Chinese Journal of Biologicals 2023;36(3):269-
Objective To express and purify Cc PT1 protein from Aspongopus chinensis in prokaryotic cell. Methods Thesynthesized Cc PT1 gene was cloned to vector p GEX-4T-1 to construct recombinant expression plasmid p GEX-4T1-Cc PT1,which was then transformed to competent E.coli Rosetta strain and induced by IPTG.The induction temperature(20 ℃ and37 ℃),final concentration of IPTG(0.25,0.5,0.75 and 1 mmol/L)and induction time(6,8,10,12 h)were opti-mized.The obtained protein was purified by GST protein purification system, which was then analyzed by 10% SDS-PAGEand identified by Western blot.GST tags were removed by Pre Scission Protease during purification. Results The recombi-nant protein GST-Cc PT1 was expressed in the form of inclusion body with a concentration of 0.026 9 mg/ml,of which therelative molecular mass was 29 800,consistent with the expectation. The optimum induction condition was induction withIPTG of final concentration of 0.75 mol/L for 12 h at 20 ℃.The purified protein was more than 90% in purity and boundspecifically to mouse monoclonal antibody against GST.After remove of GST tags,Cc PT1 protein showed a relative molecu-lar mass of about 2 830 and the yield was 11.15%.Conclusion A.chinensis Cc PT1 protein was expressed by prokaryoticexpression system, and the purity of Cc PT1 protein was high after purification, which laid a foundation of the in-depth studyof anticancer peptides of A.chinensis.

Chinese Journal of General Surgery 2009;24(11):889-891
Objective To explore the safety and effectiveness of using low molecular weight heparin (LMWH) in place of warfarin for anticoagulation in patients with a previous cardiac valve replacement. Methods We reviewed 70 patients hospitalized from Jan 2002 to Apr 2009 undergoing abdominal surgery who have had a previous cardiac valve replacement and had been on warfarin anticoagulation therapy. LMWH began to replace warfarin for anticoagulation 3 clay before the surgery in 33 cases, while the other 37 patients simply ceased to use any anticoagulant since 3 days before the surgery. Postoperative bleeding and embolism risk were compared between these two groups. Results There were 2 patients suffering from postoperative bleeding in the operative field in LMWH group, while there was no postoperative bleeding in patients on anticoagulant suspension group (χ~2=0.641, P>0.05). There was no valve embolism occurred in these two groups. Conclusions It is a safe way to stop using warfarin with LMWH taking the task for perioperative anticoagulation in patients with a history of cardiac valve replacement 3 days before an elective abdominal surgery. This protocol decreases the risk rate of valve embolism not at the expense of increasing postoperative bleeding.

Objective To investigate the best pathologically diagnostic method for breast cancer before surgery. Methods Wet fine needle aspiration cytology was conducted in 68 cases of probable breast cancer by using a 10 ml syringe with size 7 needle. Results Malignant cells were found in 19 cases, without any false positive cases.Of the remaining 49 cases of negative cytological findings: 46 cases underwent segmental mastectomy,1 of which was postoperatively diagnosed as having fibrosarcoma by paraffin section examination and the rest of which were all demonstrated the presence of benign tumors;3 cases without surgical intervention were followed for (11~20) months and no evidences of breast cancer were found. The sensitivity was 95 0% (19/20),the specificity was 100% (48/48),the accuracy rate was 98 5% (67/68),and the false negative rate and false positive rate were 5 0% (1/20) and 0 (0/48),respectively.Of the 19 cases of positive cytological findings,no implantation metastasis in needle passage occurred. There were no complications in the study. Conclusions Wet fine needle aspiration cytology is safe, economical, convenient, quick, almost painless, and accurate, being the method of first choice in the diagnosis of breast cancer.

Objective To observe the effects of cyclooxygenase (COX) inhibitors and prostaglandin (PG) receptor blocker on hyperalgesia of diabetic rats. Methods Nociceptive paw-withdrawal threshold (NPWT) and thermal latency (TL) of diabetic hyperalgesic rats were tested after subcutaneous injections of COX inhibitors indomethacin and meloxicam, and EP 1 PG receptor blocker SC-19220 into the dorsum of the hindpaw of the rats. Results Indomethacin, a classic inhibitor of COX, produced a significant and long-lasting relief of mechanical hyperalgesia in diabetic hyperalgesic rats. Meloxicam, a selective cyclooxygenase 2 (COX-2) inhibitor, alleviated mechanical and thermal hyperalgesia, but with a shorter duration. SC-19220, a selective EP 1 PG receptor blocker, produced significant relief of mechanical and thermal hyperalgesia. Conclusion COX inhibitors and EP 1 PG receptor blocker relieve hyperalgesia of diabetic rats, which suggests that PGs play an important role in inducing and maintaining hyperalgesia in diabatic rats.

Objective To understand the perceptions of normal and ideal body size in registered nurses and nursing students, and to analyze the impacts of individual weight levels and misperception of body weight on their such cognitions. Methods A cross-sectional survey of 407 nursing students from two hospitals and 466 registered nurses from one hospital was conducted with the questionnaire survey.Sociodemographic information, actual weight, perceived weight levels and scores for figure rating scale were included. Results FRS scores for normal and ideal body size to male were significantly higher than to female either in registered nurses or in nursing students; FRS scores for ideal body size to both sexes were significantly lower than those for normal body size among registered nurses and nursing students. There were no statistically significant associations between FRS scores for normal and ideal body size and individuals' weight levels and misperceptions of body weight among registered nurses and nursing students. Conclusions There existed differences in rating female and male body size among registered nurses and nursing students, with “bigger” normal and ideal body sizes to male,Nurses preferred to “thinner” ideal body size in comparison to normal body size.
