Objective To study the effects of repeated hypoxic exposures (HEs) on glycolysis, mitochondrial oxi-dative phosphorylation and energy charge in mouse brain. Methods Adult BALB/c mice were repeatedly exposed to hypoxia for 5 times and the standard tolerant time and body temperature were recorded. The activities of PFK, PK and mitochondrial complex Ⅰ in the brain were assayed. Phosphoadenosines and energy charge were measured. Results Repeated HEs prolonged the hypoxic tolerance and reduced the body temperature. The activities of PFK and PK experienced regular changes, with an increase in 1st and 3rd HEs and a decline to control levels in 5th HE. The complex Ⅰ activity continued to decrease during HEs. The energy charge was stable. Conclusion HEs lead to a regular change of glycolysis, a continued inhibition of mitochondrial oxidative phosphorylation, and a main-tained energy charge in the brains of mouse.

Objective To explore the effect of hypoxic preconditioning on the activity and gene expressions of glu-cose transporters in the cultured rat hippocampal neurons and astrocytes under anoxic condition. Methods The cultured rat hippocampal neurons and astrocytes were treated for 6 days by intermittently exposing to hypoxic gas mixture (1% O_2, 10% CO_2, 89% N_2) for20 min each day. 24 h after the last hypoxic exposure, the cells were exposed to anoxic gas mixture (10% CO_2, 90% N_2) for 6 h, and the uptake rate of [~3H]-2-deoxyglucose (2-DG), the levels of glucose transporter GLUT1 and GLUT3 mRNAs and the cell survival rate were examined im-mediately after anoxic exposure. Results Neurons and astrocytes preconditioned with hypoxia showed higher 2-DG uptake rates and increased expressions of GLUT 1 mRNA in the astrocytes and GLUT 1 and GLUT 3 mRNA in the neurons. The preconditioned neurons also showed an increased tolerance to anoxia. Conclusion Hypoxic precon-ditioning up-regulates the activity and gene expressions of glucose transporters of hippocampal neurons and astro-cytes under anoxic condition.

Objective To identify the host-cell death pathways (apoptosis, autophagy or necrosis) in L929 cells at the time point of 48 hours post infection (h.p.i.) with Chlamydia muridarum. Methods L929 cells were infected with Chlamydia muridarum at a multiplicity of infection (MOI) of 0.85 for 48 hours.Nuclear fragmentation was observed under fluorescence microscopy following staining L929 cells with DAPI (4′,6-diamidino-2-phenylindole).L929 cells were stained with propidium iodide (PI) plus Annexin Ⅴ and then analyzed by fluorescence-activated cell sorting (FACS) to clarify whether apoptosis or necrosis occurred after Chlamydia muridarum infection.L929 cells were transiently transfected with GFP-LC3 and observed under fluorescent microscopy to analyze cell autophagy.Western blot assay was performed to detect LC3 protein for further analysis of autophagy. Results Apoptosis was not induced in L929 cells by Chlamydia muridarum infection at 48 h.p.i.as no significant nuclear fragmentation was observed. Results of FACS showed that most cells died due to necrosis. Moreover, fluorescent dots of GFP-LC3 formed after infecting transfected L929 cells with Chlamydia muridarum. An increased ratio of LC3Ⅰ to LC3Ⅱ in the L929 cells infected with Chlamydia muridarum was detected by Western blot assay, indicating that autophagy occurred during Chlamydia muridarum infection. Conclusion Necrosis and autophagy rather than apoptosis are induced in L929 cells 48 hours after infection with Chlamydia muridarum.

Objective To evaluate the value of stereotactic core needle biopsy(SCNB)in diagnosis of breast lesions. Methods Forty-seven cases were punctured with computer-assisted stereotactic system, spring-loaded biopsy guns and 16 G core needles. The record of each item was collected, including clinical manifestations, descriptions of the mammographic characteristics(such as calcification, mass and architectural distortion),the pathology of the SCNB and the surgical pathology or mammographic follow-up data. Then the results of SCNB were analyzed based on the comparison of SCNB pathology and the surgical pathology. The reason that SCNB failed and misdioagnosed was inferred from the relationship of SCNB accuracy and the X-ray characteristics. Results Forty-four cases were punctured successfully,3 cases failed.Thirty-one patients were operated soon after biopsy. The results of 27 SCNB cases agreed well with the final pathology but the other 4 did not. Conclusions SCNB as an accurate,time-saving and cost- effective method,is also minimally invasive and hardly changes the architecture of the breast.SCNB can diagnose breast lesions in advance, reduces the number of surgical biopsy, and is promising in clinical application.
