1.DETERMINATIONS OF ENDOTHELIN IN NORMAL PREGNANCY

We measured Plasma endothelin 1 (ET-1) concentrations in 110 normal pregnant women (NPW) and 23 normal non pregnant women (NNPW), that of maternal venous (MV), umbilical artery (UA) and umbilical vein (UV) in 20 normal parturient women at term (vaginal), and the tissue ET-1 concentrations of the placenta, fetal membrane, decidua and myometrium in 12 normotensive women at term (caesarean section) by radioimmunoassay. Results: compared with NNPW, plasma ET-1 levels were significantly decreased in NPW, and the lowest levels was in the second trimester. The plasma ET-1 levels were significantly increased in the labour, and were similar to that in NNPW. There were significant positive correlations between plasma ET-1 levels and mean arterial pressure in NNPW and NPW during the first, second and third trimester. UA and UV (ET -1) were 3.7 and 6.5 times of MV (ET 1), the UV (ET 1) was higher than UA (EF-1) significantly. The ET-1 concentrations of fetal membrane, placenta, decidua and myometrium were 38, 26, 19 and 11 times of MV (ET 1). Conclusions: ET 1 may play an important role in the initiation and process of labour and the regulation of blood pressure.
2.Enhanced effect of recombinant IL-2 on the development of thymocytes in thymus and the recovery of immunologic functions of sublethally irradiated mice

Sublethally irradiated BALB/c mice undergo thymic regeneration which follows a pheno-typic pattern of events similar to that observed during normal fatal development the regenerationafter irradiation is the result of a limited pool of intrathymic radioresistant stem cells.In this study we showed that 6?10~5u/kg IL-2 can accelerate the proliferation of thymo-cytes,improve the development of thymocytes from CD4~-CD8~-to CD4~+CD8~+ and CD4~+CD8~-/CD4~- CD8~+.At 30th day after irradiation,the proliferative responses of the splenocytes to ConAand LPS,the mixed lymphocyte reaction (MLR) to allogeneic spleen cells (C57BL/6),and thenumbers of plaque forming cells to SRBC were significantly enhanced in IL-2 treated mice,ascompared to that in the irradiated mice without giving IL-2.The above immune functions on day30 after irradiation were recovered to the condition of irradiated control mice on day 45,suggest-ing that the IL-2 is important cytokin in the development of thymocytes in thymus and is able toaccelerate the recovery of immune functions of irradiated mice.
3.Effect of Danggui Buxue Tang on immunological reconstitution of mice following bone marrow transplantation

Objective To study the effect of Danggui Buxue Tang (DBT) on immunological reconstitution of bone marrow transplantation (BMT) in mice. Methods BALB/c mice were irradiated by 137Cs γ once 8.5 Gy, then the mice were engrafted with bone marrow cells (107 cells/mouse) within 4 hours lethal irradiation. And the mice were fed by DBT every day for 15 days. Flow cytometry technique combined with immunological methods were performed to evaluate immunological reconstitution of BMT mice in 30 and 60 days pest-transplantation. Peripheral blood RBC and WBC were counted, and nucleated cells were assayed in recipient bone marrow. Lymphocyte numbers in thymus and periphery were counted and subpopulations of the two origins were observed respectively. Lymphocyte proliferation induced by Con A and LPS, plaque-forming cell (PFC), delayed type hypersensitivity (DTH) were also examined in 30,60 days in post-transplantation respectively. Results Compared with BMT mice, BMT mice treated with a certain dose of DBT could increase the number of peripheral blood RBC and WBC in the recipients, and also could increase that of nucleated cells significantly. BMT mice treated with DBT could increase lymphocyte numbers in thymus and periphery, and improve thymocyte subpopulations, resulting in enhancement in immune function. Conclusion DBT can enhance the immunological reconstitution of BMT mice.
4.Secular trends of premarital medical examination in China during 1996 and 2013

Objective:To describe the secular trends of premarital medical examination ( PME ) in China during 1996 and 2013 and to assess the impacts of national health policies on the PME rate. Methods:The information on marriage and PME for districts and counties in 31 provinces of China was annually collected by the Office for National Maternal & Child Health Statistics of China, and the infor-mation on the health policies was from official governmental websites. According to the main health poli-cies, the calendar years were categorized into 3 periods:1996 to 2003 was mandatory PME period;2004 to 2008 was encouraged voluntary PME period; and 2009 to 2013 was free-paid voluntary PME period. Results: During the 18-year period, 284 242 719 people were registered for a marriage in which 107 198 795 were examined, giving the PME rate of 37. 7%. During the mandatory PME period, the rate ranged 52 . 7% -67 . 7% with an average of 60 . 9% ( urban 71 . 5%, and rural 51 . 7%) . In 2004 , the first year when the PME became voluntary, the rate was abruptly dropped to 2. 6%, and thereafter gradually increased to 11 . 5% in 2008 . As the policies of the free-paid voluntary PME were subsequently issued, the rate was quickly increased to 52. 3% (urban 49. 8%, and rural 54. 6%) in 2013. The in-creasing trend was consistently observed both in urban and rural areas, and across East, Middle, West, and Northeast economical regions. However, the rates differed greatly among provinces. In 2013, 5 pro-vinces had rates of >90% ( Guangxi 97 . 5%, Fujian 96 . 0%, Ningxia 95 . 4%, Zhejiang 93 . 4% and Anhui 90. 1%), whereas some provinces were stuck at a low rate, including developed and underdeve-loped provinces/cities. The PME rate in 2013 was 27. 4% for Shanghai, 25. 5% for Guangdong, 12. 4%for Chongqing, 5. 8% for Beijing and 4. 6% for Tianjin. Underdeveloped provinces were Guizhou (6. 4%) and Qinghai (1. 8%). Conclusion:As various national policies to promote voluntary PME were issued, the PME rate was significantly increased after a sharp decline, though it varied greatly by provinces. For provinces with high PME rate, PME-related health benefits need to be evaluated;for provinces with low rate, it is of important practical significance to explore a cost-effective health service model that is likely incorporated with pre-pregnancy examination.
5.Induction of skin allograft tolerance in mice by using anti-?? T cell receptor and anti-CD80 monoclonal antibodies combined with bone marrow transfusion

Objective: To explore the role of anti-?? T cell receptor(TCR) and anti-CD80 monoclonal antibodies(mAbs) combined with donor bone marrow cells(BMCs) infusion in the induction of murine skin allografts tolerance.Methods: On day 0,2?10~8 BMCs of BALB/c mice were injected into recipient C57BL/6 mice via the tail vein,meanwhile,an intraperitoneal injection of TCR?? mAb(500 ?g) was given.On day 2,CD80 mAb was administered intraperitoneally.Skin grafting was performed on day 6.Delayed type hypersensitivity(DTH),mixed lymphocyte reaction(MLR),IL-2 reverse assay of MLR,adoptive transfer assay and chimerism detection were performed at different time points and tolerance mechanisms were investigated.Results: The mean survival time(MST) of BALB/c skin allografts in C57BL/6 recipients that were treated by anti-TCR?? and anti-CD80 mAbs combined with donor BMCs infusion was 70 days.DTH and MLR assay indicated that the tolerant mice displayed significant hyporesponsiveness.The result of IL-2 reverse test showed that clone anergy was probably involved in the formation of tolerance in the tolerant C57BL/6 mice.In vivo and in vitro adoptive transfer assay,suppressive activity in the spleens of tolerant C57BL/6 mice was observed.Chimerism existed in both the thymus and spleen of the tolerant C57BL/6 mice.The chimerism level gradually declined with time.Conclusion: Treatment of anti-TCR?? and anti-CD80 mAbs combined with donor BMCs infusion can successfully induce a long-term tolerance in BALB/c mice to C57BL/6 skin graft.Multiple mechanisms,including clone anergy,suppressor cells and chimerism are involved in the tolerance.
6.Preparation of arsenic trioxide albumin microspheres and its release characteristics in vitro.

Arsenic trioxide albumin microspheres (As2O3-BSA-NS) were prepared by using methods of chemical cross-linking. The desirability function (DF), calculated according to the size (<1 microm) distribution, drug loading and drug trapping efficiency, was introduced as a total index for the microspheres formulation. Four factors, inculding W/O ratio, decentralization speed, BSA concentration and stirring stabilization time, were selected and arranged in an orthogonal experimental table. The release characteristic was studied by the drug release experiment in vitro. The four factors affected DF differently. Decentralization speed behaved as the maximum (P<0.01), followed by BSA concentration (P<0.05) and the W/O ratio dose (P<0.05). Stirring stabilization time did not influence DF (P>0.05). The release experiment in vitro showed that As2O3 in As2O3-BSA-NS was released more slower than pure As2O3. It was concluded that regular As2O3-BSA-NS may be prepared by the methods of chemical cross-linking, which was optimized by orthogonal experimental analysis of different factors, and the microspheres can release As2O3 slowly.

7.Distribution of Pathogens and Characteristics of Drug Resistance in Intensive Care Unit 2007-2008

OBJECTIVE To investigate the distribution of pathogenic bacteria and the status of drug resistance in intensive care unit(ICU) of Anhui Provinceical Hospital,to provide reference for rational use of the antibiotics in clinical practice and effective control of hospital infection.METHODS From Jan 2007 to Dec 2008,a total of 873 clinical isolates were collected from different samples of infective patients in ICU,drug sensitivity test was conducted based on disk diffusion testing(K-B),to analyze the drug resistance of the pathogenic bacteria.RESULTS The highest isolating rate came from the sample of respiratory tract,which was 68.96%.Among 873 clinical isolates,Gram-negative bacilli and Gram-positive cocci accounted for 65.95% and 19.22%,respectively.Gram positive cocci rised obviously in 2008 compared to 2007.The rate of bacterial drug resistance was relatively high,meticillin-resistant Staphylococcus aureus(MRSA) detection rate was 56.58%;The detection rates Escherichia coli and Klebsiella ESBLs were 75.00% and 57.41%.CONCLUSIONS There is a higher rate of infection for ICU patients,and the condition of drug resistance is serious.To strengthen the pathogen distribution and drug resistance monitoring is of great guiding significance for rational clinical use of drug,reducing multi-drug resistance and nosocomial infection control.

9.Using ESR technology observe the change of free radicals in cochlea after acute acoustic trauma.

OBJECTIVE:
To observe the changes of free radicals in the cochlea of guinea pigs after noise exposure directly using electron spin resonance (ESR) technology.
METHOD:
Forty-two guinea pigs as experimental group were given (125 +/- 1) dB SPL noise exposure for 2 hours, and then investigated auditory function immediately, at 2, 6, 12, 24, 48 and 72 hour. After ABR examinations, 21 animals decollated and extracted cochlea immediately and then placed the cochleas to liquid nitrogen for deep freezing and measuring free radicals using ESR technology. Another 21 animals observed hair cells morphology by AgNO3 staining. Meantime, 6 animals without noise exposure were served as negative control group.
RESULT:
A few free radicals were detected in the cochlea at control group and the relative value of free radicals were (21.68 +/- 1.27) dB SPL. After noise exposure, the relative value of free radicals increased obviously and achieved to the max of (147.01 +/- 4.95) dB SPL at 2 h and gradually decreased near the normal level.
CONCLUSION
Free radicals in the cochlea increase evidently and have a concentration-time rule after acute acoustic trauma. The ESR method can be used to examine the content of free radicals in cochlea for its direct, objective and sensitive characters.

10.Accuracy evaluation of cardiac models of cases with complex congenital heart disease printed by domestic 3D printers

Objective To evaluate the accuracy of 3D models of patients with complex congenital heart disease(CHD) printed by domestic 3D priuters based on cardiac CT data.Methods From April 2018 to June 2018,our hospital used domestic 3D printers to print the hearts of 50 patients with complex CHD.The median age of the patients was 24 months(1 month to 61 years),and the diseases included pulmonary atresia,right ventricular double outlet and transposition of aorta.3 measurement sites(150 in total) were selected for each patient.Pearson correlation coefficient calculation,paired t test and Bland-Altman analysis were performed.Results Pearson correlation coefficient is 0.997.The difference of the measured value of CT-model was tested for normality.P was 0.2 of the D test.The Q-Q graph showed that the data point and the theoretical line were highly overlapped.The mean difference was (-0.07 ± 0.67) mm,P =0.196.In Bland-Altman analysis,the consistency boundary value interval of the difference was(-1.29 mm,1.16 mm) between which there were 143/150(95.33%) points.Conclusion 3D models of patients with complex CHD printed by domestic 3D printers based on cardiac CT data have good accuracy.
