1.Clinical efficacy of the correction of midface depression with structural fat grafting

Objective To explore the clinical effects of correction the midface depression.Methods From Aug.2010 to Aug.2015,185 cases of midface depression (aged from 18 to 61 years)were corrected by using structural fat grafting technique,the site and volume were placed as follows,tear trough 1ml,lid cheek groove 2 ml,midcheek groove 0.5 ml,anteriorcheek 4-8 ml,nasolabial fold 1-2 ml,nasal alar base 1-2 ml,evator labii superioris 1-2 ml,zygomaticus major 1-2 ml,buccinator 1-3 ml,and upper lip 3-5 ml.Results 185 cases were followed from 3 months to 3 years and the results were evaluated by visual analogue scales;156 cases of midface depression were corrected after the first structural fat grafting,the midface changed from concave to convex,and acquired prominent malar became more attractive;satisfaction of visual analogue scales were more than 80 %;only 12 cases needed a second grafting.Conclusions The midface concavity can be corrected effectively by using structural fat grafting technique,the results are satisfied and the technique is safe,simple and effective.
2.Rehabilitative effects and pathological changes in rats after cerebral infarction

ObjectiveTo study the rehabilitative effects and pathological changes in rats after acute cerebral infarction. Methods 30 male Wister rats were randomly divided into 3 groups :Group A(sham group),Group B(model group) and Group C(rehabilitation group). Animal model was made by the middle cerebral artery occlusion(MCAO).For each group, Bederson Neural Function was scored and balancing wood,rotating bar,and net screen were tested at 24 hours,3 days and 7 days after operation respectively. On the 7th day after operation, pathological change of brain tissue was observed. ResultsCompared with Group A, Bederson Neural Function scores of Group B,C indicated significant differences at each time (P<0.01).The abilities of grasping, walking and coordinating of all the groups after 24 hours by operation handicapped, however with time continuing,every function of Group A restored and that of Group B or C improved partly,but showed significant difference compared with Group A(P<0.01).After 3 days by operation,balancing wood test of Group C was better than that of group B(P<0.05),after 7 days,except Bederson Neural Function score,there were difference between Group C and B(P<0.05 or P<0.01).After 7 days by operation,compared with Group A,brain tissue of infarction area in Group B indicated significantly edema, neural cells decreased. Compared with Group B, the edema in Group C decreased and neural cells increased.ConclusionsRehabilitation therapy can improve the MCAO rats' ability of equilibrating, grasping and walking, and decrease edema, increase neural cells in infarction areas.
3.Reliability and validity of Chinese Rehabilitation Research Center Standard Aphasia Examination

ObjectiveTo investigate the reliability and validity of Chinese Rehabilitation Research Center Standard Aphasia Examination (CRRCAE) among patients with aphasia after cerebral injury. Methods40 patients were measured using the CRRCAE. For reliability, interclass correlation coefficients were analyzed. For validity, the correlation coefficients of the aphasia quotients and 4 items of CRRCAE and Western Aphasia Battery (WAB) were analyzed.ResultsThe interclass correlation coefficients>0.9, while the Cronbach α of total scale was 0.9411, the scale of the comprehension was 0.8677,the scale of express was 0.8889. CRRCAE had positive relationship with WAB(with the aphasia quotients, r=0.948,P<0.01). The effect size statistics scale was 0.94.ConclusionCRRCAE shows a good retest reliability and good internal consistency. The total scale of CRRCAE can be correlated with serious level of aphasia effectively.
4.Effects of rehabilitation training on behavior and sensory evoked potential of rats with acute cerebral infarction

The sensory evoked potential (SEP) latency of each group was also tested right after operation and on the 7th day after operation.ResultsComparing with group A, Bederson Neural Function scores of groups B and C indicated significant differences at each time (P<0.01). The ability of grasping, walking and coordinating of all groups handicapped 24 hours after operation, however, with time continuing, every function of group A restored and that of group B or C improved partly, but showed significant difference compared with group A (P<0.01). On the 3rd day after operation, balancing wood test of group C was better than that of group B (P<0.05), on the 7th day after operation, except Bederson Neural Function scores, there were significant difference between groups C and B (P<0.01 or P<0.05). Right after operation, compared with group A, the SEP latency of group B and C showed significant difference (P<0.01), on the 7th day after operation, the SEP latency of group C improved significantly compared with itself right after operation (P<0.01).ConclusionThe rehabilitation training can improve the MCAO rats' ability of equilibrating,grasping and walking and adjust the abnormal latency of SEP after MCAO, and prevent the nerve cells from damage caused by ischemic trauma.
5.DNA location embB 306 regarded as the molecular marker for drug resistant Mycobacterium tuberculosis

Objective To investigate the prevalence of mutation in the locus 306 of embB gene in multi-drug resistant (MDR) Mycobacterium tuberculosis (TB) and evaluate the prospects for using it as a molecular marker to detect MDR-TB.Methods The 291 strains enrolled in this study were from the reference laboratory of Shanghai municipal centers for disease control and prevention, all of which had been tested for drug susceptibility.Mutation in embB 306 was screened both by amplification refractory mutation system (ARMS) and DNA sequencing.The mutation frequencies of embB 306 in the sample groups varied in drug resistance were statistically analyzed.Results 38(51.4% ) of the 74 MDR-TB were embB 306-mutant (X2 =93.8,P<0.01).Of the 24 TB resistant to at least two drugs but not MDR, 9(37.5% ) were embB 306 mutant (X2 =60.1 ,P<0.01 ).But only two(4.9% ) embB 306-mutant strains were found in 41 strains resistant to only one drug (X2 =6.8,P=0.0093).None embB 306-mutant strains were found in 152 pansensitive strains.The specificity of using embB 306 as a molecular marker for detecting multi-drug resistant TB was 94.9% (206/217).Conclusions As a molecular marker for screening drug resistant TB,especially MDR-TB, the gene locus embB 306 shows a relatively high sensitivity and specificity, promising a sound future for its application in clinics to realize fast screening of patients infected with MDR-TB and to provide evidence for appropriate medication.
6.Effects of different skin interventions in the result of penicillin skin test:a network Meta-analysis

Objective To evaluate the effects of seven different skin interventions in the result of penicillin skin test by network Meta-analysis. Methods A systematic search of PubMed, Cochrane Library, Web of Science, EBSCO, Chinese Biomedical Literature Database, China National Knowledge Infrastructure and Wanfang Database from construction to July 2017 was carried out. Control trials examining the effects of skin interventions on the false positive rate of penicillin skin test were included. Methodological quality of included studies was assessed according to the Cochrane Handbook for Systematic Reviews of Interventions 5.1.0.All data was screened,extracted and reviewed by 2 researchers independently and analyzed by WinBUGS, R and Stata. Results Totally 18 trials involving 25 077 patients were included.The results of network Meta-analysis showed the statistically significant difference in favor of saline, 0.5% glutaral and iodine-containing disinfector when compared to alcohol (P<0.05). However,there was no difference in effects between other different skin interventions on the false positive rate (P>0.05). Probability ranking showed that there was a great possibility for saline, direct injection, 0.5% glutaral and iodine-containing disinfector in the reduction of false positive rate. Conclusions Saline, 0.5% glutaral and iodine-containing disinfector were superior to other skin interventions in reducing false positive rate of penicillin skin test.
7.Association study of interleukin-1 genotypes with Alzheimer′s disease

Objective To explore the association of interleukin (IL)-1 genotypes with Alzheimer′s disease (AD). Methods Using polymerase chain reaction and restriction fragment length polymorphism, the IL-1A (-889) and IL-1B (+3953) genotypes in 84 cases of AD and 139 controls were detected and analyzed. Results The frequencies of IL-1A(- 889) C/C, C/T and T/T genotypes were 72.6% and 84.2%, 23.8% and 14.4%, 3.6% and 1.4% in AD cases and controls respectively. The genotypes frequencies of IL-1A (-889) C/C, C/T and T/T in AD cases were similar to that of controls (χ2=4.53, P>0.05), but the frequencies of IL-1A (-889) T allele were significantly higher in AD cases than in controls (15.5% vs. 8.6%, χ2=4.93, P<0.05). The frequencies of IL-1B (+3953) C/T genotypes and T allele were also significantly higher in AD cases than in controls (16.7% vs. 6.5%, 8.3% vs. 3.2%, χ2=5.88,5.56, both P<0.05). Conclusions IL-1 genotypes are associated with AD. IL-1 genotypes may play an important role in the development of AD.
8.Changes of T cell immune mechanism in patients with acute exacerbation of chronic obstructive pulmonary disease

Objective To investigate the changes of T lymphocyte immune balance in the patients with COPD.Methods Forty-eightpatients with AECOPD from January 2015 to December 2016 in this hospital were selected as study group and 30 healthy subjects as control.T cells and cytokines in peripheral blood were detected by flow cytometry and ELISA,respectively.The levels of them were compared between the two groups.The correlations of them and principal components were analyzed among the two groups.Results (1) CD8+T cells in the study group decreased [23.8%(17.5%,30.1%) vs.28.2%(23.8%,31.5%),P<0.05],TregandTh17increased[8.5%(8.3%,9.0%) vs.5.6%(4.9%,6.1%),P<0.01;2.9%(2.8%,3.0%) vs.1.2%(1.2%,1.4%),P<0.01],respectively.(2)In the study group,IL-12 [54.97 pg/mL(31.20,161.59) pg/mL],IL-22 [17.70 pg/mL(15.60,35.99) pg/mL],IL-23[120.28 pg/mL(82.97,169.27) pg/mL],IL-23R [0.15 pg/mL(0.15,0.71)pg/mL]and TNF-α[1 000 pg/mL(1 000,1 000)pg/mL] were higher than that of the control group[31.20 pg/mL(31.20,56.23)pg/mL,15.60 pg/mL(15.60,15.60)pg/mL,78.10 pg/mL(78.10,99.08)pg/mL,0.15 pg/mL(0.15,0.15)pg/mL,722.16 pg/mL(494.25,941.44)pg/mL,P<0.05].(3)Correlation analysis showed that there was a negative correlation between Th17 and Treg (r=-0.883,P<0.01) in the study group (r=-0.883,P<0.01),and IL-22 was positively correlated with IL-23 (r=0.340,P<0.05).In the control group,there was a negative correlation between CD4+T cells and CD8+ T cells,Th17 and CD3+ T cells,Th17 and CD8+T cells (r was-0.578,-0.393 and-0.569,respectively,P<0.05),and a positive correlation between Th17 and Treg,IL-23 and CD4+T cells,TNF-α and DNT(r=0.403,0.440 and 0.392,respectively.(4) There were 5 main components among each group,but the variable factors differed in the study group from the controls,as that:Th17/Treg,Th17,Treg vs.CD4+ T/CD8+ T cells,CD4+ T cells,CD8+ T cells and CD4+T/CD8+T cells,CD8+ T cells vs.Th17/Treg,Treg and CD4+ T cells,CD3+ T cells vs.IL-12,IL-23R andIL-12,IL-23R vs.CD3+ T cells and IL-23,IL-22 vs.TNF-α,DNT.Conclusion The increasing immune function of Th17 cells with IL-22 and IL-23 may be involved in the pathological mechanism of COPD,which accompanied with the weakened toxicity of CD8+T cells and regulation of DNT.
9.Autologous fat granule grafting in treatment of the volume lost in aging face

Objective To conclude the experience of restoration of fat compartments volume intreatment of facial volume lost aging.Methods From May 2015 to May 2018,126 patients with facialvolume lost aging,aged from 34 to 52 years (mean age 42.6 years),were corrected by restoring fatcompartments volume using different sized autologous fat granule grafting technique.Patients' satis-faction was rated using Visual Analogue Scale (VAS) ranging from 1 (most unsatisfactory) to 10(most satisfactory).Results 126 patients with volume lost aging face were followed up from 3months to 2.5 years,the sculpted appearance became more fullness,the laxity,wrinkles and textureof skin were improved obviously,the patients' VAS score was (8.2±1.4).Conclusions Restorationof fat compartments by using different sized autologous fat granule grafting can augment the facicalfullness,reshape the facial contour and improve the aging skin.Therefore it proves to be a safe,multi-effecive and satisfying technique in the treatment of facial volume lost aging.
10.Logic-gated tumor-microenvironment nanoamplifier enables targeted delivery of CRISPR/Cas9 for multimodal cancer therapy.

Recent innovations in nanomaterials inspire abundant novel tumor-targeting CRISPR-based gene therapies. However, the therapeutic efficiency of traditional targeted nanotherapeutic strategies is limited by that the biomarkers vary in a spatiotemporal-dependent manner with tumor progression. Here, we propose a self-amplifying logic-gated gene editing strategy for gene/H2O2-mediated/starvation multimodal cancer therapy. In this approach, a hypoxia-degradable covalent-organic framework (COF) is synthesized to coat a-ZIF-8 in which glucose oxidase (GOx) and CRISPR system are packaged. To intensify intracellular redox dyshomeostasis, DNAzymes which can cleave catalase mRNA are loaded as well. When the nanosystem gets into the tumor, the weakly acidic and hypoxic microenvironment degrades the ZIF-8@COF to activate GOx, which amplifies intracellular H+ and hypoxia, accelerating the nanocarrier degradation to guarantee available CRISPR plasmid and GOx release in target cells. These tandem reactions deplete glucose and oxygen, leading to logic-gated-triggered gene editing as well as synergistic gene/H2O2-mediated/starvation therapy. Overall, this approach highlights the biocomputing-based CRISPR delivery and underscores the great potential of precise cancer therapy.
