2.Cloning, expression, distribution and subcellular localization of AK078438 gene.

OBJECTIVETo investigate the molecular cloning, tissue distribution, tumor distribution and the subcellular localization of AK078438 gene.
METHODSReverse transcriptase-polymerase chain reaction (RT-PCR) technique was applied to amplify the open reading frame of AK078438 gene. Prokaryotic expressing vector and affinity purification were used to get the fusion protein. At mRNA expressing levels semi-quantitative RT-PCR was employed for the investigation of its distribution in normal rat tissues, tumor cell and mesenchymal stem cell (MSC) lines. The eukaryotic expression green fluorescence protein (GFP) fusion protein vector was transfected into cells and identified subcellular localization.
RESULTSThe full open reading frame of AK078438 gene with 1065 bp, 355aa were identified. mRNA of the gene distributed in brain, uterus, colon, bone marrow, testis and kidney, but not in stomach, liver, the lung, spleen and heart. Murine colon adenocarcinoma C26, melanoma B16, Lewis lung carcinoma LL/2 and MSC had the gene and mouse myeloma cell line NS-1 and Hepa mouse hepatoma cell line had no the gene. The protein was localized in cytoplasm.
CONCLUSIONProtein expression, expressing profile and subcellular localization of AK078438 gene set the basis for further exploration of its functions.

4.Application of intraoperative ultrasound combined with neuronavigation for resection of intracranial cavernous malformations in minimally invasive neurosurgery.

OBJECTIVESTo analyze the reliability and clinical value of intraoperative ultrasound combined with neuronavigation for resection of intracranial cavernous malformations.
METHODSFrom January 2007 to December 2009, 40 cases of intracranial cavernous malformations were operated under the application of intraoperative ultrasound combined with neuronavigation. There were 18 male and 22 female, aged 18 to 58 years, with a mean age of 34.5 years. Neuronavigation was used for all patients before operation to display the three-dimensional model of nervous system and lesions, so to design the operative approach and determine the scope of the incision. Lesions were allocated by real-time neuronavigation in order to continuously verify the accuracy of operative approach during the operation, supplemented by real-time monitoring of intraoperative ultrasound to guide the process of surgery and determine the extent of resection of lesions.
RESULTSThe registration error of neuronavigation was 1.3 - 3.2 mm, with an average of 2.0 mm. All the patients' three-dimensional model of nervous system and lesions were satisfactorily displayed, and the area of lesions were all accurately located. Structural brain-shifts occurred in 4 cases in the remove process of the lesion, with shift degree 5.0 - 10.0 mm, and were corrected by intraoperative ultrasound. All lesions were well displayed by intraoperative ultrasound. Gross total resection was achieved in all patients, with no patient infected or dead. Neurological deterioration was seen in 2 patients, the morbidity was 5.0%.
CONCLUSIONSThe combination of neuronavigation and intraoperative ultrasound for resection of intracranial cavernous malformations can provide valuable intraoperative informations of the location and resection level of the lesion, thereby maximize the accuracy of lesion localization and the extent of resection, with less complications and enhanced efficacy of the surgery.

5.Metabonomics and its applications.

The concept, characteristics and history of metabonomics are introduced. The techniques used in data acquisition and data analysis in metabonomics including their advantages and disadvantages are summarized. In data acquisition platform, NMR, GC/MS, LC/MS (/MS) are the prevalent techniques although at present, none of them is a perfect technique that could meet with the requirement of the metabonomics for measuring all metabolites. While in data analysis, the PCA, PLS and ANN are the major techniques. The researchers could select them according to the research destination. Recent advances and applications of metabonomics in disease diagnosis, drug toxicity evaluation, plant metabolomics and microbial metabolomics are reviewed. In addition, by giving the situation on the establishment of the related corporations, the conferences about metabonomics and proclamation of NIH roadmap the current boom of the metabonomics is reflected. It can be expected that with the development of the function genomics, metabonomics will play a major role in the discovery of the phynotype of the genome and searching for the disease diagnostic biomarkers, and it will also bring much benefit to the drug discovery, clinical diagnosis and nutrition science.

As a classical external treatment of traditional Chinese medicine, blistering moxibustion has significant curative effect on asthma, rheumatoid arthritis and other immune disorders, which suggests that it has certain immunoregulation effect. When stimulated, skin immune system evokes innate immune or acquired immune systems, including dendritic cells (DC), a type of crucial cell related to acquired immunity. From the classification and function of DC, especially the differentiation, migration and maturation of DC in local skin after blistering moxibustion. This article was to discuss the possible ways of immunoregulation of DC in local skin after blistering moxibustion, so as to provide reference for the study on immunoregulatory mechanism of blistering moxibustion.
9.Effects of Heyutai Fuzhu Jiangtang Tablets Combined with Metformin on Insulin Resistance in Skeletal Muscle of Diabetic Rats

Objective To observe effects of Heyutai Fuzhu Jiangtang Tablets combined with metformin in insulin resistance (IR); To discuss its mechanism of action. Methods 6–7 week old male ZDF (fa/fa) rats were randomly divided into model group,metformin group,Heyutai Fuzhu Jiangtang Tablets group(Jiangtang Tablets group),and metformin combined with Heyutai Fuzhu Jiangtang Tablets group.ZDF(fa/+)rats were chosen as normal group.Each medication group was given relevant medicine for gavage for 6 weeks. Body weight, FBG, TG, TC, FFA, FINS, HOMA-IR, OGTT and HE staining were tested. HE staining was used to observe the pathological changes of skeletal muscle. RT-PCR and Western blot were used to detect skeletal muscle corresponding gene and protein expression. Results Compared with Jiangtang Tablets group and metformin group, TC, FFA, FBG, and HOMA-IR in metformin combined with Heyutai Fuzhu Jiangtang Tablets group decreased significantly (P<0.05, P<0.01). Blood glucose level and AUC significantly decreased at each time point in OGTT. HE staining of skeletal muscle fibers arranged in order; nucleus increased and internal movement was not significant, without obvious infiltration of inflammatory cells. Expressions of skeletal muscle InsR, Akt, and Glut4 mRNA expression increased (P<0.05, P<0.01). Expressions of skeletal muscle p-InsR, p-Akt, and Glut4 protein expression increased (P<0.05, P<0.01). Conclusion Heyutai Fuzhu Jiangtang Tablets combined with metformin can improve IR in type 2 diabetic rats, and the effect is better than single-application.
10.Analysis of the incidence of gastriccancer in Qido ng,Jian gsu Province from 1993 to 2012

Objective To analyze the incidence of gastric cancer in Qidong , Jiangsu Province from 1993 to 2012 and analyze the harmful levels and the clinical characteristics thereof , thus providing bases of implementing the prevention , research and control measures for gastric cancer . Methods The clinical data of gastric cancer from January 1993 to December 2012 for patients with census registration in Qidong were sorted out from the cancer registration center in Qidong .The annual percent change ( APC) model was used to analyze the trend over time for the incidence of gastric cancer .The incidence was standardized by utilizing the standard population age composition based on the nationwide census in 2000 . Results A total of 8 272 new cases of gastric cancer were diagnosed in Qidong from 1993 to 2012, and the median age for these patients was 67.92(57.50-76.00).Among them, 5 269 were male cases and the median age for these male patients was 67.25(57.75-74.75);3 003 were female cases and the median age for these female patients was 69.16(57.00-77.91).The annual average for the crude incidence rate of gastric cancer was 40.37/100 000, and the standardized incidence rate thereof was 25.22.37/100 000.The crude incidence rate of gastric cancer decreased significantly from 1993 to 2002(for the crude incidence rate thereof , APC=-1.59, t=-4.6, P<0.01);the crude incidence rate of gastric cancer increased significantly from 2002 to 2007(for the crude incidence rate thereof , APC=-8.88, t=-5.3, P<0.01).Both the standardized incidence rate of gastric cancer and the crude incidence rate thereof had the same trend for the period from 1993 to 2002 as well as the period from 2002 to 2007.The crude incidence rate of gastric cancer rose with the growth of ages(for male patients, APC=734.8, t=-511.2, P<0.01; for female patients, APC=43.6, t=17.9, P<0.01).The crude incidence rate of gastric cancer reached a climax for both males and females of the age group between 75 and 80. Conclusion The incidence of gastric cancer was relatively high in Qidong from 1993 to 2012;the male and the old-aged groups are high-risk groups .
