1.Clinical efficacy of Danshen injection combined with triple therapy in treatment of gastric ulcer

Objective To investigate the effect of triple therapy treatment combined danshen injection in treatnent of gastric ulcer. Methods Period gastroscopy 1 Hp-positive gastric ulcer diagnosed 70 patients as subjects,34 patients were randomly divided into two groups,control group 36 cases. Control group,omeprazole 20mg + amoxicillin clarithromycin 1.0g + 0.5g,2 times / d and treated for 7d,the observation group based in the control group given Danshen injection fluid 250ml, 1 times/d treatment intravenous infusion 7d,observe and compare the efficacy and side effects two groups. Results The total effective rate was observed 97.06% (34/33), significantly higher than the total effective rate 75.00% ( 27/36), by statistical analysis, the total effective difference between the two groups was significant (P < 0. 05). Before treatment, clinical symptom score difference between the two groups not significantly,after 6 weeks of treatment,clinical symptom score than those in the two groups before treatment significantly reduced( P <0. 01 ) ,and the difference between the two groups was significant( P <0. 05 ). Diminished appetite and the observation group 2 patients,nausea in 3 cases the control group, 1 case of anorexia, dizziness, headache in 1 case,were mild and tolerable, without treatment, treatment of and withdrawal go away. Conclusion Triple therapy omeprazole combined Danshen injection treatment of gastric ulcer lidocainefor with good ulcer symptoms disappeared and quick, was worthy of promotion and application.
2.Evaluation of the quality of life of Hazak patients with hypertension in Urumqi Nanshan pasturing area

Objective To evaluate the quality of life(QOL)of Hazak patients with hypertension in Urumqi Nanshan pasturing area.Methods Totally 900 Hazak hypertensive patients were investigated by using questionnaire of SF-36 quality of life,Stepwise regression analysis was used to study the correlated factors.Results The scores of the quality of life was (413.98±146.31),Their physical function,bodily pain,general health and mental health dimensions had lower scores.The factors which had influenced the QOL of patients was age,education level,the per capita monthly income,the payment of medical expenses,medical treatment,the nearest clinique,energy,sleep quality,availability of negative life events,compliance,whether or not having chronic diseases,social support,pain and others.Conclusions The quality of life of Hazak hypertensive patients in Urumqi Nanshan pasturing area was poor,medical workers should pay high attention to health eduction for these patients,and improve their life quality through increasing cognition of hypertension-related knowledge and simplify treatment protocol.
3.A survey of patients compliance and its influencing factors in Hazak hypertensive patients in Urumqi nanshan pasturing area

Objective To investigate the patients compliance and its influencing factors in Hazak hypertensive patients in Urumqi nanshan pasturing area and put forward corresponding nursing measures. Methods We investigated 625 Hazak hypertensive patients in their compliance behaviors by self-designed questionnaires and analyzed the results. Results 4.3% patients could maintain medication treatment,90.2% patients could keep physical exercises,2.5% patients could maintain reasonable diet,14.4% patients could quit smoking,30.3%patients could stop drinking and 3.8% patients could persist clinical follow-up. The patients compliance was related with their age, forgetting to take medicine, stop taking medicine because of hypertension alleviation and alteration of living pattern(P<0.01).Conclusion The patients compliance could be improved by increasing knowledge about hypertension, strengthened health education, predigested treatment scheme and support from society and families.

Objective: To investigate the relationship between senile degenerative valvular heart disease (SDVHD) and heart failure. Methods:86 cases of SDVHD consisted of 67 males and 19 females, with the mean age of 60 -93(78?9)yr . 86 cases of controls with comparable gender, age and baseline conditions were include in the control group. Morbidity rate of heart failure were compared between the two groups during 12 years follow-up. Heart function was determined by color Doppler ultrasonography. Results:Prevalence of left heart failure and whole heart failure in SDVHD group were 75. 5% and 22. 0% , respectively, significantly higher than the control group ( 16. 2% and 7. 0% , respectively). 6.Expression of vascular basement membrane-derived multifunctional peptide in Pichia pastoris and identification of its bioactivity

Objective: To express vascular basement membrane-derived multiple peptide (VBMDMP) in Pichia pastoris and to identify its bioactivity. Methods: PCR technique was used to obtain the target fragment GST-VBMDMP, which was then cloned into pPIC9K vector. The resultant vector pPIC9K-GST-VBMDMP was transfected into Pichia pastoris cells with spheroplast and the positive recons was identified. The His + Muts transformant was shake-cultured in MGY at 30 ℃. The culture supernatant (1 ml) was collected for preservation at-70 ℃ every 24 hours while maintaining the concentration of methanol at 0.5% in the culture medium. SDS-PAGE analysis was used to detect the protein expression after 8 days and then animal and cell experiments were performed with GST-VBMDMP. Results: SDS-PAGE analysis found that protein express increased during 1-7 days and decreased after 8 days in MGY medium; GST-VBMDMP fused protein was obtained by Glutathione Sepharose 4B and it inhibited the artery endothelial cell tube structure formation in C57BL/6 mouse; GST-VBMDMP( 2, 6, 10 mg/kg)also significantly inhibited the primary cancer Lewis mouse (tumor inhibitor rates being 96.6%, 82.1%, and 61.2%, respectively)and metastatic lung tumors(tumor inhibitor rates being 96.8 %, 87.9 %, and 75.3%, respectively)(P
7.Effects of sarsasapogenin on the activity of osteoblasts and the differentiation and the function of osteoclasts

Aim: To observe the effects of sarsasapogenin ( SAR) on osteoblasts and osteoclasts cultured in vitro. Methods: Colonal murine calvarial osteoblast-like cell line MC3T3-E1 cells were cultured in vitro. MTT,p-nitropheneye phosphate and tinctorial method of alizarin Bordeaux were used to investigate the effects of SAR on the proliferation, ALP expression, and mineralization tuberculation of MC3T3-E1 cells. Mature osteoclasts were i-solated from the long bone of one-day rat. Meanwhile, marrow cells of mouse bone were cultured with induction of 1,25( OH)_2VitD_3. During the culturing of osteoclasts or marrow cells, SAR of different concentrations was added into the medium. The number of osteoclasts was recognized as tartrate resistant acid phosphatase( TRAP) ( +) multinucleate cells and the resorption lacuna on bone slice were examined with toluidine blue staining. Results: Comparing with the control group, SAR (0.01, 0. 1, 1μg/mL) significanthy increased the proliferation of MC3T3-E1 cells (P <0. 05, P <0. 01). There was no significant difference in the expression of ALP in early pro-liferating MC3T3-E1 cells exposed to SAR of 0.01,0. 1, 1μg/mL, but in the differentiation phase MC3T3-E1 cells, SAR improved ALP activity very significantly if compared with the control group, of which SAR of 1 μg/mL had the most promotion effect(P <0. 01). In addition, compared to the control group, there were, to various ex-tents, increased in the number of mineral nodes in MC3T3-E1 cells after 15day incubation with SAR of different conentrations. Furthermore, no obvious effects of 0.01-1μg/mL SAR on mature osteoclast were observed. But typical osteoclasts were formed when marrow cells were cultured with the induction of 1,25(OH)_2D_3 in medium for 7 days while little or no osteoclasts were induced from marrow cells in the presence of SAR. Conclusion: The results suggest that SAR can effectively promote the proliferation, differentiation and mineralization of osteoblasts cultured in vitro. Besides, SAR can inhibit the generation of osteoclasts from marrow cells.
8.Characteristics of facioscapulohumeral muscular dystrophy's vascular inflammatory pathological changes

Objective To investigate the pathological features of blood vessel inflammation in facioscapulohumeral muscular dystrophy ( FSHD ) and the role of vasculitis on the pathogenesis of FSHD. Methods The clinical manifestations and myopathological features of 26 FSHD patients were retrospectively analyzed and summarized. All of the patients were divided into 2 groups; inflammatory infiltration group and non-inflammatory infiltration group. The latter was further divided into 3 subgroups;endomysial inflammation subgroup, perivasculitis subgroup and transmural vasculitis subgroup.Immunohistochemical staining were carried out in inflammatory infiltration group with anti-CD3, anti-CD4,anti-CD8,anti-CD20 and anti-SMA antibody. The control group was composed of 10 dermatomyositis ( DM)cases and 10 polymyositis ( PM) cases. Results The age of onset was (25. 2 ± 12. 6) years old and the average course was (7. 8 ±7. 3) years. The sex ratio of male to female was 1.6: 1. Five of them had family history. The main clinical features were progressive weakness and atrophy of facial, shoulder girdles and proximal upper limbs muscles. The lower distal limbs and (or) lower distal limbs and pelvic girdle muscles were involved in 18 cases. The main pathological features were shown as followed. Seventeen of them had focal inflammatory cell infiltration, including endomysial inflammation (4/17) , perivasculitis (7/17) , and transmural vasculitis (6/17). Immunohistochemical staining confirmed the major types of inflammatory cells were CD4* T lymphocytes and CD20B lymphocytes, which was familiar with DM. While in PM, CD8+ T lymphocytes were dominant The proportionality of residual muscle fibers obviously decreased in inflammatory infiltration group ( 48. 0% ± 23. 6% ) than non-inflammatory infiltration group ( 94. 3% ±3. 1% , T = 198. 000, P = 0. 000). As to CK levels, there were no significant deviation. Conclusions Obvious inflammatory cell infiltration can be seen in FSHD, the locations of inflammatory cells are endomyosium inflammation, perivasculitis and transmural vasculitis. Transmural vasculitis indicates vascular pathological factor may have something to do with pathogenesis of FSHD.
9.Clinical and pathological features of the 5 Limb-girdle muscular dystrophy 2A patients

Objective To investigate the clinical and molecular pathological features of limb-girdle muscular dystrophy 2A (LGMD2A) of Chinese patients. Methods Thirty cases of LGMD with excluding LGMD2B were included in this study. The muscle specimens were performed by a standard series methods of histochemistry, enzymohistochemistry, immunohistochemistry and Western blot. The clinical and molecular pathological features of LGMD2A were retrospective analyzed. Results Five cases with no or only trace expression of calpain-3 protein were diagnosed as calpainopathy (LGMD2A) by Western blot analysis. The age of onset of these 5 patients ranged from 10 to 45 years and the duration of the disease were about 2-10 years. Proximal muscles weakness and atrophy of lower limbs were predominantly involved. In all patients,symptoms progressed slowly. The ambulation could be retained for many years but running and jumping were impaired early. The serum creatine kinase level was elevated moderately to markedly. Electromyography showed myopathic patterns in all cases. Two siblings had similar symptoms indicating autosomai recessive inherited pattern. Pathologically, there was marked variation in fibre size and most small fibres were round. Some necrotic and regenerating fibers were seen. Fibres with centrally placed nuclei can be found frequently. No infiltrations of inflammatory cells were seen. Lobulated fibers were observed in 2 patients by NADH-TR stain. The expression of dystrophin, caveolin-3, α-, β-, γ- and δ-sarcoglycan protein were normally staining of 5 LGMD2A patients' specimens by immunohistochemistry. Two patients had reduced staining of dysferlin by immunohistochemistry study. Conclusions Clinical and pathological characteristics of our 5 LGMD2A patients are consistent with typical muscular dystrophy features reported in other countries. Identification of calpian-3 deletion by Western blot is essential for the diagnosis of calpainopathy.

Objective To study the histological features of plasmacytoid dendritic cells (pDC) in muscle tissue affected by dermatomyositis (DM) and to discuss the pathological significations of pDC.Methods Muscle tissues from 30 cases of DM and 25 cases of polymyositis (PM) were collected.HE stain, immunohistochemistry studies were carried out in all muscle samples.Results Pathological features of DM included: perifascicular atrophy (25/30); punched-out fiber (14/30); perivasculitis (17/30),inflammatory infiltration in the endomysium(6/30).Using immunohistochemistry study, 19 cases from DM were infiltrated by macrophages which are CD68 positive and CD303 negative, 20 cases with DM were infiltrated by pDC which are CD303 positive.The location of pDC were: perivascular of interfascicular septae only (15/20); endomysium only (3/20) and both (2/20).Myopathic damage such as necrotic and regenerating fibers and inflammatory infiltration could be seen in PM.There was few pDC infiltration in PM.Conclusions There is few pDC in muscle tissue affected by PM and many pDC in muscle tissue affected by DM with infiltration mainly in the wide interfascicular septae.pDC may be connected to perifascicular atrophy and play a roll in the pathogenesis of DM.
