of fixed orthodontic treatment with the history of anterior teeth trauma in order to exclude the root-alveolar bone adhesion cases.
7.Flipping moxibustion of medicine at acupoints in governor vessel combined with acupuncture for vascular dementia.

OBJECTIVE:
To observe the difference for vascular dementia among flipping moxibustion of medicine at acupoints in governor vessel combined with acupuncture, simple medicine and simple acupuncture.
METHODS:
A total of 120 patients with vascular dementia were randomly assigned into a combination group, a flipping moxibustion group and an acupuncture group, 40 cases in each one, with 2 patients dropping respectively. Flipping moxibustion was used at the acupoints of group A on Monday and at the acupoints of group B on Friday in the flipping moxibustion group. The acupoints of group A were Baihui (GV 20), Dazhui (GV 14), Shenzhu (GV 12), Zhiyang (GV 9), Jizhong (GV 6), Mingmen (GV 4), Yaoyangguan (GV 3), and the acupoints of group B were Fengfu (GV 16), Taodao (GV 13), Shendao (GV 11), Jinsuo (GV 8), Xuanshu (GV 5), Changqiang (GV 1). Acupuncture was used in the acupuncture group at the main acupoints of Baihui (GV 20), Sishencong (EX-HN 1), Neiguan (PC 6), Zusanli (ST 36), Taixi (KI 3) and Xuanzhong (GB 39), matched with the acupoints based on syndrome differentiation, 30 min a time, once a day, continuous 5 times a week. The above two methods were applied in the combination group. All the treaments were for 4 weeks. The indexes were observed before and after treatment, including syndrome differentiationof TCM scale for vascular dementia (SDSVD), simple mental state scale (MMSE), self-care ability of daily life scale (ADL). The clinical effects and safety were evaluated.
RESULTS:
After treatment, the total effective rate in the combination group was 89.5% (34/38); the rate in the flipping moxibustion group was 65.8% (25/38); the rate in the acupuncture group was 63.2% (24/38). The difference among groups was statistically significant (<0.05). The effect in the combination group was better than those in the flipping moxibustion group and in the acupuncture group (both <0.05). There was no statistically significant difference between the flipping moxibustion group and the acupuncture group (>0.05). The SDSVD scores after treatment were lower and the MMSE and ADL scores after treatment were higher than those before treatment in the three groups (all <0.01), with better results on the above three scores in the combination group than those in the other two groups (<0.05, <0.01), and the differences on the three scores between the flipping moxibustion group and the acupuncture group were not statiatically significant (all >0.05). The treatment in the three groups was safe, without stastical significance (>0.05).
CONCLUSION
The effect of flipping moxibustion combined with acupuncture for vascular dementia is better than those of simple flipping moxibustion and simple acupuncture. The combination treatment achieves better effect on TCM syndrome, cognitive function and daily activity ability than the other two simple treatment.

8. Microglial Exosome miR-7239-3p Promotes Glioma Progression by Regulating Circadian Genes

Glioma-associated microglial cells, a key component of the tumor microenvironment, play an important role in glioma progression. In this study, the mouse glioma cell line GL261 and the mouse microglia cell line BV2 were chosen. First, circadian gene expression in glioma cells co-cultured with either M1 or M2 microglia was assessed and the exosomes of M2-polarized and unpolarized BV-2 microglia were extracted. Subsequently, we labeled the exosomes with PKH67 and treated GL261 cells with them to investigate the exosome distribution. GL261 cell phenotypes and related protein expression were used to explore the role of M2 microglial exosomes in gliomas. Then a specific miR-7239-3p inhibitor was added to verify miR-7239-3p functions. Finally, the mouse subcutaneous tumorigenic model was used to verify the tumorigenic effect of M2 microglial exosomes in vivo. Our results showed that in gliomas co-cultured with M2 microglia, the expression of the BMAL1 protein was decreased (P < 0.01), while the expression of the CLOCK protein was increased (P < 0.05); opposite results were obtained in gliomas co-cultured with M1 microglia. After treatment with M2 microglial exosomes, the apoptosis of GL261 cells decreased (P < 0.001), while the viability, proliferation, and migration of GL261 cells increased. Increased expression of N-cadherin and Vimentin, and decreased E-cadherin expression occurred upon treatment with M2 microglial exosomes. Addition of an miR-7239-3p inhibitor to M2 microglial exosomes reversed these results. In summary, we found that miR-7239-3p in the glioma microenvironment is recruited to glioma cells by exosomes and inhibits Bmal1 expression. M2 microglial exosomes promote the proliferation and migration of gliomas by regulating tumor-related protein expression and reducing apoptosis.
9. Characteristics of the human adenovirus infections from influenza-like illness samples in Xi′an

Objective:
To analyze the epidemiological characteristics and the molecular types of human adenovirus (HAdV) from influenza-like illness (ILI) samples with negative influenza virus in Xi'an from January 2013 to December 2015.
Methods:
Samples from patients with ILI were collected from two national influenza sentinel surveillance hospitals during 2013—2015 in Xi′an. HAdV was detected by real-time PCR, and then the positive samples were inoculated into Hep-2 cells to isolate the viruses. The amplified products were purified and sequenced of hexon gene, and the sequences were compared with the Genebank data and phylogenetic trees were constructed.
Results:
In 2367 samples, 88 samples were positive for HAdV, the positive rate was 3.72%. There were 7 subtypes detected, and the rates of each subtype are as follows: HAdV-1 was 9.09%, HAdV-2 was 22.73%, HAdV-3 was 23.86%, HAdV-4 was 5.68%, HAdV-5 was 7.95%, HAdV-6 was 3.41% and HAdV-7 was 1.14%. Males had higher infection rate than females, but there was no significant difference. The patients were divided into 6 groups according to age. There were 3 positive samples among those under 1 year of age, 36 positive samples among those 1 to 3 years old, 26 positive samples among those 4 to 6 years old, 16 positive samples in those 7 to 18 years old, 5 positive samples in 19 to 59 years old and 2 positive samples in those older than sixty years of age. HAdV infection was primarily confined to children under 7 years of age.
Conclusions
HAdV-3 and HAdV-2 were the dominant epidemic strains during 2013—2015 in Xi′an. Children younger than 7 years were the main susceptible population. HAdV infections circulate all year-round and there was no considerable seasonal variation.
