1.Anxiolytic effect of antianxietic compound prescription capsule on acute stress in rats and influence upon expression of ERK/CREB signaling pathway and BDNF in the brain of rats

Aim To study the pharmacodynamics of antianxietic compound prescription capsule ( ACPC ) on acute stress in rats and the influence upon the ex-pression of ERK/CREB signal pathway and brain-de-rived neurotrophic factor ( BDNF) in the cerebral cor-tex and hippocampus of rats. Methods The elevated plus maze ( EPM ) test was applied to observe the effects of ACPC on acute stress rats administered 7 d low-, medium- and high-dose ( 0. 75 , 1. 5 , 3 g · kg-1 ) . The expression of ERK/CREB signal pathway and BDNF in the cerebral cortex and hippocampus of rats were studied by using Western blot method. Re-sults In EPM, high-dose of ACPC increased signifi-cantly the rat open arm time ( OT%) ( P<0 . 05 ) and the percentage of open arm entries ( OE%) ( P <0. 05). In Western blot, the medium-dose of ACPC reduced significantly p-ERK1/2 expression in hippo-campus ( P <0. 05 ) , and high-dose group decreased significantly the expression of p-ERK1/2 and p-CREB in the cortex and hippocampus of rats ( P <0. 05 ) . High-dose group increased significantly the expression of BDNF in the cortex and hippocampus of rats ( P<0. 05 , P<0. 01 ) . Conclusion ACPC has anti-anxie-ty effect in the model of EPM, and its mechanism may be related to the ERK/CREB signal pathway and in-creased BDNF expression.
2.Clinical efficacy and safety of oxycodone hydrochloride controlled-release tablets in treatment of advanced cancer pain

Objective To investigate the clinical efficacy and safety of oxycodone hydrochloride controlled-release tablets in treatment of advanced cancer pain.Methods 130 patients with advanced cancer pain were selected and randomly divided into two groups,65 patients received oxycodone hydrochloride controlled-release tablets as observation group,65 patients received morphine sulfate controlled-release tablets as control group.Quality of life score was evaluated, clinical efficacy and safety were compared between two groups.Results After treatment, cancer pain of patients in two groups were relieved than pre-treatment (P<0.05), and patients in observation group were better than control group (P<0.05).After treatment, the quality of life in two groups were all improved ( P <0.05 ) , and patients in observation group was more obvious than control group ( P <0.05 ) .During the treatment, adverse reactions occurred in different degrees, and the incidence of adverse reactions in the observation group was lower than control group (P<0.05).Conclusion Oxycodone hydrochloride controlled-release tablets are effective drug in treatment of advanced cancer pain,which can significantly relieve pain and improve quality of life.
3.Determination of Valepotriates and Their Degradation Products in Valeriana Jatamansi Jones by HPLC

This study was aimed to establish an HPLC method for simultaneous content determination of valtrate, acevaltrate, and their degradation products, which were baldrinal and 11-ethoxyviburtinal, in Valeriana jatamansi Jones. The separation and quantification of 4 constituents mentioned above were performed on an Agilent ZORBAX SB-C18 (4.6 mm×250 mm, 5 μm). The mobile phase consisted of water (A) - acetonitrile (B) with an optimized gradient program. The flow rate was 1 mL·min-1. The column temperature was set at 25℃. The wavelength was set at 241 nm. And the injection volume was 10μL. The results showed that among 14 different places of V. jatamansi, the 4 contents determined were different. The contents of valtrate, acevaltrate, and baldrinal in the Yunnan Baoshan Mount were the highest. And the content of 11-ethoxyviburtinal was the highest in Yunnan Dali. It was concluded that the method was with good precision, reproducibility and stability. And it was suitable for the determination of 4 valepotriates ingredients in V. jatamansi. It also provided references for the quality control and exploitation of V. jatamansi.
4.Metaanalysis of effectiveness of different exercises on inflammatory response in obese adolescents

Objective:
To systematically evaluate effectiveness of exercises on inflammatory response among obese adolescents.
Methods:
By searching CNKI, PubMed, Web of Science, EBSCOhost and other databases, randomized controlled trials (RCTs) on the influence of different exercises on inflammatory indicators in obese adolescents were collected according to the inclusion and exclusion criteria. Cochrane evaluation tools were conducted methodological studies on the included literature quality evaluation, and RevMan 5.3 analysis software was used for statistical analysis.
Results:
A total of 21 RCTs were included, involving 1 020 subjects. The results of Meta analysis showed that aerobic training (AT) and aerobic plus resistance training (AT+RT) could significantly reduce BMI, serum C reactive protein (CRP) and leptin levels in obese adolescents. In addition, the effects of AT+RT on pro inflammatory CRP ( MD=-0.52, 95%CI=-0.75--0.30, P <0.01), leptin ( MD=-7.20, 95%CI=-10.45--3.94, P <0.01) and anti inflammatory adiponectin ( MD=1.28, 95%CI=1.01-1.55, P <0.01) were stronger than AT. High intensity interval training (HIIT) was associated with BMI reduction ( MD=-1.14, 95%CI=-1.92--0.36, P <0.01) and increased adiponectin in obese adolescents ( MD=1.79, 95%CI=1.09-2.50, P <0.01), and HIIT was superior in improving adiponectin level in obese adolescents than AT and AT+RT.
Conclusion
Long term and regular AT, AT+RT and HIIT can reduce inflammation in obese adolescents. HIIT is a notable exercise method for obese adolescents to resist inflammation.
5.Metabonomic study on protective effect of ethanol extracts of drynariae rhizoma on osteoporosis in rats urine by using UPLC-MS/MS.

This paper was designed to study metabonomic characters of the osteoporosis induced by high dose of hydrocortisone and the protective effects of Drynariae Rhizoma, which can replenish the kidney and strengthen the bones. A urinary metabonomics method based on ultra-performance liquid chromatography coupled with mass spectrometry (UPLC-MS/MS) was developed. Clear separation of healthy control group, model group and treatment group was achieved by using the principal components analysis (PCA) and 9 significantly changed metabolites were identified as potential biomarkers of osteoporosis. Compared with the health control group, the model group rats showed lower levels of creatinine, citric acid, azelaic acid, hippurate, tryptophan and indoxyl sulfate together with higher levels of phenylalanine, cresol sulfate and phenaceturic acid. These changes in urinary metabolites suggest that the disorders of amino acid metabolism, energy metabolism, gut microflora and anti-oxidative damage are related to osteoporosis induced by high dose of hydrocortisone and the potential effect of Drynariae Rhizoma on all the four metabolic pathways.

6.Practice of constructing high-quality hospital in Zhejiang Provincial People′s Hospital

7.Predictive value of monocyte to lymphocyte ratio,neutrophil percentage to albumin ratio for diabetic macular edema

Objective To investigate the value of monocyte to lymphocyte ratio(MLR)and neutrophil percentage to albumin ratio(NPAR)in predicting diabetic macular edema(DME).Methods One hundred and one diabetic retinopathy patients admitted to the Third Affiliated Hospital of Xinxiang Medical University from January 2018 to February 2023 were selected as the research subjects,and they were divided into DME group(n=56)and non-DME group(n=45)based on fun-dus examination results.The general data such as gender,age,course of diabetes and laboratory indicators were collected by consulting medical records.Fasting elbow venous blood was collected early in the morning of the next day after the diagnosis of DME in both groups,the monocytes(MONO)count,lymphocyte(LYM)count,white blood cell(WBC)count,percentage of neutrophils(NEUT),plasma albumin(ALB),glycosylated haemoglobin(HbA1c)were measured by full automatic blood routine analyzer,and MLR,NPAR were calculated.General information and laboratory indexes of patients in the two groups were compared,and risk factors for DME were analyzed by multivariate logistic regression,and receiver operator characteristic(ROC)curve was applied to evaluate the predictive value of MLR and NPAR for DME.Results The course of diabetes,MONO count,NEUT,MLR,NPAR,WBC count,and HbA1c level of patients between the DME group were significantly higher than those in the non-DME group(P<0.05);there was no statistically significant difference in gender,age,LYM count,and ALB level of patients between the two groups(P>0.05).Multivariate logistic regression analysis showed that increased levels of WBC,MLR,and NPAR were independent risk factors for the occurrence of DME(P<0.05).The ROC curve showed that the best cut-off value of MLR was 0.192,and the area under the curve(AUC)for the prediction of DME was 0.729(95%confidence interval:0.631-0.826),with a sensitivity of 58.9%and a specificity of 82.2%;while the best cut-off value of NPAR was 1.404,and the AUC for predicting DME occurrence was 0.884(95%confidence interval:0.820-0.949),with a sensitivity of 75.0%and a specificity of 91.1%;the AUC of MLR and NPAP for predicting the occurrence of DME was 0.906(95%confidence interval:0.851-0.906),with a sensitivity of 69.6%and a specificity of 93.3%.With MLR>0.192 as positive and NPAR>1.404 as positive,the parallel test of MLR and NPAR predicted the occurrence of DME with a sensitivity of 87.5%,a specificity of 71.1%,and an accuracy of 80.2%;while the tandem test of MLR and NPAR predicted the occurrence of DME with a sensitivity of 46.4%,a specificity of 97.8%,and an accuracy of 69.3%.Conclusion Increased levels of MLR and NPAR are independent risk factors for the occurrence of DME and have certain predictive value for DME.The predictive value of combined MLR and NPAR test for DME is higher than that of separate test,and parallel experiment is more helpful for the early prediction of DME.
8.Acute myeloid leukemia with NPM-MLF1 fusion gene: report of one case and review of literature

Objective To investigate the characteristics of NPM-MLF1 fusion gene in acute myeloid leukemia (AML). Methods The data of one AML patient with NPM-MLF1 fusion gene was analyzed,and literatures were reviewed. Results A female patient was diagnosed as AML M6. In the course of the disease, 2 hematologic relapsed, and 2 recurrences were associated with NPM-MLF1 fusion gene positive. After inductive treatment, hematologic complete remission was achieved, and NPM-MLF1 fusion genes were all negative. Survival time surpassed 6 years when the chemotherapy was performed alone. Conclusion The incidence of NPM-MLF1 fusion gene in AML is low. It is necessary to collect more clinical data to judge whether an independent disease type or not.
9. Prognostic significance of flow cytometric minimal residual disease in acute myeloid leukemia during aplasia

Objective:
To investigate the impact of minimal residual disease (MRD) by multiparameter flow cytometry (MPFC) during aplasia on efficacy and prognosis of de novo acute myeloid leukemia (AML) (non M3) patients.
Methods:
The MRD data by 8-color MPFC during aplasia (day 14-15 of induction therapy) in 85 de novo AML (non M3) patients and the MRD impact on efficacy and prognosis were retrospectively analyzed.
Results:
Data of 85 patients, including 42 males (49.4%) and 43 females (50.6%) , were collected, with a median age of 35 (15-54) years. The median MRD by MPFC during aplasia was 0.58% (0-81.11%) , and 70 (82.4%) patients achieved complete remission (CR) after first induction chemotherapy. The cutoff of MRD by receiver operating characteristic (ROC) analysis was 2.305% (Se= 0.867, Sp=0.800) . The CR rate after one course was significantly higher in patients with MRD<2.305% [96.6% (56/58) ]than in patients with MRD≥2.305%[51.9% (14/27) ] (

Objective To establish the effect of curcumin on PTX-resistant esophageal cancer cell line EC9706/PTX and to investigate the mechanism of curcumin on the epithelial stromalization (EMT) of EC9706/PTX cells.Methods EC9706/PTX cells were established by medium concentration intermittent method.The drug resistance index and cross resistance were measured by MTT assay.The inhibitory effects of different concentrations of curcumin on EC9706/PTX cell proliferation were detected.The effects of curcumin on the morphological changes,migration and invasion of EC9706/PTX cells were examined by cytostatic staining,scratching and transwell invasion assay.The effects of curcumin on the expression of E-cadherin,N-cadherin,vimentin and fibronectin in EC9706/PTX cells at mRNA and protein levels were detected by fluorescence quantitative PCR and Western blot.The effect of curcumin on the expressions of NF-κB p65 and Snail in EC9706/PTX cells were detected by Western blot.Results The drug resistance index of EC9706/PTX was 28.4,which was cross-resistant to cisplatin and doxorubicin.Curcumin could inhibit the proliferation of EC9706/PTX cells.The migration and invasion of EC9706/PTX cells were significantly decreased under the action of curcumin at 20 μmol/L concentration.Fluorescence quantitative PCR and Western blot analysis showed that the expression of Ecadherin was down-regulated and the expression of N-cadherin was up-regulated,and curcumin reversed this phenomenon.Western blot analysis showed that the expression of NF-κB p65 and Snail protein was enhanced after PTX-resistant generated in EC cell,but curcumin reversed this phenomenon.Conclusion Curcumin can inhibit the proliferation,migration and invasion of EC9706/PTX cells.The mechanism maybe that curcumin inhibits the NF-κB-Snail pathway.
